Effect of Sodium in Artificial substrate on the Growth, Gas Exchange and Leaf Water Status of Cucumber (Cucumis sativa L.) and Korea Melon(Cucumis melo L.) (상토에 함유된 Na함량이 오이와 참외의 생육, 광합성 및 잎의 수분상태에 미치는 영향)

  • Sodium is known to reduce a plant growth and yields. However, the relationships between physiological response of seedling and salinity stress caused by growing media are not well understood yet. We conducted experiments to investigate change of some parameters including Na, EC, moisture content in media under different air temperature ($15^{\circ}C$, $25^{\circ}C$), and the response of fruit-vegetables such as cucumber, oriental melon on saline conditions originated from horticultural substrate. Volumetric moisture content of media at $15^{\circ}C$ was 70%, but at $25^{\circ}C$ was decreased by 45% within 22 hrs, showing below optimal matric potential, approximately. During reaction time, the increase of Na concentration was significantly greater in saline substrate than in control. The decrease rate of Na concentration according to supplying irrigation water was higher in saline substrate than in control. $CO_2$ assimilation rate and transpiration rate of Korea melon grown in low temperature were decreased with a Na/cation ratio in hydroponic solution. Water saturation deficit was also increased significantly at $15^{\circ}C$ as compare to $25^{\circ}C$. Saline stress during nursery stage induced a reduction of seedling quality, growth and cucumber yield. The results suggest that the relationship between uncontrolled Na uptake of seedling from saline substrate and meteological condition is responsible for saline stress.

  • A rotting bacterium was isolated from decayed root of ginseng (Panax ginseng Meyer), cultured purely, and its pathogenicity was confirmed by reinoculation test. The strain causing ginseng root rot was identified as Pseudomonas fluoresens biotype II. The strain was somewhat different from P.marginalis and P.talaasii, considering the number of flagella, pathotype and ability of indole production. The strain did not exhibit pathogenicity to other plants tested, such as red kidney bean(Phasolus vulgaris L.), soy bean (Glycine max Merr.), cucumber (Cucumis sativus L.) lettuce (Lactuca sativa L.) and cowpea bean (Vigna sinensis Savi.).

  • Objectives: The objective of this study was to teach patients clinically useful external application of acne. Methods: We investigated the effects and manufacturing methods of the medical herbs published in the woman's monthly magazines according to "Zhong-Yao-Da-Ci-Dian(中藥大辭典)". Results: We investigated that Saururus chinensis Baill.(三白草), Houttuynia cordata Thunb.(魚腥草), Coix lachrymajobi L.(薏苡仁) were discovered 3 times, Brassica oleracea L.(甘藍), Raphanus sativus L.(래복), Daucus carota L. var. Sativa DC.(홍래복), Oryza sativa L.(米皮糠), Spinacia oleracea L.(파래), Cucumis sativus L.(黃爪) 2 times, and Glycyrrhiza uralensis Fisch.(甘草), Gallus gallus domesticus Braisson.(?子白), Aloe vera L.(노회), Phascolus radiatus L.(綠豆), Prunus persica Batsch.(桃葉), Lycopersicon esculentum Mill.(番茄), Taraxacum mongolicum Hand.-Mazz.(蒲公英), Prunus armeniaca L.(杏仁), Musa paradisiaca L. var. Sapientum O. ktze.(香蕉) and Malus pumila Mill.(평과) were discovered once in woman's monthly magazine during last two years. According to the effects, we classificated four groups which were defervescence(淸熱), subsidence of edema(消腫), pyorrhea(排膿) and gloss(滋潤) group.   • A brown leaf and stem spot disease of bayonet-grass (Scirpus maritimus) was epidemic in reclaimed paddy fields of Chunbuk province, Korea. A fungal pathogen was repeatedly isolated from the necrotic lesions of the bayonet-grass and identified as Nimbya scirpicola. The pathogen induced disease symptoms only in bayonet-grass but not in 8 other plants tested; Brassica pathogen induced disease symptoms only in bayonet-grass but not in 8 other plants tested; Brassica compestris subsp. napus var. pekinensis, Cucumis sativus, Glycine max, Hordeum vulgare, Lycopersicon esculentum, Oryza sativa, Sesamum indicum and Triticum aestivum. The fungus has potential to be developed as a mycoherbicide.

Economic Threshold of Meloidogyne incognita for Greenhouse Grown Cucumber in Korea (오이 시설재배시 고구마뿌리혹선충의 경제적피해한계)

  • To determine the economic threshold level of Meloidogyne incognita on cucumber in greenhouse conditions, cucumber seedlings(Cucumis sativa L. cv. 'Super Manchon') grafted on 'Jangsushintozoa'(Cucurbita maxima x Cu. moschata) were planted in wooden boxes($30{\times}40{\times}15cm,\;L{\times}W{\times}D$) under a plastic house in August 01 and harvested from Sep. 01-Oct. 30, 2006. The initial nematode population densities(Pi) in the wooden boxes were adjusted to 0, 10, 30, 100, 300, and 600 second-stage juveniles(J2)/100 $cm^3$ soil. The relationship of total fruit yield to Pi level could be adequately described by a linear regression equation, $Y=0.82-0.04{\cdot}Log_{10}$(Pi+1). Initial nematode densities(Pi) before planting in excess of 5 J2/1,000 $cm^3$ soil caused in total yields loss that is equivalent to the costs of granular nematicide application; Pi level in excess of 25 J2/1,000 $cm^3$ soil caused in total yields loss that exceed the costs for application of fumigants at current control costs. We propose two different management strategies depending on nematode densities, (i) application of non-fumigant granular nematicides for M. incognita Pi level of 5 J2/1,000 $cm^3$ soil and (ii) fumigant treatment with Pi level over 25 J2/1,000 $cm^3$ soil. Soil samples to determine population density of Meloidogyne spp. for advisory purposes should be taken several months before planting time, which allows a period of time enough for implementing management procedures.

Effect of Plant-growth-promoting Bacteria Inoculation on the Growth and Yield of Cucumber(Cucumis sativa L.) (식물생육촉진 세균이 오이 생육 및 수량에 미치는 영향)

  • We studied the effect of inoculation of microorganisms known to produce plant growth promoting substances, on the growth and yield of cucumber(Cucumis sativa L.), through a field experiment. The microorganisms used were isolated from the forest soil and consisted of Micrococus sp., Baccilus sustilis, Enterobacter agglomerans, Baccilus megaterium, Pseudomonas putida, Cellulomonas sp. and Staphylococus xyposus. Fotr the multiplication, microorganisms were cultured in liquid media of Pseudomonas P and Sabouraud dextrose. Inoculation of microorganisms was done by spraying the culture media after the culture of them to soil and cucumber plants, three times during the growth of cucumber at the rate of 10l/ha. The inoculation of microorganisms tended to promote the growth of cucumber plant and increase the yield of it. No sign of significant improvement of soil chemical and physical properties were observed after the harvest of crop. The population of bacteria and actinomycetes tended to be higher in the inoculated plots than in not inoculated plots, while opposite was the case in the population of fungi.

Thermostability of Superoxide Dismutase from Cucumber(Cucumis sativa) (오이 추출물에 존재하는 Superoxide Dismutase의 열안정성)

  • The superoxide dismutase(SOD) in peeled pericarp of cucumber was most stable at pH 8.0 and relatively stabe between pH 5.0 and 9.0. The enzyme was stable up to 6$0^{\circ}C$ and retained 12% by heat treatment at 10$0^{\circ}C$ for 5 min. At pH 2.0, the peeled pericarp enzyme activity was decreased to 10% by incubation for 3 hrs. However, the enzyme activity was increased above 25% after incubating the enzyme at pH 7.0 for 6 hrs. Retention of SOD activity in cucumber by various heating methods was also measured. The residual SOD activities of peeled pericarp and whole cucumber was estimated to be 25% and 27% after blanching(2 min), respectively. The skin enzyme retained 53% of its activity after steaming (3 min). When the peeled pericarp enzyme was incubated at 4$^{\circ}C$ for 20 days, the enzyme activity remained about 81%. However, when the enzyme incubated at 3$0^{\circ}C$ for 20 days, the peeled pericarp enzyme activity decreased to 17% of its original activity. The enzyme activity of peeled pericarp cucumber was not changed after exhaustive dialysis for 3 days, which indicated that the SOD activity in cucumber seems to have molecular weight above 12,000.

  • The effects of seawater on growth of lettuce(Lactuca sativa L.), leaf perilla(Perilla frutescens var. japonica Hara), red pepper(Capsicum annuum L.) and cucumber(Cucumis sativus L.) seedlings were investigated in the glass greenhouse. These effects were studied on seedlings, and diluted seawater (1%, 5%, 10%, 20%, 50%, 100% v/v) was sprayed enough on leaves. The tested four vegetable crops have well grown up to 10% diluted seawater, but the tested vegetable crops were damaged from increasing salt levels. Of these, lettuce was provided salt-tolerant vegetable crop and red pepper was considered salt-sensitive vegetable crop. The salt tolerance of vegetable crops is different between crops and complicated because of additional detrimental effects caused by accumulated ions or specific ion toxicities in their leaves. These results show that agricultural use of seawater may be benefit crop cultivation in organic farming system as well as in conventional farming system.

Allelopathic Potential and Antioxidant Activity of Leaf Extracts from Several Wild Plant Species

  • Several wild plant species are known to contain biologically active substances that are allelopathic to weed species as well as antioxidant to foods. Plant extracts or residues from leaves of 4 species, Achyranthes japonica (speedwell), Cucumis sativus (Cucumber), Trifolium repens (white clover), and Vicia angustifolia (narrowleaf vetch) were bioassayed against Medicago sativa (alfalfa) or Echinochloa crus-galli (barnyard grass) to determine their allelopathic effects, and used for measurement of antioxidant activities. The aqueous extracts applied on filter paper significantly inhibited root growth of alfalfa. Aqueous extracts or residues from V. angustifolia showed the most inhibitory effect on alfalfa or barnyard grass seedling growth and followed by A. japonica and T. repens. Oxidative stability by Rancimat method, antioxidant activity by TBA (2-thiobarbituric acid) method and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity for the ground samples or methanol extracts were the greatest in V. angustifolia, although were less than those of commonly used antioxidants, BHT (butylated hydroxytoluene) and ascorbic acid. These results suggest that the wild plant species had potent allelopathic and antioxidant activities, and that their activities differed depending on plant species.

Suppression of Meloidogyne incognita in Lettuce and Oriental Melon by Pasteuria penetrans KW1

  • Pasteuria penetrans KW1 (PP), parasitic bacterium of nematode, was isolated from oriental melon greenhouse soil in Korea and evaluated for the suppression effect on the reproduction of southern root-knot nematode, Meloidogyne incognita (MI), in lettuce (Lactuca sativa L. var. Chungchima) and oriental melon (Cucumis melo L. var. Eunchun). Pot experiments were conducted by planting the lettuce seedlings in medium inoculated with 5,000 MI juveniles/pot (J), J +100,000 PP endospores/l g medium, and J +200,000 PP endospores/1 g medium. After 11 weeks of plantation, number of root galls in J +200,000 PP endospores/1 g medium was decreased to 92/root (38.9%, control effect), compared to the J of 150/root. In the second plantation of lettuce in the same pots, the numbers of root gall were significantly decreased in PP treated pots with 75 (77.2%, control effect) and 150/root (54.4%, control effect) in J +200,000 and J +100,000 PP endospores/1 g medium, respectively, compared to the J of 330/root when harvested at 10 weeks after planting. In oriental melon, root gall percentages were 32.1 (60.2%, control effect) and 52.9% (34.5%, control effect) in J +200,000 and J + 1(10,000 endospores/l g medium which were significantly lower than that of 80.7% in J.
