돼지의 소장 점막하 조직은 면역반응이 없는 재료로 널리 사용되고 있다. 본 연구에서는 소장 점막하 조직을 1-ethyl-(3,3-dimethyl aminopropyl) carboimide hydrochloride(EDC)를 이용하여 경화시켰으며 동시에 스폰지 형태로 제조하여 조직공학적 담체와 상처 드레싱재료로서의 응용 가능성을 검토하였다. 다세포 구성분을 제거한 소장 점막하 조직 분말을 3% 아세트산에 용해하여 일정한 형태의 몰드 내에 붓고 동결 건조하였다 이 소장 점막하 조직 스폰지를 여러 농도의 EDC용액에 침지하여 가교반응을 실시하였다 이를 전자주사 현미경, 시차 주사열량계, 적외선 분광분석기, 다공도 테스트 및 흡수성 실험을 수행하여 특성을 결정하였다. 소장 점막하 조직 스폰지는 경화제의 농도가 50mM 이상에서 물리적 특성이 우수한 것으로 판단되었다. 일련의 실험을 통하여 경화된 소장 점막하 조직 스폰지가 조직공학적 담체와 상처치료 드레싱재료로 유용하게 사용될 수 있음을 확인하였다.
천연고분자인 chitin은 인체에 무해무독하며 생분해성이 있는 것으로 알려져 있다. 이 chitin을 탈아세틸화시켜 얻은 chitosan에 propionyl group을 도입하여 porous bead의 형태로 제조하고 이들의 흡수성수지로서의 가능성을 검토하였다. 실험에 사용된 propionyl chitosan porous bead의 합성은 chitosan을 acetic acid 수용액에 녹인 후 emulsion을 이용한 방법으로 아실화 반응을 일으켜 제조하였으며, crosslinked propionyl chitosan porous bead는 ethyleneglycol diglycidyl ether로 부분가교시킨 chitosan을 사용하여 상기와 같은 방법으로 제조하였다. 이때 propionic anhydride의 몰수와 반응시간 그리고 반응온도를 변화시킨 결과, propionyl chitosan의 합성조건은 propionic anhydride 5 moles, 반응시간 10시간 그리고 반응온도가 $22^{\circ}C$일 때 흡수능이 가장 좋은 것을 알 수 있었다. 제조된 porous bead에 대하여 증류수 및 생리식염수, 각종 염용액, 인공뇨 및 인공혈액에 대한 흡수도를 검토하였고, 흡수속도 및 보수력에 대한 교환 사항을 검토하였으며, 나아가 가교에 따른 기계적 강도의 변화를 고찰하였다.
스티렌-무수말레인산 공중합체로 가교된 PVA막을 용액법으로 제조하였다. 투과증발공정에서 가교된 PVA/ PSMAn막에 대한 분리거동을 연구하기 위해 30~90 wt % 에탄올-물 혼합 조성에서 막에 대한 팽윤 및 수착 실험을 막내부로 수착되는 용액의 양과 조성을 측정할 수 있는 장치를 이용하여 수행하였다. 본 연구에서 제조된 막의 경우, 수착과 투과단계에서 물에 대해 좋은 선택성을 나타냈고, 공급액의 농도 50 wt % 이상에서 막의 투과선택도는 가교제의 함량에 따른 팽윤비 변화보다 수착선택도와 보다 많은 상관 관계를 나타내었다. 이러한 거동을 투과 선택도와 수착선택도과 관련된 용해-확산 모델로 고찰한 결과, 투과선택도는 막 내에서의 확산단계에서 보다 오히려 수착단계에서 결정됨을 결론지을 수 있다.
초임계 유체 공정을 이용하여 가교 폴리에틸렌 수지를 탈가교화하여 재활용하는 방법을 조사하였다. 가교 폴리에틸렌 수지는 메탄올을 이용한 초임계 반응 시 탈가교화가 이루어져 재활용 될 수 있다. 초임계를 이용한 탈가교화 반응은 반응온도에 의해 크게 영향을 받으며 $360^{\circ}C$부터 $400^{\circ}C$ 구간에서 온도의 증가에 따라 탈가교화가 급격히 촉진되었다. 반응온도의 증가에 따라 생성물인 탈가교 폴리에틸렌의 가교도, 분자량, 기계적 물성은 감소했지만, $360^{\circ}C$에서 탈가교된 폴리에틸렌은 가교전의 원료로 사용된 폴리에틸렌과 비교할만한 연신강도와 내충격강도를 보였다. 본 연구의 모든 반응조건에서 폴리에틸렌의 주쇄의 화학구조는 반응조건에 거의 영향을 받지 않았다.
In order to construct a controlled release system of drugs and to reduce toxic side effects of 5-fluorouracil, the novel ramose chitosan-based-5-fluorouracil microspheres (CS-FU-MS) were prepared. Firstly, using chitosan (CS) as carriers and 5-fluorouracil (5-FU) as a model drug, ramose chitosan-based-5-fluorouracil (CS-FU) was efciently synthesized by chemical crosslinking method through microwave irradiation, drug loading was 10.6%; Secondly, CS-FU-MS were prepared by CS-FU self-assembled under the dialysis conditions and the free 5-FU was encapsulated further at the same time. The size dispersivity of particles is uniform, and the average diameter of the CS-FU-MS was $4{\mu}m$. The drug encapsulation efficiency was 76.1%, and the drug loading was increased to 26.22%. CS-FU-MS maintain the zero-order release time in PBS (pH = 7.4) and HCl/KCl (pH = 1.2) dialysis medium was 40h and 34h respectively, and the cumulative release were 58.89% and 79.33% in 182 h. The results showed that CS-FU-MS have excellent sustained release properties.
The imidization and cure reaction of a thermosetting phenylethynyl-terminated amic acid (LaRC PETI-5) in film form have been monitored as a function of temperature by means of a steady-state fluorescence technique using a front-face illumination method. The variation of the fluorescence emission spectra of LaRC PETI-5 can be divided into four temperature regions; Region I: below 15$0^{\circ}C$, Region II: 150-25$0^{\circ}C$, Region III: 250-35$0^{\circ}C$, and Region IV: above 35$0^{\circ}C$. The fluorescence spectra in Region I are largely influenced by residual N-methyl-2pyrrolidinone in the polymer and also slightly by partial imidization of the polymer. There is a combined effect of imidization and solvent removal on the fluorescence behavior in Region II. The spectra in Regions III and IV are due significantly to the cure reaction of LaRC PETI-5 and to a post-cure effect of the polyimide, respectively. This spectroscopic evidence indicating the transformation of the amic acid imide oligomer into the corresponding polyimide via imidization and cure, agrees well with thermal analysis results obtained previously. The intermediate stage of cure in the range of 250-30$0^{\circ}C$ predominantly influences the change of the fluorescence intensity. The later stage above 30$0^{\circ}C$ significantly influences the position of the spectrum. This fluorescence study also supports the mechanism proposed in earlier work that the crosslinking reaction takes place at the reaction sites in the conjugated polyene and the phenylethynyl end group in the polyimide chain.
Background: The root of Angelica gigas Nakai is used as a traditional herbal medicine in Korea for the treatment of many diseases. However, the poor water solubility of the active components in A. gigas Nakai is a major obstacle to its bioavailability. Methods and Results: This work aimed at enhancing the solubility of the active compounds of A. gigas Nakai by a chemical (using a surfactant) and physical (hot melt extrusion, HME) crosslinking method. Fourier transform infrared spectroscopy revealed multiple peaks in the case of the extrudate solids, attributable to new functional groups including carboxylic acid, alkynes, and benzene derivatives. Differential scanning calorimetry analysis showed that the extrudate soilid had a lower glass transition temperature ($T_g$) and enthalpy (${\Delta}H$) ($T_g:43^{\circ}C$, ${\Delta}H$ : < 6 J/g) as compared to the non-extrudate ($T_g:68.5^{\circ}C$, ${\Delta}H:123.2$) formulations. X-ray powder diffraction analysis revealed the amorphization of crystalline materials in the extrudate solid. In addition, enhanced solubility (53%), nanonization (403 nm), and a higher amount of extracted phenolic compounds were achieved in the extrudate solid than in the non-extrudate (solubility : 36%, nanonization : 1,499 nm) formulation. Among the different extrudates, acetic acid and span 80 mediated formulations showed superior extractions efficiency. Conclusions: HME successfully enhanced the production of amorphous nano dispersions of phenolic compound including decursin from extrudate solid formulations.
본 연구에서는 타이로신이 풍부한 펩타이드, Tyr-Tyr-Leu-Tyr-Tyr (YYLYY)를 이용하여 금 나노입자를 담지한 균일한 금-펩타이드 계층적 초분자 구조체의 합성에 대해 연구하였다. 펩타이드의 광가교 반응을 통해 다이타이로신 결합으로 자기조립된 펩타이드 나노입자를 합성하였고, 타이로신의 생체 광물화 특성을 이용하여 금-펩타이드 하이브리드 나노입자를 친환경적 방법으로 합성하였다. 합성된 금-펩타이드 하이브리드 나노입자는 투과 전자 현미경(TEM), 주사투과 전자 현미경(SEM), 동적 광산란(DLS), 자외선-가시광선 분광광도계(UV-Vis spectroscopy), 에너지 분산 X선 분광법(STEM-EDS), X선 회절 분석법(XRD)을 통해 분석하였다. 또한 합성된 금-펩타이드 하이브리드 나노입자는 메틸렌블루의 환원 반응에서 13.4 × 10-3 s-1의 반응속도 상수를 가지는 촉매 특성을 확인하였다.
