Efficient Synthesis of Novel 3-Substituted Coumarin-3-carboxamide

  • A series of novel pseudopeptides contained coumarin skeleton were synthesized through the Ugi-four-component reaction. The 3-substituted coumarin-3-carboxamides were formed through reaction of benzaldehyde derivatives, anilines, coumarin-3-carboxylic acid and isocyanides with high yields and high bond-forming efficiency at room temperature. These novel amidated coumarins exhibit brilliant fluorescence in range of 535-547 nm in chloroform.

  • The chemical behaviours of 4-methyl-2-oxo-2H-benzopyran-7-yl oxoacetyl hydrazine (2) towards some different reagents such as anhydride compounds, aromatic aldehydes, carb on disulphide, and nitrous acid yielded the corresponding pathalazine derivatives (3, 4, 5), hydrazone derivative (6), 1,3,4-oxadiazole derivative (7, 8, 9) and acid azide (10) respectively. Treatmen of 10 with absolute alcohols, amines and ethyl amino acid ester gave the corresponding carbamate derivative (11), substituted urea derivative (12) and ethyl substituted alkyl acetate (13) respectively. The biological activity of some synthesized compounds was evaluated.

  • The ether extract of Dang-Gui, the dried root of Angelica gigas N$_{AKAI}$, was placed into a column with aluminium oxide gel. The elution of it with a mixture of n-hexane and ethyl acetate afforded Nodakenetin (I) and the elution with a mixture of n-hexane, ethyl acetate, acetone and methyl alcohol yielded Umbelliferon (II) and Nodakenin (III). Three coumarin derivatives were identified by comparing with authentic samples. The existence of sucrose and glucose was also proved.

  • The reaction of either Digitoxin or Digoxin with cyanoacetic acid hydrazide gave the hydrazone derivatives 3a and 3b respectively. The reactivity of the latter products towards chemical reagents was studied to give heterocyclic derivatives with potential biological activities.

  • The cycloaddition reaction of N-phenyl-C-cinnamonitrilimine 4 to coumarin leads to the for-mation of 3-styrylbenzopyrano[4,3-c]pyrazole derivative 6, whereas 3-phenylsulfonylcoumarin 9 or 3-bromocoumain 10 or 3-cyanocoumarin 11 gives 1-styrylbenzopyrano[3,4]pyrazole derivative 7. Also, the cycloaddition of 4 to 3-acetylocoumarin 15 and 3-benzoylocumain 16 gives the corresponding dihydropyrano[3,4-c]pyrazole adducts 17 and 18 respectively. Oxidation of 17 and 18 gives 7.

  • Three coumarin derivatives, which stimulate collagen biosynthesis in human fibroblasts were isolated from Angelica dahurica root which has been used in traditional medicine for cold, headache, discharging blood and women's diseases. Three coumarin derivatives were identified as 5-benzofuranacrylic acid, $6,7-dihydroxy-4-(3-methyl-2-butenyl)-{\delta}-lactone\;(prangenidin)$, 5-benzofuranacrylic acid, $6,7-dihydroxy-4-methoxy-{\delta}-lactone\;(8-hydroxybergapten)$ and 5-benzofuranacrylic acid, 8-hydroxypsoralen(xanthotoxol) from their spectral data.

  • Two quinazolone alkaloids, (+)-febrifugine (1) and isofebrifugine (2), along with three coumarin derivatives, 6-hydroxy coumarin (3), skimmin (5), and $umbelliferone-7-O-{\alpha}-L-rhamnopyranosyl(1{\rightarrow}4)-{\beta}-D-glucopyranoside$ (6), were isolated from the roots of Hydrangea chinensis. Compound 6 is a new compound. In addition, umbelliferone (4), linoleic acid (7), two steroidal glycosides (8, 9), three furfural derivatives (10-12), and butyl-$\beta$-D-fructofuranoside (13) were isolated from the leaves of the same plant. The structures of all isolates were elucidated by spectral methods.

  • From the root of Angelica gigas $N_{AKAI}$ (Umbelliferae), known as "Tang Gui" in Korea, substance A (colorless needle crystal, $C_{14}H_{14}O_{4}$, m.p. $178^{\circ}$) and B (white plate crystal, $C_{19}H_{20}O_{5}$, m.p. $111^{\circ}$) were obtained. Acetylation of "A" gave its monoacetate, white needles, $C_{16}H_{16}O_{5}$. Saponification of "B" with 5% sodium hydroxide produced "A" and senecionic acid. Examination using I.R. and N.M.R. spectra shows that "A" is identical with decursinol and that "B" with decursin. The root also appears to contain five coumarin derivatives and two steroidal compounds.

  • $^{14}C$-warfarin을 흰쥐에 경구투여 후 4시간 경과시 혈액에서 최고치를 나타내었으며, coumarin과 그 유도체인 warfarin에 의한 간조직 microsome 내의 Cyt. p-450은 이들 화합물에 의해 microsomal electron transport system이 증가됨과 Cyt. p-450의 isozyme들이 유도됨을 확인할 수 있었다.
