In this study, an optical fiber detector was constructed by using a Ce:GAGG scintillator, optical fiber, and photomultiplier. The single crystal size of the scintillator was set to $3{\times}3{\times}20mm^3$ after simulating the counting efficiency of gamma rays in the scintillator by using the MCNPX code. The constructed detector used the standard gamma ray sources $^{137}Cs$ and $^{133}Ba$ to measure radiation and analyze the spectral characteristics of gamma rays. The resulting trend curve showed excellent linearity with an R-squared value of 0.99741, and the detector characteristics were found to vary 2% or less with distance based on comparison with the MCNPX value. Furthermore, the spectroscopic analysis of the gamma ray energy from the single-ray and mixed-ray sources showed that $^{137}Cs$ had its peak energy at 662 keV, and $^{133}Ba$ had at 356 keV. It seems that if the fiber-optics detector is used, working hours and exposure of worker can be reduced.
Human UHRF1 (ubiquitin-like PHD and RING finger domain-containing 1) has been reported to be over-expressed in many cancers, but its role in ovarian cancer remains elusive. Here, we determined whether knockdown of UHRF1 by lentivirus-mediated shRNA could inhibit ovarian cancer cell growth. Lentivirus-mediated short hairpin RNAs (lv-shRNAs-UHRF1) were designed to trigger the gene silencing RNA interference (RNAi) pathway. The efficiency of lentivirus-mediated shRNA infection into HO-8910 and HO-8910 PM cells was determined using fluorescence microscopy to observe lentivirus-mediated GFP expression and was confirmed to be over 80 percent. UHRF1 expression in infected HO-8910 and HO-8910 PM was evaluated by real-time PCR and Western blot analysis. The Cell Counting Kit-8 (CCK-8) assay was used to measure cell viability; flow cytometry and Hoechst 33342 assay was applied to measure cell cycle arrest and apoptosis. Cell invasion was assessed using transwell chambers. Our results demonstrated that the loss of UHRF1 promoted HO-8910 and HO-8910 PM cell apoptosis, while inhibiting cell proliferation. In addition, UHRF1 knockdown significantly inhibited the invasion of human ovarian cancer cells. In the present study, we also showed that depleting HO-8910 cells of UHRF1 caused activation of the DNA damage response pathway, with the cell cycle arrested in G2/M-phase. The DNA damage response in cells depleted of UHRF1 was illustrated by phosphorylation of CHK (checkpoint kinase) 2 on Thr68, phosphorylation of CDC25 (cell division control 25) on Ser 216 and phosphorylation of CDK1 (cyclin-dependent kinase 1) on Tyr 15.
In line with the "mandatory seismic design of fire protection facilities," development of design automation software is indispensable for improving the reliability and efficiency of seismic design. The seismic design automation software developed in this study is an automated S/W for seismic design of fire-fighting facilities, and functions such as automatic arrangement of anti-shake braces according to Korea National Fire Agency's Seismic Design Standards for fire-fighting facilities, output of seismic bracing calculation bills and automatic quantities counting. In addition, the seismic design automation software not only reduces the work speed by three times compared to the manual design of the designer, but also improves the reliability of the design by reducing the human error related to the design quantity such as the brace. In addition, in the seismic design method of fire protection facilities that have been approached conservatively, it was possible to perform the optimal seismic design by using computer algorithms for at least in the use of braces.
Human embryonic stem (hES) cell lines have been derived from human blastocysts and are expected to have far-reaching applications in regenerative medicine. The objective of this study is to improve freezing method with less cryo-injuries and best survival rates in hES cells by comparing various vitrification conditions. For the vitrifications, ES cells are exposed to the 4 different cryoprotectants, ethylene glycol (EG), 1,2-propanediol (PROH), EG with dime-thylsulfoxide (DMSO) and EG with PROH. We compared to types of vehicles, such as open pulled straw (OPS) or electron microscopic cooper grids (EM grids). Thawed hES cells were dipped into sequentially holding media with 0.2 M sucrose for 1 min, 0.1 M sucrose for 5 min and holding media for 5 min twice and plated onto a fresh feeder layer. Survival rates of vitrified hES cells were assessed by counting of undifferentiated colonies. It shows high survival rates of hES cells frozen with EG and DMSO (60.8%), or EG and PROH(65.8%) on EM grids better than those of OPS, compared to those frozen with EG alone (2.4%) or PROH alone (0%) alone. The hES cells vitrified with EM grid showed relatively constant colony forming efficiency and survival rates, compared to those of unverified hES cells. The vitrified hES cells retained the normal morphology, alkaline phosphates activity, and the expression of SSEA-3 and 4. Through RT-PCR analysis showed Oct-4 gene expression was down-regulated and embryonic germ layer markers were up-regulated in the vitrified hES cells during spontaneous differentiation. These results show that vitrification method by using EM grid supplemented with EG and PROH in hES cells may be most efficient at present to minimize cyto-toxicity and cellular damage derived by ice crystal formation and furthermore may be employed for clinical application.
The analysis of 41Ca in concrete generated from the nuclear facilities decommissioning is critical for ensuring the safe management of radioactive waste. An analytical method for the determination of 41Ca in concrete is described. 41Ca is a neutron-activated long radionuclide, and hence, for accurate analysis, it is necessary to completely extract Ca from the concrete sample where it exists as the predominant element. The decomposition methods employed were the acid leaching, microwave digestion, and alkali fusion. A comparison of the results indicated that the alkali fusion is the most suitable way for the separation of Ca from the concrete sample. Several processes of hydroxide and carbonate precipitation were employed to separate 41Ca from interferences. The method relies on the differences in the solubility of the generated products. The behavior of Ca and the interfering elements such as Fe, Ni, Co, Eu, Ba, and Sr is examined at each separation step. The purified 41Ca was measured by a liquid scintillation counter, and the quench curve and counting efficiency were determined by using a certified reference material of known 41Ca activity. The recoveries in this study ranged from 56 to 68%, and the minimum detectable activity was 50 mBq g-1 with 0.5 g of concrete sample.
This study aimed at determining the genetic and virulence characteristics of the Listeria monocytogenes from smoked ducks. L. monocytogenes was isolated by plating, and the isolated colonies were identified by PCR. All the obtained seven L. monocytogenes isolates possessed the virulence genes (inlA, inlB, plcB, and hlyA) and a 385 bp actA amplicon. The L. monocytogenes isolates (SMFM2018 SD 1-1, SMFM 2018 SD 4-1, SMFM 2018 SD 4-2, SMFM 2018 SD 5-2, SMFM 2018 SD 5-3, SMFM 2018 SD 6-2, and SMFM 2018 SD 7-1) were inoculated in tryptic soy broth (TSB) containing 0.6% yeast extract at 60℃, followed by cell counting on tryptic soy agar (TSA) containing 0.6% yeast extract at 0, 2, 5, 8, and 10 min. We identified five heat resistant isolates compared to the standard strain (L. monocytogenes ATCC13932), among which three exhibited the serotype 1/2b and D-values of 5.41, 6.48, and 6.71, respectively at 60℃. The optical densities of the cultures were regulated to a 0.5 McFarland standard to assess resistance against nine antibiotics after an incubation at 30℃ for 24 h. All isolates were penicillin G resistant, possessing the virulence genes (inlA, inlB, plcB, and hlyA) and the 385-bp actA amplicon, moreover, three isolates showed clindamycin resistance. In conclusion, this study allowed us to characterize L. monocytogenes isolates from smoked ducks, exhibiting clindamycin and penicillin G resistance, along with the 385-bp actA amplicon, representing higher invasion efficiency than the 268-bp actA, and the higher heat resistance serotype 1/2b.
The purpose of this comprehensive research is to observe the impact of scintillator crystal type on entire detection process. For this aim, MCNPX (version 2.6.0) is used for designing of a physical plastic scintillation detector available in our laboratory. The modelled detector structure is validated using previous studies in the literature. Next, different types of plastic scintillation crystals were assessed in the same geometry. Several fundamental detector properties are determined for six different plastic scintillation crystals. Additionally, the deposited energy quantities were computed using the MCNPX code. Although six scintillation crystals have comparable compositions, the findings clearly indicate that the crystal composed of PVT 80% + PPO 20% has superior counting and detecting characteristics when compared to the other crystals investigated. Moreover, it is observed that the highest deposited energy amount, which is a result of the highest collision number in the crystal volume, corresponds to a PVT 80% + PPO 20% crystal. Despite the fact that plastic detector crystals have similar chemical structures, this study found that performing advanced Monte Carlo simulations on the detection discrepancies within the structures can aid in the development of the most effective spectroscopy procedures by ensuring maximum efficiency prior to and during use.

Electronic voting machines (EVMs) are replacing research ballots due to the errors involved in the manual counting process and the lengthy time required to count the votes. Even though these digital recording electronic systems are advancements, they are vulnerable to tampering and electoral fraud. The suspected vulnerabilities in EVMs are the possibility of tampering with the EVM's memory chip or replacing it with a fake one, their simplicity, which allows them to be tampered with without requiring much skill, and the possibility of double voting. The vote data is shared among all network devices, and peer-to-peer verification is performed to ensure the vote data's authenticity. To successfully tamper with the system, all of the data stored in the nodes must be changed. This improves the proposed system's efficiency and dependability. Elections and voting are fundamental components of a democratic system. Various attempts have been made to make modern elections more flexible by utilizing digital technologies. The fundamental characteristics of free and fair elections are intractability, immutability, transparency, and the privacy of the actors involved. This corresponds to a few of the many characteristics of blockchain-like decentralized ownership, such as chain immutability, anonymity, and distributed ledger. This working research attempts to conduct a comparative analysis of various blockchain technologies in development and propose a 'Blockchain-based Electronic Voting System' solution by weighing these technologies based on the need for the proposed solution. The primary goal of this research is to present a robust blockchain-based election mechanism that is not only reliable but also adaptable to current needs.
This study is aimed to evaluate the effect of $T_{1/2}$ upon count rates in the analysis of dynamic scan using NaI (Tl) scintillation camera, and suggest a new quality control method with this effects. We producted a point source with $^{99m}TcO_4^-$ of 18.5 to 185 MBq in the 2 mL syringes, and acquired 30 frames of dynamic images with 10 to 60 seconds each using Infinia gamma camera (GE, USA). In the second experiment, 90 frames of dynamic images were acquired from 74 MBq point source by 5 gamma cameras (Infinia 2, Forte 2, Argus 1). There were not significant differences in average count rates of the sources with 18.5 to 92.5 MBq in the analysis of 10 to 60 seconds/frame with 10 seconds interval in the first experiment (p>0.05). But there were significantly low average count rates with the sources over 111 MBq activity at 60 seconds/frame (p<0.01). According to the second analysis results of linear regression by count rates of 5 gamma cameras those were acquired during 90 minutes, counting efficiency of fourth gamma camera was most low as 0.0064%, and gradient and coefficient of variation was high as 0.0042 and 0.229 each. We could not find abnormal fluctuation in $x^2$ test with count rates (p>0.02), and we could find the homogeneity of variance in Levene's F-test among the gamma cameras (p>0.05). At the correlation analysis, there was only correlation between counting efficiency and gradient as significant negative correlation (r=-0.90, p<0.05). Lastly, according to the results of calculation of $T_{1/2}$ error from change of gradient with -0.25% to +0.25%, if $T_{1/2}$ is relatively long, or gradient is high, the error increase relationally. When estimate the value of 4th camera which has highest gradient from the above mentioned result, we could not see $T_{1/2}$ error within 60 minutes at that value. In conclusion, it is necessary for the scintillation gamma camera in medical field to manage hard for the quality of radiation measurement. Especially, we found a tendency that count rate changes over time at this study, and we proved that it can effect $T_{1/2}$. And also, there is need of appropriate phantoms and the method of quality management like this study, because there are not any advice or limitation degrees for domestic medical purpose scintillation camera.

The muiltiwire proportional counter for the measurement of low-level and environmental $\alpha$ particles emitting nuclides was developed. External dimension of the devloped multiwire proportional counter is $350{\times}290{\times}30mm$ and the sensitivity area is $250{\times}200mm$. The wall material of the detector was selected the stainless steel to prevent the deformation by external impact and to obtain minimum background. The anode and cathode wires were used the stainless steel material of diameter $50{\mu}m$. The spacing of each wires are 10.0mm, 5.0mm and the numbers of total wire are 21, 42 lines, respectively. The multiwire proportional counter was designed that the measurement source is placed within the detector to prevent the wall absorption effect and the efficiency variation by various source heights. The characteristics of the developed detector have been investigated to obtain the plateau, operating voltage, background, counting efficiency, position sensitivity and energy resolution etc. For the $^{241}Am$ nuclide, the calculated LLD(Lower Limit of Detection) is 5.0mBq/L which is lower than 40mBq/L of recommended LLD value by ISO(International Organization for Standardization).
