Anti-Melanogenic Effect of Dendropanax Morbiferus and Its Active Components via Protein Kinas e A/Cyclic Adenos ine Monophosphate-Responsive Binding Protein-and p38 Mitogen-Activated Protein Kinase-Mediated Microphthalmia-Associated Transcription Factor Downregulation

  • Dendropanax morbiferus H. Lev has been reported to have some pharmacologic activities and also interested in functional cosmetics. We found that the water extract of D. morbiferus leaves significantly inhibited tyrosinase activity and melanin formation in α-melanocyte stimulating hormone (MSH)-induced B16-F10 cells. D. morbiferus reduced melanogenesis-related protein levels, such as microphthalmia? associated transcription factor (MITF), TRP-1, and TRP-2, without any cytotoxicity. Two active ingredients of D. morbiferus, (10E)-9,16-dihydroxyoctadeca-10,17-dien-12,14-diynoate (DMW-1) and (10E)-(?)-10,17-octadecadiene-12,14-diyne-1,9,16-triol (DMW-2) were identified by testing the anti-melanogenic effects and then by liquid chromatography-tandem mass spectrometry (LC/MS/MS) analysis. DMW-1 and DMW-2 significantly inhibited melanogenesis by the suppression of protein kinase A (PKA)/cyclic AMP (cAMP)-responsive binding protein (CREB) and p38 MAPK phosphorylation. DMW-1 showed a better inhibitory effect than DMW-2 in α-MSH-induced B16-F10 cells. D. morbiferus and its active component DMW-1 inhibited melanogenesis through the downregulation of cAMP, p-PKA/CREB, p-p38, MITF, TRP-1, TRP-2, and tyrosinase. These results indicate that D. morbiferus and DMW-1 may be useful ingredients for cosmetics and therapeutic agents for skin hyperpigmentation disorders.

Ameliorating Effect of Water Extract from Dendropanax morbifera Lev. on Memory Dysfunction in Streptozotocin-induced Diabetic Rats (스트렙토조토신(Streptozotocin) 유발 당뇨 동물모델에서 황칠나무 잎 추출물의 학습 및 기억력 개선 효과)

  • An anti-amnesic effect of water extract from Dendropanax morbifera Lev. leaves (DMW) on memory dysfunction in streptozotocin-induced diabetic rats was investigated to assess its potential industrial value. Daily administration of DMW (11 weeks) significantly reduced serum glucose, insulin, and blood urea nitrogen (BUN) levels increased by an intraperitoneal injection of streptozotocin (STZ, 55 mg/kg). In addition, the administration of DMW decreased escape latency and increased the time spent in the platform quadrant in the Morris water maze test. Step-through latency in a passive avoidance test was also improved. Finally, DMW produced ameliorating effects on STZ-induced cholinergic deficit through an inhibitory effect on acetylcholinesterase and the increment of acetylcholine level in the hippocampus. These results suggest that DMW might be used as a natural substance for improving diabetic induced cognitive impairment.

A Round-Robin Analysis of Temperature and Residual Stresses in Dissimilar Metal Weld (이종금속용접부 온도 및 잔류응력의 라운드로빈 해석)

  • DMWs are common feature of the PWR in the welded connections between carbon steel and stainless steel piping. The nickel-based weld metal, Alloy 82/182, is used for welding the dissimilar metals and is known to be susceptible to PWSCC. A round-robin program has been implemented to benchmark the numerical simulation of the transient temperature and weld residual stresses in the DMWs. To solve the round-robin problem related to Pressurizer Safety & Relief nozzle, the thermal elasto-plastic analysis is performed in the DMW by using the FEM. The welding includes both the DMW of the nozzle to safe-end and the SMW of the safe-end and piping. Major results of the analyses are discussed: The axial and circumferential residual stresses are found to be -88MPa(225MPa) and -38MPa(293MPa) on the inner surface of the DMW; where the values in parenthesis are the residual stresses after the DMW. Thermo-mechanical interaction by the SMW has a significant effect on the residual stress fields in the DMW.

Analysis of SCC Behavior of Alloy 600 Nozzle Penetration According to Residual Stress Induced by Dissimilar Metal Welding (Alloy 600 노즐관통부의 이종금속용접 잔류응력에 따른 응력부식균열 거동 분석)

  • This work is concerned with the analysis of stress corrosion cracking(SCC) behavior of Alloy 600 nozzle penetration mock-up according to a residual stress induced by a dissimilar metal welding(DMW) in a nuclear reactor pressure vessel. The effects of the dimension and materials of the nozzle penetration on the deformation and the residual stress induced by DMW were investigated using a finite element analysis(FEA). The inner diameter(ID) change of the nozzle by DMW and its dependance on the design variables, calculated by FEA, were well consistent with those measured from the mock-up. Accelerated SCC tests were performed for three mock-ups with different wall thicknesses in a highly acidic solution to investigate mainly the effect of the residual stress on the SCC behavior of Alloy 600 nozzle. From a destructive examination of the mock-up after the tests, the SCC behavior of the nozzle was fairly related with the residual stress induced by DMW : axial cracks were found in the ID surface of the nozzle within the J-weld region where the highest tensile hoop stress was predicted by FEA, while circumferential cracks were observed beyond both J-weld root and toe where the highest tensile axial stress was expected.

Melanin Production Inhibitory Activity of the Dendropanax morbifera Leaf Extract Fermented by Lactobacillus plantarum (Lactobacillus plantarum으로 발효시킨 황칠나무 잎 추출물의 피부 미백 관련 효과)

  • This study was conducted to investigate the tyrosinase inhibitory and melanin production inhibitory activity of the distilled water extract of Dendropanax morbifera leaf (DMW) and the fermented extract by Lactobacillus plantarum (DMF). DMF was prepared by inoculation of L. plantarum after the extraction procedure with distilled water. Fermentation for 48 hours at $37^{\circ}C$ is the most effective condition in this study. In DPPH radical scavenging ability, $SC_{50}$ values of the fermented DMF was $37.9{\mu}g/ml$ as a result of more effective than DMW extract ($52.6{\mu}g/ml$). Moreover, tyrosinase inhibitory activity of DMF showed higher activity than DMW. In nontoxic concentration range, DMF showed strong melanin production inhibitory effect in ${\alpha}$-melanocyte stimulating hormone-stimulated B16F10 cell. As a result, the fermentation of the distilled water extract of D. morbifera leaf by L. plantarum could be applicable to functional materials production for skin-whitening agents.

Effect of preemptive weld overlay sequence on residual stress distribution for dissimilar metal weld of Kori nuclear power plant pressurizer (고리 원전 가압기 PWOL의 용접 방향이 이종금속용접부 잔류응력 분포에 미치는 영향)

  • Weld overlay is one of the residual stress mitigation method which arrest crack. An overlay weld sued in this manner is termed a preemptive weld overlay(PWOL). PWOL was good for distribution of residual stress of dissimilar metal weld(DMW) by previous research. Because range of overlay welding is wide relatively, residual stress distribution on PWR is affected by welding sequence. In order to examine the effect of welding sequence, PWOL was applied to a specific DMW of KORI nuclear power plant by finite element analysis method. As a result, the welding direction that from nozzle to pipe is better good for residual stress distribution on PWR.

  • Fracture mechanics evaluation of stress corrosion cracking (SCC) in the dissimilar metal weld (DMW) for the nuclear piping system is performed; simulating the transition joint of the ferritic nozzle to austenitic safe-end fabricated with the Inconel Alloy A82/182 buttering and welds. Residual stresses in the DMW are computed by the finite element (FE) analyses Then, to investigate the SCC in the weld root under the combined residual and system operation stresses, the fracture mechanics parameters for a semi-elliptical surface crack are evaluated using the finite element alternating method (FEAM). As a result, it is found that the effect of weld residual stresses on the crack-driving forces is dominant, as high as three times or more than the operation stresses.

A Case History of Confinement of the Contaminated Landfill Using a Vortical Barrier (연직 차수벽을 이용한 폐기물매립지 침출수 오염 제어 사례 연구)

  • This paper presents a case history of remedial action adopted for contaminated groundwater in a landfill. The objectives of the projects are : (1) to effectively confine contaminated groundwater with an economically reasonable means, (2) to prevent further contamination of soil by collecting and treat the contaminant. and (3) to assure the environmental safety of the landfill during its operating period. Reported are the process from site investigation, through design and construction of an appropriate remedial action, to the monitoring of the selected confinement system. In view of the results of site investigation, deep soil mixing cutoff wall using the DMW(deep soil mixing cutoff wall) method and specially produced HEC soil stabilizer were used for the construction of deep soil mixing cutoff wall. For rock foundation with sever fractures, chemical grout curtain with urethane was installed. The monitoring results to date indicate that the selected vertical barrier performed satisfactorily.

Stress Distributions at the Dissimilar Metal Weld of Safety Injection Nozzles According to Safe-end Length and SMW Thickness (안전단 길이 및 동종금속용접부 두께 변화에 따른 안전주입노즐 이종금속용접부의 응력분포)

  • In the present paper, we evaluate the effects of the safe-end length and thickness of the similar metal weld (SMW) of safety injection nozzles on stress distributions at the dissimilar metal weld (DMW). For this evaluation, we carry out detailed 2-D axisymmetric finite element analyses by considering four different values of the safe-end length and four different values of the thickness of SMW. Based on the results obtained, we found that the SMW thickness affects the axial stresses at the center of the DMW for the shorter safe-end length; on the other hand, it does not affect the hoop stresses. In terms of the safe-end length, the values of the axial and hoop stresses at the inner surface of the DMW center increase as the safe-end length increases. In particular, for the cases considered in the present study, the stress distributions at the DMW center can be categorized according to certain values of safe-end length.

  • This paper is concerned with numerical analyses of residual stresses in welds and material's susceptibility to stress corrosion cracking (SCC) for the primary piping system in nuclear power plants: Both the dissimilar metal weld (DMW) for stainless steel to low alloy steel joints and the similar metal weld (SMW) for forged stainless steel to cast stainless steel joints are considered. Thermal elasto-plastic analyses using the finite element method (FEM) are performed to predict residual stresses generated in fabrication welding and its related processes for both the DMW and SMW, including effects of quenching for cast stainless steel piping, machining of the DMW root, and grinding of the SMW root. As a result, the effect of quenching should be included in the evaluation of residual stresses in the SMW for the cast stainless steel piping. It is deemed that residual stresses in both the DMW and SMW would not affect the SCC susceptibility of the welds providing that the welding processes are completed without any weld repair on the inside wall of the joint. However, the grinding process if performed on the safe-end to piping weld, would produce a high level of residual stresses in the inner surface region and thus a stress improvement process (e.g. buffing) should be considered to reduce susceptibilities to SCC.
