Renal effect of experimental feeding of melamine and cyanuric acid in different concentrations on Japanese catfish (Silurus asotus) (멜라민과 cyanuric acid의 농도별 혼합투여에 따른 메기(Silurus asotus) 신장에서의 조직병리학적 소견)

  • The recent outbreak of renal failure in infants in China and in animals in USA and Europe has been determined to be caused by melamine adulterated in the food. In the course of the investigation, cyanuric acid was identified in addition to melamine in the offending food. Fish feeds were also recently found to be contaminated with melamine. The purpose of this study was to characterize the histopathological effect and toxicity potential of different concentrations of melamine and cyanuric acid in the kidney of Japanese catfish (Silurus asotus). The fish were administered melamine and cyanuric acid in combination at the concentrations of 12.5, 25, 50, 100 and 200 mg/kg/day for 3 days by single oral administration dissolved in carboxymethyl cellulose. The results showed that renal crystals were observed in renal tubules and collecting ducts at the concentration over 25 mg/kg dose group and the number of crystals in kidney were in proportion to the concentrations of melamine and cyanuric acid.

  • In this study, we investigated the existence of melamine and cyanuric acid in cigarette smoke because leaf tobacco contains lots of nitrogen compounds and they are capable of pyrolytic synthesis while smoking. The extraction and derivatization conditions of two compounds were performed according to the procedure of U.S. Food and Drug Administration(FDA). Ky3R4F as reference cigarette was smoked on ISO condition(Puff volumne : 35 ml, duration : 60 sec, Interval : 2 sec). Compared with the results Ky3R4F and standard solution, retention time of two compounds was not corresponded. As a result, we concluded that melamine and cyanuric acid were not existed in cigarette smoke.

  • Simultaneous analytical method has developed for the determination of anion biocides in soil by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Chlorite and chlorate in soil were extracted with pure water, and cyanuric acid and sodium dodecylbenzenesulfonate (Na-DBS) were extracted with mobile phase (0.25 mM ammonium formate in 20 mM formic acid : acetonitrile (1:1)). The extract was injected into the LC-MS/MS system after filtration. The method detection limits in this study were 0.04 mg/kg for chlorite, 0.04 mg/kg for chlorate, 0.27 mg/kg for cyanuric acid, and 0.05 mg/kg for Na-DBS, respectively. The method was applied to the analysis of 50 soil samples collected from 40 sites sprayed with biocides and 10 background sites. As a result, anion biocides were not detected in all sites.

  • Melamine is an organic base and a trimer of cyanamide which contains 66% nitrogen by mass, but it was sometimes illegally added to food products in order to enhance the apparent protein content for the bigger profit. Many Chinese infants and pet animals fed melamine-containing formulas suffered acute kidney failure with renal crystal formation. Fish feeds were also recently found to be contaminated with melamine. This experiment has been designed to determine whether renal crystals can be experimentally induced in crucian carp (Carassius carassius) fed melamine and cyanuric acid (each at 400 mg/kg/day for 3 days) in combination, and to compare experimentally induced crystals with those from pet animals with triazine related renal failure. The results showed that all fish developed gold brown renal crystals arranged in radial spherulites in the renal tubules, similar to those detected in the pet animals.

  • Melamine has raised international concerns for its catastrophic health effects from tainted infant formula. This report concerns the developmental validation of a sensitive HPLC/MS/MS and GC/MS methods about melamine and cyanuric acid in human urine and serum. Analytical detection ranges of LC/MS was from 0.2 to 5.0 ng/mL and 2.0 to 60.0 ng/mL about melamine and cyanuric acid, respectively. The limits of quantification and confirmation are 0.2 ng/mL for both analytes in human urine and serum by LC/MS/MS. The range of recovery was 91.6%, and 107.6% for cyanuric acid and melamine in urine, respectively. The range of precision coefficient variation was from 2.0%, to 11.8% for cyanuric acid and melamine in urine. The range of recovery was from 94.9%, to 119.0% about cyanuric acid and melamine in serum, respectively. The range of precision coefficient variation from was 3.7%, and 13.5% about cyanuric acid and melamine in serum. Analytical detection ranges of GC/MS were 5.0 to 100.0 ng/mL about melamine and cyanuric acid, respectively. The limits of quantification and confirmation are 5.0 ng/mL for both analytes in human urine and serum by GC/MS. The range of recovery was from 83.7%, to 114.5% for cyanuric acid and melamine in urine, respectively. The range of precision coefficient variation was 3.5%, and 10.7% for cyanuric acid and melamine in urine. The range of recovery was 94.4%, and 110.7% for cyanuric acid and melamine in serum, respectively. The range of precision coefficient variation from was 3.9%, and 13.8% for cyanuric acid and melamine in serum. Several changes were taken to optimize performance by this method.

  • Melamincyanurate(MC), as an non halogen flame retardant are used as the polymer and plastic materials. In this study, melamine and cyanuric acid were used for the synthesis of MC. The optimum condition of synthetic MC were controlled by different molar ratio of melamine to cyanuric acid. MC was modified by coupling reaction with four different agents. The influences of modified MC were based on the coupling agent types. Preparation methods are available to offer the prospect of improved morphology control deposit stability in polyol. The results reveal that glycidoxypropyltrimethoxysilane(GDS) has the best storage stability. The best properties were obtained with melamine and cyanuric acid from 1:1 molar ratio. Modification of MC through coupling agent can efficiently enhanced the deposit stability in polyol up to 30 %.

  • From Oct. 2008 to Oct. 2009, 619 livestock products sold in Incheon were examined for melamine contamination. HPLC was used to detect the melamine concentration from various products. $C_{18}$ column $(3.9\times150mm,\;4{\mu}m)$ was applied with a phase composed of 10mM citric acid and 10mM sodium octane sulfonate : acetonitrile (in ratio 90:10) pumped isocratically at 1.0ml/min. Melamine was not detected from any of the products at the level of LOD 0.03mg/kg and LOQ 0.08mg/kg, suggesting that no melamine contamination was ascertained in livestock products in Incheon area. However, further tests should be done to detect other melamine analogues for the evaluation of toxicity and safety of melamine and cyanuric acid in the future.

Evaluation of Renal Toxicity by Combination Exposure to Melamine and Cyanuric Acid in Male Sprague-Dawley Rats

  • Melamine-induced nephrotoxicity is closely associated with crystal formation in the kidney caused by combined exposure to melamine (Mel) and cyanuric acid (CA). However, there are few dosage-finding studies for toxicological evaluation of chronic co-exposure to Mel and CA. The objective of this study was to investigate the possible mechanism by which a Mel and CA mixture lead to renal toxicity in rats. Mel and CA were co-administered to rats via oral gavage for 50 days. Nephrotoxicity was determined by measuring blood urea nitrogen (BUN) and serum creatinine (sCr) levels. Relative kidney weights were significantly increased in rats after co-exposure to Mel+CA (63/6.3 or 630/6.3 mg/kg) mixtures. BUN and sCr levels were significantly increased after Mel and CA co-exposure. Taken together, significant increase in KIM-1, NGAL, and calbindin levels were observed in the urine of rats exposed to Mel+CA (63/6.3 or 630/6.3 mg/kg) compared with the corresponding control group. Histological analysis revealed epithelial degeneration and necrotic cell death in the proximal tubules of the kidney after co-exposure to Mel+CA (63/6.3 or 630/6.3 mg/kg). Our data suggest that Mel-mediated renal toxicity may be influenced by CA concentrations in Mel-contaminated milk or foods.

High Throughput Screening for Searching a New Inhibitors of Acetolactate Synthase (Acetolactate synthase에 대한 고효율 활성 측정방법 및 신규 저해제 탐색)

  • This study was conducted to develop a high throughput system for screening acetolactate synthase(ALS) inhibitors, and to detect basic mother molecules for developing new novel herbicide candidates. The high throughput screening (HTS) method using 96-well plate and microplate reader was developed. This method is 8 times more effective than basic technique in one cycle per person. Futhermore, considering for less than 1/10 volume of materials required for ALS test and enzyme kinetics with 16 times faster speed compared to those of former procedure, this HTS method has more than 100 times higher efficacy than basic system in a consecutive procedure. We discovered 11 new ALS inhibitors such as 2-oxoglutaric acid, aminooxyacetic acid, azelaic acid, citric acid, cyanuric fluoride, itaconic acid, malonic acid, niclosamide, oxalic acid, glyoxylic acid, and suramin from 107 commercial plant-specific inhibitors using this technique. We hope these results might be useful to discover lead compounds for developing new novel herbicide candidate.

Preparation of $\beta$-Cyclodextrinized Cellulosic Fiber and Deodouring Property ($\beta$-시클로덱스트린화 셀룰로오스 섬유의 제조 및 소취성)

  • $\beta$-Cyclodextrine/benzoic acid complex was prepared and reacted with cyanuric chloride (2,4,6-trichloro-1,3,5-triazine). Identification of complex formation and reaction was checked by FT-IR, UV-Vis, and EDX. By reacting this material with cotton fiber, the deodourant fiber was prepared. The deodourizing property was evaluated by the concentration changes of aqueous ammonia solution after flowing ammonia gas through the column titled with deodourant fiber prepared. The deodourizing property was increased with an increase of concentration of $\beta$-cyclodextrine unit in the fiber. In the case of $\beta$-cyclodextrine/benzoic acid complex, the deodourzing property was much increased, comparing with the $\beta$-cyclodextrine only. It was considered to be the binding of aamonia gas caused by benzoic acid in the complex.
