Identification of Oocyte-Specific Diva-Associated Proteins using Mass Spectrometry (Mass Spectrometry를 이용한 난자 특이적인 Diva와 상호작용하는 단백질의 동정)

  • Objective: We previously described that Diva is highly expressed in matured metaphase II (MII) oocytes compared to immature germinal vesicle (GV) oocytes in mouse. We report here that the expression of Diva transcript as well as protein is oocyte-specific. To elucidate its physiological role in oocyte, the binding partner(s) of Diva has been identified by using immunoprecipitation (IP) followed by Mass Spectrometry. Methods: NIH/3T3 cells were transiently transfected for 24 h with either empty vector for control or FLAG-tagged mouse Diva construct, and IP was performed with anti-FLAG antibody. The immuno-isolated complexes were resolved by SDS-PAGE on a 12% gel followed by Coomassie Blue staining. For in-gel digestion, 15 bands of interest were excised manually and digested with trypsin. All mass spectra were acquired at a positive reflector mode by a 4700 Proteomics Analyzer (Applied Biosystems, Framingham, MA). Proteins were identified by searching the NCBI nonredundant database using MASCOT Peptide Mass Fingerprint software (Matrixscience, London). Results: Diva-associated complexes were formed in FLAG-tagged mouse Diva-overexpressed NIH/3T3 cells via IP using anti-FLAG-conjugated beads. Among the excised 15 bands, actin and actin-binding proteins such as tropomyosin, tropomodulin 3, and ${\alpha}$-actinin were identified. Binding between Diva and actin or tropomyosin was confirmed by IP followed by Western blot analysis. Both bindings were also detected endogenously in mouse ovaries, indicating that Diva works with actin and tropomyosin. Conclusions: This is the first report that immuno-isolated Diva-associated complexes are related to actin filament of the cytoskeletal system. When we consider the association of Diva with actin and tropomyosin, oocyte-specific Diva may play a role in modulating the cytoskeletal system during oocyte maturation.

Study on Integrated Workflow for Designing Sustainable Tall Building - With Parametric method using Rhino Grasshopper and DIVA for Daylight Optimization

  • Purpose: The Objective of this study is to explore the capabilities of an integrated modelling and simulation workflow when applied to an experiment-based research process, aimed at deriving daylight optimization strategies specific to tall buildings. Methods: Two methods were devised to apply this workflow with the help of DIVA and Rhino/Grasshopper. The first method is a multiple variant analysis by setting up an appropriate base case and analysing its daylight and energy performance, forming the basis of comparison for subsequent cases for design variants. The second method involved setting up the base case within a site context and conducting a solar irradiation study. An architectural variables such as overhang and shading device, were then defined as inputs in the parametric definition in Grasshopper to control the selected variable. Results: While the first method took advantage of the speed and efficiency of the integrated workflow, the second method was derived based on the ability to directly process simulation data within the integrated, single-software platform of the proposed workflow. Through these methods, different architectural strategies were explored, both to increase daylight penetration and to reduce radiant heat gain. The focus is on methods by which this workflow can be applied to facilitate the experimental derivation of daylight optimization strategies that are specific to tall building design.

A Study of Simplified Calculation Methods for Outside Vertical Illuminance using VBA (VBA(Visual Basic for Applications)를 활용한 실외 수직면 조도 간이계산법에 관한 연구)

  • The purpose of this study is to predict vertical illuminance accurately at the design stage of a building without the help of simulation tools. Comparing two well-known vertical illuminance prediction algorithms with measured values, it is verified that the Igawa model is more consistent with the measured values than the Perez model. Using the DIVA program, we simulated the vertical illuminance at 30-degree intervals from south to north, compared with the vertical illuminance calculated with the Igawa model. The result of calculation values were verified from 120 degrees east to 120 degrees west. The vertical illuminance values with each of three shade devices were calculated using the Igawa model, and compared with the vertical illuminance simulated by DIVA program. As a result, all the errors when installing horizontal / vertical / grid shade divices were included in the error standard specified by ASHRAE.

  • Wi-Fi Direct standard technology(Wi-Fi Peer to Peer Technical Specifications v1.2, 2010) was designed for allowing peer to peer communication between two or more devices and various products which have been currently manufactured such as smart phone and smart TV have already provided this technical function. In this technology the role of Coordinator in Wi-Fi infrastructure would be matched to GO(Group Owner) which needs relatively high energy resource and computation power due to the high probability for allowing 3rd-party connection, however, the current standard specification would be limited in terms of energy distribution because it has not included a process to determine the role of GO. To address above problem, this paper classified considerable parameters into the general parameters for the physicality of devices and the specific parameters for considering the role position depending on use-case scenario, and proposed a new method called DIVA to help efficiently determining GO role from the member devices of Wi-Fi Direct network. Furthermore the effect of this mechanism was proved via simulation-based experiments.

A new species of the genus Eurycletodes Sars G.O., 1909 (Copepoda: Harpacticoida: Argestidae) from South Sea of Korea

  • The genus Eurycletodes Sars, 1909 is reported for the first time in Korea. A new species of the genus Eurycletodes was collected from Hansando Island, South Sea of Korea. So far, Eurycletodes comprises 27 species, with no previously recorded species reported in East Asia, including Korea. Eurycletodes (Oligocletodes) vadumus sp. nov. is morphologically most closely related to E. (O.) denticulatus Por, 1967, E. (O.) aculeatus Sars, 1920, and E. (O.) diva Menzel, 2011, but clearly distinguishable from the others based on the following morphological characteristics: absence of A2 exopodal seta, A1 last segment with two inner setae, P4 exp-3 proximal inner seta modified, the ratio of length and of caudal rami (1.6 times), and mandibular palp with three setae. Together with the new species, the number of valid species in this genus has risen to 28. A key to species of the genus Eurycletodes is provided.

Differentially Expressed mRNA Profiles between Immature Germinal Vesicle(GV) and Mature Metaphase II(MII) Mouse Oocytes (미성숙 난자와 성숙 난자에서 서로 다르게 발현하는 유전자에 관한 연구)

  • Oocyte maturation refers to the process that prophase I arrested germinal vesicle(GV) drives the progression of meiosis to metaphase II(MII) to have the capacity for fertilization and embryo development. To better understand the molecular mechanism(s) involved in oocyte maturation, we identified differentially expressed genes(DEGs) between GV and MII mouse oocytes using a new innovative annealing control primer (ACP) technology. Using 20 ACPs, we successfully cloned 32 DEGs between GV and Mll oocytes, and 26 out of these 32 DEGs were functionally known genes. Four genes including Pscd2 were GV-specific, 10 genes including PKD2 and CSN3 were highly expressed in GV oocytes(GV-selective), and 12 genes including Diva were highly expressed in MII oocytes (MII-selective). Ail of the genes identified in this study were first reported in the oocyte expression using ACP system and especially, we could characterize the existence of PKD-CSW signaling pathwayin the mouse oocytes. Results of the present study would provide insight for studying molecular mechanisms regulating oocyte maturation.

A Study on Daesoon Jinrihoe's Participation and Role in International Development Cooperations (대순진리회의 국제개발협력 참여와 역할에 관한 시론적 연구)

  • This study aims to examine the International Development Cooperations (IDCs) of Daesoon Jinrihoe and discuss the future prospects of these programs in terms of Daesoon Thought. The international community calls for the active participation of various donors, and this is not limited to only traditional donors such as governments, public institutions, and multilateral organizations. In this regard, the role of Faith-Based Organizations (FBOs) and Non-Governmental Organizations (NGOs) for religious communities is recognized as important. Daesoon Jinrihoe has systematized its IDCs by expanding its overseas services to implement the three major works with the official establishment of the Daejin International Volunteers Association (DIVA) in 2013. In addition to these activities, Daesoon Jinrihoe was selected by Korea International Cooperation Agency (KOICA) in 2022. As a new religion, Daesoon Jinrihoe does not provide as much support via FBOs when compared to other traditional religions (Christianity, Buddhism, Catholicism, etc.), but it is able to conduct strategic IDCs by utilizing its expertise and capabilities (education, healthcare, etc.). In fact, the nature and content of the projects carried out by these FBOs and Daesoon Jinrihoe are similar, and the results have been remarkable considering that these programs are still in their infancy. Above all, Daesoon Jinrihoe's foreign aid is more meaningful because it does not have a religious purpose such as missionary work, and it is funded by donations from believers. In carrying out IDCs, it is necessary to promote the professionalism of IDCs based on the strategic linkage of the three major works (charity aid, social welfare, and education). It is also necessary to recognize the value and spirit of IDCs for peace and coexistence based on mutual beneficence.
