브로콜리 유래 Sulforaphane의 혈관 수축성 조절 효과 (The Inhibitory Effect of Broccoli in Cruciferous Vegetables Derived-Sulforaphane on Vascular Tension)

  • The present study was undertaken to investigate the influence of sulforaphane on vascular smooth muscle contractility and to determine the mechanism involved. We hypothesized that sulforaphane, the primary ingredient of broccoli of cruciferous vegetables, plays a role in vascular relaxation through inhibition of Rho-kinase in rat aortae. Intact of denuded arterial rings from male Sprague-Dawley rats were used and isometric tensions were recorded using a computerized data acquisition system. Interestingly, sulforaphane significantly inhibited fluoride, phorbol ester or thromboxane $A_2$ mimetic-induced contraction in denuded muscles suggesting that additional pathways different from endothelial nitric oxide synthesis such as inhibition of Rho-kinase or MEK might be involved in the vasorelaxation. Furthermore, sulforaphane inhibited thromboxane $A_2$-induced increases in pERK1/2 levels suggesting the mechanism including inhibition of thromboxane $A_2$-induced increases in ERK1/2 phosphorylation. This study provides evidence that sulforaphane induces vascular relaxation through inhibition of Rho-kinase or MEK in rat aortae.

초음파 처리가 채소 중의 sulforaphane 분석에 미치는 영향 (Effects of ultrasonication on the analysis of sulforaphane content in vegetables)

  • 초음파처리가 십자화과 채소의 sulforaphane 함량 분석에 미치는 영향을 GC/MS를 이용하여 검토하였다. 무, 알타리무, 순무, 양배추, 브로콜리를 각각 dichloromethane을 추출용매로 하여 초음파처리 후 용매 추출 및 질소 농축을 하고, m/z 72, 160, 55, 114을 선택이온측정법(selective ion monitoring, SIM)을 이용하여 분석하였다. 각 채소류의 sulforaphane 함량은 브로콜리가 149.0 ppm으로 가장 많았으며, 양배추(67.9 ppm), 무(35.4 ppm)의 순으로 높았다. Sulforaphane 추출율은 진탕교반(shaking)만 할 경우보다 초음파처리(sonication)후 진탕교반(shaking)을 할 경우 2.7배 높은 것으로 나타났다. 따라서 초음파추출공정이 sulforaphane의 추출효율 향상에 유효한 방법으로 판단된다.

  • Korean cabbage (Brassica campestris L.ssp.pekinensis) is one of the major cruciferous vegetables. Cruciferous vegetables contain a rare series of secondary metabolites of amino acids called glucosinolates, as well as carotenoids, tocopherol, vitamin C and fibers. This study evaluated the effect of common cooking methods (boiling, microwaving, steaming and frying) on the phytochemical content (lutein, ${\beta}$-carotene, ${\gamma}$-tocopherol, and ${\alpha}$-tocopherol), and total antioxidant capacity of Korean cabbages, determined by DPPH assay and ABTS assay. Boiling caused a decrease in carotenoids, lutein and tocopherols. Microwaving and steaming were relatively good cooking methods for maintaining lutein, ${\beta}$-carotene, ${\gamma}$-tocopherol, and ${\alpha}$-tocopherol. The overall results of this study demonstrate that some domestic cooking procedures, specifically microwave and steaming, increased the bioaccessibility of carotenoids and tocopherol, highlighting the positive role of the nutritional properties of Korean cabbage.

Determination of Goitrogenic Metabolites in the Serum of Male Wistar Rat Fed Structurally Different Glucosinolates

  • Glucosinolates (GLSs) are abundant in cruciferous vegetables and reported to have anti thyroidal effects. Four GLSs (sinigrin, progoitrin, glucoerucin, and glucotropaeolin) were administered orally to rats, and the breakdown products of GLSs (GLS-BPs: thiocyanate ions, cyanide ions, organic isothiocyanates, organic nitriles, and organic thiocyanates) were measured in serum. Thiocyanate ions were measured by colorimetric method, and cyanide ions were measured with CI-GC-MS. Organic isothiocyanates and their metabolites were measured with the cyclocondensation assay. Organic nitriles and organic thiocyanates were measured with EI-GC-MS. In all treatment groups except for progoitrin, thiocyanate ions were the highest among the five GLS-BPs. In the progoitrin treated group, a high concentration of organic isothiocyanates (goitrin) was detected. In the glucoerucin treated group, a relatively low amount of goitrogenic substances was observed. The metabolism to thiocyanate ions happened within five hours of the administration, and the distribution of GLSs varied with the side chain. GLSs with side chains that can form stable carbocation seemed to facilitate the degradation reaction and produce a large amount of goitrogenic thiocyanate ions. Because goitrogenic metabolites can be formed without myrosinase, the inactivation of myrosinase during cooking would have no effect on the anti-nutritional effect of GLSs in cruciferous vegetables.

  • The consumption of cruciferous vegetables such as cabbage and broccoli have been shown to have a chemopreventive effect in human and in experimental animals. Indole-3-carbinol (I3C), one component of cruciferous vegetables, has been shown to exert its chemopreventive effect in liver, colon and mammary tissue before or concurrent exposure of carcinogen, but in some reports, there have been several evidence that consumption of I3C after carcinogen treatment induced tumor promotion in some tissues. There have been no reports about the effect of I3C after carcinogen exposure in N-Nitroso-N-methylurea (MNU)-induced mammary tumor model of rats. (omitted)

캐러웨이 Sulfhydryl Oxidase를 이용한 십자화과 채소의 함황 불쾌취 억압 (Suppression of Undesirable Sulfurous Aromas of Cruciferous Vegetables with Caraway Sulfhydryl Oxidase)

  • 십자화과의 양배추와 브로콜리에서 함황 휘발성분 함량과 캐러웨이 종자의 sulfhydryl oxidase를 이용한 십자화과 채소의 함황 불쾌취 억압을 FID와 FPD가 동시에 연결된 개스 크로마토그래피로써 조사하였다. 양배추와 브로콜리에서 생성되는 휘발성 유황화합물은 캐러웨이 종자의 첨가량과 항온처리 시간에 따라 함량 및 생성률에 차이가 있었는데, 양배추와 브로콜리에 캐러웨이 종자를 첨가한 것이 대조구보다 methanethiol과 dimethyl disulfide 생성량이 훨씬 적었다. 또한 thiol oxidase 활성을 가진 캐러웨이 종자의 액상 슬러리를 이용하여 methanethiol과 dimethyl disulfide를 제거하는데 효과가 있었으며, 이들의 제거율은 캐러웨이 종자의 첨가량에 비례하였고, 특히 2.5% 캐러웨이 종자를 함유한 액상슬러리에서 효과적이었다.

십자화과 채소의 가공처리 중 Indole Glucosinolate의 분해산물 형성 (Formation of Indole Glucosinolates Breakdown Products during Processing Treatment in Cruciferous Vegetables)

  • 십자화과 채소를 습열처리한 thiocyanate ion의 방출량은 열처리시간이 증가함에 따라 점차 증가하여 30분 열처리시 최대치를 나타내었으나, 건열처리시는 유의적인 변화가 없었다. 또한, 내생의 효소 myrosinase에 의한 자가분해시는 방치시간이 경과함에 따라 점차 증가하여 3시간 후에 최대치를 나타내었는데, 이는 습열처리시의 최대 방출량보다 많은 양을 나타내었으며, 시료별 방출량은 양배추, 배추, 무, 케일 및 겨자의 순으로 높게 나타났다. 열처리에 따른 indoleacetonitrile의 생성량은 시간이 경과함에 따라 증가함을 나타내었으며, 시료별 생성량은 양배추 및 무 순으로 높게 나타났다.

Association between food consumption and serum aryl hydrocarbon receptor ligand activity among middle-aged Korean adults

  • BACKGROUND/OBJECTIVES: The diet is an important route of exposure to endocrine-disrupting chemicals (EDCs). However, few studies have investigated the association between dietary intake and EDC exposure levels among Koreans. In an earlier study, we showed that the bioactivity of serum aryl hydrocarbon receptor ligands (AhRLs) could be a surrogate biomarker to indicate exposure to EDCs and that they inhibit mitochondrial function. We also found that the mitochondria-inhibiting substances (MIS) in serum ascertained by intracellular adenosine triphosphate (MIS-ATP) and reactive oxygen species (MIS-ROS) levels could be biomarkers of exposure to EDCs, as they showed a strong correlation with AhRL and the levels of EDCs in the blood. Here, we investigated the association between the consumption of specific foods and surrogate serum biomarkers for EDCs, namely AhRL, MIS-ATP, and MIS-ROS, among middle-aged Korean adults. SUBJECTS/METHODS: A total of 1,466 participants aged 45-76 yrs from the Ansung cohort of the Korean Genome and Epidemiology Study were included. Food consumption, including that of meat, fish, vegetables, and fruits, was measured using a semiquantitative food frequency questionnaire. RESULTS: Fish intake was positively associated with AhRL (β = 0.0035, P = 0.0166), whereas cruciferous vegetable intake was negatively associated with AhRL (β = -0.0007, P = 0.0488). Cruciferous vegetable intake was positively associated with the MIS-ATP levels (β = 0.0051, P = 0.0420). A higher intake of fish was significantly associated with an increased risk of high AhRL (tertile: odds ratio [OR], 1.49; 95% confidence intervals (CIs), 1.08-2.06; P for trend = 0.0305). In addition, the second-highest tertile of cruciferous vegetable intake had lower odds of high AhRL than the lowest tertile (OR, 0.73; 95% CIs, 0.54-0.97), although no significant linear trend was observed. CONCLUSION: Consumption of different types of foods may be differentially associated with EDC exposure in middle-aged Korean adults.

사람구강편명상피암세포주인 KB세포에서 십자화과채소 유래물질인 DIM-pPhBr과 DIM-pPhF의 세포사멸유도를 통한 항종양효능에 관한 연구 (Anti-tumorigenic Effect of DIM-pPhBr and DIM-pPhF Originating from Cruciferous Vegetables in KB Human Oral Squamous Cell Carcinoma Through Apoptotic Cell Death)

  • 십자화과 채소는 Diindolylmethane (DIM)을 포함한 Isothiocyanates계 화합물을 함유하고 있는데, 이들이 암을 예방하는 효과가 있는 것으로 알려져 있다. 특히 이번 연구에 사용되어진 DIM-C-pPhBr과 DIM-C-pPhF를 기존에 알려진 DIM보다 독성은 최소화되고, 효능은 높은 물질로 보고되어져 있다 하지만, 아직까지 구강편평상피세포암종에서의 세포증식 및 세포사멸에 대한 효능연구는 보고된 바 가 없다. 따라서, 이번 연구에서는 C-DIM 중에 DIM-pPhBr과 DIM-pPhF의 구강편평상피세포암종 세포주인 KB세포에서의 세포증식 및 세포사멸유도효능을 확인함으로써 C-DIM 의 항종양능력을 확인하고자 하였다. 결과를 분석해보면, DIM-C-pPhBr과 DIM-C-pPhF은 구강편평상피세포암종 세포주인 KB의 세포증식을 감소시키고 Western blot분석법, DAPI염색법, 및 Sub-$G_1$축적분석결과에서 알수 있듯이 세포증식을 억제하기 위해 세포사멸을 유도하는 것으로 나타났다. 따라서, DIM-C-pPhBr과 DIM-C-pPhF에 의해 유도되는 세포사멸 및 세포증식억제는 구강암 예방 및 치료를 위한 좋은 전략이 될 수 있을 것으로 사료된다.
