Optimization of Culture Conditions for Production of Pneumococcal Capsular Polysaccharide Type I

  • Streptoccus Pneumoniae (pneumococcus), the most common cause of bacterial pneumonia, has an ample polysaccharide (PS) capsule that is highly antigenic and is the source of PS vaccine. This investigation was undertaken to optimize the culture conditions for the production of capsulard PS by type 1 pneumococcus. Among several culture media, brain heart infusion (BHI) and Casitone based media were found to support luxuriant growth of pneumococcus type 1 at the same level. Because BHI medium is rather expensive and more complex than the Casitone based media, the Casitone based media was uwed to study optimization of the culture condition. The phase of growth which accomodated maximum PS production was logarithmic phase. Concentrations of glucose greater than 0.2% did not ehnahce growth or PS production. Substitution of netrogen sources with other resources or supplementation of various concentrations of metal ion (with the exception of calcium ion) had adverse affects on growth and PS production. On the other hand, low level aeration was beneficial for increased PS production. Addition of 3 mg/1 concentration of methionine, phenylalanine, and threonine were found to enhance growth and PS production. The synerigistic effect of all the favorable conditions observed in pneumococcal growth assays provided a two-fold cummulative increase in capsular PS production.

  • Aspergillus luchuensis is known as an industrially important fungal species used for making fermented foods such as awamori and shochu in Japan, makgeolli and Meju in Korea, and Pu-erh tea in China. Nonetheless, this species has not yet been widely studied regarding mating-type genes. In this study, we examined the MAT1-1 and MAT1-2 gene ratio in black koji molds (A. luchuensis, Aspergillus niger, and Aspergillus tubingensis) and in Aspergillus welwitschiae isolated from Meju, a fermented soybean starting material for traditional soy sauce and soybean paste in Korea. The number of strains with the MAT1-1 locus was 2 of 23 (A. luchuensis), 6 of 13 (A. tubingensis), 21 of 28 (A. niger), and 5 of 10 (A. welwitschiae). Fungal species A. tubingensis and A. welwitschiae showed a 1 : 1 ratio of MAT1-1 and MAT1-2 mating-type loci. In contrast, A. luchuensis revealed predominance of MAT1-2 (91.3%) and A. niger of MAT1-1 (75%). We isolated and identified 2 A. luchuensis MAT1-1 strains from Meju, although all strains for making shochu in Japan are of the MAT1-2 type. These strains may be a good resource for breeding of A. luchuensis to be used in the Asian fermented-food industry.

  • The purpose of this study was to examine the effects of VM components(facade color and show window type) on store image and purchasing intention of fashion stores. The subjects were 428 male and female university students in Daejeon and Chungnam province. The measuring instruments were 6 stimuli manipulated by $3{\times}2$ factorial design of facade color and show window type, and a self-administrated questionnaire consisted of store image, purchasing intention, and subjects' demographic characteristics. The results were as follows. First, facade color mainly affected store image and purchasing intention. White color was preferred as more positive images(pleasurable, comfortable, neat, and modern), and showed higher purchasing intention than brown color. Second, show window type affected some store images. Semi-open type was perceived as more pleasurable image than enclosed type, whereas enclosed type was perceived as more luxurious, neat, and attractive image than semi-open type. However, show window type didn't affect purchasing intention. Third, subject' sex affected store image and purchasing intention. Male subjects perceived more positively on store image and had higher purchasing intention than females. This study suggested that facade color and show window type are important VM components affecting store image and purchasing intention.

딸기 전조재배 온실의 인공광 설치실태 및 조도분포 조사분석 (Current Status of Lighting System and Illumination Distribution in the Greenhouse for Light Culture of Strawberries)

  • Motivated by the need for developing the more efficient lighting system for light culture of strawberries in the greenhouse, this paper aims at acquiring and suggesting more concrete and scientific foundation of illuminating position, numbers of light source by investigating the types of lighting system and illumination distribution in the greenhouse for light culture of strawberries. The results of investigation and experiment are summarized as following below: 1. The types of lighting system used in the greenhouse producing strawberries were classified as 1 line and 2 lines lighting system. 2. As for the arranging types of artificial light, 2 lines lighting system, were classified as Z-type, N-type and W-type. (Refer Fig. 3) 3. The results of illumination distribution for Z-type, N-type and W-type of 2 lines illuminating system in the greenhouse with a small size tunnel measured at the height of 1.5m from the ground with 220V, 100W lamp in 6m light gap showed that maximum illuminance are 961x, 1211x, 1251x, minimum illuminance are 4.41x, 4.71x, average illuminance are 33.71x, 43.11x, 44.51x and standard deviations are 28.31x, 35.41x, 38.31x at each types. 4. Proportion of the area below optimal illuminance to floor area at the two lines illuminating system of Z-, N-, and W-type in greenhouse were appeared as 39.4%, 26.0% and 26.3%, respectively. Also proportion of the area over optimal illuminance to floor area at the two lines illuminating system of Z-, N-, W-type in greenhouse were appeared as 16.8%, 14% and 14.7%, respectively. Thus N-type was superior to the others from the view points of optimal illumination distribution and energy saving.

융합기업의 기업문화형태와 성과창출과의 영향 관계 분석 (Analysis of the impact relationship between business culture's type and creating performance of convergence corporate)

  • 본 연구는 융합기업을 대상으로 하여 기업문화형태와 성과간의 관계를 실증적으로 분석하였다. 또한 본 연구를 수행하기 위해 기업문화와 형태를 종합적으로 분석하여 본 연구에 맞게 모형을 개발하였다. 기업문화의 중요성은 일상생활에서 조직적 문화가 경쟁력의 원천이 된다는 인식하에서 조직의 효과를 증진시키는 중요한 요인이 된다고 보기 때문이다. 즉 조직문화를 바탕으로 개인의 태도와 행동을 설명할 수 있을 뿐만 아니라, 조직문화는 조직행동의 미시적 거시적 분석사이를 개념적으로 연결시켜 주는 역할을 하며 이를 통하여 성과창출에 기여한다고 볼 수 있다. 따라서 연구모형을 토대로 하여 기업문화형태와 성과간의 영향관계를 분석, 검증하였다. 연구결과 융합기업의 기업문화형태는 직무만족과 조직몰입에 영향을 미치는 것으로 나타났다. 구체적으로 기업문화형태와 직무만족간의 영향관계에서는 친화적, 진취적 기업문화가 영향을 미쳤고, 조직몰입과의 영향관계에서는 진취적 기업문화가 영향을 미쳤다. 본 연구의 결과는 향후 융합기업의 기업문화 관리를 통한 기업의 성과창출의 기초자료로서 유용한 지침이 될 수 있을 것으로 기대하고 있다.

  • 현대는 어느 시대보다도 문화에 대한 관심이 높아지고 있으며, 이러한 시대의 조류에 맞추어 자국의 정체성을 찾으려는 노력이 여러 분야에서 다양하게 나타나고, 타문화와 우리의 문화를 비교하는 연구도 활발하게 이루어지고 있다. 한 지역의 문화는 독립적으로 자연발생된 것이 아니라 인접 국가나 문화와 관련을 가지면서 변화, 발전하는 것이므로, 문화적 특성을 제대로 파악하기 위해서는 자국의 문화에 대한 깊이 있는 연구와 더불어, 타문화와의 상호비교연구를 통해서 각각의 문화의 유사점과 차이점을 분명히 이해하는 것이 필요하다고 할 수 있다. 특히, 한국 중국 일본은 예로부터 사회문화적으로 활발한 교류를 통해 서로 밀접한 영향을 주고받으면서도 독자적인 문화적 특징을 가지고 있으나, 아시아권 중에서 한국 문화의 차별화된 정체성이 제대로 인식되지 못하고 있는 것은 안타까운 현실이라 할 수 있다. 본 연구에서는 $14{\sim}19C$ 한국 중국 일본의 전통가구의 유형을 분류하고, 가구의 유형별 특성과 조형적 특성을 비교분석하고자 한다. 이를 통해 보다 객관적으로 우리의 문화를 이해할 수 있을 것이며, 동양적인 것과 한국적인 것에 대한 혼돈으로부터 보다 명확한 우리문화의 재해석을 가능하게 하는 기반이 될 것이라 기대한다.

  • The dress culture of Korea and Turkey should be necessary to study in view of a similar culture of Korea and Turkey due to similarity of oriental culture. Thus, as two nation's dress is investigated, cultural characteristics and dress form of two nations are analyzed with cultural background in detail. Also, comprehension is determinated for the dress color and texture as well as dress culture. In this paper, It is investigated theoretically for the function, type, and class of dress. Also, The form of Korean Po are compared to background of Turkish dress and Sulltans' dress form. To unify a cultural propagation and dress form, technical, inductive contents analytical method as well as literature review are used in positivism. Subject is limited to the Korean Po and Sultans' dress of Turkey. As a results, as the nation's dress reflect to culture and society of the nation, Korean dress have some similarity and differences comparing Turkish dress as followings. In dress form of two nation, rectangular Gil's shape was very similar. Mu's shape was similar too. However, Sultans' dress had outside shape of caftan type and Po had full or partial wrinkles by transforming Mu. Also, Sultans' dress did not have Sup. Korean Po have side slits or back side slits and Jun-Sam but Sultans' dress had only some side slits.

유튜브 플랫폼을 통한 외식 문화 수용에 대한 소비자 주관성 연구 (A Study on the Subjectivity of Consumers to Accept the Culture of Eating out through the YouTube Platform)

  • 본 연구는 현재 많이 사용하고 있는 플랫폼 중 하나인 유튜브를 통한 외식문화 수용이 소비자 인식에 관한 주관성 연구를 가지고 유튜브 플랫폼을 사용하는 소비자 입장에서 어떤 효과가 있는지를 파악하고 유튜브 플랫폼을 통한 외식 문화 수용으로 인하여 얻을 수 있는 마케팅 효과를 설립하기 위하여 소비자 주관적 관점을 파악하고 분석하고자 Q-방법론을 선택하였고 탐색적 연구를 진행하였다. 이를 위해 진술문카드를 응답자가 분류하는 방식으로 분석 작업을 행하였고 Q모집단을 선정하여 구성한 후 이를 통해 진술문을 작성하여 P샘플을 선정한 후 분류작업을 통해 얻어지는 Q-sort를 PC QUANL 프로그램을 사용하여 Q요인분석을 통해 분석 실행하였다. 분석결과 총 4가지의 유형으로 분류 되었으며 제1유형[(N=6): 선호음식과 대리만족형], 제2유형[(N=3): 비감성 선호음식형], 제3유형[(N=14): 정보획득 선호음식형], 제4유형[(N=2): 부정형] 등으로 각 유형마다 각각의 특징이 있는 것으로 나타났다. 이러한 분석을 통하여 나온 각각의 주관적 의견은 추후 다양한 연구에 도움이 될 것이며 향후 유튜브 관련 마케팅 및 소비자 외식 문화 수용에 참고자료로 활용될 수 있을 것이다.
