A taxonomic study was carried out on the Perinereis nuntia species group of Korea by using morphological and molecular data (mitochondrial cytochrome c oxidase subunit I: mtCOI). Two species, P. mictodonta (Marenzeller, 1879) and P. wilsoni (Glasby and Hsieh, 2006), are recognized and redescribed. In this study, mtCOI gene showed a good resolution as molecular marker for species identification of the P. nuntia species group of Korea.
We describe three feather mites recovered from a wood sandpiper Tringa glareola that was stored in a $-20^{\circ}C$ freezer at the Chungnam Wild Animal Rescue Center. These feather mites are reported for the first time in South Korea: Avenzoaria totani (Canestrini, 1978), Ingrassia veligera Oudemans, 1904 and Montchadskiana glareolae Dabert and Ehrnsberger, 1999. In this study, we provide morphological diagnoses and illustrations. Additionally, we provide partial sequences of the mitochondrial cytochrome c oxidase subunit I(COI) gene as molecular characteristics of three species.

Interspecific hybrids between tiger grouper Epinephelus fuscoguttatus and giant grouper E. lanceolatus were genetically identified based on the partial sequence analysis of mitochondrial cytochrome c oxidase I (COX I) gene and nuclear recombination activating gene 2 (RAG 2) gene. Out of 585 base positions of RAG 2, a total of five nucleotide substitutions were detected between the two parental species (E. fuscoguttatus and E. lanceolatus). The hybrids had two distinct types of RAG 2 sequences corresponding to those of both parental species. Mitochondrial COX I gene sequencing showed that hybrids had sequences identical to E. fuscoguttatus. Molecular data clearly demonstrate that hybridization does occur between E. fuscoguttatus and E. lanceolatus, but with E. fuscoguttatus as the maternal parent.
A natural hybrid between the striped spine loach Cobitis tetralineata and the king spine loach Iksookimia longicorpa was genetically identified by sequence analyses of nuclear recombination activating gene 1 (RAG1) and mitochondrial cytochrome c oxidase I (COI) genes. Out of 850 base positions of RAG1, a total of 23 nucleotide substitutions were detected between the two parental species, whereas the electropherogram of the natural hybrid displayed double peaks at all of the 23 positions, which reflects their simple Mendelian inheritance pattern. Meanwhile, comparison of partial sequences of mitochondrial genes (COI in this study), which are well characterized by the maternal inheritance pattern, revealed that the maternal species of the hybrid was C. tetralineata because of their 100% sequence identity.
One of the mitochondrial genes, called cytochrome c oxidase I (COI), has been widely used for the species identification (called bio-barcode) in birds. In this study, the bio-barcode has been applied to chicken breeds in Korea whether it also can be used as a molecular marker for breed identification. Data indicated that Korean native chicken has the mixed SNP (single nucleotide polymorphism) patterns between White Leghorn (Layer) and Cornish (Broiler) and ultimately, it can not be used as the marker for breed identification. However, this result indicates the mixed use of the Korean native chicken, since it has been used for dual purpose for producing meat and egg for a long time. In order to use as a marker for species identification, more reliable mitochondrial and/or nuclear DNA markers need to be developed.
Euryalid specimens were collected from Gonghyeonjin and Daejin, Gangwon-do in the East Sea, Korea at a depth of 250-300 m by fishing nets on November 2013 and August 2014. They were identified as Gorgonocephalus arcticus Leach, 1819 belonging to family Gorgonocephalidae of order Euryalida, which was new to the Korean fauna. Nucleotide sequences of partial mitochondrial cytochrome c oxidase I (mt-COI) gene, which was 569 bp in length, were compared among four Gorgonocephalus species, and were subsequently employed to reconstruct phylogenetic trees using the MP, ML, and BI methods. As a result, no sequence difference was found between the G. arcticus mt-COI gene sequences from Korea and Canada, and the two made a strong monophyletic group. With the newly recorded G. arcticus in Korea, in total, four Gorgonocephalus species have been reported in Korea.
Barbel steed (Hemibarbus labeo) is a small freshwater fish species as semi-bottom dwellers distributed in eastern Asia. We carried out characterization of the cytochrome c oxidase subunit I (COI) gene from the mitochondrial DNA of H. labeo in the Sumjin River to identify the phylogenetic location of H. labeo in the genus Hemibarbus and Cyprinidae. Multiple alignment of the 577 bp COI sequence revealed high sequence homology (99~100%) between Seomjin River H. labeo. The nucleotide sequence similarity between H. labeo (HD1) and H. mylodon was 88.91% and that of H. longirostis was 88.81% among the three species found in Korea. In addition, the nucleotide sequence similarities of H. maculatus, H. meditus, H. umbrifer and H. barbus showed 98.97%, 97.20%, 96.87% and 98.85%, respectively. Phylogenetic analysis on seven species of the genus Hemibarbus showed that the H. labeo collected in this study formed two clades. One of which consisted of Hadong, Imsil, Kangjin. The other one formed a step with HD2, HD8 and HD9 of Hadong and the H. labeo reported in Busan, Asan and Seoul, Korea. Phylogenetic position of the H. labeo among Cyprinidae showed 0.143 for the evolutionary distance from Zacco platypus and 0.006 for the H. maculatus. In addition, the genetic position of the H. labeo among 28 species of Cyprinidae was found to be located in Group I, including Gobioninae fishes. The results of this study will provide key genetic information for the taxonomic comparison in Cyprinidae and study of model fish for pollution monitoring in freshwater environments.
Grouper is one of the most economically important fishes with various morphological forms and characteristics, meaning it is often difficult to identify species and distinguish between life stages, sometimes leading to morphological misidentification. Therefore, identification using a molecular deoxyribose nucleic acid (DNA) approach was needed as an alternative means to identify closely related species. This study aims to determine the molecular phylogeny of grouper from the northern part of the Bird's Head Seascape of Papua. The DNA sequence of each cytochrome oxidase I (COI) gene was used to study the molecular relationship among closely related species of grouper. The results showed that there were 16 Epinephelinae that have been compared to a gene bank (National Centre for Biotechnology Information, NCBI) in the sequence length of 623 base pairs. The closest genetic distance was found between Cephalopholis miniata and Cephalopholis sexmaculata (0.036), while the furthest genetic distance was observed between Plectropomus laevis and Cephalopholis spiloparaea (0.247). This finding was further reinforced by the morphological characters of each species. This finding highlighted that five genera were represented as a monophyletic group (clade), i.e., Epinephelus, Cephalopholis, Plectropomus, Saloptia and Variola.
Asteroid specimens of the genus Luidia were collected at a depth of 95-100 m in the Korea Strait by bottom trawling in April 2016. The specimens were identified as Luidia avicularia Fisher, 1913 (Luidiidae: Paxillosida) based on morphological characteristics and molecular phylogenetic analyses, and the species is new to the Korean fauna. A 648-bp partial nucleotide sequence of mitochondrial cytochrome c oxidase I (mt-COI) gene was obtained from Korea, and then was compared to sequences of related species stored in GenBank using molecular phylogenetic analyses. No sequence differences were detected between the L. avicularia mt-COI gene sequences from Korea and China, and the species described in this report was clearly distinct from L. maculata, which was previously reported in Korean fauna. Three Luidia species have been reported in Korea.
Thamnaconus modestus, distributed in the Northwest Pacific, has high economic value and is used in various seafood. In this study, the morphological and genetic characteristics of T. modestus and T. septentrionalis were compared and analyzed. We observed the external and internal morphology of T. modestus, sketched skeletal elements, and analyzed phylogenetic evolutionary relationships using the cytochrome c oxidase subunit I (COI) gene on mitochondrial DNA compared to T. septentrionalis. The T. modestus observed in this study had blackish-brown patterns irregularly scattered on the gray-brown body, and the fins were blue-green. Genetic analysis results based on the COI sequences of T. modestus showed seven types of base sequence variation; however, the homology was more than 98.8%. In addition, as a result of comparison of the COI nucleotide sequences and phylogenetic analysis in Tetraodontiformes, two T. septentrionalis sequences (JN813099, MW485059) were similar to T. modestus with 99% homology, and the other two T. septentrionalis sequences (EF607583, KP267619) were similar to those of species belonging to another genus Thamnaconus with 95% homology with T. modestus. It was not easy to classify the species based on morphological characteristics, and phylogenetic analysis between T. modestus and T. septentrionalis confirmed the difference in classification. These results provide the external and internal morphology of T. modestus and will be used as important information for the taxonomic study of T. modestus and T. septentrionalis.
