Agrobacterium을 이용한 토마토 떡잎에서 CRISPR-Cas9 시스템의 임시발현 시 토마토 떡잎 발달 단계에 따른 유전자교정 효율 변화 (Observation of Gene Edition by the Transient Expression of CRISPR-Cas9 System During the Development of Tomato Cotyledon)

  • BACKGROUND: Before generating transgenic plant using the CRISPR-Cas9 system, the efficiency test of sgRNAs is recommended to reduce the time and effort for plant transformation and regeneration process. The efficiency of the sgRNA can be measured through the transient expression of sgRNA and Cas9 gene in tomato cotyledon; however, we found that the calculated efficiency showed a large variation. It is necessary to increase the precision of the experiment to obtain reliable sgRNA efficiency data from transient expression. METHODS AND RESULTS: The cotyledon of 11th, 15th, 19th, and 23rd-day-old tomato (Solanum lycopersicum cv. Micro-Tom) were used for expressing CRISPR-Cas9 transiently. The agrobacterium harboring sgRNA for targeting ALS2 gene of tomato was injected through the stomata of leaf adaxial side and the genomic DNA was extracted in 5 days after injection. The target gene edition was identified by amplifying DNA fragment of target region and analyzing with Illumina sequencing method. The target gene editing efficiency was calculated by counting base deletion and insertion events from total target sequence read. CONCLUSION: The CRISPR-Cas9 editing efficiency varied with tomato cotyledon age. The highest efficiency was observed at the 19-day-old cotyledons. Both the median and mean were the highest at this stage and the sample variability was also minimized. We found that the transgene of CRISPR-Cas9 system was strongly correlated with plant leaf development and suggested the optimum cotyledon leaf age for Agrobacterium-mediated transfection in tomato.

광산화시스템을 이용한 염색폐수의 유기물 처리효율 및 잔류미생물의 독성 평가 (Organic Removal Efficiency and Toxicity Evaluation of Persistent Microorganism from Dye Wastewater Treatment using a Photooxidation system)

  • The removal efficiency of organic compounds and the toxicity evaluation of microorganism have been studied in dye wastewater treatment using $UV/TiO_2$ and $UV/H_2O_2$ photooxidation system. Sample waters tested in this work were raw dye wastewater and dye wastewater treated in $UV/TiO_2$ and $UV/H_2O_2$ photooxidation system respectively. Total organic carbon(TOC) removal rate was 50% in $UV/TiO_2$ process and 80% in $UV/H_2O_2$ process. It has been investigated with colony counting agar method and paper disk method whether the type of treatment process has affected the microorganism growth. In the raw wastewater, more than four types of microorganisms have survived. But, little of microorganisms were alive at TOC removal rate of 50% in $UV/TiO_2$ system. In contrast to that, two types of microorganisms were found at TOC removal rate of 80% in $UV/H_2O_2$ system.

탄소섬유 이오나이저를 적용한 활성탄소섬유 필터의 바이오에어로졸 항균 및 집진 성능평가 (Inactivation and Filtration of Bioaerosols Using Carbon Fiber Ionizer Assisted Activated Carbon Fiber Filter)

  • This paper reports that the installation of a carbon fiber ionizer in front of an activated carbon fiber(ACF) filter enhanced the antibacterial efficiency. In addition, the effect of the ionizer on the filtration of bioaerosols is reported. Negative air ions from the ionizer were used as antibacterial agent. The test bacteria(Escherichia coli) were aerosolized using an atomizer and were deposited on the ACF filter media for 10 minutes. E. coli deposited on the filter were exposed to negative air ions for 0, 1, 5 and 10 minutes. Then they were separated from the ACF filter by shaking incubation with nutrient broth for 4 hours. The separated E. coli were spread on nutrient agar plates and incubated at $37^{\circ}C$ for 1~3 days. The antibacterial efficiency of E. coli was measured using a colony counting method. The antibacterial efficiencies of E. coli exposed to negative air ions for 1, 5 and 10 minutes were 14%, 48% and 71%, respectively. The filtration efficiency was evaluated by measuring the number concentration of bioaerosols at the upstream and downstream of the filter media. The increase of filtration efficiency by air ions was 14%, that is similar to the 17% filtration efficiency by none air ions. The ozone concentration was below the detection limit (under 0.01ppm) when the carbon fiber ionizers were on.

환경시료의 방사능 분석에서 Monte Carlo 방법을 이용한 자체흡수 효과 보정 (Corrections of Self-Absorption Effect Using the Monte Carlo Method in the Radioactivity Analysis of Environmental Samples)

  • 환경방사능과 같은 저준위 방사능 측정에서는 원통형과 Marinelli형 측정용기가 가장 일반적으로 사용된다. 효율교정용 표준선원과 측정시료의 높이 또는 매질의 밀도가 다르면 자체흡수 효과의 차이로 인한 보정이 필요하다. 본 연구에서는 Monte Carlo 방법을 이용하여 HPGe 검출기의 전에너지 피크 효율을 계산하여 측정치와 비교하였다. 원통형 용기에 대해서는 높이에 대한 효율변화 정도를 계산하였고, 원통형 및 Marinelli 측정용기에 대해서는 밀도변화에 따른 효율을 계산하였다. 밀도에 따른 효율의 감소 정도는 1000keV 이하의 에너지 영역에 대해 자체흡수 효과의 보정치 필요하다는 것을 알았다. 또한 계산의 타당성을 검증하기 위하여 NIST SRM 4353 표준물질을 이용하여 계산값과 인증값을 상호비교한 결과 오차범위 이내로 잘 맞는 다는 것을 확인하였다.

Comparison of knife-edge and multi-slit camera for proton beam range verification by Monte Carlo simulation

  • The mechanical-collimation imaging is the most mature technology in prompt gamma (PG) imaging which is considered the most promising technology for beam range verification in proton therapy. The purpose of the present study is to compare the performances of two mechanical-collimation PG cameras, knife-edge (KE) camera and multi-slit (MS) camera. For this, the PG cameras were modeled by Geant4 Monte Carlo code, and the performances of the cameras were compared for imaginary point and line sources and for proton beams incident on a cylindrical PMMA phantom. From the simulation results, the KE camera was found to show higher counting efficiency than the MS camera, being able to estimate the beam range even for $10^7$ protons. Our results, however, confirmed that in order to estimate the beam range correctly, the KE camera should be aligned, at least approximately, to the location of the proton beam range. The MS camera was found to show lower efficiency, being able to estimate the beam range correctly only when the number of the protons is at least $10^8$. For enough number of protons, however, the MS camera estimated the beam range correctly, errors being less than 1.2 mm, regardless of the location of the camera.

$4{\pi}{\beta}-{\gamma}$ 동시계수기술에 의한 $^{56}Mn$방사능 절대측정 (Absolute $^{56}Mn$ Activity Measurement by $4{\pi}{\beta}-{\gamma}$ Conincidence Counting Technique)

  • 황산망간 용액조장치의 $^{56}Mn\;{\gamma}$선 검출효율을 결정하는데 $^{56}Mn$용액의 방사능을 절대측정하는 것은 필수적이다 $^{56}Mn$시료를 제작하기 위하여 99.99%의 순도를 갖는 Mn금속조각 13.718mg되는 시료를 한국에너지연구소 TRIGA MARK-II 원자로의 중성자선속이 약 $10^{13}n/cm^2{\cdot}s$되는 열중성자장에서 12분간 조사시켰다. 중성자 방사화된 $^{56}Mn$금속시료를 0.1N-HCI 용액 50ml 용해시켜서 $^{56}Mn$시료를 제작하여 $4{\pi}{\beta}-{\gamma}$ 동시계수기술로 방사능을 측정한 결과 불확도 0.366%를 갖는 값으로서 1987년 10월 15일 0 시를 기준하여 408.070kBq/mg을 얻었다.

액정성 고분자 블렌드의 편발광 (Polarized Light Emission of Liquid Crystalline Polymer Blends)

  • 액정성을 가지는 플로렌계 발광고분자에 비버정성 플로렌계 발광고분자를 첨가한 블렌드를 제조하여 에너지전달 효과와 흡광 및 발광 이방성에 대하여 고찰하였다. 비액정성 고분자를 액정성 고분자에 0.5wt% 첨가하고 360nm로 여기한 결과, 420nm에서 관찰되었던 액정성 고분자의 발광피크는 거의 사라졌으며 대신 비액정성 고분자에 의한 480nm에서의 새로운 발광피크가 관찰되었다. 블렌드 시료의 480nm 발광은 비액정성 고분자가 2.0wt% 첨가되었을때 가장 강했으며, 발광강도는 블렌딩 이전의 각 고분자보다 13배 증자하였다. 블렌드내의 분자들을 마찰 폴리이미드 기판상에서 가열-냉각시켜 배향시키면 시료의 발광 이방성과 order parameter는 각각 2.0 및 0.25를 나타내었다. 시간 상관 단광자 계수법 (TCSPC)에 의해 고찰한 결과, 마찰 폴리이미드 기판상에서의 배향에 의해 두 발광고분자간의 에너지전달에 필요한 시간은 93 ps만큼 단축되며 에너지전달 효율은 9% 증가함을 알 수 있었다.

A novel ceramic GEM used for neutron detection

  • A novel ceramic Gas Electron Multiplier (GEM) has been developed to meet the demand of high counting rate for the neutron detection which is an alternative to 3He-based detector at China Spallation Neutron Source (CSNS). An experiment was performed to measure the neutron transmittance of ceramic-GEM and FR4-GEM at the small angle neutron scattering (SANS) instrument. The result showed the ceramic-GEM has higher transmittance and less self-scattering especially for cold neutrons. One single ceramic GEM could give a gain of 102-104 in the mixture gas of Ar and CO2 (90%:10%) and its energy resolution was about 27.7% by using 55Fe X ray of 5.9 keV. A prototype has been developed in order to investigate the performances of the ceramic GEM-based neutron detector. Several neutron beam tests, including detection efficiency, spatial resolution, two-dimensional imaging, and wavelength spectrum, were carried out at CSNS and China Mianyang Research Reactor (CMRR). The results show that the ceramic GEM-based neutron detector is a good candidate to measure the high intensity neutrons.

추적자 방법에 의한 $^{137}Cs$용액의 방사능 절대측정 (Absolute Activity Measurement of $^{137}Cs$ Solution by the Tracer Method)

  • $^{137}$Cs 용액을 추적자로 사용하여 동시와 역동시 계수방법에 의해 $^{137}$Cs 용액의 방사능을 측정하였다. 본 연구에서의 측정선원은 $^{137}$Cs과 추적자를 혼합하여 제작하였으며 혼합 비율은 동시와 역동시 계수 방법에서 각각 1/3과 1/5로 하였다. $^{137}$Cs용액의 비 방사능은 $\beta$채널 저에너지 문턱 준위 변화와 효율외삽 방법에 의해 산출되었다. 동시와 역동시 계수 방법에 의해 산출된 결과는 기준일 현재 552.78kBq/g과 554.32kBq/g이었으며 합성오차는 각각 1.60kbq/g과 1.51kBq/g이었다. 두 개의 결과는 오차범위 내에서 잘 일치함을 보여준다.

액체섬광계수기를 이용한 저준위 트리튬 분석법연구 (Low level tritium analysis using liquid scintillation counter)

  • 환경준위의 삼중수소 측정법을 확립코자 극저준위 액체섬광계수기(LSC)와 1 TU(Tritium Unit) 미만까지 측정이 가능한 전기분해 농축 장비의 특성을 연구하였다. 삼중수소의 측정한계를 알기위해 다양한 측정용기에 의한 장비의 바탕값을 조사한 결과 테프론 코팅된 폴리에틸렌 측정용기에서 1.86 cpm 수준이었으며 5 시간동안 계측할 경우 2.01 Bq/L로 17 TU의 검출한계를 얻었다. 400 mL 시료를 전기분해하여 20 mL까지 농축할 경우 0.8 TU의 검출한계를 얻었다. 전해농축시 수소와 삼중수소의 분리효율은 20 이상이었으며, 전해농축과정에 따른 삼중수소 회수율은 약 90%정도였다. NIST 삼중수소 표준시료를 사용하여 LSC의 측정효율을 검증한 결과 10 mL의 시료와 10 mL의 cocktail 용액을 사용한 경우 $28.70{\pm}0.27%$의 측정효율을 얻었다.
