Possibility of breeding super rice cultivars using gene linkage

  • The rapid growth of world population, reduction of cultivated areas for crop production, and detrimental effects of pests, diseases, and climate changes have required to breed new rice cultivars with high yield, accepted quality, but strong resistance to abiotic and biotic stresses. However, traditional breeding needs much time to breed a new cultivar, whereas the successful use of molecular breeding is still questionable. We have developed a novel mutation which allow to cross many rice cultivars together with low segregation, that allow to breed a new cultivar in only several cropping. The mechanism has been unknown, but we suggest that gene linkage may play a crucial role, of which the semi dwarf gene might be the center gene for gene linkage occurrence. The phenomenon of this possible gene linkage is contrary to Mendel rules, but it is promising to breed new rice cultivars, of which, the most elite genes in rice might be able to gather in a targeted rice variety.

  • The wild Pyrus plants belong to the deciduous woody plants in Rosaceae family. Most are cultivated for fruit productions, but some for ornamental plants. There are 5 Pyrus species including Pyrus ussuriensis and 8 varieties cultivated in Korea. In Korea, these plants are populated near villages or on the hills, and they are easily propagated since their fruit are very Juicy and quite big In size. Plant breeding to get improved cultivars Is not difficult because the Pyrus plants are easy to cross breeding among its genus. Because not only this reason but there have been shown lots of variation in wild groups, the Pyrus plants inhabited In Korea are excellent deciduous woody plants and favorable fruit trees to develop ornamental plants. It is necessary to develop many kinds of fruit trees and ornamental trees containing genetically different character, so that it will be satisfied for many different people's taste and demands Collecting lots of wild plant resource with diverse genes is necessary for plant breeding.

  • KB103 was developed from a cross between Burley 21 and Va 528 in 1983. It was developed by haploid method derived from anther culture method. It has moderate resistance to black shank(Phytophhorn nicotianae Breda de Haan Var. nictiana Waterhouse). KB103 produced good yields and quality in 1988-1990. It flowered about four or five days later than standard variety Burley 21, and plant height, leaf width, leaves per plants were similar to those of Burley 21. In maturing and curing properties as well as in chemical constituents of cured leaves, KB 103 was very similar to that of Burley 21. Regional farm test with the breeding line, KB 8304- ADH 65 named as KB 103 in 1986, was performed for 4 crop years from 1987 at 29 locations. It was released on burley tobacco area in 1992.

An Asiatic Lily New Cultivar "Hyehwa" with Unspotted Orange Petals

  • "Hyehwa" an Asiatic lily cultivar was released in 1998 at National Institute of Horticultural and Herbal Science (RDA), Suwon, Korea. The cross was made in 1991 between Asiatic lily "White Bird", a white colored, and "Avignon", an unspotted scarlet red colored. It was preliminarily selected as A93-20 in 1993. Its multiplication, bulbing growth and flowering characteristic tests were conducted from 1994 to 1998. A new cultivar "Hyehwa" flowers in middle of June and grows 98.7 cm in height. Flowers bloom upward-facing, thick orange (RHS, 28A). Year-round flowering is possible by storage of the bulb under $-1.5^{\circ}C$ conditions. For forcing culture, it is necessary to add calcium to the fertilizer or remove side scales to prevent leaf scorch. Botrytis disease control is needed in the wet season.

A New Gerbera Cultivar "Pink Swan"; High-yielding Cultivar with Pink and Semi-double for Cut-flower

  • A new gerbera variety "Pink Swan" was released by the National Horticultural Research Institute (NHRI) in 2007. A cross was made between "Royal" with orange and single and 'Himalaya' with white and semi-double in 2004. After investigation of the characteristics for 4 years (from 2004 to 2007), it was selected specially for the use of cut-flower. "Pink Swan" is a vigorous medium-sized cultivar with pink (RHS R48D), yellow green center and semi-double. The average flower diameter is 10.6 cm. And inner ray floret and disc diameter is 5.99cm and 1.83cm, respectively. It produces a large yield with continuous supply of flowers and has adequate peduncle height. We believe that "Pink Swan" has a great potential for expanding cultivated area of domestic gerbera cultivar and increasing farmers-income.

Multiplication and bulbing, and characteristic tests were performed from 1997 to 2000. The evaluation of characteristics was made investigated as 'Wongyo C1-31' in 2001 at Suwon. 'Honghwa' flowers at the beginning of June and grows to 111.4 cm stem length. Flowers are upward-facing, unspotted with orange petals. Year-round flowering is possible by storing the bulb at $-1.5^{\circ}C$ conditions. It is necessary to add calcium to the fertilizer or remove side scales to prevent leaf scorch. It is needed to control Botrytis disease during wet season.

We believe that 'Yellow Wing' has a great potential for expanding cultivated area of domestic gerbera cultivar and increasing farmers-income.

Non-pungent Capsicum Contains a Deletion in the Capsaicinoid Synthetase Gene, which Allows Early Detection of Pungency with SCAR Markers

  • The capsaicinoid synthetase (CS) gene cosegregated perfectly with the C locus, which controls the presence of pungency, in 121 $F_2$ individuals from a cross between 'ECW123R' and 'CM334', both of Capsicum annuum. We concluded that CS and C are tightly linked. Sequence analysis of the genes of four pungent and four non-pungent pepper lines showed that the non-pungent peppers had a 2,529 bp-deletion in the 5' upstream region of CS. We have developed molecular markers of the C locus to detect pungency at the seedling stage. Based on the deleted sequence, we developed five SCAR markers, two of them being codominant. These SCAR markers will be useful for easy, accurate, and early detection of non-pungent individuals in breeding programs.

Cloning and Characterization of Liver cDNAs That Are Differentially Expressed between Chicken Hybrids and Their Parents

  • Using mRNA differential display technique, we investigated differential gene expression in hybrids relative to their parents in a diallel cross involving four chicken breeds in order to provide an insight into the molecular basis of heterosis in chicken. The results indicated that there was extensive differential gene expression between chicken F1 hybrids and their parents which was classified into four kinds of patterns as following: (1) bands only detected in hybrid F1; (2) bands only absent in hybrid F1; (3) bands only detected in parent P1 or P2; (4) bands absent in parent P1 or P2. Forty-two differentially expressed cDNAs were cloned and sequenced, and their expression patterns were confirmed by Reverse-Northern dot blot. Sequence analysis and database searches revealed that genes showed differential expression between hybrid and parents were regulatory and functional genes involved in metabolism, mRNA splicing, transcriptional regulation, cell cycles and protein modification. These results indicated that hybridization between two parents can cause changes in expression of a variety of genes. In conclusion, that the altered pattern of gene expression in hybrids may be responsible for heterosis in chickens.
