고추의 비가림재배 시 온도와 토양수분 환경이 생육 및 수량에 미치는 영향 (Influence of Air Temperature and Soil Moisture Conditions on the Growth and Yield of Hot Pepper under a Plastic Tunnel Culture)

  • This study was conducted to determine the effects of high temperature and deficit irrigation on growth and yield of hot pepper. Hot pepper was subjected to four irrigation treatments: fully irrigation (FI), 10, 20, and 30 days deficit irrigation (DI) combination with high temperature treatment. Control plants were grown natural environment and conventional culture methods. The plant height treated with high temperature was significantly higher than that of control plant. At FI combination with high temperature treatment, growth parameters such as stem diameter, leaf area, fresh and dry weight were the greatest. The yield was the greatest (2,036 kg/10a) under control, DI combination with high temperature treatment decreased by approximately 42% compare with FI combination with high temperature treatment. The number of abnormal fruits was approximately 38/plant under control, which was the smallest and that of 30 days DI combination with high temperature was higher 3.3 times compare with control. Flower abscission and calcium deficiency induced by DI treatments, especially those physiological disorder promoted by increasing DI treatments period. Results indicated that yield of hot pepper reduced by DI treatments, these results suggest that the growers should irrigate to proper soil moisture for preventing reduction of total fruit yield.

  • Alkaline bacterium producing a high pro-tease activity at low temperature was isolated by using enrichment culture from soil samples and identified as Bacil-lus sp. WRD-1 Cell growth was maximal at 10 hours and the optimal initial pH and culture time of culture condition for enzyme production was pH 7 and 10 hours, respectively. Temperature range of high enzyme activity were $10~40^{\circ}C$. The optimal pH and temperature for the enzyme activity were pH9 and $30^{\circ}C$.

섬유소분해균의 분리 및 그의 생리학적 특성 (Isolation of Cellulolytic Microorganisms and their Physiological Characteristics)

  • Celluloytic microorgnasims were isolated form the various sources and four of them were identified as Trichoderma roningi, Aspergillus niger, Penicillium chrysogenum, and Streptomyces sp. The induction of extracellular5 cellulase of these species in the liquid culture media containing carboxymethylcellulose (CMC) or Avicel as inducer showed that CMC was a better effective inducer for the production of CMCase(Cx cellulase component) as well as Avicelase(C$_{1}$ cellulase component) than Avicel. It is believed that certain hydrolysis products of cellulose(CMC) could serve as an inducer for an enzyme synthesis. In T. roningi, Asp. niger, and Strptomyces sp., the optimum temperature of CNCase on CMC-culture medium was 50.deg. but temperature around 40.deg.C was found to be optimum for the activities of CMCase prepared from P.ehrysogenum. The optimum temperature for Avicelase activitiles on Avicel-culture media of T. roningi and P. chrysogenum was $40{\circ}C$ whereas temperature $50{\circ}C$ was found to be optimum for Avicelase from A.niger and Streptomyces sp. The optimal activities of these CNCase and Avicelase prepared from. T. ronigi, Pen.chrysogenum and Streptomyces sp. were found similarly to be at pH's around 5.4 and 6.0 while pH 4.8 was optimum for the activities of CMCase and Avicelase from A.niger, indicating that A.niger in acidic media would yield an enzyme of high activity.

  • Honey is one of the most significant ingredients in conventional food production in different regions of the world. Honey is commonly used as an ingredient in ethnic food. Beekeeping is performed in various locations as part of the local food culture and an occupation related to pollinator production. It is important to conduct beekeeping so that it generates food culture and helps regulate the regional environment in an integrated manner in preserving and improving local food culture. This study analyzes different types of environmental factors of a smart bee farm. The major goal of this study is to determine the best prediction model between the linear regression model (LM) and the support vector regression model (SVR) based on the environmental factors of a smart bee farm. The performance of prediction models is measured by R2 value, root mean squared error (RMSE), and mean absolute error (MAE). From all analysis reports, the best prediction model is the support vector regression model (SVR) with a low coefficient of variation, and the R2 values for Farm inside temperature, bee box inside temperature, and Farm inside humidity are 0.97, 0.96, and 0.44.

  • 농산 폐자원 중에서도 감귤 가공공정에 따라 부생되는 감귤과피를 발효 기질로서의 이용을 목적으로 분리균 Asp. niger를 사용하여 구연산 발효에 미치는 배양조건 및 영향인자를 조사하였다. 그 결과 고 체 배양에서의 최적조건 즉 감귤과피와 물을 동량 혼합하고 여기에 NH$_4$NO$_3$0.2%, MgSO$_4$7$H_2O$ 0.1%, Methanol 2.5%를 첨가하여 PH4.5로 조절한 다음포자 현탁액(1x$10^8$ spore/$m\ell$) 2%를 접종하여 균체증식 초기에 35$^{\circ}C$, 20시간 배양하고 4시간 저온 처리한 다음 구연산 생성기에서 3$0^{\circ}C$로 48시간 배양했을 때 당에 대해 80.4%의 높은 수율의 구연산을 얻었다.

Design and Performance of an Automated Bioreactor for Cell Culture Experiments in a Microgravity Environment

  • In this paper, we describe the development of a bioreactor for a cell-culture experiment on the International Space Station (ISS). The bioreactor is an experimental device for culturing mouse muscle cells in a microgravity environment. The purpose of the experiment was to assess the impact of microgravity on the muscles to address the possibility of long-term human residence in space. After investigation of previously developed bioreactors, and analysis of the requirements for microgravity cell culture experiments, a bioreactor design is herein proposed that is able to automatically culture 32 samples simultaneously. This reactor design is capable of automatic control of temperature, humidity, and culture-medium injection rate; and satisfies the interface requirements of the ISS. Since bioreactors are vulnerable to cell contamination, the medium-circulation modules were designed to be a completely replaceable, in order to reuse the bioreactor after each experiment. The bioreactor control system is designed to circulate culture media to 32 culture chambers at a maximum speed of 1 ml/min, to maintain the temperature of the reactor at $36{\pm}1^{\circ}C$, and to keep the relative humidity of the reactor above 70%. Because bubbles in the culture media negatively affect cell culture, a de-bubbler unit was provided to eliminate such bubbles. A working model of the reactor was built according to the new design, to verify its performance, and was used to perform a cell culture experiment that confirmed the feasibility of this device.

Evaluation of Thermal Comfort on Protective Clothing Worn in an Radiation Power Plant

  • The purpose of this research was to device thermal comfort model for radiation power plant workers in protective clothing. Three fabrics commonly used in protective workwear were made into coveralls of identical design and were evaluated by adult healthy males in four simulated work environment. It was investigated between the physiological response and subjects comfort according to environmental variance and clothing types. The of simulated work enviro mensent was controlled under four different humidity and temperature of each type. (Temperature 20±1℃, RH 40∼70%±5%, Temperature 30±1℃, RH 40∼70%±5%) An index of physiological response was connected with the thermal comfort designed. Mean skin temperature, skim temperature, Axillasy temperature ear canal temperature, clothing climate, total sweat, blood pressure, and R-R interval were be evaluated. Skin temperature difference ocurring during exercise and rest were significant only with respect to time and regions of the body, This despite physical differences in the three coveralls, particulary mass statistically experiment. Also, an index of subject wearing sensation was designed for thermal comfort after investigation determined the kind of clothes and the type of environment. As a result of this research, two types of multiple regressions was deviced to estimate thermal comfort of the protective clothing.

담금액 온도에 따른 히카마 피클의 저장 중 품질변화 (Quality Changes in Jicama (Pachyrhizus erosus) Pickle Due to Variations in Soaking Solution Temperature During Storage)

  • This study was carried out to determine the optimum temperature of the jicama pickle solution by evaluating physicochemical and sensory characteristics at various temperatures. The soaking solution of the jicama pickle was prepared at different temperatures (95, 75, 50, and 25℃). During storage, the titratable acidity, Brix, and the salinity of the jicama pickles increased, and these values increased with increasing temperature of the soaking solution. The higher the temperature of the soaking solution, the lower the L and b values. In all samples, the hardness increased rapidly until the 5th day of storage, and then gradually decreased. During storage, the jicama pickles showed a high tendency to harden at soaking solution temperatures of 75 and 95℃. Sensory evaluation indicated that the taste, flavor, crispness, and overall preference of the jicama pickle were higher at a soaking solution temperature of 75℃ and above. The higher the temperature of the soaking solution, the higher the acceptability. Based on these results, we conclude that soaking solution temperature of 75°¦95℃ in the manufacture of jicama pickles had a positive effect on the reduction in tissue softening and the sensory properties of the jicama pickle.

Effect of Thermal Discharge from Semiconductor Factory into Stream on Freshwater Fish

  • The study was conducted in Manu-stream, located in Paju, Gyeonggi-do, from January 2021 to December 2021. The survey points were selected in the upper and lower streams based on where thermal discharged to investigate water temperature and fish species and biological community analysis and growth rate were analyzed. The average annual water temperature difference between the upper and lower stream is about 11.0℃, and in the case of the lower stream area, the water temperature is maintained at 20.0℃ or more per year. Fish that appeared during the survey period decreased lower stream compared to the upper stream, which is believed to be the result of a decrease in temperature-sensitive species as the simple riverbed structure and water temperature increased compared to the upper stream. As a result of biological community analysis, it showed a relatively stable community state at the upper stream. The growth rate of fish has a high regression coefficient b value in lower streams throughout the four seasons. It showed relatively good growth lower stream, with a high water temperature. However, the results of each survey point are similar from season to season. The indicator species is a resistant intermediate species, and the range of resistance to water temperature is wide, so it is judged that water temperature's effect on the indicator species' growth is low.

가수분해방법에 의한 식물세포배양여액으로부터 Paclitaxel 수율증가 (method of Using Hydrolysis to Increase Paclitaxel Yield from plant Cell Culture)

  • This work is method that uses a hydrolysis for increasing yield of paclitaxel in plant cell cultures. The best pH is 3.0 to obtain a maximum yield at fixed reaction temperature and time t pH 3.0 reaction temperature 80$^{\circ}C$ and reaction time 8 hr give the highest yield which is three time of control. This is very simple and efficient method to increase paclitaxel yield in plant cell cultures.
