Incidence and Distribution of Virus Diseases on Cucumber in Jeonnam Province During 1999-2002

  • Disease incidences of cucumber virus diseases in Jeonnam province were 52.5%, 16.1%, 35.2%, and 50.9% in 1999, 2000, 2001, and 2002, respectively. Rod- and flexuous rod-shaped virus particles were observed with the frequencies of 63.2% and 10.5%, respectively from the samples collected in 1999 under EM observation. Rod-shaped virus particles are considered as tobamovirus while flexuous rod shaped particles are considered as potyviruses. To further confirm their nature, total of 312 diseased virus samples were collected from 2000 to 2002, and tested by RT-PCR. Disease incidences of tobamoviruses including Cucumber green mottle mosaic virus and Kyuri green mottle mosaic virus were 48.7% and 3.8%, respectively while those of potyviruses including Zucchini yellow mosaic virus, Papaya ringspot virus, and Watermelon mosaic virus were 15.7%, 9.3%, and 5.1%, respectively. Interestingly, Cucumber mosaic virus was hardly detected. About 5.8% of tested samples were infected with more than one virus. Tobamovirus infection was consistently observed from September to December regardless of planting time, whereas infection of potyviruses was observed in many cucumber cultivating areas where it was planted in September and October.

  • Cucumber mosaic cucumovirus (CMV) is an isometric plant virus with functionally divided genomic RNAs and a broad host range. RNA 1 and RNA 2 each encode one protein, both of which are essential for replication. RNA 3 encodes the viral coat protein and an additional protein thought to be involved in potentiating the cell-to-cell movement of the virus. Functions of the RNAs have been confirmed using a pseudorecombinant virus constructed with infectious cDNA-derived transcripts of the RNAs. Generally, CMV produces different symptoms in various host plants depending on the virus strains. In this mini-review, we describe the potential role of the genes associated with symptom expression of CMV RNAs.

Occurrence of Two Tobamovirus Diseases in Cucurbits and Control Measures in Korea

  • Two Tobamoviruses, Cucumber green mottle mosaic virus (CGMMV) and Zucchini green mottle mosaic virus (ZGMMV), occurred in Korea in 463 ha in 1998, 33.9 ha in 1999, and 44.2 ha in 2000. CGMMV was detected in watermelon, cucumber, oriental melon, and melon, whereas ZGMMV was mainly detected in zucchini squash. Thirty-six CGMMV isolates wee classified into three types by analysis of single strand cDNA conformational polymorphism (SSCP) of the coat protein gene. In a comparison of serological relationships among CGMMV, ZGMMV, and Kyuri green mottle mosaic virus (KGMMV), the three tobamoviruses specifically reacted with each homologous antibody in the double-antibody sandwich enzyme-linked immunosorbent assay and rapid imunofilter paper assay (RIPA), although ZGMMV and KGMMV were slightly biologcially similar. In a survey of the three tobamoviruses in cucurbitgrowing field in Korea by RIPA, CGMMV and ZGMMV were detected but KGMMV was not found in commercially growing cucurbit crops so far. Seed contamination ratio of CGMMV in bottle gourd seeds tested was 84%, while seed trasmission ratio from the virus-contaminated seeds was 2.0%. Soil transmission ratio was 0-3.5% in fields naturally infested with CGMMV or ZGMMV. Control measures of the virus diseases are roguing and sanitation. These suggest that it is important to rogue the first infected crops, which include the seed and soil, especially early in the season. This may be practicable to control the diseases because CGMMV and ZGMMV have a narrow host range restricted to cucurbitaceous crops.

경북지역 오이에 발생하는 주요 바이러스 종류 및 발생실태 (Occurrence of Virus Diseases on Cucumber in Gyeongbuk Province)

  • 경상북도 오이 주산단지인 상주와 군위지역의 바이러스 병 발생을 조사한 결과 포장 발생율은 $55.0{\sim}88.6%$였으며, 상주지역이 군위보다 약간 높게 나타났다. 바이러스 발병 포장의 이병주율은 2006년 상주 지역에서 평균 25.6%로 매우 높게 나타났으며, 발생범위는 최소 1.5%에서 최대 90%로 농가별로 편차가 매우 심하였다. 포장에서 바이러스 증상으로 관찰되는 시료 217점을 RT-PCR을 이용하여 바이러스 종류를 분석한 결과 ZYMV가 85% 이상으로 가장 많이 검출되어 오이에 피해를 주는 주요 바이러스로 나타났다. ZYMV에 감염된 오이는 잎에 황화 및 심한 모자이크 증상이 나타났으며 일부 과일은 곡과 증상과 함께 표면이 울퉁불퉁하게 나타났다.

  • In 1995, we collected stock (Matthiola incana) plants causing mosaic symptoms on leaves and color breakings on flowers in Daekwallyong, Korea. Two viruses were isolated from the infected plants, and identified as cucumber mosaic virus (CMV) and turnip mosaic virus (TuMV) by experiments of host range, serology and electron microscopy. Each of the virus did not produce the same symptoms on the stock seedlings as naturally infected plants caused. When the viruses were coinoculated to the stock seedlings, however, severe mosaic symptoms were observed on leaves, and then the color breakings were expressed on flowers.

  • A single-step multiplex reverse transcription polymerase chain reaction (RT-PCR) assay was developed for the simultaneous detection of five cucurbit-infecting viruses: cucumber mosaic virus (CMV), watermelon mosaic virus 2 (WMV2), zucchini yellow mosaic virus (ZYMV), cucumber green mottle mosaic virus (CGMMV), and kyuri green mottle mosaic virus (KGMMV). The multiplex RT-PCR provides a simple and rapid method for detecting various viruses in cucurbit plants, which will help diagnose many cucurbit plants at a time.

고랭지 나리의 바이러스 발생과 RT-PCR에 의한 검정 (Occurence of Viruses in Lilies (Lilium spp.) in Highland Areas and Their Detection by One-step RT-PCR)

  • 이 연구는 고랭지 나리에서 발생하는 바이러스의 병징, 종류 및 계통별 방병률을 조사 분석하고 효과적인 검정방법을 개발하고자 수행하였다. 고랭지 나리에서 발생하는 바이러스의 병징은 모자이크, 축엽, 퇴록반점, 줄무의, 라인패턴을 나타내었으며, 증상별 분포는 모자이크가 43.8%, 축엽이 29.2%, 퇴록반점이 10.9%였다. 바이러스 종류별로는 Lily symptomless virus(LSV), Cucumber mosaic virus(CMV), Lily mottle virus(LMoV) 등 6가지 바이러스가 전자현미경으로 검정되었다. 지역별로는 강릉(왕산)이 대관령보다 바이러스 이병률이 높았으며, 계통별 바이러스 이병률은 오리엔탈 계통(카사블랑카, 마르코폴로)이 아시아틱 계통(솔레미오, 플라토)보다 2~4배 높았다. 바이러스 진단방법으로는 기존의 PT-PCR보다 개선된 one-step RT-PCR 검정이 시간을 줄이면서 민감도가 뛰어나 가장 효과적이었다.

A Subpopulation of RNA3 of Cucumber mosaic virus Quasispecies

  • This study examined the existence of genetically diverse population of Cucumber mosaic virus (CMV), known as quasispecies, from lily, Nicotiana benthamiana and from purified virions. Based on the conserved sequences of CMV lily isolates in intergenic region (IR) on RNA3, the genetic variation of IR from three different sources was investigated by a specific restriction endonuclease hydrolysis of amplified reverse transcription-polymerase chain reaction (RT-PCR) products using virus-specific primers, and was compared with IR sequences. The IR nucleotide sequences of CMV lily isolates were highly conserved, however, quasispecies was detected from all three sources in low level, containing sub-populations of RNA3. These subpopulations of RNA3 were inoculated onto zucchini squash by in vitro transcripts from corresponding full-length cDNA clones together with Eny RNA1 and 2 transcripts. The systemic symptom of zucchini plants infected by these quasispecies was chlorotic spotting, which was milder than severe mosaic and stunt symptom caused by Eny-CMV. The severity of symptom was correlated with RNA accumulation of viruses. These results suggest that the genome of CMV lily isolates consists of quasispecies populations.

국내 토끼풀에서 분리한 Cucumber mosaic virus의 특성 (Characterization of Cucumber mosaic virus Isolated from Trifolium repens in Korea)

  • 고추포장 주변의 모자이크 및 기형의 병징을 보이는 토끼풀에서 CMV를 분리하여 CMV-Tr1으로 명명하였다. 토끼풀로부터 RT-PCR, PCR-restriction fragment length polymorphism 및 외피단백질 유전자의 염기서열 분석을 통해 CMV를 동정하였다. 기주식물 반응결과 N. benthamiana를 포함한 5종의 담배와 오이, 꽈리 및 토마토의 상엽에서 모자이크 병징을 나타내었고, 명아주와 동부에서는 접종엽에 국부괴사 병반을 나타냈다. CMV-Tr1의 외피단백질을 CMV-As와 비교한 결과 99.5%의 상동성을 보였으며, 계통분석 결과 CMV subgroup IB에 속하는 CMV의 한 계통으로 확인되었다.

과꽃에서 분리한 Cucumber mosaic virus의 성질 (Characterization of an Isolate of Cucumber mosaic virus Isolated from Chinese aster (Callistephus chinensis))

  • 모자이크 증상의 과꽃(Callistephus chinensis L.)으로부터 Cucumber mosaic virus (CMV)의 한 계통(Cas-CMV)를 분리하고, Fny-CMV와 As-CMV를 대조로 기주반응, dsRNA, RT-PCR 및 RFLP분석을 통하여 바이러스를 동정하였다. Cas-CMV의 특징적인 기주반응의 차이는 박과 식물에서 발현되는 강한 병정이었으며, 특히 쥬키니호박에 접종하였을 때에는 접종 15-20일 후에 심한 모자이크 증상과 함께 어린 식물이 고사되는 괴저현상을 나타냈다. DsRNA분석과 RT-PCR실험의 결과는 Cas-CMV가 서브그룹 I의 CMV에 속하는 것으로 나타났으며, 더욱이 HindIII를 이용한 RFLP 분석은 Cas-CMV가 서브그룹 IA 구분되었다.
