This report provides the complete nucleotide sequences of the full-length cDNA encoding squalene synthase (SQS) and its genomic DNA sequence from a triterpene-producing fungus, Ganoderma lucidum. The cDNA of the squalene synthase (SQS) (GenBank Accession Number: DQ494674) was found to contain an open reading frame (ORF) of 1,404 bp encoding a 468-amino-acid polypeptide, whereas the SQS genomic DNA sequence (GenBank Accession Number: DQ494675) consisted of 1,984 bp and contained four exons and three introns. Only one gene copy was present in the G. lucidum genome. The deduced amino acid sequence of Ganoderma lucidum squalene synthase (GI-SQS) exhibited a high homology with other fungal squalene synthase genes and contained six conserved domains. A phylogenetic analysis revealed that G. lucidum SQS belonged to the fungi SQS group, and was more closely related to the SQS of U. maydis than to those of other fungi. A gene expression analysis showed that the expression level was relatively low in mycelia incubated for 12 days, increased after 14 to 20 days of incubation, and reached a relatively high level in the mushroom primordia. Functional complementation of GI-SQS in a SQS-deficient strain of Saccharomyces cerevisiae confirmed that the cloned cDNA encoded a squalene synthase.

Macrophage colony-stimulating factor (M-CSF) is a growth factor required for growth and differentiation of mononuclear phagocyte lineage. Total and 16 poly (A) mRNA of bovine M-CSF were isolated from healthy bovine peripheral mononuclear cells stimulated by phobol 12-myristste 13-acetate (TPA). The more compatible cultured mononuclear cells were 5${\times}$10/ml for RNA isolation. TPA-activated mononuclear cells increased the level of M-CSF-mRNA more than concanavalin A (Con A) and lipopolysaccharide (LPS). The optimal analysis of reverse transcriptase-polymerase chain reaction (RT-PCR) for14 Macrophage colonystimulating factor (M-CSF) as a growth factor required for bovine M-CSF was denaturation at 94$^{\circ}C$ for 1 minute, annealing at 57$^{\circ}C$ for 1 minute, extension at 72$^{\circ}C$ for 1 minute for 30 cycles. The size of cDNA of bovine M-CSF by RT-PCR was 774 base pairs. A 774 base pairs cDNA encoding bovine M-CSF was synthesized by reverse transcriptase polymerase chain reaction (RT-PCR). Ligated cDNA was transformed to competent cells and then plasmid isolation and digestion was performed. Molecular cloning and sequencing were performed for cDNA of bovine M-CSF. The size of cloned cDNA of bovine M-CSF was 774base pairs. The homology of base sequence and amino acid sequence was 88% and 86% compared with known human M-CSF, respectively. From a high degree of sequence similarity, the obtained cDNA of bovine M-CSF is thought be a specific gene of bovine M-CSF.

To develop a functional cDNA chip, specific genes expressed in the tissues of swine Kagoshima Berkshire were screened. A total of 4,434 ESTs were obtained by constructing a cDNA library from total RNA isolated from the muscle and fat tissues, affirming their functions by investigating similarity of nucleotide sequences with the database at the NCBI. Among them, 1,230 ESTs were confirmed as novel genes, which, to date, have not been identified. Attaching the genes to a cDNA microarray slide revealed expression patterns of genes in muscle and fat according to the growth stages of swine. As specific genes expressed in the muscle tissues of swine with body weight of 30 kg, 60 genes including actin, myosin, tropomysin, transfer RNA-trp synthetase, Kel-like protein 23, KIAA0182 and COI, Foocen-m, etc were obtained. In addition, 18 novel genes were obtained. As specific genes expressed in fat tissues of swine with body weight of 30 kg, 47 genes including annexin II, Collagen, Fibronectin, Pleckstrin homology domain, serine protease, etc were obtained. 21 novel genes were also obtained. The genes specifically expressed in the muscle and fat tissues of swine affect contraction and relaxation of the muscle and the fat. However, studies on the expression mechanisms of the genes are insufficient. To reveal species of structural genes in swine muscle and fat tissue, interrelation studies in expression and function of genes by using the cDNA chip should be conducted.
본 연구는 분열형 효모 Schizosaccharomyces pombe에서 여러 가지 DNA 절제회복 및 유전자 발현에 관여하는 SNF2/SW12유전자의 기능을 연구하기 위하여 이에 관련되는 유전자를 분리하고 그 특성을 연구하였다. SNF2 motif의 conserved sequence를 primer로 하여 중합효소 연쇄반응 (PCR) 방법으로 480 bp 크기의 DNA fragment를 분리하여, 이를 probe로 하여 효모에서 hrp2+ 유전자를 분리하였다. 분리한 hrp+ 유전자의 sequence homology를 비교한 결과 3개의 SNF2 motif를 포함하고 있었다. hrp2+ 유전자의 전사체 크기는 4.7kb임을 Northern hybridization으로 확인하였다. 분리한 유전자의 특성 연구를 위하여 Northern hybridization 으로 hrp2+ 유전자의 UV와 MMS에 대한 유도성을 조사한 결과 자외선에 대해서만 유전자의 발현이 유도되었다. 이 결과 분리한 hrp2+는 UV-inducible 유전자임을 확인하였다. 또한 분리한 유전자의 특성연구 중 하나로 hrp2+ 단백질을 분리하여 helicase activity를 측정하였다. 이 결과 분리한 hrp2+ 유전자는 전혀 helicase activity를 나타내지 않았다.

A cDNA encoding a cathepsin D homologue was cloned from a cDNA library of the mulberry longicorn beetle, Apriona germari. Sequence analysis of the cDNA encoding the cathepsin D homologue of A. germari revealed that the 1,158 bp cDNA has an open reading frame of 386 amino acid residues. The deduced protein sequence of the A. germari cathepsin D homologue shows high homology with cathepsin D in insects, Aedes aegypti (68.2% amino acid similarity) and Drosophila melanogaster (67.2% amino acid similarity). Two aspartic residues and six cystein residues in the A. germari cathepsin D homologue are present at identical locations in all of the other catepsins D. Unlike cathepsins D in two insect species, A. gemari cathepsin D homologue appears to have two putative glycosylation sites, rather than one. Phylogenetic analysis revealed the A. germari cathepsin D homologue is more closely related to insect cathepsins D than to the other animal cathepsins D. Northern blot analysis suggests that A. germari cathepsin D homologue gene is expressed in most if not all, body tissues.
섬유소 분해균 Cellulomonas sp. CS 1-1에 대하여 종 수준으로 분류학적 위치를 규명하기 위하여 7개 type strain 균주와 함께 생리적 및 생화학적 특성을 조사하고 DNA 상등성 및 지방산의 조성 등을 분석하여 비교한 결과, CS 1-1의 콜로니 형태는 circular, entire, smooth, convex하며, 담황색을 띤 $0.3{\sim}0.5{\times}0.8{\sim}1.2{\mu}m$ 크기의 간균이었다. 생리학적 특징으로서 비운동성의 통성혐기성 중온균으로서 Gram양성, catalase양성, oxidase음성, 탄수화물 발효성 등의 표현형은 Cellulomonas 속의 타종과 동일하였으며, 특히 D-ribose, raffinose, rhamnose, xylitol, acetate, L-lactate, propionate, aspartate, proline 등 조사한 모든 탄소원의 이용성이 없었으며, 반면에 sacchrose, arabinose 및 amylose의 이용성은 양성으로 판정되었다. G+C 함량 74.76 mol %, 주요 지방산과 quinone 성분은 전형적인 Cellulomonas의 12-methyltetradecanoic acid (anteiso-$C_{15:1}$)과 MK-$9(H_4)$이었으며, DNA의 상동성 비율은 C. uda ATCC 491과 70%, C. fimi ATCC 15724와 54~59 %, C. gelida 및 C. bibula와도 46~48%의 상동성을 나타내었다. 이상의 결과는 CS1-1이 현재 Cellulomonas 속에 인정된 7개의 type species 중 C. uda ATCC 491 균주와 가장 높은 근연성을 나타냄으로서 C. uda에 속하는 novel species로 분류될 수 있다.
자연계로부터 4-chlorobiphenyl (4CB) 을 분해하는 P08, P20, 027 그리고 P1242 균주를 불리하였다. 이들 분해 균주들의 4CB 분해 과정을 UV-spectrophotometry 방법으로 분석한 결과, 4-CB 로 부터 2-hydroxy-6-oxo-6-(4'-chlorophenyl)hexa-2, 4-dienoic acid 와 4-chlorobenzoate(4CBA) 가 생성되었다. 따라서 분해균주들은 공통적으로 meta-cleavage pathway에 의하여 4CB 를 분해하는 것으로 확인되었다. 그러나 DJ-12, P08 그리고 P27 균주는 4CBA 를 계속 분해하여 4-hydroxybenzoate 를 생성하였으나, P20 과 P1242 균주들은 4CBA 를 더이상 분해하지 못 하였다. 각 분해 균주에서 cbp 유전자군의 상동성을 분석하기 위하여 P. pseudoalcaligenes KF707 의 bphABC 유전자군을 DNA probe 로 이용하여 Southern hybridization 을 실시한 결과, DJ-12, P08 그리고 P27 균주들은 XhoI 에 의한 2.2kb 와 1.8 kb, 그리고 EcoRI 에 의한 11 kb 의 genomic DNA 의 절편에서 hybridization 이 일어났다. 따라서 본 연구에서 분리한 4CB 분해 균주들의 cbp 유전자군은 분해경로 및 bph 유전자군과의 상동성에 의거하여 부 group 으로 구분되었다.
Cyclodextrin을 합성하는 효소 CGTase를 호염기성 Bacillus sp. E1으로부터 분리하기 위하여 PCR을 실시하였다. PCR을 위하여 합성한 primer의 염기서열은 현재까지 보고된 CGTase 유전자의 염기서열을 비교 분석하여 가장 높게 보존된 영역을 찾아내어 선택하였다. PCR 증폭 결과 1.2 kbp 크기의 DNA 절편을 얻을 수 있었고 이를 molecular probe로 이용하여 Southern blot 분석을 실시하였다. Southern blot 분석결과 CGTase 유전자는 염색체 DNA를 제한효소 XbaI으로 절단한 5.3 kbp 절편내에 존재한다는 사실을 알아내었다. CGTase 유전자를 분리하기 위하여 유전자 은행을 제조한 후 선별작업을 실시하여 genomic clone인 pCGTE1을 얻을 수 있었다. pCGTEl의 염기서열을 결정한 결과 분리한 CGTase 유전자는 2109 bp의 open reading frame을 가지며 이는 703개의 아미노산으로 구성된 단백질을 coding하는 것으로 나타났다. 아미노산 서열의 유사성을 비교한 결과 Bacillus sp. KC201의 CGTase 와 가장 높은 94.3% 동질성을 나타내었다.
흑두로 제조한 청국으로부터 혈전용해력이 우수한 균을 선발하여 동정하였으며, 그를 Bacillus subtilis BB-1로 명명하였다. 이 균은 혈전용해효소 isozyme을 적어도 5개이상 생성하는 균주로 확인되었다. 이 균으로부터 크로모좀을 분리하여 shot gun법으로 혈전용해효소 유전자를 크로닝하였으며, 이 유전자를 BSF-1이라 명명하였다. 이 유전자는 714개의 아미노산을 암호화하고 있으며 기존에 밝혀진 혈전용해효소 유전자와 상동성은 검출되지 않은 새로운 혈전용해효소 유전자였다. 혈전용해효소활성 최적 pH 및 온도는 5.0과 $35^{\circ}C$였다. 기질특이성은 적혈구 배지 또는 skim milk, gelatin등에 전혀 분해활성이 없었다. 이는 혈전만을 특이적으로 분해하는 기질특이성을 보였으며, 혈전분해효소로서의 이용가능성이 충분한 것으로 판단된다.
여름 느타리 Pleurotus sajor-caju로부터 Chitin synthase(CHS) gene 특이 primer를 이용한 PCR을 통해 3개의 DNA 단편을 분리하여 cloning하였다. 분리된 DNA 단편들을 기존에 보고된 CHS 유전자들과의 염기서열을 분석한 결과, 이들 DNA 단편들 3개가 모두 CHS 유전자의 단편임을 확인하였고, 또한 이들은 각각 서로 다른 종류의 CHS 유전자들임을 알 수 있었다. 한편, RT-PCR 방법을 이용하여 분리된 유전자의 발현 실험을 실시해본 결과, 이들중 하나인 PsCHS3 유전자는 갓과 균사에서만 발현되는 기관특이 발현 특성을 보였으며, 또한 이 유전자는 상처 처리에 의해 그 발현이 증가되는 것을 확인하였다. 이러한 실험결과로 볼 때 p. sajor-caju의 경우, 다른 균류들의 경우처럼 다양한 기능을 가진 여러 종류(최소 3종류)의 CHS 유전자를 보유하고 있으며, 이들 각각은 다른 기관, 또는 다른 생육 단계에 작용하고 있을 것으로 생각되고, 특히 병 방어 기작에도 관여할 것으로 추측되어진다.
