  • 쇼트피닝 기술은 크게 피로강도 향상을 위한 쇼트피닝(shot peening), 재료의 청정 및 도장 품질 향상을 위한 쇼트블라스트(shot blast) 그리고 쇼트피닝 시 판재의 변형되는 성질을 이용한 핀포밍(peen forming) 등으로 구분할 수 있다. 그 중 본 연구에서는 해양산업 분야에서 널리 사용되는 회주철의 효과적인 내구성 향상을 위해 쇼트피닝 기술을 적용하였다. 그러나 쇼트피닝 기술 적용에 있어서 가장 중요한 것은 제품의 균일성, 정확성, 신뢰성을 확보하기 위해 쇼트피닝 강도를 제어하는 여러 가지 변수들에 대하여 최적 상태를 유지하는 것이다. 따라서 회주철에 대한 최적 쇼트피닝 분사조건 규명작업은 반드시 쇼트피닝 가공 전에 수반되어야만 한다. 그 일환으로 실험은 쇼트피닝 분사시간과 분사압력을 변수로 하여 회주철 표면에 적용하였으며, 기계적 특성 평가를 통해 최적의 쇼트피닝 조건을 규명하고자 하였다. 쇼트피닝 분사조건에 따른 회주철의 내구성을 평가하기 위해 캐비테이션 실험을 실시하였으며, 경도 측정, 횡단면 관찰 및 표면의 3D 현미경 관찰 등을 통해 기계적 특성을 분석하였다. 캐비테이션 실험은 ASTM G32 규정에 의거하여 천연해수 내 $30^{\circ}C$에서 $50{\mu}m$의 진폭으로 실시하였다. 실험 후에는 주사전자현미경으로 손상표면을 관찰하였으며, 손상 정도를 비교하기 위해 무게 감소량을 계측하여 상호 비교/분석하였다.

C-Ring Cleavage of Isoflavones Daidzein and Genistein by a Newly-Isolated Human Intestinal Bacterium Eubacterium ramulus Julong 601

  • Julong 601, a Gram-positive anaerobic bacterium strain capable of cleaving the C-ring of isoflavones daidzein and genistein, was isolated from human feces. BLAST search revealed that its complete 16S rDNA gene sequence has 99% similarity to Eubacterium ramulus. Metabolites of daidzein and genistein were determined as O-desmethylangolensin (O-Dma) and 2-(4-hydroxyphenyl) propionic acid (2-HPPA), respectively, based on UV, EI-MS, and $^1H$ and ^{13}C$ NMR spectral analyses. Enantiomers of O-Dma and 2-HPPA were isolated by chiral stationary-phase HPLC (CSP HPLC). Cleavage of the C-ring of daidzein and genistein by strain Julong 601 was highly enantioselective. Specific rotation ([$\alpha]_D$) and circular dichroism (CD) spectra of the enantiomers are reported here for the first time. Biotransformation kinetics of daidzein and genistein indicated that the C-ring of genistein has a higher susceptibility to bacterial degradation than that of daidzein.

Experimental study on rheology, strength and durability properties of high strength self-compacting concrete

  • The rheological behaviour of high strength self compacting concrete (HS-SCC) studied through an experimental investigation is presented in this paper. The effect of variation in supplementary cementitious materials (SCM) $vis-{\grave{a}}-vis$ four different types of processed crushed sand as fine aggregates is studied. Apart from the ordinary Portland cement (OPC), the SCMs such as fly ash (FA), ground granulated blast furnace slag (GGBS) ultrafine slag (UFS) and micro-silica (MS) are used in different percentages keeping the mix -paste volume and flow of concrete, constant. The combinations of rheology, strength and durability are equally important for selection of mixes in respect of high-rise building constructions. These combinations are referred to as the rheo-strength and rheo-durability which is scientifically linked to performance based rating. The findings show that the fineness of the sands and types of SCM affects the rheo-strength and rheo-durability performance of HS-SCC. The high amount of fines often seen in fine aggregates contributes to the higher yield stress. Further, the mixes with processed sand is found to offer better rheology as compared to that of mixes made using unwashed crushed sand, washed plaster sand, washed fine natural sand. The micro silica and ultra-fine slag conjunction with washed crushed sand can be a good solution for high rise construction in terms of rheo-strength and rheo-durability performance.

  • Many methods and techniques to reduce ground vibrations are well known. Some of them are to adopt electric milisecond detonators with a sequential blasting machine or an initiating system with an adequate number of delay intervals. The types of electric detonators munufactured in Korea include instantaneous, decisecond and milisecond delays byt numbers of delay intervals are only limite from No.1 to No.20 respectively. It is not sufficient to control accurately milisecond time with these detonators in tunnel excavation. Sequential fire time refers to adding an external time delay to a detonators norminal firing time to obtain sequential initiation and it is determined by sequential timer setting. To reduce the vibration level, sequential blasting machine with decisecond detonatore was adopted. A total of 134 blasting was recorded at various sites. Blast-to-structure distances ranged from 20.3 to 42.0 meter, where charge weight varied from 0.25 to 0.75 kg per delay. The results can be summarized as follow : 1. The effects of sequential blasting machine on the vibration level are discussed. The vibration level by S.B.M. are decreased approximately 14.38~18.05 to compare to level of conventional blasting and cycle time per round can be saved. 2. The empirical equations of particle velocity were obtained in S,B.M. and conventional blastin. $V=K(D/W^{1/3})-n$. where the values for n and k are estimated to be 1.665 to 1.710 and 93.59 to 137 respectively. 3. The growth of cracks due to vibrations are found but the level fall to within allowable value.

Evaluation of Rock Fragmentation due to Artificial Joint Effect (인공절리에 의한 암석의 파쇄도 평가)

  • Since the rock fragmentation by blasting can affect the subsequent processes including loading, hauling and secondary crushing, its control is essential for the assessment of blasting efficiency as well as production cost. In this study, we were analyzed the rock fragmentation by the direction of artificial joint. The underground blasting experiments were performed after forming the vertical and horizontal artificial joints. The blast fragmentation was conducted by the split-desktop which is a 2D image processing program. As a result, it was found that the horizontal artificial joint was evaluated to have lower overall the size of muck pile than the vertical artificial joint and the distribution of the size of muck pile was varied. It is possible that the direction of artificial joint could suppress the occur of oversize muck pile and control to a certain size or less.

  • Objective: To understand the domestication and spread of horses in history, genetic information is essential. However, mitogenetic traits of ancient or medieval horses have yet to be comprehensively revealed, especially for East Asia. This study thus set out to reveal the maternal lineage of skeletal horse remains retrieved from a 15th century archaeological site (Gongpyeongdong) at Old Seoul City in South Korea. Methods: We extracted DNA from the femur of Equus caballus (SNU-A001) from Joseon period Gongpyeongdong site. Mitochondrial (mt) DNA (HRS 15128-16116) of E. caballus was amplified by polymerase chain reaction. Cloning and sequencing were conducted for the mtDNA amplicons. The sequencing results were analyzed by NCBI/BLAST and phylogenetic tool of MEGA7 software. Results: By means of mtDNA cytochrome b and D-loop analysis, we found that the 15th century Korean horse belonged to haplogroup Q representing those horses that have historically been raised widely in East Asia. Conclusion: The horse is unique among domesticated animals for the remarkable impact it has on human civilization in terms of transportation and trade. Utilizing the Joseon-period horse remains, we can obtain clues to reveal the genetic traits of Korean horse that existed before the introduction of Western horses.

Comparative proteomic analysis of Celastrus hindsii Benth. phenotypes reveals an intraspecific variation

  • In Vietnam, Celastrus hindsii Benth, a medicinal plant rich in secondary metabolites, has been used to alleviate distress caused by ulcers, tumors, and inflammation for generations. The occurrence of two phenotypes, Broad Leaf (BL) and Narrow Leaf (NL), has raised questions about the selection of appropriate varieties for conservation and crop improvement to enhance medicinal properties. This study examined molecular differences in C. hindsii by comparing protein profiles between the NL and BL types using 2D-PAGE and MS. Peptide sequences and proteins were identified by matching MS data against the MSPnr100 databases and verified using the MultiIdent tool on ExPASy and the Blast2GO software. Our results revealed notable variations in protein abundance between the NL and BL proteomes. Selected proteins were confidently identified from 12 protein spots, thereby highlighting the molecular variation between NL and BL proteomes. Upregulated proteins in BL were found to be associated with flavonoid and amino acid biosynthesis as well as nuclease metabolism, which probably attributed to the intraspecific variations. Several bioactive proteins identified in this study can have applications in cancer therapeutics. Therefore, the BL phenotype characterized by healthier external morphological features has higher levels of bioactive compounds and could be better suited for medicinal use.

Bending and Shear Capacity of Reinforced Concrete Protective Wall (휨과 전단을 고려한 철근콘크리트 방호벽 성능에 관한 연구)

  • With the recent increase in gas energy use, risk management for explosion accidents has been emphasized. Protective walls can be used to reduce damage from explosions. The KOSHA GUIDE D-65-2018 suggests the minimum thickness and height of protective walls, minimum reinforcement diameter, and maximum spacing of reinforcements for the structural safety of the protective walls. However, no related evidence has been presented. In this study, the blast load carrying capacity of the protective wall was analyzed by the pressure-impulse diagrams while changing the yield strength of the reinforcement, concrete compressive strength, reinforcement ratio, protective wall height, and thickness, to check the adequacy of the KOSHA GUIDE. Results show that failure may occur even with design based on the criteria presented by KOSHA GUIDE. In order to achieve structural safety of protective walls, additional criteria for minimum reinforcement yield strength and maximum height of protective wall are suggested for inclusion in KOSHA GUIDE. Moreover, the existing value for minimum reinforcement ratio and the thickness of the protective wall should be increased.

Deletion of GSTM1 and T1 Genes as a Risk Factor for Development of Acute Leukemia

  • The glutathione S-transferases (GSTs) are a family of enzymes involved in the detoxification of a wide range of chemicals, including important environmental carcinogens, as well as chemotherapeutic agents. In the present study 294 acute leukemia cases, comprising 152 of acute lymphocytic leukemia (ALL) and 142 of acute myeloid leukemia, and 251 control samples were analyzed for GSTM1 and GSTT1 polymorphisms through multiplex PCR methods. Significantly increased frequencies of GSTM1 null genotype (M0), GSTT1 null genotype (T0) and GST double null genotype (T0M0) were observed in the both ALL and AML cases as compared to controls. When data were analyzed with respect to clinical variables, increased mean levels of WBC, Blast %, LDH and significant reduction in DFS were observed in both ALL and AML cases with T0 genotype. In conclusion, absence of both GST M & GST T might confer increased risk of developing ALL or AML. The absence of GST enzyme might lead to oxidative stress and subsequent DNA damage resulting in genomic instability, a hallmark of acute leukemia. The GST enzyme deficiency might also exert impact on clinical prognosis leading to poorer DFS. Hence GST genotyping can be made mandatory in management of acute leukemia so that more aggressive therapy such as allogenic stem cell transplantation may be planned in the case of patients with a null genotype.

Isolation and Identification of Yeasts from Wild Flowers in Deogyu Mountain and their Physiological Functionalities (전라북도 덕유산 일대의 야생화에서 효모의 분리 동정 및 생리기능성)

  • Several kinds of yeasts were isolated from wild flowers collected from Muju resort of Deogyu mountain in Korea and identified by comparison of nucleotide sequences of D1/D2 region of 26S rDNA using BLAST. Totally twenty-two strains of eight species were isolated and Cryptococcus magnus (9 strains) and Cryptococcus flavescenes (6 strains) were dominant species. The supernatant and cell-free extracts were prepared and their antioxidant activity, anti-gout xanthine oxidase inhibitory activity and whitening tyrosinase inhibitory activity were investigated. Anti-gout xanthine oxidase inhibitory activities of Sporobolomyces phaffii JS00583 and Rhodotorula graminis JS00581 were 40.4% and 39.1%, respectively and also was 36.1% whitening tyrosinase inhibitory activity in the supernatant of Cryptococcus magnus JS00570.
