알카리 내성 Bacillus sp. YA-14의 xylanase 유전자 cloning (Cloning and Expression of a Xylanase Gene from Alkali-tolerant Bacillus sp. YA-14 in Escherichia coli)

  • Chromosomal DNA fragments of Bacillus sp. YA-14 isolated from soil as a potent xylan hydrolyzing bacterium, were ligated to a vector plasmid, pBR322, and used to transfer Escherichia coli HB101 cells. The recombinant plasmid pYDC21 was found to enable the transformants to produce xylanase. pYDC21 was found to contain the 3 kb HindIII fragment originated from the Bacillus sp. YA-14 chromosomal DNA by southern hybridization. The optimum temperature and pH for the reaction of xylanse produced by E. coli (pYDC21) were appeared at 50$_7$C and pH 7.0, respectiveiy. the xylanase enzyme was stable between pH 5.0 and 7.0 and maintained stably up to 4$0^{\circ}C$.

  • A gene coding for a $\beta$-galactosidase of Bacillus subtilis HP-4 was cloned in E. coli JM109 by inserting HindIII digested fragment of B. subtilis HP-4 chromosomal DNA into the site of pBR322 and selecting recombinant transformant showing blue color on X-gal plate. The recombinant plasmid, named pBG109, was found to contain the 1.4 Kbp HindIII fragment originated from B. subtilis HP-4 chromosomal DNA by Southern hybridization. The cloned gene was stably maintained and expressed in E. coli JM109 and the pBG109 encoded $\beta$-galactosidase had the same enzymatic properties as those of $\beta$-galactosidase produced by B. subtilis HP-4.

  • A 16 kilobase (kb) HindIII fragment of alkalophilic Bacillus sp. YC-335 containing a gene for xylanase synthesis was inserted at the HindIII site of pBR322 and cloned in Escherichia coli HB101. After subcloning of recombinant plasmid pYS52, the 1.5 kb fragment was found to code for xylanase activity, and the hybrid plasmid was named pYS55. The DNA insert of the plasmid was subjected to restriction enzyme mapping, which showed that pYS55 had single site for PuvII and SstI in the 1.5 kb insert fragment. Southern hybridization analysis revealed that the cloned gene was hybridized with chromosomal DNA from alkalophilic Bacillus sp. YC-335. About 64% of the enzyme activity was observed in the extracellular and periplasmic space of E. coli HB10l carrying pYS55.

Highly Sensitive PNA Array Platform Technology for Single Nucleotide Mismatch Discrimination

  • Reliable discrimination of a single nucleotide mismatch was demonstrated using arrays with peptide nucleic acid (PNA) probes. The newly developed PNA probes immobilization method and hybridization conditions for PNA arrays gave excellent specificity and sensitivity. In addition we compared the specificity, sensitivity, and stability obtained with the PNA and DNA arrays in discriminating single nucleotide mismatches. The PNA arrays had superior perfect match-to-mismatch signal ratios and sensitivities. The relative signal intensities of mismatch PNA probes ranged from 1.6% to 12.1% of the perfect-match PNA probes. These results demonstrated that the PNA arrays were 2.0 to 37.3 times more specific and about 10 times more sensitive than DNA arrays. The PNA array showed the same specificity and sensitivity after 12-month storage at room temperature.

A Small Cryptic Plasmid pZMO1 of Zymomonas mobilis ATCC10988

  • The nucleotide sequence of pZMO1, a small cryptic plasmid of Zymomonas mobilis ATCC10988 was determined. Analysis of 1,680 bp of sequence revealed $69\%$ identity with Shigella sonnei plasmid, pKYM and $61\%$ identity with Nostoc sp. ss DNA replicating plasmid. Analysis of a deduced amino acid sequence of an orf of pZMO1 revealed $75\%$ identity and $90\%$ similarity with the repA gene of Synechocystis sp. plasmid pCA2.4. The upstream region of the repA gene of pZMO1 possesses six directed repeat sequences and two inverted repeat sequences at downstream of the IR consensus sequence of nick region of rolling circle replication (RCR) plasmid. A typical terminator hairpin structure was found at the downstream region of repA gene. Degradation of single-stranded plasmid DNA by S1 nuclease was detected by Southern hybridization. It suggests that pZMO1 replicates by a rolling circle mechanism in Z. mobilis ATCC10988 cells.

Nanogap Array Fabrication Using Doubly Clamped Freestanding Silicon Nanowires and Angle Evaporations

  • We present a simple semiconductor process to fabricate nanogap arrays for application in molecular electronics and nano-bio electronics using a combination of freestanding silicon nanowires and angle evaporation. The gap distance is modulated using the height of the silicon dioxide, the width of the Si nanowires, and the evaporation angle. In addition, we fabricate and apply the nanogap arrays in single-electron transistors using DNA-linked Au nanoparticles for the detection of DNA hybridization.

A cDNA Clone for the 5' Exon of Chloroplast ATP Synthase Subunit I Gene (atpF) from Broccoli (Brassica oleracea L. var. Italica) and Its Expression Pattern

  • We isolated a cDNA clone, BLSC1, encoding 5' exon of ATP synthase CF0 subunit I from broccoli. BLSC1 is 285 nucleotides long which consists of a 5' noncoding region of 34 nucleotides, a 5' exon of 145 nucleotides and an intron of 106 nucleotides. The 5' exon codes for 48 amino acids which reveals mostly hydrophobic. The amino acid sequence deduced from BLSC1 shares 83%, 83% and 91% identities with the genes coding for atpF from wheat, rice and spinach, respectively. Genomic Southern blot analysis for BLSC1 showed a typically strong signal for a gene located in the chloroplast genome. Northern blot analysis identified three major classes of transcripts showing strong positive signals in the leaves, but only trace amounts of the transcripts were identified in the other organs like stems, flowr buds and roots.

  • Out of 20 primers, 6 generated a total of 1,11 major and minor RAPD bands, producing approximately 4.2 average polymorphic bands pe primer in shortnecked clam (Ruditapes philippinarum) population from Anmyeondo. The bandsharing value altered form 0.15 to 0.74, with the average of 0.5, as calculated by bandsharing analysis. The RAPD profiles obtained with DNAs of two populations from Anmyeondo and Seocheon, respectively, were considerably different (0.20 and 0.51, respectively). The varying degrees of difference among populations may also be of relevance to the retricted hybridization of wild bivalve. Besides gene mapping and breeding applications, PCR-RAPD system could be very useful for the rapid certification and quality control of seed production and for every projects based on PCR amplification of specific bivalve DNA fragments.

  • Fusarium 균 중에서 section Liseola 에 속하는 8균주에 대하여 CHEF-PFGE를 이용하여 핵형을 분석비교하였다. 0.75 Mb에서 6.45Mb 크기의 DNA band가 9~13개로 분리되었고 전체 genome 크기는 38.19 Mb에서 43.22Mb 였으며 종간, 종내에서 염색체 길이 다형성을 볼 수 있었다. F. moniliforme 로부터 얻은 IGS sequence(2.6Kb), Neurospora crassa 의 chs-2 gene(2.8Kb) 과 trp-3gene(3.8 Kb)을 probe로 하여 hybridization을 통하여 이들 gene 의 위치를 확인하고자 하였다.

  • In order to achieve poly-beta-hydroxybutyric acid (PHB) production using recombinant DNA in various host bacterial cells, the isolation of genes for PHB biosynthesis was attempted. As a result, a 5.2kb DNA fragment containing phbCAB operon of Alcaligenes eutrophus was isolated by colony hybridization using synthetic oligodeoxyribonucleotides as probes. The constructed recmbinant plasmid pSK(+)-phbCAB operon was transferred to Escherichia coli, and the obtained transformant accumulated considerable amount of PHB.
