Genetic Parameter Estimation with Normal and Poisson Error Mixed Models for Teat Number of Swine

  • The teat number of a sow plays an important role for weaning pigs and has been utilized in selection of swine breeding stock. Various linear models have been employed for genetic analyses of teat number although the teat number can be considered as a count trait. Theoretically, Poisson error mixed models are more appropriate for count traits than Normal error mixed models. In this study, the two models were compared by analyzing data simulated with Poisson error. Considering the mean square errors and correlation coefficients between observed and fitted values, the Poisson generalized linear mixed model (PGLMM) fit the data better than the Normal error mixed model. Also these two models were applied to analyzing teat numbers in four breeds of swine (Landrace, Yorkshire, crossbred of Landrace and Yorkshire, crossbred of Landrace, Yorkshire, and Chinese indigenous Min pig) collected in China. However, when analyzed with the field data, the Normal error mixed model, on the contrary, fit better for all the breeds than the PGLMM. The results from both simulated and field data indicate that teat numbers of swine might not have variance equal to mean and thus not have a Poisson distribution.

Bayesian Inference on Variance Components Using Gibbs Sampling with Various Priors

  • Data for teat number for Landrace (L), Yorkshire (Y), crossbred of Landrace and Yorkshire (LY), and crossbred of Landrace, Yorkshire and Chinese indigenous Min Pig (LYM) were analyzed using Gibbs sampling. In Bayesian inference, flat priors and some informative priors were used to examine their influence on posterior estimates. The posterior mean estimates of heritabilities with flat priors were $0.661{\pm}0.035$ for L, $0.540{\pm}0.072$ for Y, $0.789{\pm}0.074$ for LY, and $0.577{\pm}0.058$ for LYM, and they did not differ (p>0.05) from their corresponding estimates of REML. When inverse Gamma densities for variance components were used as priors with the shape parameter of 4, the posterior estimates were still corresponding (p>0.05) to REML estimates and mean estimates using Gibbs sampling with flat priors. However, when the inverse Gamma densities with the shape parameter of 10 were utilized, some posterior estimates differed (p<0.10) from REML estimates and/or from other Gibbs mean estimates. The use of moderate degree of belief was influential to the posterior estimates, especially for Y and for LY where data sizes were small. When the data size is small, REML estimates of variance components have unknown distributions. On the other hand, Bayesian approach gives exact posterior densities of variance components. However, when the data size is small and prior knowledge is lacked, researchers should be careful with even moderate priors.

Optimization of Culture Conditions for Maintaining Pig Muscle Stem Cells In Vitro

  • Muscle stem cells isolated from domestic animals, including cows and pigs, were recently spotlighted as candidates for the production of alternative protein resources, so-called cultured meat or lab-grown meat. In the present study, we aimed to optimize the in vitro culture conditions for the long-term expansion of pig muscle stem cells via the screening of various signaling molecules. Pig muscle stem cells were collected from the biceps femoris muscles of 3-d-old crossbred pigs (Landrace×Yorkshire×Duroc, LYD) and cultured in minimum essential medium-based growth media. However, the pig muscle stem cells gradually lost their proliferation ability and featured morphologies during the long-term culture over two weeks. To find suitable in vitro culture conditions for an extended period, skeletal muscle growth medium-2, including epidermal growth factor (EGF), dexamethasone, and a p38 inhibitor (SB203580), was used to support the stemness of the pig muscle stem cells. Interestingly, pig muscle stem cells were stably maintained in a long-term culture without loss of the expression of myogenic marker genes as determined by PCR analysis. Immunostaining analysis showed that the stem cells were capable of myogenic differentiation after multiple passaging. Therefore, we found that basal culture conditions containing EGF, dexamethasone, and a p38 inhibitor were suitable for maintaining pig muscle stem cells during expanded culture in vitro. This culture method may be applied for the production of cultured meat and further basic research on muscle development in the pig.

돼지 품종이 등심의 일반성분, 물리화학적, 콜레스테롤, 아미노산, 지방산 조성 및 관능적 품질 특성에 미치는 영향 (The Effects of Pig Breeds on Proximate, Physicochemical, Cholesterol, Amino Acid, Fatty Acid and Sensory Properties of Loins)

  • [Landrace×Yorkshire]×Duroc(LYD) 및 [Yorkshire×Berkshire]×Berkshire(YBB) 삼원교잡종과 흑색계 영국 Berkshire(BB; British berkshire), 가고시마 Berkshire(KB; Kagoshima Berkshire), [재래돼지×멧돼지(KNW; Native black pig×Wild boars)] 이원교잡종에서 생산된 돼지 100두를 생체중 110 kg 도달시 도축한 후 등심을 채취하여 분석하였다. 수분 함량은 KW, 조단백질은 YBB와 KW, 콜레스테롤 함량은 YBB가 다른 품종들보다 유의적으로 높았다(p<0.05). 다른 품종들에 비해 YBB는 보수력과 pH는 낮고, 전단가는 높았다(p<0.05). LYD에서 명도(L*)가 유의적으로 높게 나타났으며(p<0.05), 적색도(a*)와 파쇄성에서는 품종들 간에 유의적인 차이가 없었다(p>0.05). UFA/SFA와 EFA/UFA 비율과 EAA은 KW가 높게 나타났으나(p<0.05) SAAA는 유의적으로 낮았다(p<0.05). 총 아미노산 함량은 LYD와 BB가 각각 20.44%, 20.81%로 다른 품종에 비해 유의적으로 많았다(p<0.05). 신선육 상태에서 품종은 육색, 드립로스, 마블링과 전체적인 기호도에 영향을 미쳤지만(p<0.05), 조리육의 경우 풍미를 제외한 관능검사 항목에는 영향을 미치지 않았다(p>0.05).

동결 기간 및 해동 후 냉장이 제주 흑돼지고기의 물리화학적, 기호적 품질에 미치는 영향 (Effects of Freezing Period and Chilling Process after Thawing on Physicochemical Properties and Palatability of Loin from Jeju Island Reared Crossbred Black Pigs)

  • 개량흑돼지(105~110 kg)를 도축한 후 약 18시간 냉장하고 등심육을 분할하여 진공 포장하고 $-20^{\circ}C$에서 1개월 및12개월 동결한 후 각각 $4^{\circ}C$에서 20시간 해동하였다. 해동 직후의 것(0-TP)과 해동 후 $2^{\circ}C$에서 2일간 냉장한 것(2-TP)의 물리화학적 및 기호적 특성을 비교하여 해동 후 냉장이 품질에 미치는 영향을 검토하였다. 개량흑돼지 등심육을 1개월간 동결하여 해동한 경우, 해동 직후의 0-TP보다 해동 후 냉장한 2-TP가 보수력 및 근원섬유 소편화 정도가 높아지고, 경도 및 씹힘성이 낮아졌으며, 가열육의 다즙성 및 종합적 기호도가 향상되었으나(p<0.05) 12개월 동결한 등심육은 해동 후 냉장에 의한 효과가 유의적으로 크지 않았다.

Influence of Maize Cob Inclusion Level in Pig Diets on Growth Performance and Carcass Traits of Mukota × Large White F1 Crossbred Male Pigs

  • A trial was conducted to evaluate the growth performance and carcass characteristics of LW ${\times}$ Mukota $F_1$ crosses when fed diets containing graded levels of maize cob meal. Sixteen LW ${\times}$ Mukota $F_1$ crossbred pigs of approximately 4.5 months of age, were randomly allocated to four diets that contained 0, 100, 200 and 300 g maize cobs/kg, which corresponded to 276.4, 360.3, 402.9 and 523.5 g NDF/kg, respectively. The pigs were fed ad libitum for 14 weeks. The diets were formulated to contain similar levels of energy (ca. 9MJ ME/kg) and protein (ca.160 g CP/kg). Average daily feed intake (ADFI), daily gain (ADG) and feed conversion ratio (FCR) were monitored for 14 weeks. At slaughter, the cold dressed weight (CDM) and backfat thickness (BFT) were determined. There was no significant linear relationship (p>0.05) between level of cob inclusion and ADFI. The ADG decreased as level of maize cobs increased (p<0.001). The pigs that were on 300 g cobs/kg had the highest FCR (p<0.05) as compared to pigs on the other three diets. No differences (p>0.05) were observed in the CDM between pigs that were fed diets that had 0 and 100 g maize cobs/kg. In addition, pigs on 100 and 200 g cobs/kg diets had the same CDM (p>0.05). The diet that contained 300 g cobs/kg gave the lowest (p< 0.001) CDM. Both BFT parameters showed a decrease (p<0.05) as level of maize cobs increased. An increase in CDM was associated with an increase in BFT (p<0.001), with the correlation coefficient between K7.5 and CDM being 0.84 (p<0.001). It can, therefore, be concluded that crossbred pigs perform well on diets containing up to 200 g/kg maize cobs.

Physicochemical Traits, Fatty Acid and Free Amino Acid Compositions of Two-way Crossbred Pork Belly

  • This study was conducted to determine the meat quality characteristics of pork belly from 3 different two-way crossbreeds of Yorkshire${\times}$Landrace (YL), Yorkshire${\times}$Berkshire (YB), and Yorkshire${\times}$Chester White (YC), which were domesticated for Korean consumers. Twenty pigs from each crossbreed (total n=60) were randomly selected when they reached the 110-120 kg range of market weight, slaughtered, and cooled at $0^{\circ}C$ for 24 h. The pork bellies on the left side of the cooled carcasses were then sampled and analyzed. The pH of pork bellies was the lowest in YC among the crossbreds. There was no significant difference in fat content by crossbred, but YB bellies had the lowest moisture content (p<0.05). The cooking loss of YB bellies was lower than those of others (p<0.05). The TBARS values in YB was significantly higher than those of the others at 14 d. YL bellies had a higher percentage of stearic acid, oleic acid, and MUFA than the other breeds, while YB and YC had a higher percentage of myristic acid, linoleic acid, linolenic acid, and n-6 fatty acids than the YB (p<0.05). PUFA content and P/S were significantly higher in YC compared with YL. Except for arginine, the concentrations of most free amino acids were higher in YB bellies than in others, (p<0.05). Sensory evaluation scores of bellies were higher for YC than for other breeds (p<0.05).

Meat Quality of Crossbred Porkers without the Gene RYR1T Depending on Slaughter Weight

  • The first aim of the study was to compare selected meat quality parameters in porkers without the gene $RYR1^T$ (ryanodine receptor gene). These were porkers slaughtered at 100 to 115 kg and 116 to 130 kg live weight. The second aim of the study was to determine the occurrence frequency of standard-quality meat (red, firm, nonexudative [RFN]) and the occurence frequency of defective meat (pale, soft, exudative [PSE] and acid, soft, exudative [ASE]). The analysis was conducted on the longissimus lumborum muscle in 114 crossbred porkers. The porkers were a cross of Camborough 22 sows and boars from lines 337PIC (Pig Improvement Company), Norsvin Landrace and Pietrain. All of the animals were provided with identical environmental and nutritional conditions. The average weight of the slaughtered animals in the light and heavy groups was 110 kg and 122 kg, respectively. Both groups had the same average post-slaughter meatiness (56.5%). A statistical analysis of selected meat-quality parameters did not show any significant differences between the weight groups. On the other hand, the classification based on carcass quality showed an occurence frequency of defective meat in heavier crossbred porkers (116 to 130 kg) that was three times higher than in those cross bred animals which weighed 100 to 115 kg when slaughtered. In porkers without the gene $RYR1^T$, the defective meat types PSE and ASE occurred with a frequency of 17.54%.

Effect of Dietary Fiber Level on the Performance and Carcass Traits of Mong Cai, F1 Crossbred (Mong Cai×Yorkshire) and Landrace×Yorkshire Pigs

  • The effects of feeding diets containing 20% (L) or 30% (H) neutral detergent fiber (NDF) (DM basis) on performance and carcass traits were studied in three breeds of pig, including pure Mong Cai (MC), crossbred Landrace$\times$Yorkshire (LY) and crossbred MC$\times$Yorkshire (F1). The experiment had a factorial design with two factors, breed and diet. Eighteen piglets of each breed ($60{\pm}3days$) were randomly allocated to three treatments: L-L, low fiber diet in both growing and finishing periods; L-H, low and high fiber diet in the growing and finishing period, respectively; and H-H, high fiber diet in both periods. The diets were iso-energetic and iso-nitrogenous within feeding period. The main fibrous ingredients of the diets were rice bran and cassava residue. There were no effects of fiber level on daily dry matter feed intake (DMI), expressed as g/kg metabolic body weight (BW0.75), in both feeding periods (p>0.05). DMI was highest for MC, followed by F1 and LY (p<0.001). Average daily gain (ADG) in L-L and L-H was higher than in H-H in the growing period (p<0.001) and overall (p<0.05), while feed conversion ratio (FCR) was higher in H-H than in L-L and L-H in the growing period (p<0.05) and overall, but no significant differences between treatments were found in the finishing period. In both periods, Landrace$\times$Yorkshire had the highest ADG and the lowest FCR, followed by F1 and Mong Cai (p<0.001). There were no interactions between breed and diet for performance and carcass traits. Carcass and dressing percentage was lower for L-H and H-H than for L-L (p<0.05). There were no significant differences among treatments in back fat thickness and lean meat percentage, or in crude protein and ether extract contents of lean meat. Carcass, dressing and lean meat percentage was highest for LY, lowest for MC and intermediate for F1 (p<0.001). It can be concluded that feeding a high fiber diet in the growing period reduced pig performance, but there was no effect in the finishing period. Pure Mong Cai pigs are not particularly suitable for meat purposes, although the F1 cross with Large White had reasonably good growth performance and carcass quality.

감귤피 첨가 사료를 장기간 급여한 교잡종 돼지고기의 물리화학적 특성과 기호성 (Effects of Long Term Tangerine Peel Consumption on the Physicochemical Properties and Palatability of Crossbred Pig Meats)

  • 감귤피를 급여하지 않은 돼지고기(대조구, T0)와 임신초기부터 분만 후 자돈기, 육성기 및 비육기까지 전 사육기간 동안 8%의 감귤피를 급여한 돼지고기(감귤피 급여구, T1, 198일령, $102{\sim}118kg$)의 물리화학적 특성과 기호도를 비교하였다. 등심과 삼겹살의 pH, 명도, 보수력, 동결감량, 해동 감량, 가열감량, 경도, 탄성, 응집성, 생육의 색깔 및 가열육의 종합적 기호도는 T0와 T1 사이에 유의적 차이를 보이지 않았다(p>0.05). 등심의 적색도, 황색도, 뭉침성, 저작성 및 전단력, 그리고 삼겹살의 항산화력, 전단력, 생육의 냄새 및 가열육향은 T0보다 T1이 높게 나타났다(p<0.05).
