Partial nucleotide sequences of nuclear protein-coding recombination activating gene 1 (RAG1) gene of two Abbottina and five Microphysogobio species residing in Korea were analyzed to elucidate the molecular phylogenetic position of A. springeri Banarescu and Nalbant. In RAG1 tree A. rivularis was clearly separated from the monophyletic lineage composed of A. springeri, Biwia zezera and Microphysogobio species. Within this lineage B. zezera showed sister-group relationship to the monophyletic group composed of A. springeri and five Microphysogobio species. Thus, our phylogenetic tree revealed the polyphyletic nature of two Abbottina species from Korea, which result is well congruent with the previous phyletic assumption based on osteological features. The current classification of Abbottina and Microphysogobio based on morphological criteria, such as the presence or absence of papillae on lips and size of swim bladder with or without encapsulation, does not reflect their true evolutionary history.
The family Cobitidae of Korea was reviewed as 16 species in 5 genera with the key to species for the identification by the external features. The three genera of Iksookimia, Koreocobitis, and Kichulchoia erected from the genus Cobitis were reclassified based on the presence or absence of Gambetta’s zones on the body sides, their body pigmentation, the shape of lamina circularis in males, the number of unbranched anal fin rays, and some molecular phylogenetic data. By the results of these reexamination, the generic names of the three species were changed as Cobitis choii, Iksookimia pacifica, and Kichulchoia mutifasciata. It was remarked biogeographically that most species of genera Cobitis, Iksookimia, Koreocobitis, and Kichulchoia show the disjunct distributions with bottom-dwelling preferences as endemic to Korean peninsular. Three species of Cobitis choii, Kichulchoia brevifasciata, and Koreocobitis naktongensis are considered to be critically endangered according to the severe habitat destruction and the alteration of river topography.
Representative cDNA libraries were constructed from various tissue sources of Hemibarbus mylodon, an endangered freshwater fish species in Korea, for the mining of expressed sequence tags (ESTs). Randomized and non-normalized EST analysis was performed with 7 unidirectional cDNA libraries generated from brain, intestine, kidney, liver, muscle, ovary or testis. Of 3,383 ESTs in total, the number of singleton was 2,029, and 333 contigs containing 1,354 ESTs were assembled (percent of unigene = 70.0%). Abundantly expressed gene transcripts and broad clustering of putative gene function were tissue-specific in general, and the redundancy was also variable among those libraries. Over half of H. mylodon ESTs were matched with orthologues from other teleosts among which zebrafish gene sequences were the most frequent in those matches. This initial setting of EST libraries achieved in the present study would be a fundamental basis for the banking of gene resources from this endangered fish species.

Genomic organization, including the structural characteristics of 5'-flanking regions of two 65-kDa protein (WAP65) isoform genes associated with warm temperature acclimation, were characterized and their transcriptional responses to immune challenges were examined in the intestine, kidney and spleen of the mud loach (Misgurnus mizolepis; Cypriniformes). Both mud loach Wap65 isoform genes displayed a 10-exon structure that is common to most teleostean Wap65 genes. The two mud loach Wap65 isoforms were predicted to possess various stress- and immune-related transcription factor binding sites in their regulatory regions; however, the predicted motif profiles differed between the two isoforms, and the inflammation-related transcription factor binding motifs, such as NF-${\kappa}B$ and CREBP sites, were more highlighted in the Wap65-2 isoform than the Wap65-1 isoform. The results of qRT-PCR indicated that experimental immune challenges using Edwardsiella tarda, lipopolysaccharide or polyI:C induced the Wap65-2 isoform more than Wap65-1 isoform, although modulation patterns in response to these challenges were tissue- and stimulant-dependent. This study confirms that functional diversification between the two mud loach Wap65 isoforms (i.e., closer involvement of Wap65-2 in the acute phase of inflammation and innate immunity) occurs at the mRNA level in multiple tissues, and suggests that such differential modulation patterns between the two isoforms are related to the different transcription factor binding profiles in their regulatory regions.

Lipoproteins are complexes of lipids and specific apolipoproteins that are involved in lipid transport and redistribution among various tissues. In this study, we isolated full-length apolipoprotein cDNA sequences encoding apolipoprotein A-I (apoA-I), apoE, apoC-II, and apo-14 kDa in barred knifejaw, Oplegnathus fasciatus. In addition, we reconstructed phylogenetic trees and investigated mRNA tissue distributions. Alignment analyses of amino acid sequences revealed that secondary structures of the polypeptides apoA-I, apoE, and apoC-II in barred knifejaw are well conserved with their teleostean and mammalian counterparts in terms of characteristic tandem repetitive units forming amphipathic ${\alpha}$-helices. Both the sequence alignment data and cleavage sites of apo-14 kDa indicated a clear differentiation between Percomorpha and Cypriniformes. Meanwhile, the phylogenetic trees of apolipoprotein sub-families suggested that the common ancestor prior to the split of the Actinopterygii (ray-finned fishes) and Sarcopterygii (tetrapods) would have possessed the primordial protein-encoding genes. Tissue distribution of each apolipoprotein transcript determined by semi-quantitative RTPCR showed that barred knifejaw apoA-I transcripts were more or less ubiquitously expressed in the liver, intestines, brain, muscle, spleen, and kidney. The most striking difference from previous observations on barred knifejaw was the ubiquitous expression of apoE across all somatic tissues. Barred knifejaw apoC-II showed tissue-specific expression in the liver and intestines, while the liver and brain were the major sites of apo-14kDa mRNA synthesis.
The ultrastructure of the spermatozoa of the family Cobitidae, 6 genera and 12 species, was examinated under electron microscopes. Spermatozoa of the observed species consist of a head(nucleus), a short midpiece, and a single tai1(flagellum). It is of anacrosomal aquasperm type, lacking an acrosome. However, the spermatozoa of Nemacheilus toni has vestigal acrosome or acrosome-like vesicle in the anterior region of the nucleus during spermiogenesis, The nucleus of Cobitidae is approximately spherical except that N. toni is conic. The mid piece was under $3.0{\mu}m$ in length and contained 5-8 ring-shaped mitochodria. Genera Cobitis, Iksookimia, Niwaella, and Nemacheilus. have shorter midpiece, whereas Misgurnus and Lefua have longer midpiece. The flagellum was uniflagellate consisting of a typical 9+2 axoneme without fins.
The Nakdong nose loach, Koreocobitis naktongensis is an endangered freshwater fish endemic to Korea. The spawning behaviour of the K. naktongensis was observed after treating Ovaprim in laboratory aquarium from 20~27 April 2010. The behavioral patterns were categorized into three stages of pre-spawning, spawning and post-spawning behaviors. Before spawning K. naktongensis usually repeated taking a rest and swimming. On average, initial spawning activity started 11 to 12 hours later after injection with Ovaprim. During the spawning period, the number of spawning acts ranged from 1 to 18. The spawning ratio of female to male was 1 : 1. The male embraced the back of the female's dorsal fin with his lamina circularis. It looked like a complete circle at that time. After spawning, parental care was not observed. Individuals not participating in spawning have been observed to feed on some of the fertilized eggs.
The olfactory organ of a Korean endemic species, Pseudobagrus koreanus, was investigated to describe its morphology and histology and analyze correlation between habitat environment and its ecological habits and characters, using a stereo microscope, a light microscope, and a scanning electron microscope. Its external morphology showed tubular anterior nostril at the tip of the snout and slit posterior nostril at the base of the nasal barbel. The olfactory chamber showed the rosette structure consisting of numerous lamellae radial and parallel to the medium raphe. The olfactory lamella has the sensory epithelium with olfactory receptor nuerons, supporting cells, basal cells, lymphatic cells, plasma cells and the non-sensory epithelium with stratified epithelial cells, mucous cells, lymphatic cells. In particular, 1) 41~43 (n=20) lamellae, 2) lymphatic cells and plasma cells, 3) mucous cells including neutral polysaccharid may be olfactory characteristics relevant to a stagnant or a standing habitat environment, nocturnal habit dependent on olfaction, and taxonomic characters compared at least to other catfish species.

This study examines the osteological development of the Pike gudgeon, Pseudogobio esocinus in the cranial, vertebral, caudal, pelvic and pectoral girdle for the purpose of taxonomic and aquacultural studies. Adult Pike gudgeon collected from the Boseonggang River were reared at 18.5~19.5℃ (average 19.0℃). At 5 days after hatching (total length (TL) 4.25 mm), the parasphenoid, dentary, centrum of vertebrae and clavicle began to ossify. At 22 days after hatching (TL 6.33 mm), 36 to 38 vertebrae had ossified. At 42 days after hatching (TL 16.71 mm), the majority of the skeleton had ossified. Pike gudgeon exhibited comparable characteristics to other fishes of Gobionidae throughout its development, although it displayed differences in some areas, such as the caudal skeleton and fin supports.
The egg development, early life history and spawning site characters of Korean endemic fish, Coreoleuciscus splendidus (Gobioninae), were investigated at the part of Jo-jong stream in Korea from May 2020. The fertilized eggs were 2.05~2.23 mm (mean, 2.13 mm) in diameter and had no oil globules. The embryo began to hatching about 98 hrs after fertilization under water temperature of 20±1℃. The newly-hatched larvae were 5.03~5.68 mm (mean, 5.31 mm) in total length (TL), and their mouth and anus were not opened. 4 days after hatching, Several rod-like cupulae were observed on the head and lateral side of the body at 6.95~7.89 mm (mean, 7.51 mm). 7 days after hatching, the post-larva stage were 8.39~9.29 mm (mean, 8.78 mm) in total length, and their york were completely absorbed. The cupulae were completely distinguished. They entered the juvenile stage when all fin-rays were formed at 29 days after hatching, and their TL were 14.16~17.04 mm (mean, 14.99 mm). Squamation was initiated on the caudal body at approximately 18.21~23.74 mm (mean, 30.28 mm), 38 days after hatching, and completed at 26.82~33.33 mm (mean, 19.42 mm), 73 days after hatching, the external characteristics from of juveniles were same to adults. The spawning site was characterized by bottom structure of pebble (64~16 mm) and gravel (16~2 mm), environmental conditions of the spawning sites were 6~18 cm in water depth, 0.43~0.73 m/sec in bottom water velocity. In spawning sites, an egg mass or separated eggs were located a small gap of under the pebble and gravel.
