Purification of the Candida utilis Extracellular Invertase using Affinity Chromatography

  • The extracellular invertase (EC 3.2.1.26) (Candida utilis) preparation was obtained from the liquid medium after desalting and freeze drying. This prepared enzyme was used for the comparative purification on 4 activated matrices by liquid column affinity chromatography method. In this method there were used controlled porous glass (CPG) silanized covalently activated by keratin, silanized silica gel and silica gel covalently covered by keratin. It was found that the invertase purification process was better using both CPG matrices (silanized CPG and keratin activated CPG) than these with two silica gel supports. Also the elution coefficient of the invertase from the two CPG columns was about 93 to 94%. Two silica gel supports found to be superior in terms of purification efficiency. The invertase purification process was confirmed by PAGE electrophoresis.

The Preparation and Physicochemical Characteristics of Covalently Cross-Linked SPEEK/HPA Composite Membranes for Water Electrolysis (수전해용 공유가교 SPEEK/HPA 복합막의 제조 및 물리화학적 특성)

  • In order to improve the electrochemical, mechanical and electrocatalytic characteristics, engineering plastic of polyether ether ketone (PEEK) as polymer matrix was sulfonated (SPEEK) and the organic-inorganic blend composite membranes has been prepared by loading heteropoly acids (HPAs), including tungstophosphoric acid (TPA), molybdophosphoric acid (MoPA), and tungstosilicic acid (TSiA). And then these were covalently cross-linked (CL-SPEEK/HPA) as the electrolyte and MEA of polymer electrolyte membrane electrolysis (PEME). As a result, the optimum reaction conditions of CL-SPEEK/HPA was established and the electrochemical characteristics such as ion conductivity ($\sigma$) were in the order of magnitude: CL-SPEEK /TPA30 (${\sigma}=0.128\;S/cm^{-1}$) < /MoPA40 (${\sigma}=0.14\;S/cm^{-1})$ < /TSiA30 (${\sigma}=0.22\;S/cm^{-1}$) at $80^{\circ}C$, and mechanical characteristics such as tensile strength: CL-SPEEK /TSiA30 $\fallingdotseq$ /MoPA40 < /TPA30. Consequently, in regards of above characterisitics and oxidation durability, the CL-SPEEK/TPA30 exhibited a better performance in PEME than the others, but CL-SPEEK/MoPA40 showed the best electrocatalytic activity of cell voltage 1.71 V among the composite membranes. The dual effect of higher proton conductivity and electrocatalytic activity with the addition of HPAs, causes a synergy effect.

  • Liquid chromatographic enantiomer separation of N-tert-butoxycarbonyl (BOC) $\alpha$-amino acids and their ethyl esters was performed on covalently immobilized chiral column (Chiralpak IC) derived from polysaccharide derivative. The solvent versatility of the covalently immobilized Chiralpak IC in enantiomer separation of N-BOC $\alpha$-amino acid ethyl ester derivatives was shown and the chromatographic parameters of their enantioselectivities and resolution factors were greatly influenced by the nature of the mobile phase. Also the effect on the dissolving solvent for these analytes on the enantiomer separation using the same mobile phase and the examples of preparative enantiomer separation on analytical column were shown.

Recovery of Covalently Linked Fatty Acid Monolayer on the Hair Surface Using Biomimetic Lipid (생체모사 지질을 이용한 모발 표면에 공유 결합된 지방산 단분자층의 회복)

  • There is a unique type of fatty acid in the hair surface. 18-methyleicosanoic acid (18-MEA) is an unusual anteiso fatty acid covalently linked to the outermost surface of hair cuticle. A layer of 18-MEA is located in the upper ${\beta}$-layer of the CMC that is responsible for the low surface energy and low friction resistance of the hair's outer surface. The high mobility of 18-MEA molecule facilitates spreading of extraneous lipid by decreasing interfacial shear strength. In this study, we introduced N-hydroxyl succinimidyl ester functional group to the one end of C10 - 40 isoalkyl acid for regenerating hair surface with covalently bound fatty acid layer. The re-hydrophobicization of hair surface has been investigated by contact angle measurement. The inner moisture content of hair at different levels of humidity (40, 55, 70 %RH) was measured by electric moisture analyzer. Treatment with Hydroxysuccinimidyl C10 - 40 Isoalkyl Acidate (HCIA) was supposed to make hair surface smoother by filling the cracks between cuticles with covalently bound fatty acid monomolecular layer like cuticle glue. This glue effect was also confirmed with line profile of AFM images. Therefore, the moisture and structural components of inner hair were not easily flown out and the optimum moisture content could be kept constantly though the outside humidity level was changed. The lateral force microscopy (LFM) by using atomic force microscope showed that the friction force of hair surface treated with HCIA was decreased. It also showed the constantly sustained friction value even after shampooing repeated 15 times.

Enantiomer Separation of Chiral Amino Alcohols as 9-anthraldimine Derivatives on Coated and Covalently Bonded Chiral Stationary Phases Based on Polysaccharide Derivatives by High Performance Liquid Chromatography (고성능 액체 크로마토그래피에 의한 다당 유도체를 기초로 한 흡착되거나 공유결합된 키랄 고정상에서 키랄 아미노 알코올의 안트르알디민 유도체의 광학분리)

  • The convenient derivatization method of chiral amino alcohols as 9-anthraldimine Schiff base derivatives for chiral resolution was developed and the liquid chromatographic enantiomer separation of chiral amino alcohols as 9-anthraldimine derivatives was investigated on several coated and covalently bonded polysaccharide-derived chiral stationary phases (CSPs). In general, the performance of Chiralcel OD-H (or Chiralcel OD) (${\alpha}$ = 1.24-2.89), the coated CSP derived from cellulose derivative was superior to the other CSPs for resolution of 9-anthraldimine derivatives of several amino alcohols. The results of enantioseparation depending on the structure of 9-anthraldimine analytes like the steric bulky group and the polar moiety etc were discussed. The analytical method was applied to measure the enantiomeric purity of commercially available chiral amino alcohols. It is expected that the convenient analytical method will be very efficient for determination of enantiomeric purity of amino alcohols as 9-anthraldimine Schiff base derivatives with strong UV absorption.

Preparation and Characterization of TPA Captured CL-SPEEK Polymer Composite Membranes for Water Electrolysis (수전해용 술폰화 폴리에테르 에테르 케톤과 고정된 TPA 고분자 복합막의 제조 및 특성)

  • Polyether ether ketone (PEEK) composite including tungstophosphoric acid(TPA) membranes have been intensively investigated for polymer electrolyte membrane water electrolysis (PEMWE) and thus covalently linked sulfonated polyether ether ketone (CL-SPEEK) with captured TPA composite membranes were prepared and characterized. Sulfonated polyether ether ketone (SPEEK) was prepared in sulfonation of PEEK and was cross-linked with 1,4 diiodobutane. The carbonyl group of SPEEK was reduced with $NaBH_4$ and 3-isocyanatepropyltriethoxysilane (ICPTES) was added. The TPA captured composite was prepared in reaction of TPA with 3-mercaptopropyltrime thoxysilane (MPTMS). The polymer composite membranes showed better thermostability and electrochemical properties than SPEEK. The membranes were prepared by sol-gel casting method. The polymer composite membrane featured 0.1285 S/cm of proton conductivity at $80^{\circ}C$ and outstanding durability.
