감마선조사 대두의 수분 흡수와 조리특성 및 세포구조 (Water Absorption, Cooking Properties and Cell Structure of Gamma Irradiated Soybeans)

  • 대두의 물리적 성질을 개선할 목적으로 대두에 0, 5, 10, 20 kGy의 감마선을 조사하여 수분흡수양상, 조리특성 및 세포구조를 관찰하였다. 대두의 수분흡수는 수침온도 및 감마선 조사선량의 증가에 따라 평형에 이르는 시간이 감소하였으며, $20^{\circ}C수침온도에서$ 5~20 kGy의 감마선조사 대두는 비 조사 대두에 비 해 3~6시간의 수침시간을 단축할 수 있었다 대두의 조리정도는 대두 자엽의 최대 절단력으로 나타냈으며, 관능적으로 완전히 익은 대두의 절단력을 145g/g으로 했을 때 5~20 kGy의 감마선조사는 대두의 조리시간을 55~75% 단축시켰다. 대두 표피내면의 세포구조 관찰에서 비조사 대두의 유조직은 주름이 잡힌 치밀한 구조를 나타낸 반면, 감마선조사 대두는 유조직의 느슨함과 변형됨을 보여주었다. 자엽표면 및 그 내부구조도 감마선조사에 의해 구조의 변형을 나타내었다. 특히 자엽세포의 protein body는 감마선조사 선량이 증가됨에 따라 형태가 변형되어 20k0y조사 대두는 스파이크형을 나타내었으며, lipid body의 경우도 감마선 조사선량이 증가됨에 따라 그 크기가 현저히 감소하여 작은 구의 형태로 분해되었다.

하절기 '매향' 딸기의 삽목 번식 시 관수방법에 따른 생육 및 발근율 (Growth and Rooting Rate of 'Maehyang' Strawberry as Affected by Irrigation Method on Cutting Propagation in Summer Season)

  • 본 연구는 하절기 '매향' 딸기 자묘의 삽목 번식에 적절한 관수 방법을 구명하기 위해 수행되었다. 딸기 자묘의 삽수는 24구 딸기 전용 포트(24구, $60{\times}34{\times}10cm$)에 상업용 혼합 상토(Tosilee)를 이용하여 삽목하고, 반 밀폐형 소형비닐터널에 위치시켰다. 각각의 반 밀폐형 소형터널의 처리는 다음과 같이 처리했다; 대조구(무처리), 두상관수(하루 두 번), 매트관수(하루 두 번), 그리고 안개관수(8:00시부터 18:00시까지 30분 온/오프 설정). 딸기 자묘는 반 밀폐형 소형터널 내에서 8일 동안 발근을 하였고, 그 이후 플라스틱 필름을 제거 하였다. 삽목 후 61일째에 초장, 근장, 1차 근수, 엽병장, 엽장, 엽폭, 크라운 직경, 엽록소 값, 엽면적, 지상부와 지하부의 생체중 및 건물중을 측정하였다. 터널 내의 하루 평균 상대습도는 각각 안개관수, 두상관수, 매트관수, 대조구 순으로 72.5, 56.3, 45.8, 그리고 29%를 나타냈다. 그러나 공기온도는 모든 처리에서 유사하였다. 딸기 자묘의 4일째와 8일째 발근율은 두상관수와 안개관수 처리에서 유의적으로 높았다. 초장, 엽병장, 크라운 직경, 엽면적은 두상관수와 안개관수 처리에서 높은 값을 나타냈다. 게다가 지상부의 생체중 및 건물중은 다른 처리에 비해 두상관수와 안개관수 처리에서 무거웠다. 지하부의 건물중은 안개관수 처리에서 유의적으로 무거웠다. 하지만 근장, 1차 근수, 엽록소 값, 지하부의 생체중은 모든 처리구에서 유의적인 차이를 나타내지 않았다. 결과적으로, '매향' 딸기의 삽목 번식 시 생육과 발근율은 두상관수와 안개관수 처리에서 가장 효과적인 것으로 나타났다.

Large-scale Culture of Plant Cell and Tissue by Bioreactor System

  • Large-scale cultures of plant cell, tissue, and organ have been achieved by using BTBB. When different sized BTBBs (5 L, 20 L, 100 L, 300 L, and 500 L) were tested for the culture of yew cells (Taxus cuspidata Sieb. et Zucc.), cell growth increment reached to 94.5% in SCV after 24 days of culture with 30% of inoculation cell density. However, there were some variations in the production of taxol and its derivatives among the BTBBs of different size. Approximate 4 ㎎/l of taxol and 84 ㎎/l of total taxanes were obtained by using a 500L BTBB after 6 weeks of culture. With a 20L BTBB, about 20,000 cuttings of virus-free potatoes (cv. Dejima) could be obtained by inoculating 128 explants and maintaining 8 weeks under 16 hr light illumination. The frequency of ex vitro rooting of the cuttings revealed as more than 99% under 30% shade. By incorporating two-stage culture process consisting of multiple bulblet formation in solid medium and bulblet development in liquid medium, mass propagation of lily through bioreactor seemed to be possible. In the case of 'Marcopolo', the growth of mini-bulblets in BTBB was nearly 10 folds faster than that of the solid medium. Time course study revealed that maximum MAR yield of ginseng (Panax ginseng C. A. Meyer) in a 5 L and 20 L BTBB after 8 weeks of culture was 500 g and 2.2 ㎏, respectively. By cutting the MAR once and/or twice during the culture, the yield of root biomass could be increased more than 50% in fresh weight at the time of harvest. With initial inoculum of 500 g of sliced MAR in a 500 L BTBB, 74.8 ㎏ of adventitious root mass was obtained after 8 weeks of culture. The average content of total ginseng saponin obtained from small-scale and/or pilotscale BTBBs was approximately 1% per gram dry weight. Based on our results, we suggest that large-scale cultures of plant cell, tissue, and organ using BTBB system should be quite a feasible approach when compared with conventional method of tissue culture.

  • Obstructive sialadenitis of major salivary glands is a common entity that occurs either in sialolithiasis or in foreign-body obstruction of the excretory ducts. This is characterized histologically by the presence of duct-like structural groups in a highly fibrotic stroma. Although the pathologic features are well recognized, the various cell types involved in the atrophy and subsequent regeneration of the obstructed salivary gland have been controversial. For this reason, an animal model of obstructive sialadenitis that induced atrophy in the salivary gland was used. Experimental study was performed to observe changes of submandibular gland in rabbit and apply the results to clinical activity. Forty-five rabbits each weighing about 3Kg were used and divided into control and experimental group. In the experimental group, ducts of submandibular gland was ligated and cutted divided into each twenty rabbits. Rabbits were serially sacrificed on the 3rd, 5th, 14th, 30th day of experiment. The submandibular glands were dissected out at sacrifice and stained with H&E, MT, immunohistochemical stain and the histological examinations were carried out under the light and transmission electron microscope. After examination and comparison of all specimens, the results of this study were as follows: 1. In the features of H&E stain, moderate infiltration of inflammatory cell were present at 3rd day of experiment. The features of ductal metaplasia was observed after 7th day in the ligation group and destructive changes was continued. In the cutting group, atrophic changes were less severe than ligation group but the small ductule were separated from stroma after 7th day. 2. In the feature of MT stain, apposition of connective tissue was increased in all group, more active in ligation group. 3. In the features of immunohistochemical stain, ligation group showed increased PCNA positive response at 7th day and the higher activity of duct cells was observed. Severance group showed more PCNA positive response than ligation group at 30th day. 4. In TEM features, ductal metaplasia was started at 7th day and degenerative change with margination of nucleus had been severe. Although ductal metaplasia was seen in the severance group, more numerous granule in different size was founded than ligation group. From above results, degenerative change was identified with ductal metaplasia, apically apposition of granule, r-ER destruction in ligation group. Severance of duct elicit degenerative change of grandular cells but the change was less severe than ligation group and more PCNA positive cell was founded at acinar cell.

절피원심법으로 대규모 채취한 누에체액의 곤충세포 증식효과 (Effect of Silkworm Hemolymph Collected Large-scale Bleeding Method in Insect Cell Culture)

  • 누에체액이 동물세포 배양의 필수첨가물인 FBS를 대체할 수 있는 세포증식 효과가 있는 것으로 확인되었으나, 누에의 배발에 상처를 내어 채혈하는 방법으로는 대량 채취가 어렵다. 본 연구에서는 이러한 문제점을 해결코자 대량의 체액을 한꺼번에 얻을 수 있는 대규모 채혈법을 검토하게 되었다. 1. 누에 머리부분과 꼬리부분(배발 포함)에 상처를 낸 후 원심하여 체액을 채취하는 방법(절피원심법)이 기존의 소규모 채취한 체액과 유사한 세포증식 촉진효과를 보였다. 2. 절피원심한 체액의 원심속도에 따른 세포증식 촉진 효과는 500rpm과 1000rpm에서 양호한 값을 보였다. 3. 원심속도를 500rpm으로 고정한 경우 원심시간을 5분에서 30분까지 달리하여도 세포증식 효과는 유사하였고, 체액 회수율은 원심시간이 길어질수록 증가하였으나 일정시간 이상부터는 증가하지 않았다. 4. 절피원심법으로 대규모 채취한 체액의 액상회수율은 누에(백옥잠)생체중의 19.5%이었고, 분말회수율은 1.4% 이었으며, 분말체액에서도 세포증식 촉진 효과가 인정되었다.

전부도재관용 레진시멘트의 생체적합성에 관한 연구 (A STUDY ON BlOCOMPATABILITY OF RESIN CEMENTS FOR ALL-CERAMIC CROWN)

  • Statement of problem : Resin cements were used widely on all ceramic crowns, but the influence of resin cements on biocells was not understood clearly. Purpose : This study was investigated to evaluate the biocompatibility of resin cements for all-ceramic crowns. Material and Method : The resin cements used in this study were Panavia F (Kuraray Co., Ltd. Japan), Variolink II (Vivadent Ets., Schann / Liechtenstein), and Bistite II (Bistite dual cure resin cement-clear Tokuyama Soda Co. Japan). The viability of normal human oral keratocytes, gingival fibroblast, and gingival fibroblast immortalized by Human Papilloma virus 16 was measured in vitro for evaluation of cytotoxicity on resin cements, and the response of pulp tissue was analyzed and evaluated with light microscope after application of cements at cutting edge of incisors. Results : The normal human oral keratocytes was the most sensitive to toxicity of resin cement, and toxicity of cements was higher in Bistite II than in Variolink II. The cell viability of immortalized gingival fibroblast did not affected by type of cement and cultivation period, but there was a tendency that cytotoxicity in Bistite II was higher than in Variolink II. The cell viability of gingival fibroblast was similar to that of immortalized gingival fibroblast regardless of cement type, but Bistite II showed more toxic than others after 5 days cultivation. The responses of pulp tissue according to cement type were similar after 2 days cultivation, but revealed high toxicity in Bistite II after 10 days cultivation. Conclusion : Variolink II was more biocompatible than any other resin cements used in this study.

미역김의 제조와 이화학적 특성에 관한 연구 - 제 1 보 : 미역김의 조직화학적 특성 - (Studies on the Manufacture of Underia pinnatifida Laver and it's Physicochemical Properties - I. Histochemical Properties -)

  • 미역김의 제조와 이화학적 특성에 관한 계속 연구로 실시한 미역김의 조직화학적 특성 연구 결과는 다음과 같다. 미역잎은 표층, 피층, 속층으로 구성되어 있는데 미역김의 결합단면을 광학현미경으로 관찰한 결과 피층과 속층이 완전히 파괴되었고 표층세포끼리 결합되어 있었다. 미역김 절편을 $Na_2CO_3$용액 처리로 알진산을 추출한 후 광학현미경으로 검경한 결과 알진산이 추출된 결합부위는 periodic acid schiff reagent반응이 나타나지 않았으며, 한편 미역잎을 $Na_2CO_3$용액으로 미리 처리하여 알진산을 추출한 후에는 정상적인 미역김을 제조할 수 없었고 비정상적인 결합구조이었으므로 알진산이 미역김의 결합물질임을 확인할 수 있었다. 미역김의 결합부위를 전자현미경으로 관찰한 결과 알진산의 결합위치는 미역김의 표층세포막과 세포막사이임이 밝혀졌다. $Na_2CO_3$용액을 처리한 미역잎으로 제조한 비정상적인 미역김의 결합구조는 표층세포껍질로 인지되는 물질만이 서로 엉켜 있었다.

Micro-CT - a digital 3D microstructural voyage into scaffolds: a systematic review of the reported methods and results

  • Background: Cell behavior is the key to tissue regeneration. Given the fact that most of the cells used in tissue engineering are anchorage-dependent, their behavior including adhesion, growth, migration, matrix synthesis, and differentiation is related to the design of the scaffolds. Thus, characterization of the scaffolds is highly required. Micro-computed tomography (micro-CT) provides a powerful platform to analyze, visualize, and explore any portion of interest in the scaffold in a 3D fashion without cutting or destroying it with the benefit of almost no sample preparation need. Main body: This review highlights the relationship between the scaffold microstructure and cell behavior, and provides the basics of the micro-CT method. In this work, we also analyzed the original papers that were published in 2016 through a systematic search to address the need for specific improvements in the methods section of the papers including the amount of provided information from the obtained results. Conclusion: Micro-CT offers a unique microstructural analysis of biomaterials, notwithstanding the associated challenges and limitations. Future studies that will include micro-CT characterization of scaffolds should report the important details of the method, and the derived quantitative and qualitative information can be maximized.

  • One of the current innovation trends in the solar industry is the increase in the size of silicon wafers. As the wafer size increases, the series resistance of the module rises, highlighting the need for research on methods for cutting and bonding solar cells. Among these, the Infrared (IR) laser scribing technique has been extensively researched. However, there is still insufficient optimization research regarding the thermal damage caused by lasers on the Transparent Conductive Oxide (TCO) layer of Heterojunction (HJT) solar cells. Therefore, in this study, we systematically varied conditions such as IR laser scribing speed, frequency, power, and the number of scribes to investigate their impact on the performance of cut cells under each condition. Additionally, we conducted a comparative analysis of thermal damage effects on the TCO layer based on varying scribing depths.

초정밀 대면적 미세 형상 가공기의 구조 특성 해석 (Structural Characteristic Analysis of an Ultra-Precision Machine for Machining Large-Surface Micro-Features)

  • In recent years, research to machine large-surface micro-features has become important because of the light guide panel of a large-scale liquid crystal display and the bipolar plate of a high-capacity proton exchange membrane fuel cell. In this study, in order to realize the systematic design technology and performance improvements of an ultra-precision machine for machining the large-surface micro-features, a structural characteristic analysis was performed using its virtual prototype. The prototype consisted of gantry-type frame, hydrostatic feed mechanisms, linear motors, brushless DC servo motor, counterbalance mechanism, and so on. The loop stiffness was estimated from the relative displacement between the tool post and C-axis table, which was caused by a cutting force. Especially, the causes of structural stiffness deterioration were identified through the structural deformation analysis of sub-models.
