Effect of hCG on Connexin 43 mRNA Expression in Goldfish Ovary

  • This study examined whether the connexin (Cx) is an essential protein during oocyte maturation in the ovary of the goldfish (Carassius auratus). In mature female goldfish ovaries, at late vitellogenic stage, human insulin-like growth factor-I (IGF-I; 20 M) and human chorionic gonadotropin(hCG; 20 IU/㎖) were injected. Twelve hr after the injection, mature female goldfish ovaries were removed and stored at -80C until analysis by RT-PCR. From the goldfish Cx43 cDNA sequence (GenBank accession number AB078505), two degenerate primers were designated. In vivo, 12 hr after the treatment with hCG, goldfish Cx43 mRNA expression level was increased, while the levels of IGF-I was not changed. Goldfish Cx43 mRNA expressed after, but not before the hCG treatment. These results suggest that Cx43 mRNA was judged to be a gene, which was transcribed during oocyte maturation induced by hCG.

  • To investigate the most effective way of cellulase utilization on paper making process and the systematic way of how to combine its components for better use, purified two major cellulase were applied to bleached kraft pulp at various dosage in the refining process and estimated the synergism effect of each components in the characteristics of papermaking fiber. freeness was not changed by $C_1$ Component dosage but strongly increased by Cx Component dosage. Drop in fiber viscosity, increase in WQV, fiber shortening and fine generation was strongly influenced by Cx Component, it seems to be that Cx Component works a hydrogenate that breaks hydrogen bonding in fiber and enhances pooling effect of fiber. $C_1$ Component was little influenced these characteristics of fiber but considered to help to make fiber surface smooth.

  • Objectives : This study was designed to find The characteristics of the Moire topography in patients of HIVD of Cx-spine. Methods : The Moire topography using IBS(Intek-plus Body Scanner 2000, Seoul, Korea) were obtained in 688 people of control group and 105 patients of HIVD of Cx-spine. Each values were studied to determine the difference between controls and patients. Results : There were significantly difference between controls and patients in thoracic ratio, lumbar ratio, top of the thoracic depth ratio, top of the lumbar depth ratio, Z axis distance and DZ/DY. Conclusions : There were increase of the thoracic kyphosis and lower position in top of the thoracic kyphosis in patients of HIVD of Cx-spine than controls.

Development of Neuropathic Pain Behavior and Expression of CCL2/CCR2 and CX3CL1/CX3CR1 after Spinal Cord Hemisection

  • Purpose: The purpose of this study was to evaluate the development of pain behavior and the expression of CCL2/CCR2 and CX3CL1/CX3CR1 at above and below the level of hemisection of the spinal cord in a rat model. Methods: Spinal cords of adult female Sprague-Dawley rats (n= 16, 200~250 g, 6~8 weeks old) were hemisected at T13 on the right side to develop the spinal hemisection injury model. We compared behavioral responses of the hemisection and of a sham surgery group. Behavioral tests for motor function (by the BBB locomotor scale), and for pain response for mechanical and cold allodynia were assessed postoperatively (PO) for 21 days. Expression of mRNA for chemokines and their receptors (CCL2/CCR2 and CX3CL1/CX3CR1) below and above the level of the spinal cord dissection were examined by RT-PCR. Results: We observed gradual motor improvement and the development of mechanical and cold allodynia on the ipsilateral hindpaw after spinal hemisection injury. We also found upregulation of mRNA expression of CCL2/CCR2 both above and below the level of spinal cord dissection but CX3CL1/CX3CR1 mRNA expression. Conclusion: Upregulation of CCL2/CCR2 is associated with neuropathic pain after spinal hemisection injury. CCL2/CCR2 may play an important role in the development of neuropathic pain after SCI as well as of peripheral neuropathic pain. These findings may improve understanding of the pathophysiological mechanism of neuropathic pain after SCI.

Development of the Imaging Optical System for the 545 nm Fluorescent Plate of X-ray (X선용 545 nm 형광판 결상광학계 개발)

  • To develop an imaging optical system for the 545 nm fluorescent plate of X-ray. Methods: We designed and manufactured a new imaging optical system for the 545 nm fluorescent plate of X-ray by Sigma 2000 program after deciding the design comparison standards referred to Canon CX2-70 model. Results: The characteristics of the new imaging optical system for the 545 nm fluorescent plate of X-ray have the magnification of -0.225x, the image field size of $90mm{\times}90mm$, and the 0.033 mm resolution line width at the 30% MTF value criterion. These mean that the new model has a capability of deciphering for the more large screen and the resolution of deciphering is superior to that of Canon CX2-70 model. Also the image side NA (-0.196) of the new model is about $\sqrt{2}$ times than that (-0.139) of CX2-70 and the object side NA (0.044) of the new model is about 2 times than that (0.022) of CX2-70. These mean that the sensitivity of the film in the new design model is able to be increased to about 4 times and there is the possibility of reducing the bombed time of X-ray to 1/4 times. Conclusions: We could design and manufacture the imaging optical system for the 545 nm fluorescent plate of X-ray having the possibility of reducing the bombed time of X-ray to 1/4 times in comparision to Canon CX2-70 model, the characteristics of which have the image field size of $90mm{\times}90mm$ and the MTF of 30% or more at 15 lines/mm criterion.

Formation, Properties and Antimicrobial Activities of Cotton Xanthate-Cu(II)-Homosulfamine Complex

  • To develop a cotton derivatives with prolonged antimicrobial activities, homosulfamine (Hs) was coupled to cotton xanthate (CX) via chelate bond in the presence of Cu(II) ion by one- and two-bath processes. In one-bath process, CX was treated with Cu(II)-Hs solution. In two-bath process, CX was treated with Cu(II) ion solution to produce CX-Cu(II) complex, which was isolated and treated in turn with Hs solution. Effects of concentration, Cu(II)/Hs ratio, and pH on the binding of Hs were investigated at $10{\circ}C$ . In one-bath process, binding of hs took place readily with optimum pH around 5-6. the amount of binding increased to give a maximum within 5 min and decreased slowly to establish an equilibrium withhin an hour. In two-bath process, binding of Hs was much lower than that of one-bath process. Release of Hs from CX-Cu(II)-Hs was investigated by batch and flow method. antimicrobial activiteis of CX-Cu(II)-Hs was investigated by batch and flow method. Antimicrobial activities of CX-Cu(II)-Hs were tested against Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa and Escherichia coli and it showed prolonged activity compared to that of free Hs.

Aberrant Expression of Connexin Isoforms in the Corpus Epididymis of the Adult Rat by Exposure to Estradiol Benzoate or Flutamide at the Weaning Age

  • A proper development of the epididymis during the early postnatal development is required for successful fertility in the adult male. Direct cell-cell communication via connexin (Cx) molecules is a common way of cellular interactions to achieve normal development of a given tissue consisting of different cell types. The present research was attempted to determine the effect of exogenous exposure to estrogenic agonist or antiandrogen at the weaning age on expression of Cx isoforms in the adult corpus epididymis. Male rats were subcutaneously administrated with estradiol benzoate (EB) or flutamide (Flu) at the weaning age. The tissue was collected at 4 months of age. Expressional levels of Cx isoforms were determined by a quantitative real-time PCR. Statistical comparison showed significant increases of Cxs31, 32, 37, 40, and 43 transcript amounts by a treatment of $0.015{\mu}g$ of EB /kg body weight (BW). A treatment of $1.5{\mu}g$ of EB /kg BW caused a significant decrease of Cx43 gene expression but increases of Cxs26, 31, 32, 37, and 40 transcript levels. Exposure to $500{\mu}g$ of Flu/kg BW induced an increase of Cx37 expression but significant decreases of Cxs43 and 45 mRNA levels. Expression of Cx37 was increased by a treatment of 5 mg of Flu/kg BW, while transcript levels of Cxs26, 30.3, 31, 31.1, 32, and 43 were significantly decreased by same treatment. These results demonstrate that exposure to steroidal compounds at the early developmental age alters expression of Cx isoforms in the adult corpus epididymis.

Clinical and radiological characteristics of odontomas: A retrospective study of 90 cases

  • Purpose: Odontomas represent a common clinical entity among odontogenic tumors, but are not well-addressed in the Vietnamese population. The present study aimed to determine the clinical and preclinical characteristics of odontomas and associated factors in the Vietnamese population. Materials and Methods: This retrospective study retrieved data from histopathological diagnoses from 2 central hospitals of Odonto-Stomatology in Ho Chi Minh City, Vietnam during 2004-2017. The odontomas were classified as complex (CxOD) or compound (CpOD) subtypes. The epidemiological, clinical, and radiological characteristics of the odontomas, stratified by subtype and sex, were obtained and analyzed. Results: Ninety cases, consisting of 46 CxODs and 44 CpODs, were included. The average age of patients was 32.4 (±20.2) years. The patients with CxOD were older than those with CpOD (P<0.05). Clinically, 67% of patients showed an intraoral bone expansion. Approximately 60% of patients with CxOD exhibited a painful symptom, about 3-fold more than those with CpOD (P<0.05), whereas almost all patients with CpOD exhibited perturbations of dentition, unlike those with CxOD (P<0.05). Radiologically, CxOD was characterized by a larger dimension than CpOD in both sexes (P<0.05), and CpOD induced complications in adjacent teeth more often than CxOD (P<0.05). The development of odontoma with advancing age differed significantly in odontoma subtypes related to their pathological origins, and between the sexes, resulting from different physiological states. Conclusion: The findings of this study highlight the value of clinical and radiological features of odontomas and their associated factors for the early diagnosis and adequate treatment of younger patients.

Evidence Suggesting that the Deposition of Pigments into Yolks is Independent of Egg Production: Enhanced Pigmentation of Yolks by Feeding Hens with Canthaxanthin Biosynthesized by Microbials (난황 내 색소의 축적은 산란율과 무관함을 제시하는 증거: 균체가 생성하는 Canthaxanthin의 급여에 의해 강화된 난황의 착색)

  • Pigments in the diet affect yolk colors. Due to variations in both the bioavailability of pigments in chickens and their amounts occurring in the feed ingredients, concern about egg quality arises in terms of yolk color. In this study, the effects of pigments, produced through cell culture in the laboratory, on yolk colors were determined for 4 weeks in laying hens receiving one of the 6 dietary treatments: control diets containing 1) no synthetic pigments (CON); 2) canthaxanthin (4 ppm) purchased from BASF (BASF); 3) cultured cells so that the diet had canthaxanthin at 4 ppm (CX); 4) cultured cells so that the diet had lycopene at 30 ppm (LP); 5) canthaxanthin (4 ppm) that was purified from cultured cells (SPCX); or 6) lycopene (30 ppm) that was purified from cultured cells. Relation between deposition of pigments into yolks and egg production was also tested. Yolk color of eggs from chickens fed dietary CX was significantly enhanced, which was slightly but significantly below that of BASF. Results from other treatments were lower than those of CX. Deposit rates of pigments into yolks were: BASF > CX > SPCX > LP > SPLP. The amounts of pigments, with the exception of SPLP, in feed were not changed during the storage for 4 weeks at $25^{\circ}C$. Egg production rates varied among treatments during the initial phase of the study but became relatively uniform at the later stage, except for CON and LP groups. The results of the present study indicate that the deposition of pigments into yolks is independent of egg production.
