Whitening Effect of Biochanin A (Biochanin A의 미백 효과에 관한 연구)

  • To develop a new whitening agent for cosmetics, we investigated the inhibitory effects of biochanin A on melanogenesis. The biochanoin A is an active component in the extracts of Dalbergia odorifera T. Chen. It reduced melanin contents of B16F1 melanoma cells in a dose-dependent manner and decreased to about 48% at a concentration 10 ${\mu}g/mL$. It also inhibited the expression in microphthalmia associated transcription factor (MITF), Tyrosinase, Tyrosinase related protein 1 (Trp-1), and Tyrosinase related protein 2 (Trp-2) in melanocytes. The results suggested that biochanin A has considerable potential as a cosmetics ingredient with a whitening effect.

Development of Nano-liposome with Unsaturated Lecithin (불포화레시틴을 이용한 나노리포좀의 개발에 관한 연구)

  • In cosmetics, the saturated lecithin, one of the main surfactants to prepare liposome has been used for its stability but it has been substituting with unsaturated lecithin which has excellent skin affinity and penetration property. So we studied to prepare nano-liposome that size of particles were below than 50nm by unsaturated lecithin. It was important that many factors including solvent such as propylene glycol, pH balance, homogenizing pressure, various cosurfactants and stabilizers to make stable nano-liposome. In our experimental conditions, cosurfactants with stearate class as lipophilic part were more suitable than others for our purpose. But in liposome by saturated lecithin, cosurfactants had negative effect and appropriate amount of oil should be used to be stable. These results indicated that unsaturated lecithin were more suitable than saturated lecithin to prepare nano-liposome.

  • Manuka oil named New Zealand’s tea tree oil is oil-soluble and comes firom nature. Manuka oil and its extract $\alpha$-pinene, Oxy’less clear, R-limonene which is one of the component of Citron extracted from Grapefruit seed and Citrex were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion method was used to measure the antimicrobial activity. Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus was similar when the concentration of Manuka oil and $\alpha$-pinene are 10u/paper disk. However, antimi-crobial activity of Manuka oil fDr Escherichia coli, Staphylococcus aurem was better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene was low. Antimicrobial activity of Oxy’less clear is better than that of propyl para hydroxybenzoate(PPHB), R-limonene at all the concentration and is similar to that of $\alpha$-pinene. Inhibitory Effects of Serotonin Derivatives on Adipogenesis (홍화씨 추출물 유래 세로토닌 유도체의 지방전구세포 분화억제 효능에 대한 연구)

  • N-feruloylserotonin (FS) and N-(pcoumaroyl) serotonin (CS), serotonin derivatives, which have been isolated as major and unique phenolics of safflower seed extract (SSE), are member of hydroxycinnamic acid amides and are implicated in the defense against pathogen infection and insect feeding. In this study, we evaluate inhibitory effects of N-(p-Coumaroyl)serotonin and N-Feruloylserotonin on adipogenesis using oil-red O staining, triglyceride and GPDH activity. we found that while serotonin itself did not suppress differentiation of preadipocytes into adipocytes, N-(p-Coumaroyl)serotonin and N-Feruloylserotonin inhibited the differentiation of preadipocytes into adipocytes in a concentration-dependent manner. In addition, they showed antioxidant effects in DPPH assay. Taken together, these results show that N-feruloylserotonin (FS) and N-(pcoumaroyl) serotonin (CS) suppress differentiation of preadipocytes, suggesting the possibility that these serotonin derivatives can be utilized as an anti-obesity agent.

The Antioxidant and Skin Whitening Effect of Withania somnifera (Winter Cherry) (윈터체리 추출물의 항산화 및 미백 개선 효과)

  • Withania somnifera has been used in folk medicine to treat various ailments for centuries. In this studies to investigate the whitening effect of Withania somnifera extracts as an active ingredient for whitening cosmetics, the antioxidant capacity and the effects of Withania somnifera extracts on melanogenesis in B16-F10 melanoma cells were identified. Withania somnifera extracts significantly reduced both tyrosinase activity and melanin content in a concentration-dependent manner. Furthermore, it was found that Withania somnifera extracts decreased ${\alpha}-MSH$ (melanocyte-stimulating hormone)-induced tyrosinase activity and MITF(microphthalmia associated transcription factor) protein expression. These data indicate that Withania somnifera extracts attenuate ${\alpha}$-MSH-stimulated melanin synthesis by modulating MITF expression and that they may be a useful therapeutic agent for treating hyperpigmentation and an ingredient of whitening cosmetics.

The Protective Effects of Cornus walteri Wanger Leaves against UV Induced Cellular Damage in Human Fibroblast (자외선에 의한 세포손상에 대한 말채나무잎의 보호효과)

  • Cornus walteri Wanger has been used in folk medicine in Korea. Ultraviolet (UV) irradiation has been known as a major cause of photo damage in skin. In the present study, research on how to cure damaged cells by UVB was conducted using an extract of Cornus walteri Wanger leaves (CWE), which was treated with an enzyme. CWE was applied to human dermal fibroblasts (HDFs) affected by UVB. UVB-irradiated HS68 cells showed increased caspase-3 activity, phosphorylation of p53, ${\gamma}H2AX$, cyclobutane pyrimidine dimers (CPDs) formation, and DNA fragmentation compared with non-irradiated cells. However, all these effects were inhibited by treatment with CWE for 12 h after UVB irradiation. Furthermore, CWE has proved not to cause primary skin irritation through the human patch test. Collectively, these results suggest that CWE could be a new potential candidate as photoprotective agent against UVB-induced cellular damage in HDFs.

The Study on Antimicrobial and Antifungal Activity of the Wild Seaweeds of Jeju Island (제주 자생 해양식물들의 항균 및 항산화 활성 연구)

  • In this study, we investigated the antimicrobial and antifungal activity from the wild seaweeds of Jeju island. The active ingredients of the seaweeds were prepared by 80% methanol extraction. Antimicrobial and antifungal activity of seaweed extracts was examined. We found that 6 plant extracts among 45 plants, namely, Codium contractum, Undaria pinnatifida, Ishige sinicola, Ishige okamurai, Ishige okamuriai, Ecklonia cava, Hizikia fusiformis, Ulva fasciata, Ulva pertusa, Sargassum siliquastrum, Ecklonia kurome, Gracilaria textorii, significantly inhibited growth of harmful microorganisms. Additionally, according to DPPH assay. 2 plant extracts were found to have antioxidant activities. Taken together, these results suggest the possibility that 11 plant extracts can be utilized as an antimicrobial agent.

Reliability of In Vitro Assay for Initial Depigmenting Agent Screening (미백제 선발을 위한 In Vitro 측정법의 신뢰도)

  • Initial screening assay for depigmenting agents includes in vitro mushroom tyrosinase assay and antioxidant assay. Based on this screening result, melanin synthesis in melanocyte, in screened samples, is further measured. Measuring cellular melanin needs time, human resource, and skills. Therefore initial screening method should be reliable. We examined, 34 Chinese herbs, correlated the screening assay methods with cellular melanin. No reliable relationship was observed between factors, indicating the limitation in the use of these assays, probably due to the complexicity of melanogenesis.

Flame-retarding effects depending on the number of phosphonate groups attached to phosphorus flame-retarding compounds and coating binder resins (인계 난연화합물 및 코팅 바인더 수지에 부착된 phosphonate group에 따른 난연효과)

  • In this study, we prepared phosphorous flame-retarding coating solutions by mixing triphosphate (3 phosphonate), phytic acid (6 phosphonate), or ammonium polyphosphate (10 phosphonate) with boric acid as a crosslinking agent and acryl resin binder. Prepared phosphorous flame-retarding coating solutions were coated onto non-woven fabrics, respectively, to obtain high flame-retarding effects. These prepared flame-retardant non-woven fabrics were evaluated using smoke density standard test (ASTM E662), limit oxygen index standard test (ISO E622), and vertical burning standard test (UL 94). Their flame-retarding effects were affected by the number of phosphonate groups. Regardless of natural or synthetic binder resins, their effects showed the following order: ammonium polyphosphate > phytic acid > triphosphate. Natural hydrocarbon compounds were also examined to determine the possible retardancy of binder resins. Results showed that natural hydrocarbon binder resins could be used for preparing fire-retardant nonwoven fabrics.

Synthesis of 2',4'-Dimethoxyflavone and Its Anti-pigmentation Effect (2',4'-Dimethoxyflavone의 합성과 멜라닌 생성 저해 효과)

  • We synthesized 2',4'-dimethoxyflavone and investigated the effects on melanogenesis. To determine the effects as a whitening agent, various in uitro tests were performed such as free radical scavenging activity, melanin assay, tyrosinase activity and expression of tyrosinase, TRP-1 and TRP-2 (western blot and RT-PCR) in Bl6 melanoma cells. 2',4'-Dimethoxyflavone showed neither free radical scavenging activities against 1,1- diphenyl -2-picrylhydrazvl (DPPH) radical and inhibition of mushroom tyrosinase activity, 2',4'-dimethoxyflavone significantly inhibited melanin production in B16 melanoma cells. 2',4'-Dimethoxyflavone treatment (48h) suppressed the biosynthesis of melanin up to 27% at $5{\mu}g/mL$ and reduced tyrosinase activity up to 20% at $5{\mu}g/mL$ in B16 melanoma cells. 2',4'-Dimethoxyflavone was also able to significantly inhibit tyrosinase and TRP-1 expression in protein and mRNA level. These results suggest that 2',4'-dimethoxyflavone inhibits melanin biosynthesis at the level of enzyme activity and protein mRNA expression B16 melanoma cells. Therefore, 2',4'-dimethoxyflavone may be useful as a new whitening agent in cosmetics.
