Inactivation of Pathogenic Bacteria by Addition of Thermophilic Bacteria in the Thermophilic Aerobic Oxidation(TAO) System (고온호기산화장치의 고온미생물 첨가에 의한 병원성 미생물의 불활성화)

  • This study analyzed temperature increase, microorganism changes, and inactivation of pathogenic microorganisms in pig slurry when treated with thermophilic microorganisms in Thermophilic Aerobic Oxidation(TAO) system. An amount of $6 m^3$ of pig slurry was treated in an $18 m^3(3.0\times2.5\times2.4 m)$ reactor for 5 to 7 days in two groups: the control of pig slurry only and the treatment of pig slurry with 6 liters of thermophilic microorganism(Bacillus sp.). To study the microorganism changes in the reactor, the populations of aerobic mesophilic microorganisms, thermophilic microorganisms and general pathogens were analyzed. To study the inactivation of pathogenic microorganisms, the levels of E. coli, Salmonella sp, Crytosporidium parvum and Giardia lamblia were analyzed. The temperature inside the reactor ranged from 18 to $62^{\circ}C$ for the control while far the treatment group it ranged from 18 to $66^{\circ}C$, showing a slightly higher array. With regard to changes in microorganisms, both mesophilic and thermophilic organisms decreased from $3.1\times10^6$ to $1.2\times10^2$ CFU/ml and from $1.0\times10^4$ to $8.0\times10^1$ CFU/ml, respectively, in the control. In the treatment, on the other hand, mesophilic organisms decreased from $3.0\times10^8$ CFU/ml to $8.6\times10^5$ CFU/ml while thermophilic organisms increased sharply from $2.0\times10^6$ to $1.2\times10^8$ CFU/ml. For pathogens, Salmonella and Giardia were not detected either before or after the treatment, while E. coli and C. parvum were found to be $10^5$ CFU/ml each before treatment and negative after it. From this experiment, it was concluded that thermophilic microorganisms could effectively sanitize liquid compost by generating high temperature in the TAO system, which in turn would inhibit the growth of pathogenic organisms.

Prevalence of Cryptosporidium sp among calves and pigs in Jeonbuk province (전북지역의 소와 돼지에서 Cryptosporidium sp의 감염실태 조사)

  • Cryposporidium species have a wide host range. These coccidian parasites are found in close association with epithelial cells of many species of animals including mm. The gastrointestinal tract is most commonly affected in young ruminants and this parasite is thought to be considerable importance in calf diarrhea complex. Major outbreaks of cryptosporidiosis have been reported in calves, lambs, pigs and others including avian species. Cryptosporidiosis is transmitted by oocysts of Cryptosporidium species. Because cryptosporidiosis is common infection among animals, early literature considered it a zoonosis. Human infections contracted from calves, cats, and horse feces. However, the resrvoir host is longer considered the major source of infection. Mild cases of disease have been reported in farm workers. Immunosuppressed, very young and very old persons should avoid contact with this parasite because it may cause severe diarrhea. In order to detect of Crytosporidium sp infection from feces of cattle and pigs at Chonbuk Iksan and Kunsan area, sedimentation and modified acid fast stain were applied. The positive rate of Cryptosporidium sp infection from 1, 176 of cattle and 267 of pigs were 0.5 % and 16.8%, respectively. According to area in Iksan and Kunsan, the positive rates were 0.4% and 0.9% from cattle, and 18.9% and 12.1% from pigs, respectively. In cattle, positive detection rate was 0.6% in milking cows but not in Korean cattle.

  • Recently, there is growing interest in ultraviolet (UV) irradiation as a disinfection technic in drinking water production due to its effectiveness to inactivate microorganisms such as Crytosporidium parvum without forming disinfection byproducts. However, UV disinfection is known for its drawback such as photoreactivation. Despite many works concerning the photoreactivation, most of works were focused on indicator or non pathogenic microorganisms. The objective of this study is to examine the photoreactivation of Pseudomonas aeruginosa which is an opportunistic pathogen as UV radiation by LP and MP UV lamp was applied. The result showed that P. aeruginosa had high photo repair efficiency regardless of the type of UV irradiation. Both of the effective log repair values of LP and MP UV irradiation were found approximately 2.6 log. In addition, photo repaired P. aeruginosa was not significantly different in forming biofilm in comparison with non treated P. aeruginosa.

Seasonal Dynamics of Pathogenic Microorganisms (Cryptosporidium, Giardia and Fecal Bacteria) in an Artificial Lake Ecosystem (Sangsa Lake, Korea)

  • This study was performed for the purpose of monitoring monthly levels of two pathogenic microorganisms, Cryptosporidium and Giardia, from November 2005 to August 2007 in Sangsa Lake. Water temperatures, pH and DO fluctuated seasonally at the study site. Annual mean values of BOD, COD and SS were $0.8\;mg\;L^{-1}$, $2.3\;mg\;L^{-1}$ and $1.9\;mg\;L^{-1}$ respectively. Although there was distinct seasonal variation in water chemistry and chlorophyll $\underline{a}$ concentration, the lake generally contains low concentrations of nutrients and chlorophyll $\underline{a}$. The relative abundance of coliform bacteria was always greater than that of fecal coliform. The fecal coliform bacteria comprised $8.5{\sim}22.1%$ of total coliform bacteria. Seasonal analysis of Cryptosporidium and Giardia levels in the study site showed that in winter (November through February), Cryptosporidium oocysts and Giardia cysts were most abundant ($1.1{\sim}1.8\;{\times}\;10\;cells\;L^{-1}$ and $3.8{\sim}5.1\;{\times}\;10\;cells\;L^{-1}$, respectively), while in summer (July through September) the abundance was lowest ($0.0{\sim}0.3\;{\times}\;10\;cells\;L^{-1}$ and $0.9{\sim}2.9\;{\times}\;10\;cells\;L^{-1}$, respectively). Molecular identification revealed two subtypes of Cyrptosporidium parvum in Sangsa Lake.

Immunotherapy in calves experimentally infected with cryptosporidium parvm (크립토스포리디움에 실험적으로 감염된 송아지의 면역요법)

  • To determine the efficacy of immunotherapeutic agents, four female Holstein calves 7-day-old were inoculated per os with $1{\times}10^7$ C parvum oocysts (VRI-CN91). Each calf received twice daily oral dosage of 200-500ml of the immune bovine serum, immune bovine colostrum, mAb C6, and phosphate-buffered saline, respectively. Treatment was initiated 4 days postinfection and laster 3 days. The clinical sign of the calf treated with phosphate-buffered saline lasted 9 days after the initial treatment. The calves treated with those immunotherapeutic agents, however, showed decreased severity of diarrhea at day 3, 2, 5 after the initial administration, respectively. The calves treated with immunotherapeutic agents showed reduced parasite loads compared to control calf. These results suggest that oral passive immunotherapy with immune bovine colostrum and immune bovine serum may be a useful treatment approach.

Experimental activation of cryptosporidiosis in mice by immunosuppression (면역억제에 의한 마우스의 Cryptosporidium 발현 실험)

  • Cryptosporidium, a coccidian parasite first described by Tyzzer (1907) from a laboratory mouse, has become an important human enteric pathogen causing overwhelming diarrhea especially in immunocompromised patients such as AIDS. This parasite has been reported from over 20 countries and is recognized as a cosmopolitan species. In Korea, however, thEre has been no report on human as well as animal cryptosporidiosis. This study was performed so as to verify the presence of Cryptosporidium in Korea by activating the parasite from laboratory mice by immunosuppression. Total 65 conventionally.bred ICR mice including a control (5 mice) and 3 experimental groups (20 each) were used for this study. Group I was immunosuppressed with Prednisolone injection (1 mg IM, every other day) for 7 weeks. Group II (prednisolone injection and tetracycline administration) and Group III. (prednisolone injection and trimethoprim-sulfamethoxazole administration) were prepared to observe the effect of antibacterial agents on the activation of cryptosporidiosis. In fecal examinations of mice Cryptosporidium oocysts($4-6{\mu\textrm{m}}$ in size) were detected from 1 week after the start of immunosuppression and the mice began to die. In H-E stained tissue sections of the lower jejunum, numerous very small ($2~4{\;}{\mu\textrm{m}}$), dense, ovoid or spherical, slightly basophilic bodies were seen attached on the free border of mucosal epithelial cells. In scanning and transmission electron microscopic observations, these organisms were identified as various developmental stages of Cryptosperidium. The species is considered to be C. parvum. Cryptosporidiosis was activated not only in Group I but also in Group II and III, indicating no protective effects of the antibacterial agents used, although the mice in Group II and III lived longer than those in Group I. The present study confirmed that Cryptosporidium exists in laboratory mice bred in Korea, and predicts possible occurrence of human cryptosporidiosis in Korea.

Diagnosis of bovine cryptosporidiosis by indirect immunofluorescence assay using monoclonal antibody (단세포군항체를 이용한 간접형광항체법에 의한 송아지 작은와포자충증의 진단)

  • Two hybridoma cell lines against Cwptosporinium possum oocysts nFRl-CN911 were produced. The isotype of these 2 monoclonal antibodies (mAbs) was IgG2b (lE7.2) and and IgM (C6). Enzyme immuno-transfer blotting analysis showed that 157.2 reacted specifically to 36 kDa protein and C6 reacted to 67 and 70 kDa proteins. C. pcnlum was bound specifically to the surface region of oocysts by these mobs. No cross-reactivity was observed with tachyzoites of ToxopLosma gonnii and oocysts of Eimeria zuernii,5. bouis and E. canadensis of bovine origin. The indirect immunofluorescence assay (IIF) using mAb C6 was successful with counterstain. With the IIF using mob C6, oocysts appeared as 3 to $5{\mu}m$ spherical objects fluorescing bright apple green against a reddish dark background. The IIF using mAb C6 was agreed in specificity and sensitivity with those of a commercial diagnostic kit. These results demonstrated that the produced mAbs were specific to C. parvum and that the mAb C6 could be used for diagnosis of cryptosporidiosis.
