한국산과 중국산 박하의 항염증 효과에 관한 비교연구 (Comparative Study of the Anti-inflammatory Effects of Menthae Herba from Korea and China)

  • Menthae herba (MH) extracts exhibit anti-inflammatory effects. The purpose of this study was to determine whether the anti-inflammatory effects of MH extracts vary according to the cultivation regions. We performed a comparative analysis of MH extracts by evaluating the production of inflammatory mediators in RAW 264.7 murine macrophage cells and HaCaT human keratinocyte cells. MH extracts obtained from different cultivation regions in Korea and China significantly reduced the production of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in RAW 264.7 cells stimulated with lipopolysaccharide (LPS). No differences in these inhibitory activities were observed between MH extracts. In HaCaT cells stimulated with TNF-${\alpha}$ and interferon-${\gamma}$ (IFN-${\gamma}$), MH extracts did not inhibit the production of macrophage-derived chemokine (MDC/CCL22), but most extracts reduced the production of the regulated on activation normal T-cell expression and secreted (RANTES/CCL5). We used clustering tree analysis of the MH extracts according to the chromatographic pattern and anti-inflammatory potency of MH extracts. We observed differences in the chromatographic pattern of MH extracts but no difference in anti-inflammatory potency. Our findings suggest that MH extracts from different regions do not show any differences in their pharmacological potency in that MH extracts are used as therapeutic agents to treat inflammatory disorders.

LPWA기반의 임산물 생육환경 수집 및 빅데이터 분석 시스템 개발 (Development of LPWA-Based Farming Environment Data Collection System and Big Data Analysis System)

  • 최근 스마트 팜의 연구가 활발해지면서 시설하우스와 같은 실내 환경 제어는 높은 수준에 이르렀다. 그러나 노지에서 재배가 이루어지는 임업 분야에 ICT기술의 활용은 아직 미비한 실정이다. 본 논문에서는 ICT 기술을 적용한 LPWA 기반의 임산물 생육환경 수집 및 빅데이터 분석 시스템을 제안한다. 제안된 시스템은 oneM2M 아키텍처를 기반으로 구성하였으며 소규모 태양광 발전과 LPWA기술을 이용하여 노지에서 환경 데이터를 수집하여 서버에 전송한다. 전송된 데이터는 서버에서 빅 데이터로 구축되며 이를 활용해 임산물의 생산량과 품질을 예측한다. 제안된 시스템은 신재생 에너지와 스마트 팜의 융합을 통해 저비용, 고품질의 임산물 생산에 기여할 것으로 기대된다. 또한 노지에서 이루어지는 농작물의 생장 환경 모니터링과 oneM2M 아키텍처를 활용하는 타 산업 분야에 응용될 수 있다.

신기술 수용농가의 시범사업 타당성 및 경영성과 분석 (An analysis on the propriety of pilot project and the Management Performance in Korea's new technology adaptation Farming)

  • 본 연구는 농촌진흥청에서 기술시범사업으로 추진하고 있는 마늘주아재배 기술 사업에 대하여 농업경영체의 신기술 수용에 대한 태도, 신기술의 타당성 분석, 기술도입 전후의 경영효과를 분석하는데 그 목적이 있다. 마늘주아재배 농가들은 농업 신기술에 대한 요인들 중 기술만족도(4.30)가 가장 높았으며, 그 다음 새로운 것을 적극적으로 선호하고자 하는 혁신성(4.21) 순으로 나타났다. 반면에 신기술에 대한 위험에 대해서는 관대한 것으로 나타났다. 한편, BMO 모델을 이용하여 마늘 주아재배 기술 사업에 대한 타당성을 평가한 결과에 따르면, 사업 매력도와 적사도가 각각 44점, 45점으로 사업 성공률이 80%이상으로 높았다. 마늘주아 재배기술의 도입 전후의 경영성과 분석에서도 이 사업 참여 농가들은 신기술을 도입 한 후 종구비 절감(43%) 및 수량증가(28%) 등 전반적으로 경영이 개선된 것으로 나타났다.

Optimization of Agrobacterium tumefaciens-Mediated Transformation of Xylaria grammica EL000614, an Endolichenic Fungus Producing Grammicin

  • An endolichenic fungus Xylaria grammica EL000614 produces grammicin, a potent nematicidal pyrone derivative that can serve as a new control option for root-knot nematodes. We optimized an Agrobacterium tumefaciens-mediated transformation (ATMT) protocol for X. grammica to support genetic studies. Transformants were successfully generated after co-cultivation of homogenized young mycelia of X. grammica with A. tumefaciens strain AGL-1 carrying a binary vector that contains the bacterial hygromycin B phosphotransferase (hph) gene and the eGFP gene in T-DNA. The resulting transformants were mitotically stable, and PCR analysis showed the integratin of both genes in the genome of transformants. Expression of eGFP was confirmed via fluorescence microscopy. Southern analysis showed that 131 (78.9%) out of 166 transformants contained a single T-DNA insertion. Crucial factors for producing predominantly single T-DNA transformants include 48 h of co-cultivation, pretreatment of A. tumefaciens cells with acetosyringone before co-cultivation, and using freshly prepared mycelia. The established ATMT protocol offers an efficient tool for random insertional mutagenesis and gene transfer in studying the biology and ecology of X. grammica.

Marker compounds contents of Salvia miltiorrhiza Radix depending on the cultivation regions

  • Salvia miltiorrhiza Radix is cultivated in Korea and China and is traditionally used to treat cardiovascular diseases. In this study, we developed and validated a quantitative analysis method for S. miltiorrhiza Radix using high-performance liquid chromatography (HPLC). Identification was performed using ultra performance liquid chromatography-tandem mass spectrometry. For quantitative analysis, we used seven marker compounds. Separation conditions for HPLC were optimized using an ODS column with gradient conditions of 1% formic acid in distilled water and 1% formic acid in acetonitrile, with a flow rate of 0.8 mL/min and a detection wavelength of 280 nm. This method showed good linearity ($R^2=0.9998$), precision (relative standard deviation ${\leq}3.3%$), accuracy (recovery of 94.16-102.89%), limit of detection ($7.53{\mu}g/mL$), and limit of quantification ($23.71{\mu}g/mL$). This approach successfully quantified marker compounds in S. miltiorrhiza Radix. The individual marker compounds were identified by comparing the molecular masses and retention times with does standard compounds. Marker compound contents of S. miltiorrhiza Radix were investigated with different cultivation regions. Seven marker compounds were detected and quantified in all samples. Among them, salvianolic acid B showed the highest contents and it ranged from 4.13 to 7.15%. The salvianolic acid B content (7.15%) of marker compound was the highest in Bonghwa, and the tanshinone IIA content (1.90%) was the highest in Pohang. The results of marker compounds and developed method were intended to provide a favorable reference for the study of S. miltiorrhiza Radix from different regions of Korea.

스마트팜 채소에 대한 소비자의 지각된 자연성이 혜택과 태도 및 추가지불의도에 미치는 영향 : 저탄소 라벨의 조절효과 검증 (The Effect of Consumer Perceived Naturalness on Benefits, Attitude, and Willingness to Pay a Premium for Smart Farm Vegetables: Low Carbon Label as a Moderating Variable)

  • Purpose: Smart farming is related to the low carbon certification system as it provides many opportunities to cultivate and manage crops in an eco-friendly, thereby reducing carbon footprint. However, there is a significant lack of consumer perception research on low carbon labels for smart farms vegetables. Therefore, this study aims to investigate consumer perceptions of smart farm vegetable and low carbon labels. Methods: This study manipulated cultivation type(general vs. smart farm) and low carbon labels (yes vs. no) as experimental stimuli. Measurement questions and the research model were validated through confirmatory factor analysis and reliability analysis. Hypotheses testing were conducted using SPSS 29.0, AMOS 28.0. Results: The results of the study showed no significant difference in consumers perceived naturalness based on cultivation types, and there was also no moderating effect of the low carbon label. There was no difference between environmental benefits and health benefits according to the cultivation type. Perceived naturalness had a significant effect on both environmental and health benefits, and environmental benefits showed a higher impact relationship. These benefits positively affected attitudes and willingness to pay a premium, Environmental benefits had a higher impact on attitudes, while health benefits had a higher impact on willingness to pay a premium. Lastly, attitudes were found to have a significant impact on the willingness to pay a premium. Conclusion: This study is valuable in that it investigated consumer perceptions of smart farms and low carbon labels that have not been previously studied. It compares the environmental and health benefits, confirming their influence on attitudes and willingness to pay a premium. The results suggest a potential expansion in academic research on smart farming and low carbon labels, offering practical insights for marketing strategies and policies for relevant companies.

과채 재배농가의 경영현황 및 경영효율성 분석 : 딸기·토마토를 중심으로 (An Analysis of Economic Efficiency of Fruits and Vegetables Farms: the Case of Strawberries and Tomatoes Farms)

  • This paper analyzes economic efficiency of strawberries and tomatoes farms using the data envelopment analysis (DEA). We examine how production characteristics and farm size affect overall efficiency (OE), allocative efficiency (AE), and technological efficiency (TE). We also survey sample farm households from the Farm Income Investigation database of Rural Development Administration to investigate management and cultivation techniques and to analyze economic efficiency by technique. Implications for improving efficiency are suggested in the conclusion.

  • Plant cell culture provides a viable alternative over whole plant cultivation for the production of secondary metabolites. In order to successfully cultivate the plant cells at large scale, several engineering parameters such as, cell aggregation, mixing, aeration, and shear sensitivity are taken into account for selection of a suitable bioreactor. The media ingredients, their concentrations and the environmental factors are optimized for maximal synthesis of a desired metabolite. Increased productivity in a bioreactor can be achieved by selection of a proper cultivation strategy (batch, fed-batch, two-stage etc.), feeding of metabolic precursors and extraction of intracellular metabolites. Proper understanding and rigorous analysis of these parameters would pave the way towards the successful commercialization of plant cell bioprocesses.

Optimization of Producing Liquid Fuel from Photosynthetic Algal Growth

  • The green alga, Dunaliella salina under fed-batch cultivation produced 51.12 mg of hydrocarbon per liter with maintaining 0.313 (g dry wt/l). About 20% of hydrocarbon production yield based on dry biomass was obtained from both batch and fed-batch processes. Optimum culture conditions of light intensity, pH and salt concentration were obtained as 0.0080 (kJ/$cm^2$/h), 8.0 and 1.4 (g of NaCl/l), respectively by response surface analysis. The production of hydrocarbons in D. salina was closely correlated to cell growth. Fed-batch cultivation produced more hydrocarbons and maintained better cell growth than a batch process.

논 감수심에 영향을 주는 요인에 관한 연구 (A study of influence factors on water requirement in paddy fields)

  • The water requirement of transplanting culture(TC) consisting of 16 plots and direct seeding culture in a well-drained paddy field(DC) also having 15 plots were measured in order to clarify the influence factors on water requirement. Each plot was maintained under different cultivation conditions. Those varied cultivation conditions were classified into 7 items. Subsequently, the relationship between water requirement and 7 items were analyzed by evaluating the average water requirement in each condition to clarify the items influencing water requirement. From the analysis of the results, the water requirement of DC was determined to be 3.5 times of the TC. Furthermore, water requirement in DC and TC were influenced mainly by ponding depth and locational condition respectively.
