  • Various antibiotics, that could induce the appearance of resistant microorganisms, have been used for treatment or prevention of bacterial diseases in marine and ornamental fish. We determined and characterized the level of antibiotic-resistant bacteria and proportion of multi-drug resistant bacteria in intestinal microflora of both marine fish cultured in Korea and imported ornamental freshwater fish. For this the bacterial species and resistance to antibiotics were investigated in intestine of rock bream Oplegnathus fasciatus cultured in Korea and pearl gourami Trichogaster leeri imported from Singapore to characterise. Although the bacterial species were different, proportions of resistant bacteria to single antibiotics or multi-drug were higher in intestinal microflora of pearl gourami Trichogaster leeri imported from Singapore than in rock bream Oplegnathus fasciatus cultured in Korea. These results indicate that various antibiotics have been being used before trading without measures in the market of asian ornamental fishes, providing high risks for the emergence of multi-drug resistant bacteria.

  • Lipid classes and fatty acid compositions of eight species of wild and cultured seawater fish in Korea were investigated. Total lipid (TL) contents of wild and cultured fish were $2.64\pm1.88\%$ and $5.42\pm1.76\%$, respectively, except for rockfish and striped beakperch. Non-polar lipids (NL) in all fish samples comprised approximately $84\%$ of the TL content. The proportion equation of NL content to TL content was y=0.9296x-0.4468 $(R^2=0.98l2, p<0.001)$. The most abundant NL class was triglyceride. The prominent fatty acids in all fish samples were 16: 0, 18: 1(n-9), 22 : 6(n-3) (docosahexaenoic acid, DHA), 16: 1(n-7), 20 : 5 (n-3) (eicosapentaenoic acid, EPA), 18 : 0 and 18 : 1(n-7). The polyunsaturated fatty acid (PUFA) group was the richest among the total fatty acids (TFA). DHA and EPA were comprised of approximately $85\%$ of n-3 PUFA. There was a positive correlation between TFA content and n-3 PUFA content; y=0.292x-0.0055 $(R^2=0.9349, p<0.001)$. The n-3 PUFA content of the cultured fish was approximately twice as much as that of the wild fish. Therefore, cultured fish were proven to provide better sources of n-3 PUFA if compared to wild fish.

  • Although cultured sweet smelt production recently have been rapidly increased from 89 metric tons (M/T) in 1997 to 500M/T In 1999 in Korea, wild fish have been still used as broodstock for cultured fish seed. This is considered due to having higher sperm and e99 qualify in wild fish broodstock than cultured fish. However, capturing a number of wild sweet smelt broodstock might lead to decreasing of the production of wild fish, In truth, of which production has been greatly decreased so that a number of artificial juvenile have been stocked some rivers in Korea. • This study investigated the seasonal changes of body composition and elasticity between wild and cultured brown croaker, Miichthys miiuy. The wild fish were analyzed and compared with cultured fish in moisture, crude protein, lipid, ash and many kinds of amino acids. Cultured fish was higher in moisture content and lower in crude lipid and protein content than those of wild one. The wild fish were more abundant in the total amino acid compositions than those of cultured one. As result E/A ratio there was a little significant differences between wild and cultured. In highly unsaturated fatty acid, EPA (Eicosapentaenoic Acid) and DHA (Docosahexaenoic Acid) content of cultured fish were higher than wild one. On the other hand, the gel strength, max weight and hardness of wild fish were higher than cultured one.

  • Changes of nucleotides and their related compounds in raw, cooked and frozen fish muscle were studied with HPLC. Red sea bream(cultured and wild) and flounder(cultured, cultured with Obosan(equation omitted) and wild) were used for this study. In nucleotides, contents of ATP was similar to that of IMP and some of H$\times$R(inosine) and H$\times$(hypoxanthine) were existed in fresh muscle. ATP was decomposed rapidly and contents of IMP became different between cultured and wild fish after 6 hours. The content of IMP was lower in the cultured red sea bream(3.39$\mu$ mole/g) and flounder(3.17$\mu$ mole/g) than in the wi1d red sea bream(7.31$\mu$ mole/g) and flounder(5.03$\mu$ mole/g). But, the flounder cultured with Obosan contained the largest amounts of IMP After 24 hours, K values of cultured fish muscle(27.7%, 28.2%) were higher than that of wild ones(22.8%, 24.3%). The K value of cultured flounder fed with 0.3% Obosan(equation omitted)(25.7%) was between cultured and wild flounder. IMP was the one which existed the most in cooked and frozen muscle. Amounts of H$\times$R and H$\times$ were more in cooked and frozen muscle. than in raw muscle. From these results, we could suggest that the wild one was more palatable and fresher than the cultured one and the palatability of cultured one seemed to be improved depanding on the feed.

  • Although Vietnam has a high risk of fish borne zoonotic trematode(FZT) infections for humans, little information exists on the epidemiology of these infections in the country's fish. Because of the importance of cultured cat-fish and snakehead production in An Giang province, a major production area in the Mekong Delta of Vietnam, a survey for FZTs was carried out in randomly selected fish farms between June 2005 and March 2006. For comparison, wild fish from the same area were also surveyed. A total of 852 cultured fish from 4 districts were collected and examined by pepsin digestion to determine their FZT infection status. In Tra catfish, the prevalence of all types of metacercariae was 2.6%, of which the prevalence of Haplorchis pumilio was 0.7%. The overall prevalence of metacercariae in wild fish was 30.6%, of which 10.3% harbored zoonotic species: H. pumilio(2.8%) and Procerovum sp.(5.6%). The prevalence of Opisthorchis metacercariae, which were diagnosed as O. viverrini, was 1.9%. No metacercariae were found in cultured snake head fish, although wild-caught snakehead fish had a FZT prevalence of 10.3%: 5.1% were O. viverrini; 2.6% H. pumilio; and 2.6% were Procerovum sp. These are the first reports of H. pumilio, Procerovum sp., and O. viverrini metacercariae in Vietnamese fish. These results indicate that consumption of improperly prepared fish represents a significant risk of acquiring FZTs in this south Vietnam region.

  • Antibacterial efficacy and safety of copper sulfate pentahydrate (5% solution) was evaluated in laboratory and field studies for disinfection of some fish bacterial pathogens in cultured fish, flounder(Paralichthys olivaceus), mirror carp(Cyprinus carpio), gold fish(Carrassius auratus) and eel(Anguilla japonica). Dipping of fishes for acute toxicity was performed for a period of 48h, and the TLm value(median tolerance limit) were 4.0ppm in mirror carp and flounder, 4.4ppm in rainbow trout, and 10.0ppm in gold fish. After exposure below than TLm value for 1 month, no side effects and histopathological changes were observed. The test drug were shown high antibacterial activities against most bacterial pathogens by exposure in the laboratory and field trials. However, Aeromonas salmonicida showed a resistance to the dosage. We can prevent outbreaks of bacterial diseases of fish by the dosage of 1ppm for 6hr and the test drug is thought to be used effectively and widely as a disinfectant against most bacterial fish pathogens.

  • The control mechanism of gonadotropin-releasing hormone (GnRH) on gonadotropin (GTH) release was studied using cultured pituitary cell or cultured whole pituitary obtained from Testosterone (T) treated and control immature rainbow trout. The release of FSH was not changed by salmon type GnRH (sGnRH), chiken-II type (cGnRH-II), GnRH analogue ([des-$Gly^{10}D-Ala^6$] GnRH ethylamide) and GnRH antagonist ([Ac-3, 4-dehydro-$Pro^1$, D-p-F-$Phe^2$, D-$Trp^{3,6}$] GnRH) in cultured pituitary cells of T-treated and control fish. Indeed, FSH release was not also altered by sGnRH in cultured whole pituitary. All tested drugs had no effect on the release of LH in both culture systems of control fish. The levels of LH, in contrast, such as the pituitary content, basal release and responsiveness to GnRH were increased by T administration in both culture systems. In addition, the release of LH in response to sGnRH or cGnRH-II induced in a dose-dependent manner from cultured pituitary cells of T-treated fish, but which is not significantly different between in both GnRH at the concentration examined. Indeed, LH release was also increased by sGnRH in cultured whole pituitary of T-treated fish. GnRH antagonist suppressed the release of LH by sGnRH ($10^{-8}\;M$) and GnRH analogue ($10^{-8}\;M$) stimulation in a dose-dependent manner from cultured pituitary cells of T-treated fish, and which were totally inhibited by $10^{-7}\;M$ GnRH antagonist. These results indicate that the sensitivity of pituitary cells to GnRH is elevated probably through the T treatment, and that GnRH is involved in the regulation of LH release. GnRH-stimulated LH release is inhibited by GnRH antagonist in a dose-dependent manner. The effects of gonadal steroids on FSH levels are less clear.

  • The purpose of this study is to analyze the nutrition composition and physiological changes, and to evaluate the food quality of live fish in cultured and wild fishes which have been kept in an aquarium tank. The moisture and lipid content of wild and cultured fishes when kept in an aquarium tank for seven-days storage was found to be lower than those of the initial stage storage(zero day). The breaking strength was also rapidly decreased in all of live fishes tested in this study as the periods of storage extended. The protein content did not differ significantly. However, the content of cortisol, which is a indicator indicating a stress reaction in tissues, was apt to increasing as the periods of storage extended. The cortisol content of wild fishes were higher than those of cultured fishes. On the other hand, the activity of activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST), which is also a indicator indicating a stress reaction, were not changed in the serum of wild and cultured fish, suggesting the ALT and AST activity dose not directly related with a healthy loss originated from stress. The death ratio of wild fishes were higher than cultured ones due to limited activity and stress during the storage in a aquarium tank.

  • This study surveyed for the prevalence of parasites, bacteria and viruses in four fish species, olive flounder (Paralichthys olivaceus), red sea bream (Pagrus major), black sea bream (Acathopagrus schlegeli) and black rockfish (Sebastes schlegeli) in 2010. The survey was aimed to compare the pathogens detected from wild and cultured fish for an epidemiological study. Anisakis sp. was predominantly detected from wild olive flounder and red sea bream (58.6% and 41.7% respectively), but not from the cultured fishes, suggesting anisakid infection is rare in cultured fish. The wild fish get in contact with the anisakids through their prey such as small fishes or crustaceans which carry the anisakids; whereas the cultured fish are fed with formulated feed, free of anisakids. Bacterial detection rates from the wild fishes examined in the study were lower than those of cultured fishes. Vibrio sp. dominated among detected bacterial population in cultured olive flounder (18%). Since vibriosis is known as a secondary infection caused by other stressful factors such as parasitic infections, handling and chemical treatment, it seems that cultured olive flounder are exposed to stressful environment. Viruses diagnosed in the study showed difference in distribution between wild and cultured fishes; hirame rhabdovirus (HRV) (0.1%) and lymphocystis disease virus (LCDV) (3.9%) were detected in the cultured olive flounder, but not in the wild fish, and marine birnavirus (MBV) (1.7%) and red sea bream iridovirus (RSIV) (3.2%) were detected from the wild and cultured red sea bream, respectively. From the survey conducted, it can be concluded that even though some pathogens (Trichodina sp., Microcotyle sp., etc.) are detected from both the wild and cultured fish, pathogens such as Anisakis sp., Vibrio sp. and LCDV showed difference in distribution in the wild and cultured host of same fish species and this can be attributed to their environmental condition and feeding.
