Protective Effect of DWP-04 Against Hepatotoxicity Induced by D-galactosamine (흰쥐에서 DWP-04가 D-galactosamine에 의해 유도된 간독성의 보호효과)

  • This study was conducted to investigate the biological activity and hepatoprotective effect of DWP-04 [DDB : selenium yeast: glutathione (31.1 : 6.8 : 62.1(w/w/w)] in D-galactosamine (GaIN) intoxicated rats. The DWP-04 (50, 100 or 200 mg/kg) or its vehicle was orally administered everyday before the start of GaIN injection (400 mg/kg, ip) for two weeks and animal decapitated for 24 hrs after GaIN­injected. The activities of serum enzymes, markers of liver function, were increased in the GaIN group compared to normal group and significantly lowered in the DWP-04 pretreated group than in the GaIN group. Hepatic lipid peroxide level and activities of phase 1 enzymes were significantly higher than those of GaIN group compared to normal group and lower in the DWP-04 pretreated group than in the GaIN group, and phase II enzyme activities in liver were lower in the GaIN group than in the normal group and were increased in the DWP-04 pretreated group than in the GaIN group. Total hepatic glutathione content and glutathione biosynthesis enzymes were lower in the GaIN group than in the normal group and were increased in the DWP-04 pretreated group than in the GaIN group. Therefore, the current results indicated that DWP-04 administration alleviated the GaIN-induced adverse effect through enhancing the antioxidant enzyme activities.

Expression Study on the Scaffold Gene of CRL4 Complex in Rice (Oryza sativa L.) (벼에 존재하는 CRL4 복합체 scaffold 유전자의 발현 양상에 대한 연구)

  • The stability of diverse cellular proteins in eukaryotes is regulated via ubiquitination. Moreover, E3 ligase plays a crucial role in determining substrate specificity and transfers ubiquitins into the substrates during the ubiquitination process. As a type of multi-subunit E3 ligase, cullin4 (CUL4)-based E3 ligase (CRL4) complex is involved in a variety of cellular processes, such as hormonal and stress responses in plants. In spite of several reports on the versatile roles of CRL4 in various signalings in Arabidopsis, CRL4's function in rice has been poorly known. To learn about CRL4-mediated cellular processes in rice in more detail, OsCUL4 that exhibits the highest homology with Arabidopsis CUL4 was isolated, and its expression patterns in various tissues and in response to plant hormones and abiotic stresses were monitored. Exogenous application of ABA or cytokinin increased the transcript levels of the OsCUL4 gene. Moreover, OsCUL4 was significantly upregulated in response to drought and salt stresses. These findings imply that OsCUL4 may be functionally related to ABA- and/or cytokinin-mediated cellular responses. OsCUL4 directly interacted with OsDDB1, an adaptor protein of CRL4, indicating that OsCUL4 can act as a scaffold protein of CRL4. An expression study on the OsCUL4 gene from this report could be used as a starting point to elucidate cellular responses in which a CRL4-mediated ubiquitination process is involved in rice.

Characterization of a Novel DWD Protein that Participates in Heat Stress Response in Arabidopsis

  • Cullin4-RING ubiquitin ligase (CRL4) is a family of multi-subunit E3 ligases. To investigate the possible involvement of CRL4 in heat stress response, we screened T-DNA insertion mutants of putative CRL4 substrate receptors that exhibited altered patterns in response to heat stress. One of the mutants exhibited heat stress tolerance and was named heat stress tolerant DWD1 (htd1). Introduction of HTD1 gene into htd1-1 led to recovery of heat sensitivity to the wild type level, confirming that the decrease of HTD1 transcripts resulted in heat tolerance. Therefore, HTD1 plays a negative role in thermotolerance in Arabidopsis. Additionally, HTD1 directly interacted with DDB1a in yeast two-hybrid assays and associated with DDB1b in vivo, supporting that it could be a part of a CRL4 complex. Various heat-inducible genes such as HSP14.7, HSP21, At2g03020 and WRKY28 were hyper-induced in htd1-1, indicating that HTD1 could function as a negative regulator for the expression of such genes and that these genes might contribute to thermotolerance of htd1-1, at least in part. HTD1 was associated with HSP90-1, a crucial regulator of thermotolerance, in vivo, even though the decrease of HTD1 did not affect the accumulation pattern of HSP90-1 in Arabidopsis. These findings indicate that a negative role of HTD1 in thermotolerance might be achieved through its association with HSP90-1, possibly by disturbing the action of HSP90-1, not by the degradation of HSP90-1. This study will serve as an important step toward understanding of the functional connection between CRL4-mediated processes and plant heat stress signaling.

  • The hepatoprotective activity of six extracts (BE, EE, HH, PS-1, PS-2, KP) from Artimisia iwayomogi was investigated against experimentally produced hepatic damages. Silymarin, DDB and UDCA were used as reference compounds. Treatment with PS-1 extract reduced hepatic demages induced by $CCl_4$, acetaminophen and ANIT but it did not alter ethionine-induced hepatotoxicity In addition, PS-1 extract showed a protective effect against chronic $CCl_4$-induced hepatotoxicity as well as liver regeneration. PS-2 and KP extracts exhibited significant antihepatotoxic effects on D-galactosamine-induced hepatitis. Treatment with EE extract inhibited ethionine-induced fatty liver. These data indicate that the PS-1 extract is the roost hepato-protective constituent and rationalize the traditional use of this plant in hepatobiliary disorders.

  • 간재생 및 만성 간질환모델에 대한 인진호 추출분획의 간장 약효검색 방법 1. 간세포 재생능력에 대한 효과 : SD계 수컷 흰쥐(체중 300-350g)를 마취하에서 복부정중선을 따라 개복후 간의 중앙엽 및 좌측엽을 절제하여 봉합하고 수술조작을 끝낸 추 4시간, 익일부터 7일간 오후 5시에 약물을 경구투여하고 마지막투여 후 20시간 절식, 부검하여 간재생률 및 BSP 정체율을 측정하였다. 2. $CCl_4$ 만성간장해 모델에 대한 효자 SD계 수컷 흰쥐(체중 200-250g)에 $CCl_4$ : olive oil 40%용액을 0.5 $m\ell$/kg b.wt. 용량으로 6일간 결구투여 한 후 7일째부터 매일 시험약물을 $CCl_4$ 혼합액 투여 4시간전 및 4시간후에 2회 경구투여하였으며 이 조작을 8일간 하고 마지막 투여 24시간 후에 부검하여 혈액내 ALT 및 AST치, 간조직내 total protein, triglyceride 및 hydroxyproline 양을 측정하였으며, 간중량도 측정하였다. 3. 약물투여 음성대조 : 1% CMC-Na 용액(10$m\ell$/kg b.wt.) 양성대조 : silymarin(25mg/kg), DDB(37.5mg/kg) 인진호추출분획 : KP(180mg/kg), PS-1(300mg/kg), PS-2(300mg/kg), EE (500mg/kg ), HH(640mg/kg), BE(1500mg/kg)

Protective Effects of Chalcone Derivatives for Acute Liver Injury in Mice

  • The hepatoprotective effects of chalcone derivatives were evaluated in D-galactosamine/lipopolysaccharide (D-GaIN/LPS)-induced fulminant hepatic failure in mouse. Thirteen chalcone derivatives were synthesized for study and their hepatoprotective effects were evaluated by assessing aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in serum. Chalcone preparations were injected into mice at 12 hand 1 h before intraperitoneal injection of D-GaIN/LPS. After abdominal administration, changes in AST and ALT between the control and treated groups were observed. Ten of the synthesized chalcone derivatives exhibited inhibitory effects on D-GaIN/LPS-induced levels of AST and ALT in mice. Compounds 2, 3, 8, 9, and 12 markedly reduced serum AST and ALT at 8 h, inhibited hepatocyte necrosis and showed significant hepatoprotective activities. The activity of compound 3 was compared with the bifendate (DDB) through oral administration. Compound 3 showed much higher inhibitory effects than bifendate for decreasing AST and ALT activity. The results indicate that compound 3 has strong hepatoprotective activity through suppression of tumor necrosis factor­alpha (TNF-alpha) preduction, reduction of the histological change in the liver, and attenuated of hepatocyte apoptosis confirmed by DNA fragmentation assay.

Hepatoprotective Effect of Lactic Acid Bacteria, Inhibitors of $\beta$-Glucuronidase Production Against Intestinal Microflora

  • The hepatoprotective activity of lactic acid bacteria (Lactobacillus brevis HY7401, Lactobacillus acidophilus CSG and Bifidobacterium longum HY8001), which inhibited $\beta$-glucuronidase productivity of intestinal microflora, on t-BHP- or CCl$_4$-induced hepatotoxicity of mice were evaluated. These oral administration of lactic acid bacteria lowered $\beta$-glucuronidase production of intestinal microflora as well as Escherichia coli HGU-3. When lactic acid bacteria at a dose of 0.5 or 2 g (wet weight)/kg was orally administered on CCl$_4$-induced liver injury in mice, these bacteria significantly inhibited the increase of plasma alanine transferase and aspartate transferase activities by $17-57\%$ and $57-66\%$ of the $CCI_4$ control group, respectively. These lactic acid bacteria also showed the potent hepatoprotective effect against t-BHP-induced liver injury in mice. The inhibitory effects of these lactic acid bacteria were more potent than that of dimethyl diphenyl bicarboxylate (DDB), which have been used as a commercial hepatoprotective agent. Among these lactic acid bacteria, L. acidophilus CSG exhibited the most potent hepatoprotective effect. Based on these findings, we insist that an inhibitor of $\beta$-glucuronidase production in intestine, such as lactic acid bacteria, may be hepatoprotective.

Evaluation of a DDB design method for bridges isolated with triple pendulum bearings

  • In this study a direct displacement-based design (DDBD) procedure for a continuous deck bridge isolated with triple friction pendulum bearings (TFPB) has been proposed and the seismic demands of the bridge such as isolator's displacement and drift of piers obtained from this procedure evaluated under two-directional near-field ground motions. The structural model used here are continuous, three-span, castin-place concrete box girder bridge with a 30-degree skew which are isolated with 9 different TFPBs. By comparing the results of DDBD method with those of nonlinear time history analysis (NTHA), it can be concluded that the proposed procedure is able to predict seismic demands of similar isolated bridges with acceptable accuracy. Results of NTHA shows that dispersion of peak resultant responses for a group of ground motions increases by increasing their average value of responses. It needs to be noted that the demands parameters calculated by the DDBD procedure are almost overestimated for stiffer soil condition, but there is some underestimation in results of this method for softer soil condition.

Effect of Wave Load on the Member Force of Steel Structure of Floating Buildings

  • For floating buildings may fl oat on the water for a long time, they are constantly affected by various environmental loads such as wind and wave loads. In this study to find the wave effect on the floating building, five models are designed using steel moment resisting frame. It is assumed that the lower part of the floating building is a reinforced concrete pontoon, while the upper part is a three-story steel frame. To analyze floating buildings affected by wind and wave loads, hydro-dynamic and substructure analysis are performed. As input loads, this study set limits that the mean wind velocity is 35 m/s and the significant wave height is 0.5 m for the residential building. From the hydrodynamic analysis, the time-history acceleration of building is obtained and transformed into a base ground input for a substructure analysis of the superstructure of the building. Finally the mean of the maximum from 30 dynamic analysis of the floating buildings are used to be compared with the results of the same model on the ground. It was shown that the dynamic results with wind and wave loads are not always lesser than the static results which are calculated with static equivalent wind load for a building that is located on the ground.

  • This paper presents an efficiency enhancement for the high power amplifier using DDBS(Digitally-controlled Dynamic Bias Switching) method which dynamically provides the power amplifier with two bias voltage levels according to the input envelope signal. It is quite easy to adjust the control signal by using a digital processing. The fabricated DDBS PA was evaluated using an 64 QAM FDD LTE signal, which has a center frequency of 2.6 GHz, a bandwidth of 10 MHz and a PAPR of 9.5 dB. The DDBS increases the power amplifier's PAE(Power-Added Efficiency) from 40.9 % to 48 %, at an average output power level of 43 dBm.
