Pseudomonas putida로 부터 salicylate 분해 plasmid의 분리 (Isolation of Salicylate-Degrading Plasmid from Pseudomonas putida)

  • 자연계에서 분리한 Pseudomonas에서 크기가 매우 큰 plasmid (약 180 megadaltons:를 분리하였다. 이 plasmid는 기존의 plasmid 검출방법보다 더 온화한 조건에서 분리되었다. Alkalin., sodium dodecyl sulfate (pH 12. 45)를 사용하여 chromosomal DNA를 변성시킨 뒤 $55^{\circ}C$로 열처리 하여 covalently closed circular DNA 의 손실을 최대한으로 방지하였고, polyethylene glycol을 최종농도 10%로 첨가하여 plasmid DNA를 선택적으로 농축하였다. 한편, 이 plasmid는 mitomycin C를 처리하여 얻은 cured sitrains에서는 나타나지 않아 salicylate를 분해하는 새로운 plasmid로 확인되었다.

사과의 성숙에 따른 Pectin질의 변화 (Changes in the Pectic Substance during Maturity of Apple)

  • Fuji 과실의 성숙에 따른 pectin 질의 변화를 조사하기 위하여 LMP (low methoxy pectin), HMP (high methoxy pectin) IAP (ionically associated pectin), CBP (covalently bounded pectin), methoxyl 및 구성당의 함량변화와 DE(degree of esterification)를 조사하였다. 그 결과 LMP+HMP와 IAP+CBP는 AIS(alcohol insoluble solid)의 각각 70%, 22%를 차지하였고 이들 성분은 성숙에 따라 다소 증가하였다. Pectose 질을 구성하는 중성당중 hexose는 LMP, HMP, IAP, CBP 다같이 $20{\sim}50%$가 감소하였는데 특히 Gal의 감소율은 63%이었다. Pentose는 $1{\sim}9%$가 증가하였고 uronic acid는 $2{\sim}4%$감소하였다. 그러나 HMP, CBP에서는 uronic acid의 상대적 함유비율은$3{\sim}5%$ 증가되었다.

공유가교 CL-SPEEK/Cs-TSiA/Ceria 복합막의 고온 수전해 성능 (High Temperature Water Electrolysis of Covalently Cross-linked CL-SPEEK/Cs-TSiA/Ceria Composite Membrane)

  • The high temperature performance of PEM type electrolyser at $120^{\circ}C$ based on covalently cross-linked sulfonated poly ether ether ketone (SPEEK) composie membrane was investigated. Ion conductivity and other properties of SPEEK membrane were improved by adding heteropoly acid and Ceria. The membrane electrode assemblies were prepared using commercial PtC and nano-sized $IrRuO_2$ catalyst by electro-spraying and decal process. Voltage efficiency of MEA equipped with SPEEK membrane was slightly better than that of $Nafion^{(R)}$ membrane, due to its higher proton conductivity at high temperature. The cell performance of MEA with CL-SPEEK/Cs-TSiA/Ceria is 1.71 V at $1A/cm^2$ and $120^{\circ}C$.

Identification of Novel Irreversible Inhibitors of UDP-N-Acetylglucosamine Enolpyruvyl Transferase (MurA) from Haemophilus influenzae

  • Uridinediphospho-N-acetylglucosamine enolpyruvyl transferase (MurA, E.C. 2.5.1.7) is an essential bacterial enzyme that catalyzes the first step of the cell wall biosynthetic pathway, which involves the transfer of an enolpyruvyl group from phosphoenolpyruvate to uridinediphospho-Nacetylglucosamine. In this study, novel inhibitors of Haemophilus influenzae MurA (Hi MurA) were identified using high-throughput screening of a chemical library from the Korea Chemical Bank. The identified compounds contain a quinoline moiety and have much lower effective inhibitory concentrations ($IC_{50}$) than fosfomycin, a wellknown inhibitor of MurA. These inhibitors appear to covalently modify the sulfhydryl group of the active site cysteine (C117), since the C117D mutant Hi MurA was not inhibited by these compounds and excess dithiothreitol abolished their inhibitory activities. The increased mass value of Hi MurA after treatment with the identified inhibitor further confirmed that the active-site cysteine residue of Hi MurA is covalently modified by the inhibitor.

  • The polymer electrolyte membranes, CL-SPEEK/Cellulose composite membrane I, II, III with the improved electrochemical and mechanical properties were prepared and characterized. The engineering plastic polyether ether ketone (PEEK) and cellulose were sulfonated and cross-linked. The membranes were prepared by sol-gel casting method with different amount of cross-linking reagent. In conclusion, the composite membranes I, II, III showed improved thermostability, tensile strength and oxidative durability. Proton conductivity of the membranes was also improved and the composite membrane I showed 0.1312 S/cm at $80^{\circ}C$ which was the best of those composite membranes.

Structural Study of Monomethyl Fumarate-Bound Human GAPDH

  • Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is a core enzyme of the aerobic glycolytic pathway with versatile functions and is associated with cancer development. Recently, Kornberg et al. published the detailed correlation between GAPDH and di- or monomethyl fumarate (DMF or MMF), which are well-known GAPDH antagonists in the immune system. As an extension, herein, we report the crystal structure of MMF-bound human GAPDH at $2.29{\AA}$. The MMF molecule is covalently linked to the catalytic Cys152 of human GAPDH, and inhibits the catalytic activity of the residue and dramatically reduces the enzymatic activity of GAPDH. Structural comparisons between $NAD^+$-bound GAPDH and MMF-bound GAPDH revealed that the covalently linked MMF can block the binding of the $NAD^+$ cosubstrate due to steric hindrance of the nicotinamide portion of the $NAD^+$ molecule, illuminating the specific mechanism by which MMF inhibits GAPDH. Our data provide insights into GAPDH antagonist development for GAPDH-mediated disease treatment.

Hepatitis B Virus X Protein Stimulates Virus Replication Via DNA Methylation of the C-1619 in Covalently Closed Circular DNA

  • Methylation of HBV cccDNA has been detected in vivo and in vitro; however, the mechanism and its effects on HBV replication remain unclear. HBx derived from a 1.2-mer HBV replicon upregulated protein levels and enzyme activities of DNA methyltransferase 1 (DNMT1), 3a, and 3b, resulting in methylation of the negative regulatory region (NRE) in cccDNA, while none of these effects were observed with an HBx-null mutant. The HBx-positive HBV cccDNA expressed higher levels of HBc and produced about 4-fold higher levels of HBV particles than those from the HBx-null counterpart. For these effects, HBx interrupted the action of NRE binding protein via methylation of the C-1619 within NRE, resulting in activation of the core promoter. Treatment with 5-Aza-2′dC or DNMT1 knock-down drastically impaired the ability of HBx to activate the core promoter and stimulate HBV replication in 1.2-mer HBV replicon and in vitro infection systems, indicating the positive role of HBx-mediated cccDNA methylation in HBV replication.

$Cs^+$치환에 따른 수전해용 공유가교 SPEEK/HPA 복합막의 안정화 (Stabilization of Covalently Cross-Linked SPEEK/Cs-Substituted HPA Composite Membranes for Water Electrolysis)

  • To improve the mechanical properties, such as durabilities and antioxidative characteristics, the covalently cross-linked (CL-) SPEEK (sulfonated polyether ether ketone)/Cs-substituted HPA (heteropoly acid) organic-inorganic composite membranes (CL-SPEEK/Cs-HPAs), have been intensively investigated. The composite membrane were prepared by blending cesium-substituted HPAs (Cs-HPAs), including tungstophosphoric acid (TPA), molybdophosphoric acid (MoPA), and tungstosilicic acid (TSiA) with cross-linking agent content of 0.01 mL. And composite electrolytes composed of Cs-HPAs, prepared by immersion (imm.) and titration (titr.) methods to increase the stability of HPAs in water, were applied to polymer electrolyte membrane electrolysis (PEME). As a result, the proton conductivity of Cs-substituted composite membranes increased rapidly over $60^{\circ}C$ but mechanical properties, such as tensile strength, decreased in accordance with added Cs content. The bleeding-out of Cs-TPA membranes by titration method (50 vol.% Cs) decreased steadily to 2.15%. In the oxidative stability test by Fenton solution, the durability of membranes with Cs-HPA significantly increased. In case of CL-SPEEK/ Cs-TPA membrane, duration time increased more than 1200 hours. It is expected that even though CL-SPEEK/Cs-MoPA membrane shows the high proton conductivity, electrocatalytic activity and cell voltage of 1.80 V for water electrolysis, the CL-SPEEK/Cs-TPA (imm.) is more suitable as an alternative membrane in real system with the satisfactory proton conductivity, mechanical properties, anti-oxidative stability and cell voltage of 1.89 V.

  • Using molecular dynamics simulation method, we studied the carbon nanotube (CNT) non-covalently interacting with a polymer. As the polymer coiled around the CNT, the diameter of CNT deformed by more than 40% of its original value within 50 ps. By considering three different polymers, we conclude that the interaction between the CNT and polymer is governed by the number of repeating units in the polymer, not by the molecular weight of polymer.

Modification of Carboxyl Residues of Proteins with Pyridoxamine as a Fluorophore

  • A general procedure to quantitate the reaction of carbodiimides with carboxy groups of proteins is described. Pyridoxamine reacts with the o-acylisourea intermediate generated during the reaction of carboxyl residues with carbodiimides. The extent of the reaction is determined by measuring the spectroscopic properties, absorption and emission, of pyridoxyl residues covalently attached to the proteins. Resolved pig brain aspartate aminotransferase (apoenzyme), inactivated by 1-ethyl-3-(3-dimethylamino propyl) carbodiimide, reacts with $[^{3}H]pyridoxamine$. After trypsin digestion, one peptide labeled with radioactive pyridoxyl was separated by reverse phase HPLC.
