Effects of $Cl^-$ Channel Blockers on the Cardiac ATP-sensitive $K^+$ Channel

  • To explore whether $Cl^-$ channel blockers interact with the ATP-sensitive $K^+\;(K_{ATP})$ channel, I have examined the effect of two common $Cl^-$ channel blockers on the $K_{ATP}$ channel activity in isolated rat ventricular myocytes using patch clamp techniques. In inside-out patches, 4,4'-diisothio-cyanatostilbene- 2,2'-disulfonic acid (DIDS) and niflumic acid applied to bath solution inhibited the $K_{ATP}$ channel activity in a concentration-dependent manner with $IC_{50}$ of 0.24 and 927 ${\mu}M,$ respectively. The inhibitory action of DIDS was irreversible whereas that of niflumic acid was reversible. Furthermore, DIDS-induced block was not recovered despite exposure to ATP (1 mM). In cell-attached and inside-out patches, DIDS blocked the pinacidil- or 2,4-dinitrophenol (DNP)-induced $K_{ATP}$ channel openings. In contrast, niflumic acid did not block the pinacidil-induced $K_{ATP}$ channel openings in inside-out patches, but inhibited it in cell-attached patches. DIDS and niflumic acid produced additional block in the presence of ATP and did not affect current-voltage relationship and channel kinetics. All these results indicate that DIDS among $Cl^-$ channel blockers specifically blocks the cardiac $K_{ATP}$ channel.

Studies on the Metabolism-Independent Calium Binding of the Rat Liver Mitochondria (흰쥐 肝미토콘드리아의 非代謝依存性 칼슘 結合에 관한 연구)

  • Measurements were made of the $Ca^++$ uptake, oxygen consumption and ATPase activity of mitochondria extracted from the rat liver in sucrose-tris chloride medium. $Ca^++$ binding of mitochondria was not affected by the incubation temperature in the range of $0^\\circ - 37^\\circ C$. Succinate did not increase the amount of $Ca^++$ bound while it increased oxygen consumption highly. The presence of ATP in the incubation medium did not enhance the $Ca^++$ uptake either. Therefore, it is concluded that the initial binding of $Ca^++$ is independent on metabolism.

  • 송변전자동화시스템에서 사용하는 주장치 와 RTU간에는 무선 통신망으로 TRS(Trunked Radio System), CMDA(Code Division Multiple Access) 그리고 에어미디어망을 사용하고 있다. 특히 디지털 TRS기술인 유럽 전자 통신 표준기구인 ETSI에서 개발한 TETRA는 임대망인 CDMA와 에어미디어망과는 달리 비임대망으로 비용적인 장점을 가지고 있다. 본 논문은 송변전자동화시스템에서 주장치 기능을 하는 DNP3.0 주장치 서버와 RTU의 기능을 하는 FRTU간에 TETRA 망을 사용할 수 있도록 MFTG(Multi Function Tetra Gateway) Platform을 구현하였다.

Study on Anti-allergic Effect and Safety of Bangpung-galgeun-tang (방풍갈근탕(防風葛根湯)의 항알레르기효과에 관한 연구)

  • The purpose of this study was to examine the anti allergic effect in vivo and in vitro, and to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated anti DNP IgE-mediated passive cutaneous anaphylaxis in rodents and compound 48/80-induced active systemic anaphylatic shock in mice after oral administration with BGT of 0.4 g/kg and 0.8 g/kg for 8 days, and also examined MTT assay, ${\beta}-hexosaminidase$ activity, IL-4 and $TNF-{\alpha}$ from RBL-2H3 and $TNF-{\alpha}$ from Raw264.7 after pre-treatment with BGT of 0.25 mg/ml, 0.5 mg/ml, 1 mg/ml and 2 mg/ml. To ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg /p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. BGT inhibited passive cutaneous anaphylaxis and active systemic anaphylatic shock by oral administration. All the concentrations of BGT from 0.25 to 2 mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosaminidase$ release, IL-4 and $TNF-{\alpha}$, and in Raw264.7, $TNF-{\alpha}$ were significantly reduced by treated all concentrations of BGT. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.

  • Effects of organic compounds, photosensitizers and influx of metal ions on the light-induced mitochondrial ATP synthase in Lentinus edodes purified by stepped sucrose density gradient centrifugation were studied. In our previous work, the activation wavelength and the illumination time of mitochondrial ATP synthase were 470 nm and 15 sec, respectively. This enzyme was activated 85% by 1 mmole 2,6-dichlorophenol indopheol and inhibited by 1 mmole 2,-4-dinitrophenol, $10\;{\mu}mole$ 2-heptyl-4-hydroxyquinoline-N-oxide and $100\;{\mu}g$ oligomycin per ml of ethanol. Particularly, the enzyme was activated 414% by 10 mmole phenazine methosulfate as photosensitizer at 470 nm light. In the influx effects of $Fe^{3+}$ and $Fe^{2+}$ ion, the activity of the above enzyme increased under the optimal light condition compared with nonillumination state.

The Development of New Diffusive Sampler for Formaldehye in Air (기중 포름알데히드 측정을 위한 확산포집기의 개발)

  • To utilize diffuse sampling of formaldehyde in air, a new sampler was designed. A glass fiber filter was impegnated with 2,4-dinitrophenylhydrazine(DNP) and phosphoric acid and mounted 37mm cassette. The formaldehyde vapor was sampled in the dynamic chamber and measured by high performance liquid chromatograph and compared with solid sorbent tube. The results were as follows ; 1. The desorption efficiencies of diffusive sampler between 97.0% and 100%. 2. the sampling rate is constant as in $58{\sim}61.8m{\ell}/min$ when sampling times are between 120 and 360 min. 3. There was a significant relationship between concentrations of diffusive samples and active samples with the coefficient of determination(R2) of 0.92. 4. Desorbed amount of formaldehyde diffusive sampler was increased by high relative humidity. 5. Wheth diffusive samplers were stored at room temperature or at refrigerator there was no statistically significant difference in the accuracy of result. 6. When the diffusive samplers, which collected formaldehyde vapor, were exposed to clean air for three hours, there was no significant loss of formaldehyde due to reverse diffusion. In conclusion, this study suggest that developed diffusive samplers will be a reasonable substitute for the solid sorbent tube for sampling formaldehyde and practical comparative study of developed diffusive sampler should be performed at workplaces of manufacturing industry.

  • (1) Relatively active polyphenol oxidase influence was seen at $0{\circ}C$, and the optimal pH level for the enzyme from the seeds of a small type sweet pepper Zairaisisi is 6.5. (2) The starting stage of the brown coloration with low-temperature injuries showed a strong activity of polyphenol oxidase, and the activity drops to 0 as the entire seed became brownish. (3) The browning effect with enzyme solution of polyphenol extracts suggested that the brown coloration continues in vitro even if polyphenol oxidase activity is nil. (4) Although cytochrome oxidase activity dropped when an abnormality occurrs in electron pathways of respiration at the starting stage of the browning with low temperature injuries, there was no marked influence of it on the total respiration, indicating the fact that polyphenol oxidase can take place of terminal oxidase in the compensatory respiration process.

Bioactivity-Guided Fraction from Viscera of Abalone, Haliotis discus hannai Suppresses Cellular Basophils Activation and Anaphylaxis in Mice

  • Basophils and mast cells are specialized effector cells in allergic reactions. Haliotis discus hannai (abalone), is valuable seafood. Abalone male viscera, which has a brownish color and has not been previously reported to show anti-allergic activities, was extracted with acetone. Six different acetone/hexane fractions (0, 10, 20, 30, 40, and 100%) were obtained using a silica column via β-hexosaminidase release inhibitory activity-guided selection in phorbol myristate acetate and a calcium ionophore, A23187 (PMACI)-induced human basophils, KU812F cells. The 40% acetone/hexane fraction (A40) exhibited the strongest inhibition of PMACI-induced-β-hexosaminidase release. This fraction dose-dependently inhibited reactive oxygen species (ROS) production and calcium mobilization without cytotoxicity. Western blot analysis revealed that A40 down-regulated PMACI-induced MAPK (ERK 1/2, p-38, and JNK) phosphorylation, and the NF-κB translocation from the cytosol to membrane. Moreover, A40 inhibited PMACI-induced interleukin (IL)-1β, IL-6, and IL-8 production. Anti-allergic activities of A40 were confirmed based on inhibitory effects on IL-4 and tumor necrosis factor alpha (TNF-α) production in compound (com) 48/80-induced rat basophilic leukemia (RBL)-2H3 cells. A40 inhibited β-hexosaminidase release and cytokine production such as IL-4 and TNF-α produced by com 48/80-stimulated RBL-2H3 cells. Furthermore, it's fraction attenuated the IgE/DNP-induced passive cutaneous anaphylaxis (PCA) reaction in the ears of BALB/c mice. Our results suggest that abalone contains the active fraction, A40 is a potent therapeutic and functional material to treat allergic diseases.

Formation of D-Glucose Isomerase by Streptomyces sp. (Streptomyces sp.에 의한 포도당 이성화효소의 생성)

  • A source of D-xylose was required for the enhanced production of D-glucose isomerase of Streptomyces sp. strain K-17. D-glucose supported the luxuriant growth of the organism as well as D-xylose, but D-glucose isomerase activity was hardly detected in the D-glucose-grown cells. When the D-glucose-grown cells were incubated aerobically for a few hours in 0.5% xylose solution in 0.05 M phosphate buffer, pH 7.0, it was found that inductive formation of D-glucose isomerase occurred in the cells without multiplication. In the non-growth phase of cells the inductive formation of D-glucose isomerase occurred because a source of nitrogen for the synthesis of enzymes was obtained from turnover of protein accumulated in cells. D-ribose, L-arabinose, D-glucose, D-mannose, citrate, succinate and tartrate could not induce the formation of D-glucose isomerase, but D-xylose could induce. Inductinn of D-glucose isomerase was repressed by D-glucose and its catabolites : glycerol, succinate and citrate. Inductive formation of the enzymes in the non-growth phase was stimulated by $Ba^{2+}$, $Mg^{2+}$ and $Co^{2+}$, and inhibited by C $u^{2+}$, C $d^{2+}$, A $g^{+}$and H $g^{2+}$. The synthesis of enzymes in the induction system composed of 0.5% xylose solution was disrupted by actinomycin D, streptomycin, chloramphenicol, kanamycin, tetracycline, p-chloromercuribenzo ate, arsenate and 2, 4-dinitrophenol, but not disrupted by mitomycin C and penicillin G.icillin G.

Effects of Tribuli Fructus extract on inflammatory responses in IgE-stimulated RBL-2H3 mast cells (비만세포에서 백질려 추출물의 항염증효과에 대한 연구)

  • Objectives : Tribulus terrestris $Linn{\acute{e}}$ (Tribuli Fructus; TF) has been used to treat hypochondrium, agalactia, nebula, itching and vitiligo in traditional Korean medicine. In this study, we investigated the effects of TF 30% ethanol extract on inflammatory responses in IgE-stimulated RBL-2H3 mast cells. Methods : TF extract was prepared by 30% ethanol. RBL-2H3 cells, a rat mast cell line, were treated with TF extract at different concentrations for 1 hr and then stimulated with DNP-IgE/HSA for indicated times. Cell viability was measured by WST-1 assay. The expression of inflammatory cytokines (IL-4, IL-13 and $IFN-{\gamma}$) mRNA was determined by reverse transcriptase-PCR, and the phosphorylation of ERK1/2, p38 and JNK MAP kinases (MAPKs) was determined by Western blot. The nuclear expression of $NF-{\kappa}B$ p65 in the cells was detected by Western blot and immunocytochemistry, respectively. Results : The treatment of TF extract at 0.1 and $0.2mg/m{\ell}$ significantly decreased the expression of IL-4 and IL-13 mRNA in IgE-stimulated RBL-2H3 mast cells, while significantly increased the expression of $IFN-{\gamma}$ mRNA. TF extract treatment was also inhibited the phosphorylation of ERK1/2, p38 and JNK MAPKs in IgE-stimulated RBL-2H3 mast cells in a dose-dependent manner. In addition, TF extract significantly blocked the translocation of $NF-{\kappa}B$ p65 into the nuclear of cells after IgE stimulation. Conclusions : These results indicate that TF extract inhibits inflammatory response in IgE-stimulated mast cells through blocking MAPKs/$NF-{\kappa}B$ pathway. This suggests that TF extract has an anti-inflammatory activity in mast cell activation.
