자생식물 Essential Oil 5 종의 항 아토피피부염 활성 연구 (A Study on the Activities of Five Natural Plant Essential Oils on Atopic Dermatitis)

  • 본 연구는 개똥쑥(Artemisia annua L., AA), 유자(Citrus junos Sieb. ex TANAKA, CJ), 산국화(Chrysanthemum boreale Makino, CB), 잣나무(Pinus koraiensis, PK), 금강송(Pinus densiflora for. erecta, PD) 총 5 종의 자생식물 essential oil의 항 아토피 효능을 확인하기 위한 실험이다. 항균 효과를 확인하기 위해 자생식물 essential oil 5 종을 Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans 총 4 종류의 균에 처리했다. 항염증 효과를 확인하기 위해 TNF-α와 IFN-γ (TI)를 처리한 HaCaT 세포에 5 종의 자생식물 essential oil을 처리했다. AA, CJ, CB, PK, PD은 10 mg/mL 농도에서 Candida albicans에 대한 항균 효과를 나타냈다. AA (1 ㎍/mL), CB (1 ㎍/mL), PK (0.1, 1 ㎍/mL)에서 thymus and activation-regulated chemokine (TARC) 생성량이 억제되었고, AA 및 PK (1 ㎍/mL)에서 macrophage-derived chemokine (MDC) 생성량이 억제되었으며, AA (0.1, 1 ㎍/mL), PK (0.1, 1 ㎍/mL)에서 IL-6 생성량이 억제되었다. AA, CB, PK essential oil의 항균 및 항염증 효과가 확인되었으며, 이에 아토피 피부염 완화에 기여할 수 있음을 기대한다.

  • 아토피 피부염은 다른 말로 태열이라고도 하며, 유아 시기에서부터 성인까지 광범위한 연령층에 병변의 특징적인 분포 그리고 개인적 혹은 가족적인 병력을 가진 특히 유전적 소인을 보이는 질환으로 심한 가려움증과 건조증 등을 동반하는 일종의 재발성, 만성의 알레르기성 습진을 말한다. 그 유병율은 매우 흔하여 어린이의 약 $9~12\%$ 에서발생한다. 그러나 최근에는 사회 자연환경의 변화와 음식문화의 변화, 삶의 방식의 변화에 의해서 계속 증가하는 추세를 보이고 있다. 인간의 피부는 외부 환경으로부터의 물리적, 화학적 자극에 대한 장벽기능의 역할을 하는데 아토피 피부염 환자의 피부는 각질층에서의 피부 장벽기능 및 수분유지 기능이 감소되어 있고 이러한 기능 장애의 원인으로 최근 연구에 의하면 세라마이드의 감소에 따른 것으로 밝혀지고 있다. 세라마이드는 스핑고신에 지방산이 연결되어 있는 구조를 가지고 있는 스핑고 지질의 일종이다. 세라마이드는 피부 각질층을 구성하는 각질세포간 지질 중 약 $40\%$를 차지하며, 수분 증발을 억제하는 지질 방어벽 역할과 각질층의 정연한 구조를 유지하게 하는 기능을 가지고 있다. 피부 각질층은 각화된 세포가 벽돌모양의 다층 구조로 구성되어 있으며 이러한 각화세포는 세라마이드, 콜레스테롤, 유리 지방산에 의해 견고히 결합되어 있다 따라서 이 세라마이드가 함유된 크림이나 연고를 아토피 피부염 환자에 도포함으로써 환자의 피부 장벽기능의 복구에 도움을 주어 아토피 피부염을 치료할 수 있다는 보고가 있다. 본 연구는 아토피 피부염 전용화장품으로서 아토피 피부에서 피부장벽을 복구하는데 기여한다고 증명된 각질 세포간 지질 성분들(세라마이드, 콜레스테롤, 유리지방산)을 포함하는 유액제품과 피부 면역조절과 항 염증 효과가 있다고 보고된 베타글루칸, 신이화 추출물 감초 추출물 등을 포함하는 세럼형태의 액상제품으로 구성된 두 가지 제품을 아토피 피부염 환자에게 도포하게 하여 그 제품의 안전성과 유효성을 확인하여 보았다. 그리고 그 제품이 새로운 시장인 아토피 피부염 전용 보습화장품으로서의 시장진입 가능성을 확인하여 보았다.

Coenzyme Q10의 멜라닌 생성억제효과 (The inhibitory Effects of Coenzyme Q10 on Melanogenesis of cultured Human Melanocytes and in vivo Guinea Pig Model)

  • Coenzyme Q10(CoQ10)은 피부를 포함한 모든 생체조직에 존재하는 널리 알려진 조효소이다. 전자전달에 관여하는 퀴논링은 세포에서 에너지를 생성하기 위한 매우 중요한 기능을 가지고 있다. CoQ10은 피부에서 항산화제로서 연구되어 왔으며, 최근 외용제로써 노화억제와 주름개선작용에 대해 보고된 바 있다. 이런 보고들은CoQ10이 항산화제로서 산화-환원작용을 통해 피부의 방어기능에 중요한 역할을 한다는 점을 시사하며, 일반적으로 산화-환원작용은 피부에서 흑화과정의 조절에도 많은 영향을 미친다. 따라서 본 연구자들은 CoQ10 이 피부의 색소조절기능이 있는지 알아보고자 하였다. 인체 정상 색소세포에CoQ10을 0.05-0.5 mM 처리한 결과 0.5, 0.25mM에서 멜라닌의 생합성을 약 50% 저해하였으며 이는 알려진 미백제인 Kojic acid나 vitamin C와 유사한 수준이었다. 또한, CoQ10은 인체 정상 색소세포에서 자외선이나 세포내 cAMP 증가 유도물질에 의한 멜라닌 생성을 억제하였다. 그러나 tyrosinase inhibitor인 kojic acid와는 달리, in vitro tyrosine hydroxylase의 억제효과는 보이지 않았다. CoQ10을 자외선으로 tanning을 유도시킨 brown guinea pig에 4주간 도포하고 육안 및 chromameter를 이용하여 미백효과를 측정한 결과, vehicle처리군에 비해 미백효과가 있음을 확인할 수 있었다. 이상의 결과에서 coenzyme Q10 은 in vitro및 in vivo에서 미백효과를 지닌 물질임을 확인할 수 있었다.

Production of Carrot Pomace Fortified with Mucilage, Fibrinolytic Enzyme and Probiotics by Solid-state Fermentation Using the Mixed Culture of Bacillus subtilis and Leuconostoc mesenteroides

  • Bioactive compounds were produced from carrot pomace by solid-state fermentation using Bacillus subtilis HA and Leuconostoc mesenteroides. The carrot pomace (CP) fermented by B. subtilis HA with 3% monosodium glutamate (MSG) showed higher production of various bioactive compounds, with 1.64 Pa·sn of consistency, 2.31% of mucilage content, 16.95 unit/g of fibrinolytic enzyme activity, 35.3 unit/g of proteolytic activity and 37.5 mg% of tyrosine content. The mucilage production was greatly dependent upon the concentration of MSG added. Most MSG added in CP was converted into mucilage (2.3%) including 0.83% poly-$gamma$-glutamic acid (PGA) with 1,505 kDa of molecular weight. The CP fermented secondly by Leuc. mesenteroides showed acidic pH and lower consistency. However, the fibrinolytic and proteolytic activities were increased. The secondly fermented CP showed the viable cell counts with $2.5{\time}108$ CFU/g of B. subtilis HA and $3.7{\time}109$ CFU/g of Leuc. mesenteroides, respectively. The freeze-dried fermented CP showed 2.88 Pa·sn of consistency, 24% of mucilage content and 104.9 unit/g of fibrinolytic enzyme activity, respectively. Also, the powder of fermented CP indicated viable cell counts of $8.0{\time}107$ CFU/g of B. subtilis and $4.0{\time}108$ CFU/g of Leuc. mesenteroides. Therefore, the fermented CP that was fortified with dietary fibers, fibrinolytic enzyme and probiotics could be utilized as valuable ingredients of functional foods in food or cosmetic industries.

Triple Matrix Capsulation having Visible Effects and Stabilizing Functions

  • This study is to develop the double capsulation technology in order to increase the conservativeness and stability of unstable materials such as vitamins, polyphenols, natural active ingredients. And also, best way of triple matrix capsulation using natural polymers were detail described. As the first capsulation with w/o/w (water-in-oil-in-water) emulsifying system, our study group was especially made to soft and moisture cream using 5wt% of sucrose ester emulsifier as first capsulation. Nutrient agents are squalane, camellia oil. Triple matrix capsulation was formed with the best stabilized bead type capsules when it blended of chitosan, algin, sodium-potassium alginate. The bead diameter size was about 2.0~4.5mm (mean diameter: 3.2mm). Activity of lactobacillus containing cream for depending on various pH variations showed that alkalinity ($pH=10.8{\pm}0.5$) condition was higher than acidity ($pH=4.2{\pm}0.2$) and neutrality ($pH=7.1{\pm}0.3$) conditions. After a month, it also was certified to the activity of lactobacillus in incubated at $37{\pm}1^{\circ}C$ in culture medium. As application of food industry, we developed the containing lactobacillus capsule and 7 colored kinds of double and triple matrix capsulation in yogurt cream and active ingredients. As for above mentioned those results, one of tool to stabilize the living lactobacillus, doubled matrix capsulation greatly be expected to contribute to food industry. Furthermore, it can be expected to apply the drug delivery system (DDS) to active ingredients of stabilizing technologies at drug, pharmaceutical division and cosmetic industry, etc.

20대 대학생들의 상악중절치 색조와 구강보건 및 식습관 관련 특성 (Oral health and eating habits characteristics relating to maxillary central incisor color of the university students in their twenties)

  • Objectives : This study aims to prepare the basic data of the teeth color by measuring tooth mode using the color of the maxillary central incisor of the college students in their twenties as the dental colorimeter, and oral health and eating habit characteristics were examined. Methods : The maxillary central incisor(1 teeth) of the subjects of study as 467 students(male 89, female 378) was measured during the period from November 14, December 2 2011, so the color of total 467 teeth were examined, and oral health and eating habit characteristics were researched. Results : 1. As the results of measurement of the shade, value, hue of maxillary central incisor by the general characteristics of the subject, the major(shipbuilding information, special rehabilitation for young children) was higher at 3.3 of the shade, and the grade(1), age($$\geq_-20$$), major(beauty culture & cosmetic, occupational therapy) showed higher at +2 of the value, and the grade(1) and all major showed higher at R3 of the hue. 2. As the results of measurement of the shade, value, hue of maxillary central incisor by the oral health characteristics, the subjective teeth color(yellow) and the education of oral health(no) showed higher at 2.5 of the shade. 3. As the results of measurement of the shade, value, hue of maxillary central incisor by the eating habit characteristics of the subjects, the chocolate showed higher at 3.0 and 2.0 of the shade, and the red pepper paste showed higher at +2 and STD of the value, and the green tea showed higher at $R_3$ of the hue. Conclusions : This study demonstrates that the color of the maxillary central incisor showed differences various tendency according to the oral health and eating habit characteristics.

Purification and Characterization of Chitinase from a New Species Strain, Pseudomonas sp. TKU008

  • The chitinase-producing strain TKU008 was isolated from soil in Taiwan, and it was identified as a new species of Pseudomonas. The culture condition suitable for production of chitinase was found to be shaking at $30^{\circ}C$ for 4 days in 100 ml of medium containing 1% shrimp and crab shell powder, 0.1% $K_2HPO_4$, and 0.05% $MgSO_4{\cdot}7H_2O$ (pH 7). The TKU008 chitinase was suppressed by the simultaneously existing protease, which also showed the maximum activity at the fourth day of incubation. The molecular mass of the chitinase was estimated to be 40 kDa by SDS-PAGE. The optimum pH, optimum temperature, pH stability, and thermal stability of the chitinase were pH 7, $50^{\circ}C$, pH 6-7, and <$50^{\circ}C$, respectively. The chitinase was completely inhibited by $Mn^{2+}$ and $Cu^{2+}$. The results of peptide mass mapping showed that 11 tryptic peptides of the chitinase were identical to the chitinase CW from Bacillus cereus (GenBank Accession No. gi 45827175) with a 32% sequence coverage.

Saccharomyces cerevisiae deletion mutant의 세라마이드 생합성 (Biosynthesis of ceramide by deletion mutant of Saccharomyces cerevisiae)

  • Saccharomyces cerevisiae의 deletion mutant를 이용하여 ydc1, ypc1, scs7, sur1, csg2, ipt1, Icb4, Icb5, dpll의 deletion이 세라마이드의 생산에 미치는 영향을 고찰하였다. 세라마이드는 ELSD가 연결된 HPLC를 통하여 분석하였으며 ${\triangle}$ydc1 mutant의 세라마이드 생산량이 6 mg ceramide/g cell로 최대량을 나타내었으며 ${\triangle}$sur1 mutant, ${\triangle}$lcb5 mutant, ${\triangle}$dpll mutant의 경우 control로 사용한 BY4742와 비슷한 세라마이드 생산량을 나타내었고, 그 외 ${\triangle}$ypc1 mutant, ${\triangle}$scs7 mutant, ${\triangle}$csg2 mutant, ${\triangle}$ipt1 mutant, ${\triangle}$lcb4 mutant는 BY4742보다 낮은 세라마이드 생산량을 나타내었다.

건칠의 항주름 효과와 미백효과 연구 (The effect of wrinkle care and whitening by Rhus verniciflua Stokes)

  • Objectives : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro.Methods : Human dermis cell (HS68) was used to test the effect of Rhus verniciflua Stokes (RVS). 80% ethanol or water extracts were screened for their inhibitory activities against elastase. Elastase inhibition effect was tested by microplate reader instrument. And MMP-1 suppression effect of RVS was tested by western blot. These cells were investigated the viability by MTS assay. And also the inhibition effect of tyrosinase by RVS was tested.Results : RVS (final concentrstion 1 ㎎/㎖ appeared over 30% of inhibition of elastase activity. So we are investigated anti wrinkle effects of Rhus verniciflua Stokes look through MMP-1 inhibition activity, also Extracts of RVS showed higher anti-tyrosinase activity than arbutin as final concentration 1 mg/ml. These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.Colclusions : RVS was the best suppressor candidate of elastase activity among other control oriental drugs. It was found that RVS did not have toxicity to cells. We found proper concentration of RVS to treat in HS68 culture and investigated not only wrinkle care effect but also whitening effect of RVS.

갈대 발효추출물의 이화학적 특성 및 생리활성 연구 (Physicochemical Characteristics of Fermented Phragmites communis Extract and Its Biological Activity)

  • This study evaluates the tyrosinase, elastase inhibitory and antioxidant activities of isolated Lactobacillus rhamnosus fermented extracts of Phragmites communis Trinius. After culture for 4 days at $30^{\circ}C$ using 1% P. communis extract, the cell mass of L. rhamnosus reached $1.4{\times}10^{10}CFU/mL$. The number of cells on P. communis extract and MRS medium was similar. This results indicated that P. communis extract can be used as an economical medium for industrial lactic acid bacteria production. The fermented P. communis extract exhibited 4 fold higher tyrosinase inhibitory effect than non fermented P. communis extract. The non fermented P. communis extract has no inhibitory effect on elastase. However the fermented P. communis extract show high inhibitory effect on elastase ($IC_{50}$; $249{\mu}g/mL$). These results indicated that the fermented P. communis extract can potentially be used for developing new cosmetic or health food ingredients.
