Cytohistological Study of Abnormal Cell Division of Arabidopsis Stem Infected with Geminivirus (Geminivirus에 감염된 Arabidopsis 줄기의 이상세포분열에 관한 세포조직학적 연구)

  • The internal structures of Arabidopsis thaliana infected with beet curly top virus (BCTV) were studied by light microscopy. Hyperplasia was observed in the inflorescence stems of Arabidopsis thaliana ecotype Sei-O at 2 weeks after BCTV-Logan inoculation and callus was induced on symptomatic tissues at 4 weeks after virus inoculation. The infection processes were revealed as follows: hyperplasia of phloem tissue, necrosis of hyperplastic phloems, lacuna formation of necrotic tissues, elongation and enlargement of cortex and epidermal cells surrounding the lacuna formed phloem tissues, induction of cell division in the enlarged cortex and epidermal cells, and induction of callus tissue. Callus formation on Arabidopsis was caused by the virus infection, and virus inclusion body was observed in both phloem and callus tissue by azure-A staining.

Molecular Analysis of Geminigirus ORFs on Symptom Development

  • Mutants of the monopartite geminivirus beet curly top virus (BCTV) have been screened for infectivity, systemic movement, replication and symptom development in Arabidopsis thaliana. As known by coding for coat protein, R1 mutant was not infectious and did not move systemically. R2, R3 and L2/L3 mutants produced milder symptoms compared to wild type BCTV but the infectivity was reduced by 40% to 60%. R2 ORF is thought to be involved in the regulation of ssDNA and dsDNA accumulation because only dsDNA was accumulated on R2-infected organs. Disruption of ORF L4 resulted in reduced infections, but the viral DNA was accumulated in infected organs from roots to shoot tips as much as wild type BCTV on Sei-O. In addition, 4 mutants did not produce callus-like tissues on infected organs, suggesting that L4 ORF may play a role in the induction of host cell divisions by virus infection. This result was supported by the patterns of mRNA expression and promoter analysis of the cell cycle marker gene, cycl, on Arabidopsis. cycl mRNA was accumulated on symptomatic organs by wild type BCTV infections but not by L4 mutant. We conclude that the BCTV L4 ORF is essential for symptom developments, specially callus-like formation on infected organs.

Halloysite Formation by the Alteration of Trachytic Glass in Ulleung Island (울릉도 조면암질 유리의 변절에 의한 할로이사이트의 생성)

  • Hydrated halloysite was formed by the low temperature alteration of trachytic tuff in onion-like spherical or curly platy forms. The Si content was higher than that of ideal kaolin minerals, indicating the possible presence of amorphous materials. The high Fe content is responsible for the platy morphology of halloysite. The leached ions precipitated as halloysite filling the interparticle pores, while trachytic glass was replaced by Si-rich amorphous materials in an hemispherical form, which was further evolved into spherical halloysite. Halloysite is one of the major alteration products of the volcanic ash in Ulleung Island underwent intense trachytic volcanism.

Effect of Permanent Wave on Hair Damage and Morphological Changes after Natural Henna Treatment (천연헤나 처리 후 퍼머넌트 웨이브 시술 시 모발 손상도 및 형태학적 변화에 미치는 영향)

  • This study attempted to investigate the effects of natural henna-treated permanent wave on degree of damage and morphological changes of hair. For this, amino acid was analyzed, and hair thickness and surface were observed through SEM. In addition, hair curl formation & retention, methylene blue coloring, tensile strength and elongation were measured. The results found the followings: Total amino acid contents rose as the number of natural henna treatment increased, hair protection effects are expected. In terms of curl formation, in an experimental group hair became more elastic as the number of natural henna treatment increased, compared to a control group. In both control and experimental groups, hair retention decreased as shampooing frequency increased because of extension of (C) and (C)', According to analysis of hair thickness using SEM, hair became thick in an experimental group after natural henna treatment, compared to a control group. In morphological observation of hair surface, it was mostly clean, and cuticle was tidy and healthy. In methylene blue coloring, as the number of natural henna treatment increased, absorbance went up. In addition, tensile strength declined in an experimental group, compared to a control group. As the number of natural henna treatment increased, elongation in an experimental group increased, compared to a control group. The above results show that as the number of natural henna treatment increased, hair became thicker, and curl formation and retention improved. In addition, hair elasticity and flexibility were enhanced, reducing a risk of hair damage. Therefore, the study results would be available as basic data in reducing scalp and hair damage, proposing the natural henna treatment as a method to reduce hair damage from chemical perm in cosmetology industry.
