Effect of temperature on pharmacokinetics of nalidixic acid, piromidic acid and oxolinic acid in olive flounder Paralichthys olivaceus following oral administration (넙치, Paralichthys olivaceus에 nalidixic acid, piromidic acid, oxolinic acid의 경구투여 약물동태에 미치는 수온의 영향)

  • Effects of temperature ($13{\pm}1.5^{\circ}C$, $23{\pm}1.5^{\circ}C$) on the pharmacokinetic properties of nalidixic acid (NA), piromidic acid (PA) and oxolinic acid (OA) were studied after oral administration to cultured olive flounder, Paralichthys olivaceus. Serum concentrations of these antimicrobials were determined after oral administration of a single dosage of 60 mg/kg body weight (average 700 g). At $23{\pm}1.5^{\circ}C$, the peak serum concentrations of NA, PA and OA, which attained at 10 h, 24 h and 30 h post-dose, were 11.55, 3.79 and $1.12{\mu}g/m\ell$, respectively. At $13{\pm}1.5^{\circ}C$, the peak serum concentrations of NA, PA and OA, which attained at 10 h, 15 h and 30 h post-dose, were 6.36, 1.4 and $1.01{\mu}g/m\ell$, respectively. Better absorption of NA and PA was noted at $23{\pm}1.5^{\circ}C$ compared to $23{\pm}13^{\circ}C$. The elimination of NA from serum of olive flounder was considerably faster at $23{\pm}1.5^{\circ}C$ than at $13{\pm}1.5^{\circ}C$. However, both absorption and elimination of OA were not affected significantly by temperature. The kinetic profile of absorption, distribution and elimination of these antimicrobials in serum were analyzed by fitting to a one- and two compartment model, with WinNonlin program. In the one compartment model for NA, AUC, Tmax and Cmax at $23{\pm}1.5^{\circ}C$ were $258.26{\mu}g{\cdot}h/m\ell$, 10.67 h and $8.91{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $C_{max}$ at $13{\pm}1.5^{\circ}C$ were $341.45 {\mu}g{\cdot}h/m\ell$, 7.72 h and $6.23{\mu}g/m\ell$, respectively. In the one compartment model for PA, AUC, $T_{max}$ and $C_{max}$ at $23{\pm}1.5^{\circ}C$ were $248.12{\mu}g{\cdot}h/m\ell$, 21.15 h and $3.09{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $C_{max}$ at $13{\pm}1.5^{\circ}C$ were $103.89{\mu}g{\cdot}h/m\ell$, 12.89 h and $1.22{\mu}g/m\ell$, respectively. In the two compartment model for OA, AUC, $T_{max}$ and $C_{max}$ at $23{\pm}1.5^{\circ}C$ were $138.20{\mu}g{\cdot}h/m\ell$, 23.95 h and $1.06{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $T_{max}$ at $13{\pm}1.5^{\circ}C$ were $159.10{\mu}g{\cdot}h/m\ell$, 28.03 h and $1.02{\mu}g/m\ell$, respectively.

Changes of Stress Response and Physiological Metabolic Activity of Flounder, Paralichthys olivaceus Following to Food Deprivation and Slow Temperature Descending (먹이제한과 단기 수온하강 조건에서 넙치의 스트레스 반응과 생리학적 대사활성 변화)

  • To find the preliminary environmental conditions for a short-time transport of living olive flounder, Paralichthys olivaceus, the stress response and physiological metabolic activity of the cultured fish to feed deprivation and slow temperature descending ($15.8^{\circ}C{\rightarrow}13.3^{\circ}C$) were monitored for 8 days. The monitored variables were the plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST), glucose (GLU), total protein (TP), electrolytes ($Na^+$, $K^+$, $Cl^-$) and thyroid hormones ($TT_4$, $TT_3$, $FT_4$ and $FT_3$). In food deprivation experiment for 8 days, we did not find any statistical change of level in AST, ALT and electrolytes ($Na^+$, $K^+$, $Cl^-$), but found a significant decrease in TP and GLU. In thyroid hormones, the levels of four hormones in plasma were all showing a tendency to decrease. Especially, $FT_4$ and $TT_3$ were significantly decreased, indicating a withering of physiologic activity. In the temperature test, although no any significant change in AST, TP and electrolytes ($Na^+$, $K^+$, $Cl^-$), we observed a significant decrease of ALT and GLU following to temperature descending from $15.8^{\circ}C$ to $13.3^{\circ}C$ (P<0.05). In the levels of thyroid hormones, any significant change was not observed for experimental period. We conclude that the stress response and physiological activity of olive flounder were more influenced by feed deprivation than slow temperature descending at a transport of living fish, and plasma GLU appears to be sensitive factor to physiological metabolic activity, indicating that it could be used as a monitering mark or index for a health inspection of the fish.

Microbiological characteristics of Vibrio harveyi (Vibrio harveyi의 미생물학적 특성)

  • In 1999, Vibrio harveyi infection occurred among cultured marine finfishes including olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), and turbot (Scophtalmus maximus) in the province of Gyeongsang, Korea. We examined the various microbiological characteristics of this V. harveyi strains. V. harveyi was grown well in the 3% NaCl at 30℃. The swarming activity appeared to be an one of the characteristic properties of the V. harveyi was the highest in the 2% NaCl concentration at TSA medium. These cells were elongated and had the several lateral flagella which is not waved and shorter than polar flagella. It was not luminescent in the all isolated strains. Whole cell of the V. harveyi had a major protein of 50 kDa and presented various band around of 40 kDa in strains.

Proteases in Cell Lysate of Uronema marinum (Ciliata: Scuticociliatida), an Opportunistic Pathogen of Cultured Olive Flounder (Paralichthys olivaceus)

  • The effects of pH, temperature and various inhibitors on the proteolytic activity of the cell lysate of Uronema marium were investigated using colorimetric and substrate gel electro­phoretic methods. The cell lysate of U. marinum showed proteolytic activity over a wide range of pH, and pH optima ranged from pH 5 to 7. The proteolytic activity was increased according to a rise of temperature but decreased at $40^{\circ}$. The proteolytic activity of the parasite lysate was significantly inhibited by protease inhibitors including trans-epoxysuccinyl -L-leucylamido-(4-guanidino) butane (E-64), pepstatin A, phenyl-methanesulfonyl fluoride(PMSF), and ethylenediamine-tetraacetic acid (EDTA). Preincubation of the lysate with E-64 showed the maximum inhibition of the caseionolytic activity. Four protease bands (152, 97, 67 and 40 kDa) were detected by gelatin SDS-PAGE. Significant inhibition of caseinolytic activity and complete abolition of a 152 kDa band in gelatin SDS-PAGE by EDTA indicated that the cell lysate of U. marinum had a metalloprotease Another three proteolytic bands were inhibited by E64, a cysteine protease inhibitor. Preincubation of the cell lysate with pepstatin or PMSF had no effects on the protease bands.

Monitoring of VHS and RSIVD in Cultured Paralichthys olivaceus of Jeju in 2015 (2015년 제주도 양식 넙치(Paralichthys olivaceus)를 대상으로 한 VHS 및 RSIVD 모니터링)

  • In this study, disease surveillance was performed to monitor the prevalence of viral haemorrhagic septicaemia virus (VHSV) and red seabream iridovirus (RSIV) in olive flounder, Paralichthys olivaceus in 2015. The fish samples were collected in March (60 farms), May (55 farms) and July (52 farms) from different farms in Jeju. Reverse transcription polymerase chain reaction (RT-PCR) (VHSV) or PCR (RSIV) results showed that VHSV detected in 2 farms, but RSIV has not been detected in any farms. The sequences of the nucleocapsid protein (N) and glycoprotein (G) gene of the 2 VHSV isolates were successfully sequenced. Phylogenetic analysis was included VHSV isolates reported here together with a representative VHSV isolates available in GenBank. Phylogenetic analysis indicated that most of Korea VHSV isolates were closely related to the Japan and China genotype IVa which is clearly distinct from the North American genotype IVb.

  • A viable but non-culturable (VBNC) state is a survival strategy adopted by bacteria when faced with unfavorable environmental conditions, rendering them unable to grow on nutrient agar while maintaining low metabolic activity. This study explored the impact of temperature and nutrient availability on inducing VBNC state in Edwardsiella piscicida, the most important bacterial fish pathogen, and assessed its pathogenicity at VBNC state. E. piscicida was suspended in filtered sterile seawater and exposed to three different temperatures (4, 10, and 25℃) to induce the VBNC state. Subsequently, the induced VBNC cells were subjected to resuscitation by either raising the temperature to 28℃ or inoculating them in brain heart infusion broth supplemented with 1% NaCl. A propidium monoazide (PMA)-qPCR method was also developed to selectively quantify live (VBNC or culturable) E. piscicida cells. The results showed that the bacteria entered the VBNC state after approximately 1 month at 4℃ and 25℃, and 2 months at 10℃. The VBNC E. piscicida cells were successfully revived within 3 days in a nutrient-rich environment at 28℃, highlighting the significance of temperature and nutrition in inducing and resuscitating the VBNC state. In pathogenicity tests, resuscitated E. piscicida cells exhibited high pathogenicity in olive flounder comparable to cultured bacteria, while VBNC cells showed no signs of infection, suggesting they are unlikely to resuscitate in fish. In conclusion, this study contributes to our understanding of fish pathogen ecology by investigating the characteristics of the VBNC state under varying temperature and nutrition conditions.

Biological characteristics of Scuticociliate with a polymorphic life cycle of ciliate and cyst isolated from cultured olive flounder, Paralichthys olivaceus (양식넙치, Paralichthys olivaceus에서 분리된 Ciliate와 Cyst의 생활사를 갖는 스쿠티카섬모충의 생물학적 특성)

  • Scuticociliatosis has badly settled one of most damaging diseases during the seedling production process of olive flounder. Paralichthys olivaceus in Korea. We isolated a new type of Scuticociliate from flounder. The parasite metamorphoses to ciliate and cyst phases with each other by environmental changes and survive for a relatively long span. The ciliate was measured average 41.8 ${\mu}m$ in length and 21.0 ${\mu}m$ in width, and cyst was 17.0 ${\mu}m$ and 13.5 ${\mu}m$, respectively. Nutritional condition was determined as a major parameter of metamorphosing between ciliate and cyst stages. The ciliate transforms to a cyst stage because of food shortage, and the cyst returns to a ciliate stage with a favorite environmental condition and shows active growth and reproduction. The ciliate multiplied at the maximal density of $2.9 {\times} 10^5 {m\ell}^{-1}$cells in vitro cultivation at $15 ^{\circ}C$temperature using MS BHI medium and bacterial food sources. The ciliate could be proliferated at a 2.5 to $30 ^ \circ}C$ temperature range, pH 6 to 9, and 1 to 55 ppt salinity. Particularly, it survived over one week at $0 ^{\circ}C$temperature showing a high resistance against unfavorable environmental conditions. And the cyst survived for 320 days in the condition of $5 ^{\circ}C$with no feeding, but its survival period was markedly shortened in higher temperature conditions. The chemotherapeutants (formalin and hydrogen peroxide) were clarified as effective chemicals against the ciliate during in vitro trials, but the effect of therapeutants differed in proportion, depending upon the density and the bathing time of chemical compounds.

Pharmacokinetics of cefadroxil after oral administration in olive flounder, Paralichthys olivaceus (Cefadroxil의 경구투여에 따른 넙치(Paralichthys olivaceus)에서의 약물동태학 연구)

  • The pharmacokinetic properties of cefadroxil (CDX) were studied after oral administration for 7 days to cultured olive flounders (average 660 g), Paralichthys olivaceus. For examination of pharmaco-kinetic profiles, CDX of 45 to 225 mg/kg body weight was administered at two different water temperatures, $13{\pm}3^{\circ}C$ or $22{\pm}3^{\circ}C$. CDX concentrations were determined in muscle, plasma, gastrointestinal tract, hematopoietic organs and liver by HPLC-MS/MS. Muscle samples were taken at 0.25, 0.5, 1, 3, 7, 14 and 28 days post dose, whereas plasma, gastrointestinal tract, hematopoietic organs and liver concentrations were measured at 1, 3, 7, 14 and 28 days post-dosing. The kinetic profiles of $C_{max}$, $T_{max}$, $T_{1/2}$ of CDX were analyzed by fitting to a non-compartmental model with PKSolver program. The following pharmacokinetic parameters were obtained with oral administration of 45 and 225 mg/kg at 13 and $22^{\circ}C$ in muscle, plasma, gastrointestinal tract, hematopoietic organs and liver, respectively: $C_{max}$ (maximum tissue concentration)=$985.98-5,032.86{\mu}g/kg$, $5,670.99-38,922.23{\mu}g/l$, $2,457.27-10,192.78{\mu}g/kg$, $886.04-3,070.87{\mu}g/kg$ and $1,188.15-3,814.33{\mu}g/kg$; $T_{max}$ (time for maximum concentration)= every 1 day; $MRT_{0-{\infty}}$ (mean residence time)= 1.51-4.74, 2.12-3.06, 4.25-13.18, 1.37-18.66 and 1.78-29.76 days; $T_{1/2}$ (half-life)= 1.08-3.47, 1.14-5.42, 3.56-10.99, 1.17-14.93 and 1.25-28.55 days.

Biological characterization of Tenacibaculum maritimum isolated from cultured olive flounder in Korea and sensitivity against native plant extracts (한국의 양식넙치에서 분리한 Tenacibaculum maritimum의 특성과 자생식물 추출물에 대한 감수성)

  • Tenacibaculum maritimum (formerly Flexibacter maritimus) is the aetiological agent of an ulcerative and necrotic disease commonly called tenacibaculosis in marine fish. Tenacibaculosis is an economically important disease in a great variety species in Jeju Island cultured fish and leading to this pathogen initially affected by skin, mouth, fins, tail causing severe necrotic and ulcerative lesions on the body surface. In the present study, A-7 strain was isolated from Paralichthys olivaceus showing symptoms of tenacibaculosis and identified as T. maritimum by morphological, biochemical and molecular biological analysis. T. maritimum A-7 is experimentally infected through immersion route in Paralichthys olivaceus which the disease outbreaks in land-based fish tanks of Jeju Island. Up to data a number of treatments proposed for the tenacibaculosis outbreaks are based on the immersion administration of drugs in tank. Oxytetracycline is the most widely used disinfectants in fish farms. However, most of fish farms manager and consumers have expressed concern as bioaccumulation in tissue and its environmental. In addition, this antimicrobial compounds is expensive in fish farmers. The overcome of this problem is desired the application of natural plant derived products. To obtain as 70% EtOH extract antimicrobial compounds against tenacibaculosis from 35 species of Jeju Island native plants were screened for antimicrobial activity against T. maritimum. In the present study were identified most of the plant extracts were better antimicrobial activity against T. maritimum.

Stress in Olive Flounder (Paralichthys olivaceus) and Fat Cod (Hexagrammos otakii) by the Sudden Drop and Rise of Water Temperature (수온의 급하강과 급상승이 넙치 (Paralichthys olivaceus)와 쥐노래미(Hexagrammos otakii)에 미치는 스트레스)

  • The effects of sudden changes of water temperature (WT) on the stress response and physiological change of the cultured olive flounder in large (FL) and small (FS) size, and fat cod (FC) were examined by manipulating WT (2 types) in a flow through seawater culture system with 6 tanks (water vol. 270 L/tank). The WT was decreased from $20^{\circ}C$ to $10^{\circ}C$ within 5 hours ($2^{\circ}C/hour$) and maintained at $10^{\circ}C$ for 21 hours (Exp. I), and it was raised from $20^{\circ}C$ to $30^{\circ}C$ within 5 hours and maintained at $30^{\circ}C$ for 21 hours (Exp.II). In Exp. I, the levels of blood hematocrit at 5 hours ($10^{\circ}C$) in FS was significantly decreased from $13.5\pm2.0\%\;to\;11.3\pm2.3\%$, but FC at 2.5 hours ($15^{\circ}C$) ($19.0\pm0.3\%\;to\;23.2\pm3.8\%$) was increased, The blood hemoglobin concentration of all fish in Exp, II was significantly increased until 8 hours after raising WT from $20^{\circ}C$ to $30^{\circ}C$. In Exp. I and II , the levels of plasma cortisol in FL, FS and FC was changed from $5.2\pm8.5ng/mL,\;4.4\pm4.5ng/mL\;and\;2.7\pm0.4ng/mL$, respectively, before sudden drop and rise of WT. The levels of plasma cortisol of in FL ($164.0\pm53.1ng/mL$) and FC ($207.9\pm25.4ng/mL$) were significantly increased by the lowering WT sharply during whole experiment. The FL ($12.6\pm2.0ng/mL$) and FS ($4.0\pm3.9ng/mL$) showed no significant differences in cortisol level according to sudden rise of WT (5 hours). But it in FC ($44.7\pm18.2ng/mL$) was increased. In Exp. I, the plasma glucose levels of all fish groups were decreased after 5 hours ($10^{\circ}C$), The plasma lactic acid concentration of FL and FS showed no significant differences until 5 hours after raising WT from $20^{\circ}C$ to $30^{\circ}C$, But it in FC was significantly increased with WT raise.
