In Vitro and In Vivo Anti-Tobacco Mosaic Virus Activities of Essential Oils and Individual Compounds

  • Essential oils are increasingly of interest for use as novel drugs acting as antimicrobial and antiviral agents. In the present study, we report the in vitro antiviral activities of 29 essential oils, extracted from Chinese indigenous aromatic plants, against the tobacco mosaic virus (TMV). Of these essential oils, those oils from ginger, lemon, tea tree, tangerine peel, artemisia, and lemongrass effected a more than 50% inhibition of TMV at 100 ${\mu}g/ml$. In addition, the mode of antiviral action of the active essential oils was also determined. Essential oils isolated from artemisia and lemongrass possessed potent inactivation and curative effects in vivo and had a directly passivating effect on TMV infection in a dose-dependent manner. However, all other active essential oils exhibited a moderate protective effect in vivo. The chemical constitutions of the essential oils from ginger, lemon, tea tree, tangerine peel, artemisia, and lemongrass were identified by gas chromatography and gas chromatography-mass spectrometry. The major components of these essential oils were ${\alpha}$-zingiberene (35.21%), limonene (76.25%), terpinen-4-ol (41.20%), limonene (80.95%), 1,8-cineole (27.45%), and terpinolene (10.67%). The curative effects of 10 individual compounds from the active essential oils on TMV infection were also examined in vivo. The compounds from citronellal, limonene, 1,8-cineole, and ${\alpha}$-zingiberene effected a more than 40% inhibition rate for TMV infection, and the other compounds demonstrated moderate activities at 320 ${\mu}g/ml$ in vivo. There results indicate that the essential oils isolated from artemisia and lemongrass, and the individual compound citronellal, have the potential to be used as an effective alternative for the treatment of tobacco plants infected with TMV under greenhouse conditions.

  • Aplastic anemia is a rare disease, which is characterized by pancytopenia and hypocellular bone marrow without infiltration of abnormal cells or fibrosis. The incidence in Asia is higher than in the West and new cases are diagnosed at a rate of 5.1 per million pediatric populations per year in Korea. The pathophysiology is understood roughly by defective hematopoiesis, impaired bone marrow micro-environment and immune mechanism. Treatments are performed on basis of pathogenesis and selected depending on the severity. Immunosuppressive therapy with antilymphocyte or antithymocyte globulin and cyclosporine is effective in the majority of patients but has some problems including relapse or clonal evolution. Recently, there have been clinical trials of immunosuppression with hematopoietic growth factors or other drugs. Allogeneic hematopoietic stem cell transplantation (HSCT) is curative in children with severe aplastic anemia. The overall survival in HSCT from HLA-identical sibling is higher than alternative donor, including HLA matched unrelated donor or cord blood. We have to consider quality of life after HSCT because of high survival rate. However, chronic graft versus host disease and graft failure are important factors that affect the quality of life and overall survival. We need further investigation to make new regimens aimed at overcoming these risk factors and perform clinical trials.

Cestrum tomentosum L.f. Extracts against Colletotrichum scovillei by Altering Cell Membrane Permeability and Inducing ROS Accumulation

  • Chili pepper anthracnose, caused by Colletotrichum spp., is a significant biotic stress affecting chili fruits globally. While fungicide application is commonly used for disease management due to its efficiency and cost-effectiveness, excessive use poses risks to human health and the environment. Botanical fungicides offer advantages such as rapid degradation and low toxicity to mammals, making them increasingly popular for sustainable plant disease control. This study investigated the antifungal properties of Cestrum tomentosum L.f. crude extracts (CTCE) against Colletotrichum scovillei. The results demonstrated that CTCE effectively inhibited conidia germination and germ tube elongation at 40 ㎍/ml concentrations. Moreover, CTCE exhibited strong antifungal activity against C. scovillei mycelial growth, with an EC50 value of 18.81 ㎍/ml. In vivo experiments confirmed the protective and curative effects of CTCE on chili pepper fruits infected with C. scovillei. XTT analysis showed that the CTCE could significantly inhibit the cell viability of C. scovillei. Mechanistic studies revealed that CTCE disrupted the plasma membrane integrity of C. scovillei and induced the accumulation of reactive oxygen species in hyphal cells. These findings highlight CTCE as a promising eco-friendly botanical fungicide for managing C. scovillei infections in chili peppers.

Characteristics and control activity of copper hydroxide against pepper Phytophthora blight caused by Phytophthora capsici (고추 역병에 대한 Copper hydroxide의 방제 특성)

  • Characteristics and control activity of copper hydroxide against pepper phytophthora blight caused by Phytophthora capsici were investigated in a greenhouse and a pepper field Copper hydroxide strongly inhibited the germination of zoosporangia and zoospores of P. capsici JHCS 2-5, showing $EC_{50}$ value by 0.6 and 0.3 ${\mu}g\;mL^{-1}$. With 1,040 ${\mu}g\;mL^{-1}$ of copper hydroxide in a greenhouse it showed 80% of the control value by soil-drenching application, while 16% by leaf-spraying. However, when it was treated enough to runoff to soil by leaf-spraying application with 50 ml per a pepper plant, it controlled a pepper phytophthora blight by 94.6 % of control value. Copper hydroxide showed a high protective activity at 1 and 3 days before application, while no curative activity. In a field it showed a high activity of 91%, when pepper plants were treated with copper hydorxide 4 times with a intervals of 10 days.

Selection of Fungicides for the Control of Soybean Black Root Rot Caused by Calonectria ilicicola (콩 검은뿌리썩음병 방제를 위한 살균제 선발)

  • Fungicidal screening was performed to control soybean black root rot caused by Calonectria ilicicola through in vitro and greenhouse assays. In in vitro assay, 25 fungicides were assessed by an agar dilution method and a 96-well microtiter plate method. While protective fungicides including dithianon, dichlofluanid, mancozeb, and captan showed a very low activity against the mycelial growth C. ilicicola SC03-15 in the agar dilution method, they displayed potent inhibitory activity against spore germination in a 96-well microtiter plate method with $EC_{50}$ values of 4.65, 0.61, 4.64, and $0.29{\mu}g\;mL^{-1}$, respectively. Ergosterol biosynthesis-inhibiting (EBI) fungicides showed different antifungal activity against mycelial growth and spore germination according to molecules. Difenconazole displayed higher antifungal activity against spore germination rather than mycelial growth, and prochloraz inhibited potently both mycelial growth and spore germination with EC50 values less than $1.8{\mu}g\;ml^{-1}$. In contrast, the other EBI fungicides inhibited more highly mycelial growth than spore germination. Carbendazim+diethofencarb and dazomet also inhibited both mycelial growth and spore germination of C. ilicicola SC03-15 at very low concentrations. In greenhouse assay, carbendazim+diethofencarb effectively controlled a soybean black root rot by drenching 2 days before or after inoculation. In addition, tebuconazole showed potent curative activity against soybean black root rot.

Control Efficacy of Carboxylic Acid Amide Fungicides against Pepper Phytophthora Blight Causing Phytophthora capsici (고추 역병에 대한 Carboxylic Acid Amide계 살균제의 방제 효과)

  • It was conducted to investigate the control efficacy of carboxylic acid amide (CAA) fungicides, such as benthiavalicarb, iprovalicarb, dimethomorph and mandipropamid, against pepper Phytophthora blight caused by P. capsid in the laboratory and the field. The fungicides inhibited mycelial growth and direct sporangium germination of P. capsid strongly, while there was no activity of all fungicides against zoospore release from sporangium. In greenhouse test, they showed the good protective and curative effect against pepper Phytophthora blight. Benthiavalicarb applied at $100{\mu}g\;mL^{-1}$ 7 days before inoculation prevented pepper Phytophthora blight by 100%, even though the zoosporangiurn suspension of P. capsid adjusted to not only $5{\times}10^3$ zoosporangia $mL^{-1}$ but also $1{\times}10^5$ zoosporangia $mL^{-1}$ was inoculated by soil-drenching. Except for dimethomorph, the other fungicides showed an excellent control activity over 2 years from 2009 to 2010 in the field test. The control value of dimethomorph applied at $250{\mu}g\;mL^{-1}$ was low, 27.2% in 2009, but that of dimethomorph applied even at $125{\mu}g\;mL^{-1}$ was high, 89.5% in 2010. All the fungicides showed good inhibitory effect on the mycelial growth and the direct germination of zoosporangiurn, and controlled pepper Phytophthora blight preventively and curatively, can be used to establish the spray system for control1ing the pepper disease.

Controlling Effect of Agricultural Organic Materials on Phytophthora Blight and Anthracnose in Red Pepper (고추 역병과 탄저병에 대한 친환경유기농자재의 방제 효과)

  • A total of 20 agricultural organic materials including inorganic compounds, plant oils and plant extracts were used in the study for assessing the control efficacy on pepper diseases. Among inorganic compounds, only copper hydroxide showed inhibitory effect on both Phytophthora capsici causing Phytophthora blight and Colletotrichum acutatum causing anthracnose. Phosphorous acid inhibited the growth of P. capsici on PDA, and Sulfur/quicklime had it on that of C. acutatum. Plant essential oil, rosemary oil, and rapeseed oil among plant oils and plant extract of Japanese apricot/ginkgo nut inhibited the mycelial growth of the two pathogens. In the screening using pepper plant seedlings, the control efficacy on Phytophthora blight in 6-leaf stage of seedling was superior to that in 4-leaf stage of seedling. A protective effect on Phytophthora blight was displayed by copper hydroxide, sulfur/quicklime, water soluble calcium, phosphorous acid, plant essential oil, and cloves extract. When C. acutatum was inoculated by the non-wound method, copper hydroxide and rapeseed oil showed excellent protective activities with control values of 91.3% and 82.6%, respectively. However, copper hydroxide did not show any activity, when C. acutatum was inoculated after wounding pepper fruits. All organic materials never showed the curative effect on Phytophthora blight and anthracnose in pepper seedling assay and fruit assay.

Control effect of the Mixture of Bacillus amyloliquefaciens M27 and Plant Extract against Cucumber Powdery Mildew (Bacillus amyloliquefaciens M27과 식물천연물 혼합제에 의한 오이 흰가루병의 방제 효과)

  • Bacillus amyloliquefaciens M27 was selected as a control agent for the biological control of cucumber powdery mildew. The new mixture of B.amyloliquefaciens M27 and plant (Eucalytus) extract was developed to improve the control activity of B.amyloliquefaciens M27 against cucumber powdery mildew. The mixed formulation showed the high preventive and curative control effect against cucumber powdery mildew when it was diluted at 500 times and foliar-sprayed. Its control effect was higher in preventive spraying than curative spraying. When 500-fold diluted solution of the formulation was sprayed preventively four times at five-day intervals, three times at seven-day intervals and twice at ten-day intervals, the diseased leaf area was shown to be 4.4%, 8.0%, 27.9%, respectively; Whereas the diseased leaf area in the control plot was 45.4%. When the 500-fold diluted formulation was sprayed curatively four times at five-day intervals, three times at seven-day intervals and twice at ten-day intervals after occurred cucumber powdery mildew, the diseases leaf area was 11.5%, 25.2%, 51.8%, respectively; whereas in the control plot, the diseases leaf area was 64.3%. When the 500-fold diluted formulation was treated four times at five-day intervals in the plastic house, its control effect was higher than that treated three times at seven-day intervals and twice at ten-day intervals. As the results, the mixed formulation of B.amyloliquefaciens M27 and plant extract could be a promising candidate of bio-fungicides for the environment-friendly control of powdery mildew of cucumber.

FACTORS OF MENTALLY HANDICAPPED CHILDREN AND THEIR FAMILY ASSOCIATED WITH THE QUALITY OF LIFE AND THE EMOTIONAL WELLBEING OF THEIR MOTHERS (장애 아동의 행동 특성과 가족환경이 어머니의 정서적 안녕감과 삶의 질에 미치는 영향)

  • Objective:In life-long disabilities like autism and mental retardation, the authors thought that it is important for clinician to consider the quality of life of a primary caregiver for long-term management and prognosis. This study was to investigate the factors of children and family environment affecting the quality of life and depression in mothers with autistic and/or mentally retarded children. Methods:41 autistic and/or mentally retarded children aged 5-12 years with their mothers were surveyed from September, 1998 to January, 1999, with K-CBCL, K-BDI, K-FES, and K-SBQOL scale and compared with data from 35 normal control subjects. Results:1) Total K-BDI and K-SBQOL scores of mothers with mentally handicapped children were significantly poorer than the scores of normal control group. Independence, intellectual/cultural orientation and active recreation subscales of K-FES in mentally handicapped children were significantly decreased than those in normal control group. 2) Total K-BDI score of mothers with mentally handicapped children was correlated with their children’s behavioral problems, especially internalizing and thought symptoms, and with family cohesion, expressiveness, conflict and independence. 3) Totol K-SB quality of life score of mothers with mentally handicapped children was correlated with their children’s behavioral problems, especially attention problem, and with family cohesion, conflict, independence, intellectual/cultural orientation, and moral-religional emphasis. 4) The quality of life of mothers with mentally handicapped children was predicted by attention problem($R^2$=.36, p=.000) and social competence($R^2$=.07, p=.038) in children and family cohesion ($R^2$=.16, p=.001). 5) Depression of mothers with mentally handicapped children was predicted by internalizing symptom ($R^2$=.21, p=.003) and thought disorder($R^2$=.06, p=.048) in children and family cohesion($R^2$=.14, p=.008). Conclusion:Reducing behavioral problems and family therapeutic intervention in autistic and mentally retarded children can improve the quality of life of primary caregivers and long-term prognosis of the children, although those are not curative.
