Evaluation of Maternal Behavior between Normal Parturition and Expected Cesarean Section in Rats (자연 분만 및 예정된 제왕절개 수술 랫드에 있어서 모성 행동의 차이에 대한 검토)

  • Oxytocin is a neurohypophyseal hormone which has multiple functions in mammals. Mainly, oxytocin regulates milk ejection and has an effect on uterine contraction and is related to maternal behavior. Maternal behavior is believed to be suppressed by stress and facilitated by oxytocin. In the cesarean section, oxytocin may be administrated into uterus to promote uterine involution. The present study aimed to test the effect of oxytocin into uterus on maternal behavior of rats with cesarean section. It was measured the effects on maternal behavior of oxytocin infused into uterus in rats with cesarean section as a stressor. In the first experiment, pup survival rate of between a control group and a group with laparotomy as a stress in natural parturition rats was compared. In the second experiment, survival rate for 2 weeks and maternal pup searching behavior (MPSV) were observed in one cesarean sectioned group without oxytocin and the other cesarean sectioned group with oxytocin. Infanticide was observed in stressed group in the first experiment while a normal maternal behavior was observed in a control one. In the second experiment, MPSV was only observed in a cesarean sectioned group with oxytocin and infanticide was observed in two groups except one rat which is thought to be affected by oxytocin as operated relatively late. This is the first study to show that the administration of oxytocin into uterus in the cesarean section is not involved in the regulation of maternal behavior in rats. In conclusion, this study proves the needs of oxytocin into brain in cesarean section related rats model and further study of maternal behavior list, like MPSV.

Microbial Hazard Analysis of Astragalus membranaceus Bunge for the Good Agricultural Practices (농산물우수관리를 위한 황기(Astragalus membranaceus Bunge)의 미생물학적 위해요소 분석)

  • The objective of this study was to analyze the microbiological hazards of Astragalus membranaceus Bunge on the post-harvest processing. Samples from processing equipments (cleaner, water, cart, table, tray and packaging machine), personal hygiene (hand) and harvested crops (before washing, after washing, after sorting, and after drying) were collected from four farms (A, B, C, and D) located in Chungchengbuk-do, Korea. The samples were analyzed for sanitary indication bacteria and pathogenic bacteria. First, total aerobic bacteria and coliform in processing facilities were detected at the levels of 0.93~4.86 and 0.33~2.28 log CFU/$100cm^2$ and/mL respectively. In particular, microbial contamination in hand (5.43~6.11 and 2.52~4.12 log CFU/Hand) showed higher than processing equipments. Among the pathogenic bacteria, Bacillus cereus was detected at the levels of 0.33~2.41 log CFU/$100cm^2$, 1.48~3.27 log CFU/Hand and 0.67~3.65 log CFU/g in equipments, hands, and plants and Staphylococcus aureus were detected in cleaner, table, hand and harvested crops (before washing and after sorting) by qualitative test. Escherichia coli O157:H7, Listeria monocytogenes, and Salmonella spp. were not detected. These results indicated that personal hygiene and processing equipments should be managed to reduce the microbial contamination of A. membranaceus Bunge. Therefore, management system such as good agricultural practices (GAP) criteria is needed for hygienic agricultural products.

Drag Characteristics According to Ground Rope and Seabeds in the Bottom Trawl (저층트롤의 발줄 및 저질의 종류에 따른 저항특성)

  • The model experiments for the Eastern sea bottom trawl were conducted for the 3 types of ground ropes and 2 types of seabeds to examine drag characteristics according to towing speed. The model net was based on the net of trawler with 750ps and made by 1/78 of a full scale net. This experiments focused on the drags for 3 types of ground rope, the serving wire, the original and the chain types and 2 types of seabeds, mud and sand of bottom. The results obtained are summarized as follows 1. In the case of the sand bottom, the drags by 3 types of ground rope were orderly increased in turn of the chain, the serving wire and the original type. But, in the case of the mud bottom, the drag of the original type is higher than that of the serving wire type and less than the chain type. 2. In the case of the serving wire type of ground rope, the drags by the diameters of ground rope, ø 1.3, ø 1.9, ø 2.6 and ø 3.25 were increased respectively by 23g, 25g, 32g and 42g at the towing speed of 0.4m/s. 3. The drags of ground ropes were increased exponentially in proportion to the towing speed, in the same way in all of three types of ground rope. 4. The ratios of drag of ground rope against that of the trawl net by the type of ground rope at the towing speed of 0.4m/s on the mud bottom were 0.69 in the chain type, 0.64 in the serving wire type and 0.67 in the original type respectively. 5. The Coefficient of drag($C_d$) by type of ground rope according to the towing speed were 10.0~56.7, 3.0~l6.0 and 1.5~8.5 respectively in turn of the chain type, the serving wire type and the original type on the mud bottom, and that on the sand bottom were 10.0~60.0, 3.0~14.0 and 1.2~6.0 respectively In turn of the chain type, the serving wire type and the original type.

Characteristics of Wheat Flour Dough and Noodles with Amylopectin Content and Hydrocolloids (아밀로펙틴 함량 변화와 하이드로콜로이드 첨가에 의한 밀가루 반죽 및 국수의 특성)

  • The effects of amylopectin and hydrocolloid (locust bean gum and guar gum) content on wheat flour dough and noodle properties were investigated. As the amount of amylopectin increased, the water absorption rate (farinograph), the tension (tension test), the gel stability (freeze-thawing treatment), and the springiness and the cohesiveness (TPA) increased, but the pasting temperature (RVA), the lightness and yellowness (color measurement), and the hardness (TPA) tended to decrease. In sensory evaluations, the scores for cohesiveness, springiness, and acceptability of cooked noodle increased as the proportion of amylopectin increased. The proper combination of amylose/amylopectin ratio and hydrocolloids improved the freeze-thaw stability and the sensory acceptability of wheat flour dough and noodle.

Production of hTPO Transgenic Chickens using Tetracycline-Inducible Expression System (Tetracycline-Inducible Expression System을 이용한 Human Thrombopoietin (hTPO) 형질전환 닭의 생산)

  • It is well-known that unregulated over-expression of foreign gene may have unwanted physiological or toxic effects in transgenic animals. To circumvent these problems, we constructed retrovirus vector designed to express the foreign gene under the control of the tetracycline-inducible promoter. However, gene expressions in the tetracycline-inducible expression system (Tet system) are not completely regulated but a little leaky due to the inherent defects in conventional Tet-based systems. A more tightly controllable regulatory system can be achieved when the advanced versions ($rtTA2^SM2$) of rtTA and a minimal promoter in responsive components (pTRE-tight) are used in combination therein. In this study, we tried to produce human thrombopoietin (hTPO) from various target cells and transgenic chickens using the retrovirus vector combined with Tet system. hTPO is the primary regulator of platelet production and has an important role in the survival and expansion of hematopoietic stem cells. In a preliminary experiment in vitro, higher hTPO expression and tighter expression control were observed in chicken embryonic fibroblast (CEF) cells. We also measured the biological activity of the hTPO using Mo7e cells whose proliferation is dependant on hTPO. The biological activity of the recombinant hTPO from CEF was higher than both its commercial counterpart and hTPO from other target cells. The recombinant retrovirus was injected beneath the blastoderm of non-incubated chicken embryos (stage X). Out of 138 injected eggs, 15 chicks hatched after 21 days of incubation. Among them, 8 hatched chicks were hTPO positive. When the Go transgenic chicken was fed doxycycline (0.5 mg per 1 gram of feed), a tetracycline derivative, hTPO concentration of the transgenic chicken blood was 200 ng/mL. Germline transmission of the transgene was confirmed in sperm of the Go transgenic roosters. These results are informative to establish transgenic chickens as bioreactors for the mass production of commercially valuable and biological active human cytokine proteins.

Studies on the Duration of Immunity and Production of Antibody following Immunization with Inactivated Killed Japanese Encephalitis Vaccine (일본뇌염 백신 접종후 항 일본뇌염 항체의 생성율과 지속적인 면역반응에 대한 연구)

  • Purpose : Studies on the duration of immune response against Japanese encephalitis virus from recipients with JE vaccine (Nakayama-NIH strain) in Korea. Methods : To determinate the immune response and the duration of antibody against JE vaccine, 213 students were examined since 1994 using hemmaglutination inhibition test and plaque reduction neutralization test (PRNT). Results : 24 months after the first vaccination, haemmaglutination inhibition and neutralizing antibody maintained from the recipients 63.4% (>1:20) and 100% (>1:20), respectively. In April 1996, one dose booster to the same recipients those who were vaccinated in 1994, the GMT antibody for HI and PRNT titer were both increased from 1:11.6 to 1:13.2 and 1:275.7 to 1:348.1, respectively, after 6 months booster (after 30 months from the initial vaccination). This results showed that the antibody from the active immunity could be maintained more than 12 months after the initial vaccination. On the basis of these results, inactivated killed JE vaccine (Nakayama-NIH strain) using for preventing against JE purpose seems to produce antibody enough to protect against JE at present. Conclusions : Along with the results of this study demonstrating duration of antibody, the active immunization could be maintained as long as by initial vaccination of 2 doses, a single dose of booster vaccination made during a period of 1 month to 12 months and the successive booster vaccination by 2 or 3 year intervals. However, the immunization schedule should be concerned with both epidemiology of disease and the immune response of vaccinated individuals.

Clinical manifestation of human bocavirus infection in children (소아 human bocavirus의 임상적 고찰)

  • Purpose : Human bocavirus (HBoV) was recently identified world widely in clinical specimens from infants and children with respiratory tract illness, but the role of HBoV in respiratory tract illnesses is unknown. The aim of this study was to investigate the frequency and the clinical manifestation of HBoV in pediatric patients. Methods : We retrospectively investigated 1,777 throat swab obtained between 2005 and 2006 from pediatric in-patients with acute respiratory tract diseases at the Kwang-ju Christian Hospital. The medical records of patients with positive results were reviewed for demographic and clinical data of HBoV infections. Results : HBoV DNA was found in 84 (4.7%) of the 1,777 hospitalized children and the mean age was 19 months. The most common diagnosis were pneumonia (67.8%), bronchiolitis (35.7%). HBoV infections were found year-round, though most occurred in spring and winter months. Conclusion : HBoV is frequently found in hospitalized infants and children with acute respiratory tract diseases in Korea, but an association of HBoV with a distinct respiratory tract manifestation was not apparent. To clarify the clinical significance of HBoV, further evaluation of various age groups and clinical groups is needed.

On-line Image Guided Radiation Therapy using Cone-Beam CT (CBCT) (콘빔CT (CBCT)를 이용한 온라인 영상유도방사선치료 (On-line Image Guided Radiation Therapy))

  • $\underline{Purpose}$: Using cone beam CT, we can compare the position of the patients at the simulation and the treatment. In on-line image guided radiation therapy, one can utilize this compared data and correct the patient position before treatments. Using cone beam CT, we investigated the errors induced by setting up the patients when use only the markings on the patients' skin. $\underline{Materials\;and\;Methods}$: We obtained the data of three patients that received radiation therapy at the Department of Radiation Oncology in Chung-Ang University during August 2006 and October 2006. Just as normal radiation therapy, patients were aligned on the treatment couch after the simulation and treatment planning. Patients were aligned with lasers according to the marking on the skin that were marked at the simulation time and then cone beam CTs were obtained. Cone beam CTs were fused and compared with simulation CTs and the displacement vectors were calculated. Treatment couches were adjusted according to the displacement vector before treatments. After the treatment, positions were verified with kV X-ray (OBI system). $\underline{Results}$: In the case of head and neck patients, the average sizes of the setup error vectors, given by the cone beam CT, were 0.19 cm for the patient A and 0.18 cm for the patient B. The standard deviations were 0.15 cm and 0.21 cm, each. On the other hand, in the case of the pelvis patient, the average and the standard deviation were 0.37 cm and 0.1 cm. $\underline{Conclusion}$: Through the on-line IGRT using cone beam CT, we could correct the setup errors that could occur in the conventional radiotherapy. The importance of the on-line IGRT should be emphasized in the case of 3D conformal therapy and intensity-modulated radiotherapy, which have complex target shapes and steep dose gradients.

Competition between Transplanted Lowland Rice and Weeds as Affected by Plant Spacing and Rice Cultivar Having Different Eco-geographic Race (수도품종(水稻品種)의 생태형(生態型) 차이(差異)가 잡초(雜草)와의 경쟁력(競爭力)에 미치는 영향(影響))

  • An experiment was conducted to obtain the basic information about competitive ability of rice against weeds as affected by eco-geographic race and plant spacing. Generally, more weeds were grown at the plot of Japonica-type cultivar, (Nakdongbyeo) compared to Tongiltype cultivar (Seogwangbyeo). The degree of weed suppression was highest at the 10 ${\times}$ 10cm plant spacing while no difference was observed between 30 ${\times}$ 15cm and 40 ${\times}$ (10 ${\times}$ 10 cm) plant spacings in both cultivar;. In all treatments except the plot of 10 ${\times}$ 10 cm plant spacing of Seogwangbyeo, singnificant amount of weeds was higher for no weeding plot compared to hand weeding plot. Grain yield loss due to weed competition was the highest at the 40 ${\times}$ (10 ${\times}$ 10cm) plant spacing and followed by 30 ${\times}$ 15cm and 10 ${\times}$ 10cm plant spacing, in order, for Seogwangbyeo. However, for Nakdongbyeo, the highest grain yield loss was obtained from the 30 ${\times}$ 15cm plant spacing and grain yield was not affected at the 10 ${\times}$ 10cm plant spacing. Rice grain yield was highly correlated with weed weight for both cultivars (Seogwangbyeo; Y = 5.44 - 0.0128 X, r = -0.929$^{**}$, Nakdongbyeo; Y = 4.96 - 0.0103 X, r = -0.934$^{**}$). To reduce grain yield by 50%, weed weight of 212.5g/sq.m for Seogwangbyeo and 241g/sq.m for Nakdongbyeo was needed, respectively. This result implied that Nakdongbyeo was more competitive against weeds compared to Seogwangbyeo.

  • The experiment was studied to know the competitive ability of rice against weeds as affected by rice maturity and plant spacing. For both cultivars, Tongil (12 days later) and Yeongnamjosaeng, the least amount of weed was obtained from the 10 ${\times}$ 10cm plant spacing while no difference between 40 ${\times}$ (10 ${\times}$ 10cm) and 30 ${\times}$ 15cm plant spacings was observed. When weed was nor pulled out by hand, significant yield loss was resulted in 30 ${\times}$ 15cm and 40 ${\times}$ (10 ${\times}$ 10cm) plant spacings while no difference between weeding regime was observed at the 10 ${\times}$ 10cm plant spacing in both cultivars. Rice grain yield was unaffected by plant spacing for hand weeded plot in both rice cultivars. For noweeding plot, however, the highest grain yield was obtained from 10 ${\times}$10cm plant spacing and also this yield was not significantly different from the yield of hand weeded plot while significant yield loss was recognized at the 30 ${\times}$ 15cm and 40 ${\times}$ (10 ${\times}$ 10cm) plant spacings in noweeding plot. There was also a good relationship between weed weight and grain yield for both cultivars (Tongil; Y = 5.68 - 0.0127 X, r = -0.814$^*$; Yeongnamjosaengv Y = 5.04 - 0.0314 X, r =-0.9704$^{**}$). Based on these relationship, weed weight of 188.2g/sq.m for Yeongnamjosaeng and 223.6g/sq.m for Tongil was needed to reduce rice yield by 50%, respectively. This result implied that Tongil compete more efficiently against weed than Yeongnamjosaeng and hence, could be concluded that late maturing cultivar was more competitive against weed compared to early maturing cultivar.
