Development of Prototype Electronic Dosimeter using the Silicon PIN Diode Detector (실리콘 PIN 다이오드 검출기를 이용한 전자선량계 개발)

  • A prototype electronic dosimeter(PED) adopting a silicon PIN diode detector as a radiation detector has been developed, manufactured and test-evaluated. A radiation signal processing circuit has been electronically tested and then the radiation detection characteristics of this PED has been performance-tested by using a reference photon radiation field. As a result in a electronic performance test, radiation signals from a detector were well observed in the signal processing circuit. The radiation detection sensitivity of this PED after several test-irradiations to $^{137}Cs$ gamma radiation source appeared to be 1.85 cps/$Gy{\cdot}h^{-1}$ with 19.3% of the coefficient of variation, which satisfied the performance criteria for the active personnel radiation monitor. Further improvement of the electronic circuit and operating program will enable the PED to be used in personal monitoring purpose.

  • 본 논문에서는 고정도의 방사선 측정이 가능한 능동형 전자선량계를 제작하기 위해 필수적으로 요구되는 반도체 방사선 검출기를 ASIC으로 구현하였다. 이는 전자선량계의 소형화와 저소비전력을 실현할 수 있도록 전치증폭기와 성형증폭기를 일체화한 것으로 방사선과 방사선 검출 소자인 상용 핀 포토다이오드의 상호작용으로 생성된 수 [nA]의 전류펄스를 측정할 수 있다. MOSIS 공정을 통하여 ASIC으로 구현된 방사선검출기는 $10{\mu}Ci$${\gamma}$-선 Ba-133, Cs-137 및 Co-60의 세 핵종에 대하여 방사선 조사시험을 수행하여 구현된 방사선 검출기의 유용성을 입증하였다.

An Analysis of ${\gamma}-ray$ Energy Spectra Using the NaI(T1) Scintillation Detector in the Air and Water (NaI(T1) 섬광검출기를 이용한 공기 및 수중에서의 감마선 에너지스펙트럼 분석)

  • The energy spectra in the air and water of several ${\gamma}-ray$ sources such as Cr-51, Cs-137, Mn-54, Zn-65 have been investigated using the NaI(T1) scintillation detector. General response functions, which can curve fit the measured spectra, have been constructed. We have found that the constructed response functions can successfully represent the measured spectra in the water as well as in the air, It is possible, by comparing the relevant parameters of the response functions, to quantitatively characterize the changing features of the measured spectra as obtained with varying the water depth. Of the response function parameters, those which affect the shape of the full-energy Peak have most notably changed. Besides, those parameters which affect the shapes of the flat continuum, the Compton continuum and edge have also shown slight changes with varying the water depth.

Relationship between Radiation Induced Activation of DNA Repair Genes and Radiation Induced Apoptosis in Human Cell Line A431 (인체세포주 A431에서 방사선 조사 후 DNA수선 유전자 발현과 세포고사와의 관계에 관한 연구)

  • Purpose: The purpose of this study was to evaluate the relationship between radiation-induced activation of DNA repair genes and radiation induced apoptosis in A431 cell line. Materials and Methods: Five and 25 Gys of gamma radiation were given to A431 cells by a Cs-137 cell irradiator. Apoptosis was evaluated by flow cytometry using annexin V-fluorescein isothiocyanate and propidium iodide staining. The expression of DNA repair genes was evaluated by both Northern and Western blot analyses. Results: The number of apoptotic cells increased with the increased radiation dose. It increased most significantly at 12 hours after irradiation. Expression of p53, p21, and hRAD50 reached the highest level at 12 hours after 5 Gy irradiation. In response to 25 Gy irradiation, hRAD50 and p21 were expressed maximally at 12 hours, but p53 and GADD45 genes showed the highest expression level after 12 hours. Conclusion: Induction of apoptosis and DNA repair by ionizing radiation were closely correlated. The peak time of inducing apoptosis and DNA repair was 12 hours in this study model. hRAD50, a recently discovered DNA repair gene, was also associated with radiation-induced apoptosis.

Changes in Water Quality and Sediment Yield in the Forest Catchment - A Study of the Lake Shirarutoro Area in Northern Japan - (산림유역의 토지변형에 따른 수질과 토사생산량 변화 - 일본 시라루토로호수 지역의 연구 결과 -)

  • Sediment and nutrient loading caused by the removal of forest cover and alteration of agricultural lands in catchments have led to the deterioration in Lake Shirarutoro. To examine the effects of deforestation and agricultural activities on water quality, I examined changes in total nitrogen (TN) and total phosphorus (TP) of lake water induced by land use change, and compared them with the various research data produced over the years. Our investigation showed that the level of TN and TP in the lake water decreased when forest cover increased but increased when farmland area increased. The concentration of TN and TP was high in Lake Shirarutoro despite that its catchment was surrounded by large forests and small farmlands. This result indicates that land uses near Lake Shirarutoro have affected the quality of the lake water. I have examined the changes of sediment yield in the lake's catchment over the last approximately 300 years. Eleven core samples were obtained from the lake sediment and analyzed to establish a chronology after using two tephra layers (Ko-c2 in 1694 and Ta-a in 1739) and a $^{137}Cs$ peak (in 1963). The average sediment yield under the natural condition during the first two periods was 8.4 tons/$km^2$/year in 1694~1739 and 8.9 tons/$km^2$/year in 1739~1963 respectively. The conversion of the Shirarutoro catchment into agricultural lands and deforestation intensified, leading to an increased sediment yield of 21.1 tons/$km^2$/year during 1963~2007.

Studies on the Radiation Induced Apoptosis by Morphological and Biochemical Analysis in A431 Cells (방사선에 조사된 인체세포주(A431)에서 형태학적 분석법과 생화학적 분석법을 이용한 방사선 유도 세포고사에 관한 연구)

  • Purpose: We performed this study to evaluate the process of radiation induced apoptosis in A431 skin epithelial cancer cell line. Materials and Methods: Low to high dose radiation (0, 2, 5, 10, 25 Gy) was given to A431 cells by Cs-137 cell irradiator. Apoptosis was evaluated by cell morphology, dye exclusion test, and DNA laddering. Results: Cell viability decreased as the radiation dose increased. Number of apoptotic bodies increased as radiation dose increased. It increased most significantly at 12 hours after irradiation. Lactate dehydrogenase activity in culture medium increased according to radiation dose and time after irradiation. DNA ladders could be identified in irradiated cells, but, it had no correlation with radiation dose or time after irradiation. Conclusion: Radiation-induced apoptosis which was the main course of cell death in A431 cells could be analyzed quantitatively by counting apoptotic bodies under microscope. Apoptosis increased as radiation dose increased.

Comparison of Radiation Adaptive Responses in Peripheral Lymphocytes of Patients Undergoing Tc-99m MDP and Tc-99m DTPA Scintigraphies (Tc-99m MDP와 Tc-99m DTPA 신티그라피를 시행한 환자의 말초혈액 림프구에서 유도되는 방사선 적응반응의 비교)

  • Purpose: The purpose of this study was to compare the radiation adaptive response (RAR) in peripheral lymphocytes (PL) of patients induced by Tc-99m MDP and Tc-99m DTPA scintigraphies. Materials and Methods: Lymphocytes from 45 patients (25 males, 20 females, mean age $44{\pm}18$ years) were collected before and after scintigraphies using 740 MBq Tc-99m MDP (n=22) or Tc-99m DTPA (n=23). Lymphocytes from 20 controls (12 males, 8 females, mean age $43{\pm}7$ years) were also collected. They were exposed to challenge dose of 2 Gy ${\gamma}-rays$ using a Cs-137 cell irradiator Number of ring-form (R) and dicentric (D) chromosomes was counted under the light microscope. From them a representative score, Ydr, was calculated as Ydr=(D+R)/cells. Adaptation index (AI) was defined as difference of Ydr between unconditioned and conditioned lymphocytes. Ydr was also measured after an administration of cycloheximide (CHM), a protein synthesis inhibitor, before challenge dose. Results: RAR was induced in both groups of patients. CHM abolished the adaptive response in both groups. AI of Tc-99m MDP group was significantly higher than that of Tc-99m BTPA group. Conclusion: Tc-99m MDP induced RAR was more prominent than those induced by Tc-99m DTPA.

Radiation Adaptive Response Induced by I-131 Therapy in Patients with Differentiated Thyroid Cancer (분화 갑상선암 환자에서 I-131 치료에 의해 유도되는 방사선적응반응)

  • Purpose: The purpose of this study was to ascertain whether radiation adaptive response could be induced by high dose I-131 therapy in patients with differentiated thyroid cancer. Materials and Methods: Lymphocytes from 21 patients (7 males, 14 females, mean age $55{\pm}12$ years) were collected before and after administration of 5,550 MBq (150 mCi) I-131. They were exposed to a challenge dose of 1 Gy gamma rays using a Cs-137 cell irradiator. The number of ring-form (R) and dicentric (D) chromosomes was counted under the light microscope, and used to calculate the frequency of chromosomal aberration. Ydr, which was defined as the sum of R and D divided by the total number of counted lymphocytes. Results: Ydr in patients before I-131 therapy ($0.09{\pm}0.01$) was not different from that of controls ($0.08{\pm}0.01$). Ydr was significantly increased to $0.13{\pm}0.02$ (p<0.0001) after I-131 therapy. Increase of Ydr after the challenge irradiation of 1 Gy was significantly lower in patients after I-131 therapy than before I-131 therapy ($0.17{\pm}0.03\;vs\;0.21{\pm}0.02$, p<0.0001). Cycloheximide (CHM), an inhibitor of protein synthesis, abolished this effect. Ydr after CHM ($0.20{\pm}0.01$) was significantly higher than Ydr after I-131 therapy ($0.17{\pm}0.03$, p<0.0001), but was not different from Ydr before I-131 therapy ($0.21{\pm}0.02$).Conclusion: High dose I-131 therapy induces an adaptive response in peripheral lymphocytes of patients with well-differentiated thyroid cancer, which is associated with protein synthesis.

Materials and Methods: Lymphocytes from 22 patients (6 males, 16 females, mean age $50{\pm}14$ years) were collected before and after bone scintigraphy using 740 MBq Tc-99m-MDP Lymphocytes from 10 controls (6 males, 4 females, mean age $43{\pm}7$ years) were also collected. They were exposed to challenge dose of 2 Gy gamma rays using a cell irradiator. Number of ring-form and dicentric chromosomes per 600 cells (chromosomal aberrations) was counted under the light microscope. Results: Chromosomal aberrations in patients before bone scintigraphy ($385.1{\pm}30.5$) was not different from that of controls ($367.8{\pm}36.6$). However, chromosomal aberrations in patients after bone scintigraphy was significantly decreased to $192.6{\pm}22.1$ (p=0.0001). Conclusion: Low dose gamma-irradiation by Tc-99m-MDP used for bone scintigraphy induces a cytogenetic adaptive response in peripheral lymphocytes.

  • It was reported that radiopharamaceuticals induced radiation adaptive response (RAR) in patients undergoing nuclear medicine imaging studies. Individual variations of RAR were not studied well. The purpose of this study was to evaluate individual variation of RAR in patients undergoing nuclear medicine imaging studies. Peripheral lymphocytes were collected from 23 patients undergoing $^{99m}Tc-diethylenetriamine$ pentaacetic acid $(^{99m}Tc-DTPA)$ renal scintigraphy, 18 patients undergoing $^{99m}Tc-methylene$ diphosphonate $(^{99m}Tc-MDP)$ bone scintigraphy and 21 patients undergoing $^{99m}Tc-tetrofosmin\;(^{99m}Tc-TF)$ scintigraphy were collected before and 4 hours after injection of radiopharmaceuticals. The lymphocytes were exposed to challenge dose of 2 Gy gamma rays using a cell irradiator. Numbers of ring-form (R) and dicentric (D) chromosomes were counted under the light microscope. and used to calculate the frequency of chromosomal aberration [Ydr=(D+R)/total number of counted lymphocytes]. Adaptation index (k) was defined 3s ratio of Ydr in conditioned lymphocytes over Ydr in unconditioned lymphocytes. Coefficients of variance of k in $^{99m}Tc-DTPA,\;^{99m}Tc-MDP\;and\;^{99m}Tc-TF$ were 35%, 34% and 21%, respectively k was not dependent upon age, sex, and underlying diseases. There was a wide variation of RAR induced by radiopharmaceuticals among patients undergoing nuclear medicine procedures. It remains to be determined for causes of such variation.
