Studies on proteolytic enzyme from A New Strain in Serratia sp. (Serratia속의 신균주가 생산하는 단백질분해효소)

  • Serratia sp. Y-4 was isolated from soil. Many characteristics of the strain and optimum cultivation condition for protease production were investigated.,The protease from Serratia sp. Y-4 was purified and studied for the properties of the enzyme. The isolated strain was identified to the genus Serratia. The strain was cultivated in 1%-casein, 0.5%-Na$_{3}$PO$_{4}$.7H$_{2}$O, 0.1%-NaCl, 0.05%-KCI, 0.02%-MgSO$_{4}$.7H$_{2}$O, 0.02%-CaCl$_{2}$.2H$_{2}$O, 0.02%-ZnSO$_{4}$.7H$_{2}$O, 0.02%-MnCl$_{2}$.4H$_{2}$O and 0.5%-soy bean oil at pH 7.0 for 35 hrs. The enzyme was purified about 5.89 fold from the culture broth with 31.1% recovery and 19,613 u/mg through ultrafiltration, ammonium sulfate, DEAE-sephacel and Superose-12 chromatography. The purified enzyme was identified as one band by isoelectric focusing, SDS- and native-PAGE. It has maxium activity at $37^{\circ}C$ and pH 9.0. Molecular weight of it is approx. 50 kD and pl is about 6.70. Its Km value for casein was 20 mg/ml. 5 mM-EDTA, 5mM-SDS, Ag$^{+1}$, Cu$^{+2}$, Hg$^{+2}$ and Pb$^{+2}$ inhibited the enzyme.

  • This study was carried out to investigate the micromorphology of vessel wall sculpture such as shape of perforation plate, perforation rim, steepness of perforation plate, occurrence of vestured pit, warty layer and ray-vessel pitting on 13 species of Fagaceae in Korea. The results were summarized as follows ; 1. In the species examined, all sample species except Fagus crenata var. multinervis have simple perforation plate. But Fagus crenata var. multinervis has simple and scalariform perforation plate (it was called to combination perforation plate) in a tree stem. 2. The shapes of perforation rim could be classified into five types; Type A without tail, Type B with small tail on both sides, Type C with long tail on both sides, Type D with tail only one side and Type E with the very short interval between perforations. Among five types, Type Band C have higer frequency of distribution than the others. 3. The steepness of perforation plate measured was about 20 degree on pore zone and 43 degree on outside parts of pore zone. 4. Species with vestured pit were Q. aliena, Q, dentata, Q. variabilis, Fagus crenata var. multinervis, Castanopsis cuspidata var sieboldii, Q. acuta, Q. stenophylla, and Q. glauca. But the extent of vesturing was very slight and vestured pits were observed mainly in ray-vessel pit of some vessel elements. 5. Species with warty layer on the inner surface wall of all vessel elements were Q. acuta, Q. stenophylla, Q. glauca, Fagus crenata var. multinervis and species with warty layer of some vessel elements were Q. aliena, Q, dentata, Q. variabilis, Castanopsis cuspidata var. sieboldii. 6. Species with palisade ray-vessel pit were Q. acuta, Q. stenophylla, Q. glauca, Castanea crenata, Castanea bungeana, Q. aliena, Q. serrata, Q. mongolica, and species with scalariform ray-vessel pit was Fagus crenata var. multinervis and species with oval ray-vessel pit were Q, dentata, Castanopsis cuspidata var sieboldii.

  • The first example of lead compound from $Pb(NO_3)_2$ and $H_3L$ N-heterocyclic ligand $(H_3L\;=\;(HO_2C)_2(C_3N_2)(C_3H_7)CH_2(C_6H_4)(C_6H_3)CO_2H)$, $[Pb_4(L')_4]{\cdot}5H_2O$ 1 (L' = OOC$(C_3H_7)(C_3N_2)CH_2(C_6H_4)(C_6H_3)COO)$, has been obtained under hydrothermal condition by decarboxylation, and characterized by elemental analysis, IR, TGDTA, and single-crystal X-ray diffraction. Compound 1 possesses a rare two-dimensional upper-lower offset terrace-like layer structure. In 1, crystallographic distinct Pb(II) ion adopts five-coordination geometry, and two lattice water molecules occupy the voids between 2-D layers. Results of solid state fluorescence measurement indicate that the emission band 458 nm may be assigned to $\pi^*-n$ and $\pi^*-\pi$ electronic transitions within the aromatic systems of the ligand L', however, the emission bands centred at 555 nm, 600 nm and 719 nm may be derived from phosphorescent emission ($\lambda_{excitation}$ = 390 nm).

  • Various studies such as INS(: Inertial Navigation System) are conducting to estimate the position of UAV, because the GPS information of UAV is at risk like the GPS jamming. The position estimation using DoA and RTT are used to apply many radar systems, and that process can be applied in datalink of UAV. The general monopulse feed in UAV datalink is Multi-horn, because of the wide BW(: Band Width) and frequency range. And it needs wide SNR range of tracking because of the limited transmit power of airborne unit. The estimation error of position increase at low SNR, and the DoA is valid in only 3dB beam width but high SNR causes false of mainlobe detection because of large sidelobe. In this paper, We propose the method to achieve higher accuracy of DoA estimation on low SNR and review some idea that able to detect mainlobe.

Design of a Channel Combiner for Digital Terrestrial Television Transmission (디지털 TV 방송 송신용 채널 컴바이너 설계)

  • In this paper, a channel combiner is designed to combine two adjacent digital TV channels without interference to transmit these channels with one broadband antenna. A constant impedance type combiner, consisting of two identical bandpass filters and two identical 3 dB power combiners, is chosen as a channel combiner. The bandpass filter used in the channel combiner also suppresses out-of-band signal caused by digital transmitter's non-linearity. H-plane type bandpass filters and branch-line fewer combiners are adopted in the design. The simulated results of the designed bandpass filter and power combiner show excellent agreement with the theoretical results. Finally, a channel combiner is designed by combining filters with rover combiners and the characteristics are simulated. It is shown that the designed channel combiner shows excellent performance.

  • Cerebellar ataxia is a steadily progressive neurodegenerative disease associated with loss of motor control, leaving patients unable to walk, talk, or perform activities of daily living. Direct motor instruction in cerebella ataxia patients has limited effectiveness, presumably because an inappropriate closed-loop cerebellar response to the inevitable observed error confounds motor learning mechanisms. Recent studies have validated the age-old technique of employing motor imagery training (mental rehearsal of a movement) to boost motor performance in athletes, much as a champion downhill skier visualizes the course prior to embarking on a run. Could the use of EEG based BCI provide advanced biofeedback to improve motor imagery and provide a "backdoor" to improving motor performance in ataxia patients? In order to determine the feasibility of using EEG-based BCI control in this population, we compare the ability to modulate mu-band power (8-12 Hz) by performing a cued motor imagery task in an ataxia patient and healthy control.

  • IGRINS, the Immersion GRating INfrared Spectrometer, is a near-infrared wide-band high-resolution spectrograph jointly developed by the Korea Astronomy and Space Science Institute and the University of Texas at Austin. IGRINS employs three HAWAII-2RG focal plane array (FPA) detectors. The mechanical mounts for these detectors serves a critical function in the overall instrument design: Optically, they permit the only positional compensation in the otherwise "build to print" design. Thermally, they permit setting and control of the detector operating temperature independently of the cryostat bench. We present the design and fabrication of the mechanical mount as a single module. The detector mount includes the array housing, a housing for the SIDECAR ASIC, a field flattener lens holder, and a support base. The detector and ASIC housing will be kept at 65 K and the support base at 130 K. G10 supports thermally isolate the detector and ASIC housing from the support base. The field flattening lens holder attaches directly to the FPA array housing and holds the lens with a six-point kinematic mount. Fine adjustment features permit changes in axial position and in yaw and pitch angles. We optimized the structural stability and thermal characteristics of the mount design using computer-aided 3D modeling and finite element analysis. Based on the computer simulation, the designed detector mount meets the optical and thermal requirements very well.

  • ZnO-doped near-stoichiometric $LiNbO_3$ single crystals of $0.8{\sim}1.0mm$ diameter and $30{\sim}35mm$ length were grown by the micro-pulling down (U-PD) method. The structure of the grown crystals was confirmed by powder x-ray diffraction (XRD) patterns. Electron probe micro analysis (EPMA) showed that Zn ions were homogeneously incorporated In grown crystals. The threshold in ZnO doping level was confirmed that an abrupt change in the features of $OH^-$ absorption band as doping level reaching about 2 mol%.

  • The impedance cardiogram has been proposed as a non-invasive, continuous, operator independent, and cost-effective method for cardiac output monitoring. However, it can not be completely considered as non-restrictive method because of attached spot and band type electrodes. Therefore, we developed a improved convenient hand-held typed measurement system for cardiac output by electrical impedance technique. 80 subjects from Yonsei University and the surrounding areas, participated. All subjects measured stroke volume and cardiac output through Physioflow and developed system. To verify the developed system, statistical methods such as correlation, Wilcoxon signed ranks test, and the Bland-Altman analysis were used. The proposed system showed significant correlation in both male and female stroke volume(r=0.715, r=0.704) and cardiac output(r=0.826; r=0.804). From these results, it can be concluded that stroke volume and cardiac output could be improved convenient measurement using the both hands without the help of a specialist.

Interaction Between Acid-Labile Subunit and Insulin-like Growth Factor Binding Protein 3 Expressed in Xenopus Oocytes

  • The acid-bible subunit (ALS) associates with the insulinlike growth factor (IGF)-I or II, and the IGF binding protein-3 (IGFBP-3) in order to form a 150-kD complex in the circulation. This complex may regulate the serum IGFs by restricting them in the vascular system and promoting their endocrine actions. Little is known about how ALS binds to IGFBP3, which connects the IGFs to ALS. Xenopus oocyte was utilized to study the function of ALS in assembling IGFs into the ternary complexes. Xenopus oocyte was shown to correctly translate in vitro transcribed mRNAs of ALS and IGFBP3. IGFBP3 and ALS mRNAs were injected in a mixture, and their products were immunoprecipitated by antisera against ALS and IGFBP3. Contrary to traditional reports that ALS interacts only with IGF-bound IGFBP3, this study shows that ALS is capable of forming a binary complex with IGFBP3 in the absence of IGF When cross-linked by disuccinimidyl suberate, the band that represents the ALS-IGFBP3 complex was evident on the PAGE. IGFBP3 movement was monitored according to the distribution between the hemispheres. Following a localized translation in the vegetal hemisphere, IGFBP3 remained in the vegetal half in the presence of ALS. However, the mutant IGFBP3 freely diffused into the animal half, despite the presence of ALS, which is different from the wild type IGFBP3. This study, therefore, suggests that ALS may play an important role in sequestering IGFBP3 polypeptides via the intermolecular aggregation. Studies using this heterologous model will lead to a better understanding of the IGFBP3 and ALS that assemble into the ternary structure and circulate the IGF system.
