Plant Regeneration via in Vitro Culture of Ovule Obtain by Intergeneric Crossing Between Citrus junos Sieb. et Tanaka and Poncirus trifoliata Raf. (유자와 탱자의 속간교잡후 배주배양에 의한 식물체 유기)

  • As a basic research for breeding new varieties, reciprocal -intergeneric crosses between Citrus junos and P.trifoliata were made. F$_1$ hybrid production using in vitro ovule culture, gametogenesis, and fertilization phenomena were investigated. Frequency of fruit set resulting from crossing of Citrus junos and Poncirus Trifoliata was 16.6% while that of Poncirus Trifoliata and Citrus junos was 11.7%. Callus formation occurred well when ovules at the 6th week after pollination were cultured on MT (Murashige and Tucker) medium supplemented with zeatin 0.5 mg/L and NAA 1.0 or 3.0 mg/L. Immature ovules developed into mature embryos of the MT medium supplemented with 2,4-D 0.1 or 3.0 mg/L. Immature ovules developed into mature embryos of the MT medium supplemented with 2,4 D 0.1 or 0.5 mg/L. The invitro germination rates of 20-week-old ovules set C. junos $\times$ P. Trifoliata and P. Trifoliata $\times$ C. junos were 54.5% and 48.6%, respectively. The emergence ratios of trifoliate hybrids obtained by C. junos $\times$ P. Trifoliata and P. Trifoliata $\times$ C. junos were 56.7% and 100%, respectively. The chromosome number of C. junos and P. Trifoliata was n = 9 or 2n = 18, and the sizes of their pollen grain were 33.75 $\mu$ and 25.0 $\mu$. The length and width of embryo sac in C. junos and P. Trifoliata were 69.38~79.23 $\mu$ and 27.50~38.56 $\mu$, and those of egg cells were 17.50~41.50 $\mu$ and 6.25~8.12$\mu$. Fertilization of C. junos and P. trifoliata terminated 72 h after pollination.

Growth, Flowering, and Ripening Seed Characteristics of Mammillaria goldii, M. theresae, and M. pseudopectinata (선인장 Mammillaria goldii, M. theresae와 M. pseudopectinata의 생육, 개화 및 종자형성 특성)

  • This experiment was conducted to investigate the propagation system by seed to check growth, flowering, and making seed of three-year-old cactus of potted Mammillaria goldii. M. theresae, and M. pseudopeclinata. Plant height and width of them was 2.5~2.8 cm, and 2.8~3.1 cm, and seed formation of Mammillaria goldii and M. theresae had internal body taking130 to 135 days to ripening seeds, however, M. pseudopectinata had externals taking just 26 days to ripening seeds. The mean seeded pod from April to June of M. goldii was 50.0% and the number of seeds in a pod was 25.3. In M. theresae, the mean seeded pod from April to June was 57.3% and the number of seeds in a pod was 35.0. However, the highest rate of seeded pod on May of M. pseudopectinata was 75.0% and the number of seed in a pod was 66.0. The sib crossing was much more ripening seed fertilization than that of self cross ing in the Mammillaria. The self crossing was formed from 12.0% to 20.0% of seeded pod, from 12.0 to 16.0 number of seed in a pod. However, sib crossing obtained 58.0% of seeded pod, 30 seeds in a pod of M. goldii, 65.0% seeded pods and 40 seeds in a pod of M. theresae, and 75.0% seeded pod, 68 seeds in a pod of M. pseudopectinala as the highest of them.

Cross Fertility between the Wild Japanese Quail in the Weishan Lake Area and Domestic Quail

  • Cross fertility between wild Japanese quail and domestic quail was explored in an experiment conducted on 18(3♂, 15♀)wild Japanese quails in Weishan Lake area, 18(3♂, 15♀)medium-sized domestic quails and 18(3♂, 15♀)pint-sized domestic quails, which were divided into nine groups. This study demonstrated that wild quail could succeed in crossing with domestic quail,producing fertilized eggs and hatching first filial generation. The findings indicated that there were no reproduction isolation between the wild Japanese quail and domestic quail, and that the best cross combination was between wild male quail and medium-sized domestic female quail, in which the fertility rate and hatchability of fertilized eggs amounted to 42.86% and 29.63% respectively. Based on the results, a new way could be adopted to protect, exploit and utilize genetic resources of wild quail.

Ontogenetic Behavior and Phototactic Properties of Interspecific Hybrid Prolarvae Produced by Crossing Female Siberian Sturgeon Acipenser baerii and Male Russian Sturgeon Acipenser gueldenstaedtii (시베리아 철갑상어(Acipenser baerii) 암컷과 러시아 철갑상어(Acipenser gueldenstaedtii) 수컷간 유도된 잡종 자어의 행동 발달 및 주광성 특징)

  • Female Siberian sturgeon Acipenser baerii were crossed with male Russian sturgeon A. gueldenstaedtii and the development, ontogenetic behavior, and phototactic characteristics of the resultant hybrid prolarvae were examined. The fertilization rate of the hybrid group was similar to that of Siberian sturgeon (the maternal species), and the overall developmental characteristics of hybrid embryos were not significantly different from those of either parental species. The time window from first hatch to the completion of hatching was wider in Russian sturgeon than in Siberian and hybrid sturgeon groups. The prolarval viability of the hybrid group was similar to that of the maternal species, and significantly higher than that of the paternal species. Hybrid prolarvae displayed ontogenetic behavioral patterns that were quite similar to those of Siberian sturgeon. In an illumination preference test, hybrid prolarvae displayed significant positive phototaxis, similar to the maternal species, and clearly distinct from the negative phototactic character of the paternal species. Taken together, our data indicate that the prolarval traits of hybrid sturgeon from this cross more closely resemble those of the maternal than the paternal species.

Single Crossing Condition of Miscanthus sacchariflorus and Miscanthus sinensis to Breed Miscanthus x giganteus Cultivar (이질3배체 억새(Miscanthus x giganteus) 품종육성을 위한 물억새(M. sacchariflorus)와 참억새(M. sinensis) 단교배 조건구명)

  • This study was conducted to investigate single crossing condition of M. sacchariflrous and M. sinensis for breeding of M. ${\times}$ giganteus cultivar. Compared with natural day length condition, cultivation in short day length condition shorten days to heading to 18~27 days in both species. Pollen germination ratio of were 75.8% at 6 o'clock in M. sacchariflorus and 51.9% at 7 o'clock in M. sinensis but decreased to below 10% at 8 o'clock in both species. When cut ears immerged in 150 mL of cut-flowers conservation solution and isolated with covering of white non-woven fabric, flowering and pollen dispersal were persisted for 7 days, and the ratio of pollen germination were above 40% for 4 days. The ratio of self-fertilization of both species were below 2.5%, but open pollenation ratio were above 50%. We obtained 437 seeds with experimental single cross of 14 combinations between tetraploid M. sacchariflorus and diploid M. siensis by application of developed single crossing methods. In the single cross, numbers of seed set were different by mother plants. Thus, the newly investigated single crossing condition will be used to breed M. ${\times}$ giganteous cultivar which is sterile and has superior characteristics of biomass yield.

Production of Interspecific Hybrids Using Lilium Asiatic Hybrid 'Tiny Ghost' as Female Parent (Lilium Asiatic hybrid 'Tiny Ghost'를 모본으로 한 종간잡종 생산)

  • For effective production of interspecific hybrids of Lilium and valuable new cultivars, we used Lilium Asiatic hybrid 'Tiny Ghost' as the female parent, and Oriental group, and Longiflorum group and Martagon as the male parents. Percentage pollen germination in Oriental hybrid 'Sorbonne' was the highest with 64%. As results of using normal style pollination and cut style pollination methods, all seed sets were formed in normal style pollinated treatment, except in 'Aktiva'. Pollen germination percent of Longiflorum hybrid 'Norina' was higher than that 'Gelria'. However, all crossings using 'Norina' failed to set seeds. Corresponding fertilization ratio was different from genetic compatibility, even in the same group. From the crossing using 'Aktiva' as the male parent one bulb was obtained derived from one embryo sac. And using 'Sorbonne' as the male parent, one plantlet was obtained after four ovule cultures. Crossing using 'Gelria' as the male parent five embryos, 15 embryo sacs, and seven ovules per one ovary were obtained. Among them, 18 plantlets germinated, with the germination percent of 66.7%. Crossings using L. hansonii, Martagon group, as the male parent resulted in 40% germination from five ovules.

Studies on the Production of All-Female Populations of Olive Flounder, Paralichthys olivaceus II. Progeny Test from Sex-Reversed Gynogenetc Diploid Male (넙치 전 암컷 집단의 생산을 위한 연구 II. 성전환된 자성발생성 2배체 수컷의 자손 검정)

  • A study was carried out to test sex ratio of the progeny that were induced by the artificial fertilization between normal female and gynogenetic diploid eggs and sex-reversed gynogenetic diploid male sperm. Sex reversed gynogenetic diploid males were produced successfully in gynogenetic diploid female by immersion treatment with $17{\alpha}$-methyltestosterone at the concentration of 10 pub from $13.5{\pm}0.22$ mm to $57.6{\pm}5.1$ mm in total length. In $F_1$ progeny test, matings between normal females and sex-reversed gynogenetic males were not produced all female, but matings between gynogenetic diploid females and sex-reversed gynogenetic males were produced all female populations in six out of eight groups and two lots produced $96.9\%$ and $95\%$ female. These data showed that all-female populations were produced by crossing between gynogenetic diploid female and sex reversed gynogenetic diploid male.

Breeding of Lilium Asiatic 'Lovely LiASong' as an F1 Hybrid for Pot (일대잡종 분화용 아시아틱나리 'Lovely LiASong' 육성)

  • There is a need to breed and distribute domestic varieties for Lilium Asiatic hybrids because most of the lily bulbs cultivated in Korea imported from foreign country. The genetic resources of Lilium Asiatic hybrids, 'Buff Pixie', 'Orange Matrix' and 'Tiny Puppet', which were collected from 2001, had been self-pollinated from 2004 to 2012. In 2012, Lilium Asiatic 'Lovely LiASong' as an F1 hybrid was bred through crossing 'L2-12-1' and 'L2-35-4' obtained by 4th self-fertilization. 'Lovely LiASong' had a bowl shaped single flower with a few spots and medium-sized flower, 8.7cm in flower diameter. The number of flowers per plant was 4.9, which was relatively more flowers than other cultivars. And the flower inflorescence had umbellate and racemose while the attitude of longitudinal axis was erect. The plant height was 33.4cm with 30.4 leaves per plant and 7.2cm in leaf length. The 'Lovely LiASong' showed a high consumer preference because it had a relatively longer flowering period, larger flower diameter and number of flowers than those of the control, 'Tiny Ghost'.
