  • In this study, in order to develop renewable bio-based nanocomposites, multi-functional nanocomposites from soybean resins (AESO, MAESO) and nanoclay were prepared. Photoelectrodes for environmental friendly dye-sensitized solar cell using soybean resin were also prepared. Organo-modified nanoclay was directly dispersed in functionalized soybean resins after mixing with styrene as a comonomer and radical initiator was used to copolymerize the nanocomposites. The observed morphology was a mixture of intercalated/exfoliated structure and the physical properties were improved by adding nanoclay. A nanocomposite using MAESO, which added COOH functional group to the soybean resin, showed better dispersibility than AESO composites. Ultrasonic treatment of the nanocomposites also improved the physical properties. Nanoporous $TiO_2$ photoelectrode was also prepared using soybean resins as a binder, after acid-treatment of $TiO_2$ surface using nitric acid. Dye-sensitized solar cells were prepared after adsorbing dye molecules on it. The $TiO_2$ photoelectrode prepared using soybean binder had high current density because of increased surface area by improved dispersibility. The photoelectrochemical properties and conversion efficiency of the solar cell were significantly improved using the soybean binder.

  • Background: The purpose of this study was to assess the dosimetric and clinical feasibility of volumetric modulated arc based hypofractionated stereotactic radiotherapy (RapidArc) treatment for large acoustic schwannoma (AS >10cc). Materials and Methods: Ten AS patients were immobilized using BrainLab mask. They were subject to multimodality imaging (magnetic resonance and computed tomography) to contour target and organs at risk (brainstem and cochlea). Volumetric modulated arc therapy (VMAT) based stereotactic plans were optimized in Eclipse (V11) treatment planning system (TPS) using progressive resolution optimizer-III and final dose calculations were performed using analytical anisotropic algorithm with 1.5 mm grid resolution. All AS presented in this study were treated with VMAT based HSRT to a total dose of 25Gy in 5 fractions (5fractions/week). VMAT plan contains 2-4 non-coplanar arcs. Treatment planning was performed to achieve at least 99% of PTV volume (D99) receives 100% of prescription dose (25Gy), while dose to OAR's were kept below the tolerance limits. Dose-volume histograms (DVH) were analyzed to assess plan quality. Treatments were delivered using upgraded 6 MV un-flattened photon beam (FFF) from Clinac-iX machine. Extensive pretreatment quality assurance measurements were carried out to report on quality of delivery. Point dosimetry was performed using three different detectors, which includes CC13 ion-chamber, Exradin A14 ion-chamber and Exradin W1 plastic scintillator detector (PSD) which have measuring volume of $0.13cm^3$, $0.009cm^3$ and $0.002cm^3$ respectively. Results: Average PTV volume of AS was 11.3cc (${\pm}4.8$), and located in eloquent areas. VMAT plans provided complete PTV coverage with average conformity index of 1.06 (${\pm}0.05$). OAR's dose were kept below tolerance limit recommend by American Association of Physicist in Medicine task group-101(brainstem $V_{0.5cc}$ < 23Gy, cochlea maximum < 25Gy and Optic pathway <25Gy). PSD resulted in superior dosimetric accuracy compared with other two detectors (p=0.021 for PSD.

  • The effect of ammoniation with urea and with soybean meal (SBM) as a source of urease on the nutritive value of wheat straw was evaluated in sheep. Twenty-four male Najdi lambs were used in a 3${\times}$2 factorial design, in which the animals were allocated to three straw treatments: 0% urea-treated (NT), 6% urea-treated (UT) and 2.2% urea-supplemented (US) straws. Each straw treatment was either supplemented or non-supplemented with 70 g SBM $kg^{-1}$ straw during the treatment time with urea, giving a total of six straw treatments. Each of these treatments was individually fed ad libitum to 4 lambs, together with 300 g of barley grain/head/day. Total N content of UT and US straws increased significantly (p<0.001) as compared to NT straw. The degree of urea hydrolysis, either with or without SBM addition, was nearly similar. Lambs fed either UT or US straw based diets had significantly (p<0.01) and numerically (p>0.05) higher straw DM intake (g $d^{-1}$ $kg^{-1}$ $BW^{0.75}$), compared to those fed NT straw based diet. Apparent DM or OM digestibilities increased significantly (p = 0.014) in lambs fed UT diet, and numerically (p>0.05) in lambs fed US diet as compared to those fed NT diet. Fiber (CF, NDF, ADF, cellulose and hemicellulose) digestibility increased to a similar magnitude, averaging 20.2 (p<0.001) and 7.8% (p<0.07); this corresponds to 35 (p<0.001) and 51% (p<0.001) in N digestibility and approximately 78 (p<0.017) and 105% (p<0.002) in N retention, for UT and US diets, respectively, as compared to NT diet. However, the UT diet had higher (p<0.01) fiber digestibility over the US diet. Addition of SBM tended to improve (p = 0.09) straw DM and digestible OM intakes, while significantly increasing (p<0.001) total and digestible CP intakes across all diets. Lambs fed on US diet had higher ruminal ammonia N than those fed on UT (p<0.05) or NT (p<0.001) diets. However, ruminal pH and molar proportion of the volatile fatty acids did not differ (p>0.05) among the treatment diets. This study suggests that US and UT treatments, particularly the latter, improved straw intake, digestibility and N utilization by lambs compared to NT treatment. On the other hand, addition of SBM as a source of urease had a negligible effect on urea hydrolysis.

  • Background: Clinoptilolite is a natural zeolite with high adsorption capacity for polar mycotoxins such as aflatoxins. The efficacy of clinoptilolite in ameliorating the toxic effects of aflatoxicosis has been proven in monogastric animals, but there is no such evidence for ruminants. The aim of this study was to evaluate, under field conditions, whether the dietary administration of clinoptilolite in dairy cows could reduce the concentration of aflatoxin M1 ($AFM_1$) in bulk-tank milk, in farms with higher than or close to $0.05{\mu}g/kg$ of milk (European maximum allowed residual level). An objective of the present study was also to investigate the effect of particle size of clinoptilolite on aflatoxin binding. Methods: Fifteen commercial Greek dairy herds with AFM1 concentrations in bulk tank milk ${\geq}0.05{\mu}g/kg$ were selected. Bulk tank milk AFM1 was determined prior to the onset and on day 7 of the experiment. Clinoptilolite was added in the total mixed rations of all farms at the rate of 200 g per animal per day, throughout this period. Two different particle sizes of clinoptilolite were used; less than 0.15 mm in 9 farms (LC group) and less than 0.8 mm in 6 farms (HC group). Results: Clinoptilolite administration significantly reduced $AFM_1$ concentrations in milk in all farms tested at an average rate of 56.2 % (SD: 15.11). The mean milk $AFM_1$ concentration recorded on Day 7 was significantly (P < 0.001) lower compared to that of Day 0 ($0.036{\pm}0.0061$ vs. $0.078{\pm}0.0074{\mu}g/kg$). In LC group farms the reduction of milk $AFM_1$ concentration was significantly higher than HC group farms ($0.046{\pm}0.0074$ vs. $0.036{\pm}0.0061{\mu}g/kg$, P = 0.002). As indicated by the Pearson correlation, there was a significant and strong linear correlation among the milk $AFM_1$ concentrations on Days 0 and 7 (R = 0.95, P < 0.001). Conclusions: Dietary administration of clinoptilolite, especially of smallest particle size, at the rate of 200 g per cow per day can effectively reduce milk $AFM_1$ concentration in dairy cattle and can be used as a preventive measure for the amelioration of the risks associated with the presence of aflatoxins in the milk of dairy cows.

  • Reducing $[Mg^{2+}]_o$ to 0.1 mM can evoke repetitive $[Ca^{2+}]_i$ spikes and seizure activity, which induces neuronal cell death in a process called excitotoxicity. We examined the issue of whether cultured rat hippocampal neurons preconditioned by a brief exposure to 0.1 mM $[Mg^{2+}]_o$ are rendered resistant to excitotoxicity induced by a subsequent prolonged exposure and whether $Ca^{2+}$ spikes are involved in this process. Preconditioning by an exposure to 0.1 mM $[Mg^{2+}]_o$ for 5 min inhibited significantly subsequent 24 h exposure-induced cell death 24 h later (tolerance). Such tolerance was prevented by both the NMDA receptor antagonist D-AP5 and the L-type $Ca^{2+}$ channel antagonist nimodipine, which blocked 0.1 mM $[Mg^{2+}]_o$-induced $[Ca^{2+}]_i$ spikes. The AMPA receptor antagonist NBQX significantly inhibited both the tolerance and the $[Ca^{2+}]_i$ spikes. The intracellular $Ca^{2+}$ chelator BAPTA-AM significantly prevented the tolerance. The nonspecific PKC inhibitor staurosporin inhibited the tolerance without affecting the $[Ca^{2+}]_i$ spikes. While $G{\ddot{o}}6976$, a specific inhibitor of $PKC{\alpha}$ had no effect on the tolerance, both the $PKC{\varepsilon}$ translocation inhibitor and the $PKC{\zeta}$ pseudosubstrate inhibitor significantly inhibited the tolerance without affecting the $[Ca^{2+}]_i$ spikes. Furthermore, JAK-2 inhibitor AG490, MAPK kinase inhibitor PD98059, and CaMKII inhibitor KN-62 inhibited the tolerance, but PI-3 kinase inhibitor LY294,002 did not. The protein synthesis inhibitor cycloheximide significantly inhibited the tolerance. Collectively, these results suggest that low $[Mg^{2+}]_o$ preconditioning induced excitotoxic tolerance was directly or indirectly mediated through the $[Ca^{2+}]_i$ spike-induced activation of $PKC{\varepsilon}$ and $PKC{\xi}$, JAK-2, MAPK kinase, CaMKII and the de novo synthesis of proteins.

  • Background & Objective : Naeso-san(NSS) has been used for the treatment of functional dyspepsia, regarded as a gastric dysmotility disease. A main cause of gastric dysmotility is antral dilatation or antroduodenal uncoordination. Therefore, we investigated the effect of NSS on gastric motility and its mechanism of action, as well as the morphologic changes in antral dilatated rats. Methods : Antral dilatated rats were induced by wrapping a nonabsorbable rubber ring(D:6mm, W:4mm, T:1mm) around the 1st portion of the duodenum for 8 weeks. Then morphologic changes were investigated and compared with normal intact rats before and after 8 weeks. Gastric emptying was measured by administration of normal saline(NS) or NSS in normal intact and antral dilatated rats. In another series of experiments to evaluate the mechanism of NSS under delayed conditions, normal intact rats were treated with atropine sulfate(1mg/kg, s.c.), quinpirole HCl(0.3mg/kg, i.p.), $NAME(N^{G}-nitro-L-arginine$ methyl ester, 75mg/kg, s.c.) and cisplatin(10mg/kg, i.p.), respectively. The myoelectrical activity of the gastric smooth muscle was recorded in normal intact and antral dilatated rats. The contractile waves were measured for 30 minutes before and after administration of each solution(NS, NSS). Results : Body weight gain of antral dilatated rats was significantly lower than that of the controls. Futhermore, we found the thickness of the mucosal and muscular layers and surface area of the stomach increased significantly compared with controls. NSS 278㎎/㎏ improved gastric emptying more than normal saline or NSS 93mg/kg in normal intact(p=0.026) and antral dilatated rats(p=0.03). NSS enhanced gastric emptying significantly in the NAME treated group(p=0.002). NSS 278mg/kg increased the significant postprandial dominant power than that of NS in normal intact rats, whereas there was no statistical significance in antral dilatated rats. Conclusions : NSS stimulates gastric motility through the cholinergic pathway. We expect that pathologic model with antral dilatation can be used as an exprimental tool which is similar to dyspepsia and NSS would be effective especially in dysmotility-like functional dyspepsia with antral dilatation or impaired reservoir functions such as gastric adaptive relaxation.

  • The purpose of this study was to develop an indirect enzyme-linked immunosorbent assay (indirect ELISA) based on a monoclonal antibody (MAb) that is specific to mackerel thermal stable-soluble protein (TSSP), that can be used for the rapid detection of mackerel in processed marine foods. Among the four MAbs (3A5-1, 2, 9, and 12) developed in previous studies, the 3A5-2 MAb that showed high specificity and sensitivity were selected and used to develop the indirect ELISA method. The detection range of the indirect ELISA was 0.02%-0.001% and the detection limit of 0.001% was shown. No cross-reaction to other marine products and food ingredients was observed by the indirect ELISA. Processed marine foods containing mackerel with ${\geq}0.3$ O.D. value at 405 nm were estimated as positive samples by the indirect ELISA. Therefore, the indirect ELISA can be used as a rapid and sensitive method to identify mackerel authenticity and adulteration in processed marine foods.

  • The purpose of this study was to examine the relationships between depression, stress, self-esteem, oral health-related quality of life (OHIP-14) in middle-aged women. Data were as collected by self-reported questionnaires from 205 middle-age women. The instruments were the Center for Epidemiologic Studies Depression scale (CES-D), Brief Encounter Psychosocial Instrument (BEPSI), Self-Esteem Scale (SES). The data were analyzed using t-test, one-way ANOVA, Pearson correlation coefficients and multiple regression with IBM SPSS Statistics ver. 21.0. Pearson correlation coefficient analysis found that oral health impact profile (OHIP) was significantly associated with depression (r=-0.560, p<0.001), stress (r=-0.595, p<0.001), self-esteem (r=0.522, p<0.001). The OHIP was explained 39.4% by stress (${\beta}$=-0.362, p<0.001), self-esteem (${\beta}$=0.203, p=0.009) using multiple regression analysis. These results indicate that the intervention program of oral health promotion for community-middle- aged women is needed from now on.
