To determine the effects of exposure to formalin on the secondary stress indices, changes in haematology and blood chemistry were monitored in healthy crucian carps (Carassius auratus). Fishes were separately exposed in a concentration range of 125 to 500 ppm formalin for 60 min. After exposure, red blood cell (RBC) count and packed cell volume (PCV) were elevated in the 500 ppm formalin exposed group. However, mean corpuscular volume (MCV) and mean corpuscular haemoglobin (MCH) were decreased significantly in the 500 ppm formalin exposed group. Total protein, albumin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, total bilirubin, inorganic phosphorus (IP) and magnesium were significantly increased at a concentration of 500 ppm. Alkaline phosphatase (ALP) and glucose were increased at a concentration of 500 ppm, but this was not significant. Lactate dehydrogenase (LDH) and calcium were significantly decreased at concentrations of 250 and 500 ppm. AST, ALT, glucose and magnesium were significantly increased in the 250 ppm formalin exposed group. These results suggests that formalin exposure might cause some damage in the liver and kidney of crucian carp.
Compositions of protein and amino acid in the muscle of crucian carp, Carassius carassius, and snakehead, Channa argus, were determined by amino acid autoanalyzer, and the protein subunits composed of sarcoplasmic and myofibrillar proteins were also analyzed by sodium dodecylsulfate (SDS) polyacrylamide gel electrophoresis. The crucian carp muscle contained about $14.7\%$ of protein, and the protein was composed of $32.6\%$ in sarcoplasmic, $62.0\%$ in myofibrillar, $4.9%$ in alkali soluble and $0.6\%$ in stroma protein. The snakehead muscle, on the other hand, contained about $16.1\%$ of protein, and $30.7\%,\;64.1\%,\;4.7\%\;and\;0.4\%$ in the above order. The sarcoplasmic and myofibrillar proteins were composed of 10 and 19 subunits respectively, in the crucian carp, and 12 and 18 subunits in the snakehead. The total amino acid compositions in the muscle of the crucian carp and snakehead were found to be very similar except lysine and glutamic acid. The major amino acids of their muscle protein were lysine, glutamic acid, arginine and aspartic acid in order. In free amino acid content of the crucian carp, histidine occupied $52\%$ of the total free amino acid, and glutamic acid, glycine and taurine did $85\%$ in case of snakehead.

This study investigated cytogenetic and hematological and histological characteristics between diploid and spontaneous triploid on Cyprinidae (Crucian carp, Carassius auratus; crucian carp, C. cuvieri and common carp, Cyprinus carpio) in four major rivers of Korea. In our results, DNA contents of triploid Cyprinidae were 50% more than those of diploid Cyprinidae. Also, erythrocyte size of triploid Cyprinidae was 50% larger than those of diploid Cyprinidae. In all sampling rivers, sex ratios of C. auratus were biased to female, and especially, triploid groups of C. auratus were all female groups (p<0.05). In principle, sex ratios of C. cuvieri and common carp were equivalent between male and female.
The accumulation levels of hexabromocyclododecanes (HBCDs) were measured in samples of muscle tissue from crucian carp (Carassius auratus) and also in samples of associated sediments. Samples were collected from 15 sites along the courses of four major rivers. Widespread contamination by HBCDs in this area was identified from the results of this investigation. HBCDs were found above the detection limit from all of the sampling sites. The ${\Sigma}HBCDs$ (sum of ${\alpha}$-, ${\beta}$-, and ${\gamma}$-HBCDs) levels ranged from 1.3 to 19 ng/g (lipid wt) in the crucian carp musscle tissue samples, and from 0.17 to 30 ng/g (dry wt) in the associated sediment samples. The predominance of ${\alpha}$-diastereomer in the muscle tissue of the carp and ${\gamma}$-diastereomer in the sediments was identified, accounting for 88% and 60% of the ${\Sigma}HBCDs$, respectively. The ${\Sigma}HBCDs$ levels in the muscle tissue samples of carp collected from the upstream sections of each river were much lower than those in the samples from carp in the downstream sections. In addition, the ${\Sigma}HBCDs$ levels in carp tissue samples from an isolated site were much lower than those in carp collected from the sites along the major rivers. This suggests that HBCD contamination in the rivers is likely to be sourced from the environment adjacent to them.
Random amplified polymorphic DNA (RAPD) analysis based on numerous polymorphic bands have been used to investigate genetic similarity and diversity among and within two cultured and wild populations represented by the species crucian carp (Carassius carassius). From RAPD analysis using five primers, a total of 442 polymorphic bands were obtained in the two populations and 273 were found to be specific to a wild population. 169 polymorphic bands were also produced in wild and cultured population. According to RAPD-based estimates, the average number of polymorphic bands in the wild population was approximately 1.5 times as diverse as that in cultured. The average number of polymorphic bands in each population was found to be different and was higher in the wild than in the cultured population. Comparison of banding patterns in the cultured and wild populations revealed substantial differences supporting a previous assessment that the populations may have been subjected to a long period of geographical isolation from each other. The values in wild population altered from 0.21 to 0.51 as calculated by bandsharing analysis. Also, the average level of bandsharing values was $0.40{\pm}0.05 $ in the wild population, compared to $0.69{\pm}0.08$ in the cultured. With reference to bandsharing values and banding patterns, the wild population was considerably more diverse than the cultured. Knowledge of the genetic diversity of crucian carp could help in formulating more effective strategies for managing this aquacultural fish species and also in evaluating the potential genetic effects induced by hatchery operations.
Genomic DNAs isolated from crucian carp of four rivers, belonging to the family Cyprinidae was amplified by seven oligonucleotides primers. In the present study, we employed hierarchical clustering method in order to reveal genetic distances and variations. Crucian carp was acquired from Hangang river (CAH), Geumgang river (CAG), Nakdonggang river (CAN) and Yeongsangang river (CAY). The primer BION-12 generated the most loci (a total of 50) with an average of 10 in the CAY population. The primer BION-10 generated the least loci (a total of 19), with an average of 3.8 in the CAG population, in comparison to the other primers used. Seven oligonucleotides primers made 16.7 average no. per primer of specific loci in the CAH population, 7.4 in the CAG population, 8.6 in the CAN population and 0.9 in the CAY population, respectively. The specific loci generated by oligonucleotides primers revealed inter-individual-specific characteristics, thus disclosing DNA polymorphisms. The dendrogram obtained by the seven oligonucleotides primers indicates four genetic clusters. The genetic distance that displayed significant molecular differences was between individuals no.06 and no.08 from the CAG population (genetic distance = 0.036), while the genetic distance among the five individuals that displayed significant molecular differences was between individuals no.08 and no.09 from the CAG population (genetic distance = 0.088). With regard to average bandsharing value (BS) results, individuals from CAY population ($0.985{\pm}0.009$) exhibited higher bandsharing values than did individuals from CAH population ($0.779{\pm}0.049$) (P<0.05). Relatively, individuals of CAY population were fairly closely related to that of CAN location (genetic distance between two populations<0.016).
Exotic species in aquatic ecosystem generate various problems domestically as well as globally. Largemouth bass (Micropterus salmoides) which is a well-known exotic species globally, acts as a substantial disturbance factor on the native fish communities in Korea due to the high predation pressure and hence designated as a ecologically harmful species by Ministry of Environment. In this study, we investigated the impact of largemouth bass on Korean native fish population, crucian carp (Carassius auratus), to identify responses of the prey fish. Two fish species were collected at seven reservoirs distributed at the Nakdong River catchment area, and changes in length-frequency and length-weight relationship (LWR) were analysed depending on existence of large-mouth bass. At bass absent sites, ratio of under age 1 year individuals were small, and over age two years were dominant. Conversely, normal length-frequency distribution pattern was identified at bass absent sites. The LWR of crucian carp (fish smaller than total length of 160 mm were only considered as it is frequently consumed by bass predation) was different depending upon bass existence. The value of parameter b at bass absent reservoirs was 2.909, which was smaller than that of bass present reservoirs, 3.100. Our results imply that crucian carp at bass present reservoirs presented a different strategy to survive from predation by bass, through relatively rapid growth. We propose that other native species might have similar growth strategies like crucian carp.

Differences in larvicidal activity of complement and chemiluminescent response of phagocytes in carp (Cyprinus carpio), crucian carp (Carassius auratus) and false dace (Pseudorasbora parva) against excysted metacercariae of Clonorchis sinensis were investigated. The prevalence of C. sinensis metacercariae of false daces caught at Nakdong river (NR) was 100%. However, no C. sinensis metacercariae were found in false daces, which were collected at a reservoir in Chinyoung (CY), crucian carps and carps. The sera of false dace, which were intensively infected with C. sinensis metacercariae, killed excysted metacercariae of C. sinensis more readily than that of carp. However, the serum obtained from C. sinensis metacercariae-free false dace showed the lowest larvicidal ability. The larvicidal ability of sera collected from each fish species completely disappeared when the complement was inactivated by heating. When supernatant of excysted metacercariae homogenate were added to phagocytes of each species, the chemiluminescent responses were significantly (p<0.05) diminished in false dace and carp. The inhibition ratio of chemiluminescent responses by the supernatant was 22.9% in false dace, 9.6% in crucian carp and 12.4% in carp.
