Optimum Conditions for the Formation of Ammonia as a Precursor of Tetramethylpyrazine by Lactococcus lactis ssp. lactis biovar. diacetilactis FC1

  • To investigate the optimum conditions for the production of ammonia as a precursor of tetramethylpyrazine flavor compound from arginine by Lactococcus lactis ssp. lactis biovar. diacetilactis FC1, fermentation factors such as initial pH of culture media, fermentation temperature, concentration of arginine-HC1, and sugars were examined. The optimum conditions were initial pH 5.5 of the culture media, fermentation temperature of $34^{\circ}C$, 6% (w/v) of arginine-HC1, and 1% (w/v) of galactose as a carbon source. Under the optimum fermentation conditions, 40 mmole/l of ammonia was produced after 40 h.

Lead Tolerance Profile of Pseudomonas Stuzeri in Liquid Culture

  • Pseudomonas stutzeri strain KCCM 34719 was used in this experiment to determine the effects of increasing Pb(II) concentrations on its growth rate. To obtain optimum growth conditions, strain KCCM 34719 was cultivated in nutrient broth under various conditions, such as temperature, pH, and NaCl concentration. Optimal conditions for cell growth were $30^{\circ}C$ of temperature, 8.0 of pH, and 3% of NaCl concentration, respectively. Growth response of bacterial cell to Pb(II) showed tolerance to concentrations ranging from 10 to 100 mg ${\ell}^{-1}$ in liquid culture, following a growth pattern similar to the control. Growth rate was greatly inhibited at 200 mg ${\ell}^{-1}$ of Pb(II).

넙치(Paralichthys olivaceus) 장관의 배양 및 비배양 방법에 의한 세균의 다양성 (Diversity of Cultured and Uncultured Bacteria in the Gut of Olive Flounder Paralichthys olivaceus)

  • We determined the optimal culture conditions for obtaining the maximum number of intestinal bacteria from the olive flounder Paralichthys olivaceus, and studied bacterial diversity using both culture-dependent and culture-independent methods. Using six culture conditions, mean bacterial numbers were greater than $10^6$ per gram of gut mucus, regardless of the medium. However, the bacterial diversity, based on colony morphology, appeared much higher on Marine agar (MA) and Zobell 2216 agar than on other media. We found eight and 17 cultured bacterial phylotypes with 99% minimum similarity in gut mucus grown on MA and tryptic soy agar, respectively. Furthermore, we used genomic DNA extracted from gut mucus to generate 78 random clones, which were grouped into 25 phylotypes. Of these, six were affiliated with Firmicutes, Actinobacteria, and Verrucomicrobia, and were not found using our culture-dependent methods. Consequently, we believe that Marine agar and Zobell 2216 agar are optimal media for culturing diverse intestinal microbes; we also discovered several novel sequences not previously recognized as part of the gut microbiota of olive flounder.

  • 회분배양에서 교반속도와 통기량에 대한 영향을 살펴본 결과 400 rpm과 1.0 vvm의 조건에서 균체량은 25.1 g/L, 다당체는 2.5 g/L로 가장 높게 생성되었다. 회분배양을 기초로 균체량과 다당체 생성을 높이기 위해 glucose를 대수기에 공급한 결과 배양 6일째 균체량은 29,2 g/L, 다당체 생성은 3.3 g/L로 회분배양보다 우수한 결과를 보였다.

  • In hypoxic tissue conditions, pyruvate can not enter the Krebs cycle and lactic acid, produced from pyruvate, accumulates to induce lactic acidosis. Pyruvate, However, can also be converted to alanine by glutamate pyruvate transaminase, that could be enhanced by glutamate. Therefore, it would be a fundamental measure to treat the lactic acidosis in tissue hypoxic conditions when one can convert the accumulated lactic acid, through pyruvate, to alanine. To test the above hypothesis, we induced a lactic acidosis in cats and the effect of glutamate on recovery of acid base state and removal of the lactic acid from blood were assessed and the results were compared with those of bicarbonate administration, which is one of the most frequently used conventional measure for correction of the acid base state during lactic acidosis. The results were that glutamate and combined glutamate bicarbonate solutions not only restored the acid base status completely from the lactic acidosis in an hour or two, but also restored the blood level of lactate partially. We concluded that administration of glutamate solution to convert pyruvate into alanine is effective in preventing lactic acid accumulation and treating lactic acidosis.

  • The mass cultivation of Ecklonia cava Kjellman was studied as a potential biomass source for the extract industry in Korea. Experiments were conducted to investigate the optimal conditions for artificial seed production and mass cultivation of this species. Maximum growth and young thalli development in the nursery culture area occurred at 2 m depth, whereas maximum growth of thalli in the main culture area occurred at 1 m depth. Production of E. cava was between 2.6 and 3.6 kg wet wt. $m^{-1}$ after depth control and removal of fouling organism, etc. The relationship between optimal water depth for culture and underwater irradiance during the E. cava cultivation was calculated as: y = -0.718x + 8.042 ($r^2$=0.976). The growth rates achieved in this trial indicate that E. cava cultures could produce and supply sufficient biomass.

  • The FimH subunit of type 1-fimbriated Escherichia coli has been determined as a major cause for urinary tract infections. In our previous study, the Adhesin/CTXA2B was expressed as soluble recombinant chimaeric protein derived from the uropathogenic Escherichia coli adhesin genetically coupled to cholera toxin A2B (CTXA2B) subunit in Escherichia coli. Since it is very important to optimize IPTG concentration and culture temperature to maximize cell growth and productivity, These optimal culture factors were determined to increase the productivity of the expressed Adhesin/CTXA2B chimaeric protein in Escherichia coli TB1 carrying pMALfimH/ctxa2b. Our data demonstrate that optimal concentration of IPTG for increased production of chimaeric protein was 0.5 mM. Additionally, culture time was 10 hours and temperature, 37${\circ}C$.

Control Effects of an Antibiotic Produced by Streptomyces sp. B25 on Tobacco Mosaic Virus and Determination of Its Molecular Structure

  • The culture filtrate of Streptomyces sp. B25, which was identified in this experiment, was tested for the control of tobacco mosaic virus (TMV) with the susceptible tobacco cultivar, NC 82, under the field conditions following the preliminary examination of its characters for TMV control. Control efficacy of the culture filtrate against TMV infection continued over 50% up to 6 days after treatment, and its systemic effect was about 30% of the direct effect. In field conditions control efficacy of the culture filtrate against TMV infection was 95.3 % at 2 weeks after TMV inoculation, and decreased to 58.3 % at 3 weeks after inoculation. Five fold-dilution of the culture filtrate showed about half of the control efficacy by the stock culture filtrate. Analysis of the antibiotic material responsible for the inhibition of TMV infection through nuclear magnetic resonance experiments revealed that the antibiotic is antimycin $A_1$, which is firstly reported as an anti-phytoviral antibiotic in this experiment.

Observations on Some of the Mycelial Growth and Pigmentation Characteristics of Cordyceps militaris Isolates

  • Characteristic growth patterns of Cordyceps militaris isolates on various media, under varying light conditions and at varying incubation periods were examined. Light was found to be the most critical single factor in determining the density, texture, and pigmentation of the mycelial culture of the fungus. However, under the light condition, the degree of pigmentation and mycelial density were found to be affected by the incubation period and type of medium. Irrespective of the variations in medium type or incubation period, there was no pigmentation of the mycelium under dark condition. Radial growth of the mycelium was faster under dark incubation rather than under light incubation. Abundant mycelial density and darkest pigmentation of C. militaris isolates were produced in nutritionally rich media like SDAY, SMAY and CZYA, suggesting that these media may fulfill all the requirements for vegetative growth of the fungus. Growth characteristics of C. militaris isolates could be easily observed by the simple agar culture method, which would be useful to characterize the phenotypic characteristics of large number of pure cultures of the fungus under given conditions of growth factors such as medium, light and temperature.
