항생제 다제내성 Pseudomonas aeruginosa 및 Candida 균주에 대한 산사자의 항균 활성 (Antimicrobial Activity of Fruit of Crataegus pinnatifida Bunge against Multidrug Resistant Pathogenic Pseudomonas aeruginosa and Candida sp.)

  • 산사자는 전 세계적으로 이용되는 있는 식용/약용 생물자원 중 하나이다. 본 연구에서는 산사자의 유용 생리활성 검토를 위한 연구의 일환으로, 산사자의 methanol 추출물 및 이의 n-hexane, ethylacetate, butanol 분획물 및 물 잔류물을 조제하여 항생제 다제내성 Pseudomonas aeruginosa 및 Candida sp.를 포함하는 다양한 병원성 및 식중독 미생물에 대한 항균활성을 평가하였다. 산사자의 methan이 추출물은 그람 양성 및 음성의 다양한 세균에 대해 항균활성을 나타내였고, 이의 분획물 중 ethylacetate 및 butanol 분획물은 Listeria monocytogen, Staphylococcus epidermidis, Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Proteus vulgaris, Escherichia coli는 물론 10종의 항생제 내성 병원성 Pseudomonas aeruginosa에 대해서도 우수한 항세균 활성을 나타내었다(최소생육억제농도 1.0~7.5 mg/mL). 또한 ethylacetate 및 butan이 분획물은 일부의 Candida sp.에 대해서도 항균활성을 나타내였다. 한편 n-hexane 분획물을 제외한 산사자 methan이 추출물 및 분획물들은 $500\;{\mu}g/mL$ 농도까지 인간적혈구에 대한 용혈현상을 보이지 않았으며, n-hexane 분획물은 $500\;{\mu}g/mL$ 농도에서 약 9.9%의 미미한 용혈활성을 나타내었다. 이러한 결과는 산사자가 다양한 세균의 제어는 물론 항생제 내성 Pseudomonas aeruginosa 제어를 위한 생물자원으로 개발 기능함을 제시하고 있다.

First Report of Gymnosporangium globosum Causing American Hawthorn Rust in Korea

  • Field surveys and specimen collections of the rust fungal pathogen Gymnosporangium were carried out for 15 years from 1985 through 1999 in various locations of Korea. Macroscopic and microscopic examinations of morphological characteristics of aecia from the collected specimens revealed that Gymnosporangium globosum is the causal agent of American hawthorn rust disease on Crataegus pinnatifida and C. pinnatifida varmajor. The host plants are new for this rust fungus. G. globosum was found only in Gyeonggi and Chungbuk provinces, indicating that its distribution in Korea is limited. This is a first full description on morphological characters of aecia of G. globosum in Korea.

한약재 열수 추출물의 변이원 및 항변이원 활성의 검색 (Screening of the Mutagenicity and Antimutagenicity of the Hot Water Extracts from Medicinal Plants)

  • 130종의 식물성 한약재에서 제조한 열수 추출물의 변이원성, 항변이원성 및 세포독성을 측정하였다. 세포내재성 alkaline phosphatase의 활성을 지표로 한약재가 지시세포인 E. coli PQ37의 성장에 미치는 영향을 조사한 결과, 연교(Forsythia koreana Nakai)를 포함한 14종류의 시료자체에서 세포독성이 강하게 나타난 반면, 속단(Phlomis umbrosa Turcz.)등 22종의 시료에서는 세포성장을 촉진하는 활성이 나타났다. SOS chromotest기법을 이용하여 한약재가 가지는 변이원성 및 항변이원성을 조사한 결과, 연교(Forsythia koreana Nakai)와 황련(Coptis japonica Nakai)을 포함한 36종의 시료에서 돌연변이를 유발시키는 능력이 나타난 반면, 산사(Crataegus pinnatifida Bunge)를 포함한 12종의 한약재에서 뚜렷한 항변이원 활성이 검정되었다.

Differential role of endothelium in hawthorn fruit extract-induced relaxation of rat cerebral, coronary, carotid, and aorta

  • The present study was aimed to examine the role of endothelium in the relaxant effect of hawthorn fruit extract of Crataegus pinnatifida in four different types of rat arteries, posterior cerebral communicating artery, right descending coronary artery, common carotid artery, and aorta. In $9,11-dideoxy-11{\alpha}$, $9{\alpha}-epoxy-methanoprostaglandin$ $F_{2{\alpha}}$ (U46619)-preconstricted arterial rings except for aorta, the extract produced endothelium-independent relaxations with similar potency. This relaxation was unaffected by pretreatment with $100\;{\mu}M\;N^G-nitro-L-arginine$ methylester (L-NAME, the nitric oxide synthase inhibitor), $3\;{\mu}M$ 1H-[l,2,4]oxadiazolo$[4,2-{\alpha}]$quinoxalin-1-one (ODQ, the guanylate cyclase inhibitor), or $10\;{\mu}M$ indomethacin (the cyclooxygenase inhibitor). Putative $K^+$ channel blockers (charybdotoxin plus apamin or glibenclamide) did not affect the extract-induced relaxation in cerebral or coronary artery rings. In contrast, in rat aortic rings the extract produced significantly smaller relaxant response in endothelium-denuded rings than that in endothelium-intact rings. Pretreatment with L-NAME or ODQ abolished the extractinduced endothelium-dependent aortic relaxation, whilst indomethacin $(3\;{\mu}M)$ had no effect. The present results indicate that hawthorn fruit extract possesses a vasorelaxing effect in cerebral, coronary and carotid arteries and this effect is independent of the presence of a functional endothelium. However, the extract-induced endothelium-dependent relaxation in rat aorta was mediated through endothelial nitric oxide and cyclic GMP-dependent mechanisms, suggesting that active components in the extract may act on endothelium to stimulate release of nitric oxide in large conduit arteries of the rats.

Anti-hyperlipidemic Effect of Insamsansa-eum in Mice

  • Hypolipidemic effect of Insamsansa-eum (ISE, Ren-Shen-Shan-Zah-Yin in Chinese) consisted of Red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE) is used frequently in China and Korea as a herbal medicine to treat arteriosclerosis, was investigated. Treatment of ISE significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. ISE significantly lowered the high blood TG and TC levels as well as body and epididymal mass weights in hyperlipidemic mice induced by long-term feeding of a high-fat diet and increased blood HDL cholesterol level. ISE and its ingredients, RG and CF, inhibited pancreatic lipase and HMG-CoA reductase activities. Of its ingredients, RG reduced the blood TC level and HMG-CoA reductase activity more than CF. CF reduced blood TG level and pancreatic lipase activity more than RG. These finding suggest that the antihyperlipidemic effect of ISE may be due to synergistic inhibition of HMG-CoA reductase and pancreatic lipase by RG and CF, and that ISE may be effective hypolipidemic agents.

약용식물 혼합물의 고지혈증 개선 효과 (Antihyperlipidemic Effect of Complex of Medicinal Plant Products on Hyperlipidemic Rats Induced by High Cholesterol Diet)

  • The HWND_G02 (Salvia miltiorrhiza Bunge, Crataegus pinnatifida, Polygonum multiflorum Thunberg, Cnidium officinale Makino) and HWND_G03 (Cinnamonum cassia Blume, Salvia miltiorrhiza Bunge, Crataegus pinnatifida, Polygonum multiflorum Thunberg, Cnidium officinale Makino, Allium macrostemon Bunge) are new natural mixture composed with several oriental herbs. The aim of the present study was to investigate the effects of HWND extracts on high cholesterol diet (HCD)-induced hyperlipidemic rats. Male Sprague-Dawley rats were divided into five groups: control, HCD, atorvastatin (5 mg/kg, po), ethanolic extracts of HWND_G02 (1,000 mg/kg, po) and HWND_G03 (1,000 mg/kg, po) were administered to the HCD-induced hyperlipidemic rats for 4 weeks to evaluate their anti-hyperlipidemic activities. HWND extracts markedly decreased body and liver wight gain, and recovered serum lipid levels, such as total cholesterol (TC), triglycerides (TG), low density lipoprotein-cholesterol (LDL-C) and high density lipoprotein-cholesterol (HDL-C) in the HCD-induced hyperlipidemic rats. Furthermore, the lipid levels (TC and TG) and the lipid accumulation were significantly lowered in the liver tissue of HWND-administrated rats. After a HCD, each group had a lower atherogenic index (AI) compared to the HCD group. In conclusion, these data suggest that HWND extracts could be the candidate for the material to prevent hyperlipidemia.

산사(Crataegus pinnatifida Bunge)의 추출 용매별 항산화 활성 (Antioxidative activities of various solvent extracts from haw (Crataegus pinnatifida Bunge))

  • 산사과육을70% methanol, 70% ethanol, chloroform:methanol(CM, 2:1, v/v), n-butanol, ethyl acetate(EA)로 추출하여 시료농도 0.2 mg/mL, 0.5 mg/mL, 0.8 mg/mL 농도에서 각 추출물의 항산화 활성을 비교하였다. DPPH radical 소거 활성은 70% methanol, 70% ethanol, chloroform:methanol(2:1, v/v), ethyl acetate, n-butanol 추출물 순으로 높게 측정되었고 특히 70% methanol 및 70% ethanol 추출물은 다른 추출물에 비하여 유의적으로 높은 것으로 나타났으며(p<0.05), 0.8 mg/mL 농도에서 각각 92.89%, 91.17%의 높은 활성을 나타내었다. 환원력과 ferric reducing antioxidant power(FRAP)는 동일한 경향을 나타내었는데 활성은 70% methanol, 70% ethanol, n-butanol, ethyl acetate, chloroform: methanol(2:1, v/v) 추출물 순으로 높게 나타났으며, 70% methanol 및 70% ethanol 추출물이 다른 추출물에 비하여 높은 것으로 측정되었다(p<0.05). ABTS radical 소거 활성은 DPPH와 동일한 경향이었으며 0.8 mg/mL 농도에서 70% methanol 및 70% ethanol 추출물은 각각 75.81%, 74.73%의 활성을 나타내었다. NO radical 소거 활성은 70% methanol, 70% ethanol, ethyl acetate, n-butanol, chloroform:methanol(2:1, v/v) 추출물 순으로 나타났고, hydroxyl radical 소거 활성은 n-butanol, 70% methanol, 70% ethanol, chloroform: methanol(2:1, v/v), ethyl acetate 추출물 순으로 측정되었다. Hydroxyl radical 소거 활성을 제외한 다른 실험 결과에서 70% methanol 및 70% ethanol 추출물은 다른 용매 추출물에 비하여 유의적으로 높은 것으로 나타났고 chloroform: methanol(2:1, v/v), ethyl acetate, n-butanol 추출물 간에는 대부분 유의성이 없는 것으로 측정되었다(p<0.05). 본 실험 결과 산사 과육 추출물의 항산화 활성은 우수한 것으로 나타났고 특히, 70% methanol 및 70% ethanol 추출물에서 높은 활성을 보였다. 따라서 산사는 천연 항산화제 및 기능성 식품 소재로서 활용될 수 있을 것으로 생각된다.

Antioxidant Activity of Hawthorn Fruit in vitro

  • The antioxidant activity of hawthorn fruit (Crataegus pinnatifida Bunge var. typica Schneider) extracts was investigated by several in vitro antioxidants properties, including DPPH free radical scavenging activity, total phenolic and flavonoid contents, hydroxyl radical scavenging activity, reducing power activity, iron-chelating capacity and nitrite scavenging activity. Among the extracts in this study, the 70% EtOH extract showed higher antioxidant activity than the others. The $IC_{50}$ value of DPPH free radical scavenging activity was $99.26\;{\mu}g/mL$. Furthermore, the 70% EtOH extract also showed significantly high total phenolic and flavonoids contents and reducing power activity. However, the MeOH extract exhibited stronger effects on hydroxyl radical scavenging activity, iron-chelating capacity and nitrite scavenging activity. All the results implicated that, the hawthorn fruit may has the available potential to be utilize as a potential source of natural antioxidant.

Antioxidative Activities of 60 Plant Extracts

  • The methanol extracts from 60 plant extracts were prepared and evaluated for their antioxidative activity. Arctium lappa, Diospyros kaki, Eugenia caryophyllata, Melia azedarach and Forsythia suspensa showed the inhibitory activity against lipid peroxidation. Caesalpinia sappan, Crataegus pinnatifida, Eugenia caryophyllata, Gleditsia japonica, Osmunda japonica, Rhus javanica and Sanguisorba officinalis showed the highest inhibitory activity against DPPH radical formation. In particular, Eugenia caryophyllata demonstrated strong inhibitory activity on the lipid peroxidation and free radicals. The results suggested that selected plant extracts have a potential as natural antioxidant.

  • The methanol extracts of 132 herbal medicines were screened for the inhibitory activity against heparinase enzyme from Flavobacterium heparinum. Eleven medicinal plants, Amomum xanthiodides, Agrimonia pilosa, Paeonia lactiflora, Rubia cordifolia, Sanguisorba officinalis, Torrega grandis, Morus alba, Gleditsia sinensis, Crataegus pinnatifida, Cornus officinalis, Paeonia japonica showed potent inhibition on heparinase activity. The active substituents of those herbal medicine could be extracted into butanol fraction and the inhibitory compounds of Morus alba are now isolating.
