In this work, comparative study on antioxidative activities of extracts from Glycyrrhiza uralensis (G. uralensis) produced in Korea and in China and Glycyrrhiza glabra (G. glabra) produced in Uzbekistan was conducted. Among three origins, 50% ethanol extracts (21.15 ${\mu}g/mL$), ethyl acetate fraction (29.15 ${\mu}g/mL$) and aglycone fraction (3.26 ${\mu}g/mL$) of G. uralensis from Korea showed the higher free radical (1,1-phenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) than extracts from other origins. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of extracts from three origins on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using luminol-dependent chemiluminescence assay 50% ethanol extract (1.00 ${\mu}g/mL$) and ethyl acetate fraction (0.34 ${\mu}g/mL$) of G. uralensis from China showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of G. uralensis and G. glabra extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. 50% ethanol extract and aglycone fraction of G. uralensis and G. glabra extracts from three origins showed cellular protective effects in a concentration dependent manner (5 ~ 50 ${\mu}g/mL$). Aglycone fraction of G. uralensis from Korea (${\tau}_{50}$ = 847.4 min)especially showed cellular protective effects four times higher than that from China (${\tau}_{50}$ = 194.3 min). These results indicate that G. uralensis and G. glabra extracts, which have been used as whitening agent, could be applicable to functional cosmetic ingredient as a natural antioxidant. Judging from the prominent cellular protecitve effects, it is concluded that G. uralensis and G. glabra extracts can protect the skin from $^1O_2$ and various ROS induced by UV.
L-ascorbic acid (Vitamin C) and peptide are both useful compounds for collagen biosynthesis in cosmeceuticals (cosmetic and pharmaceutical fields). The instability of these compounds, however, limit their application in these industries. In this report, we describe the development of a novel compound, Stabilized Ascorbyl Pentapeptide (SAP), which physically is much more stable than L-ascorbic acid in water. The inhibitory effects of this SAP compound on tyrosinase and melanin synthesis is comparable to that of L-ascorbic acid. Importantly, the SAP compound displays no cytotoxicity at a high concentration (5 mM). The ability of SAP to promote collagen biosynthesis is greater than that of L-ascorbic acid or the KTTKS peptide alone. Considering the in vitro stability and functional effects, our data strongly suggest that the SAP compound is a good candidate not only as a cosmetic ingredient, but also as a wound healing agent.

Whitening agents are eagerly demanded especially by oriental women who often suffers from the pigmentary disorders such as melasma and solar lentigines. As these pigmentary disorders are exacerbated by ultraviolet (UV), the whitening agents could exert its effect not only by inhibiting melanin synthesis but also by inhibiting UV activated signals. Eumelanin protects UV-induced DNA damages so that the chemicals which could reduce UV-induced DNA damages might be the ideal lightening agents. The effect of newly synthesized antioxidants, a-tocopheryl ferulate, on protective effect for UV-induced DNA damages as well as inhibiting melanin synthesis are briefly shown. For clinical evaluation, our results of the efficacy of lightening agents on treating pigment macules in combination with chemical peeling are shown. Furthermore, newly developed facial image analyzers to quantitatively evaluate the improvement of pigment macules are introduced.

Stem bark extracts of Fraxinus rhynchophylla Hance were found to contain two major bioactive components, esculetin and esculin. Esculetin substantially inhibited melanogenesis in B16F10 melanoma cells, with an $IC_{50}$ value of $2.8{\mu}M$, and reduced melanin synthesis in Melan-A cells. Moreover, esculetin suppressed melanin biosynthesis by inhibiting mushroom tyrosinase activity, with an $IC_{50}$ value of $40{\mu}M$. Taken together, these results suggest that esculetin could serve as an effective skin-lightening agent that inhibits melanin production by regulating the activity of melanogenic enzymes.

The purpose of this study was to investigate anti-melanogenesis effects of mulberry seed extracts (MSE). MSE inhibited melanogenesis in melan-a cells at $10{\mu}g/mL$ without cytotoxicity. Also, MSE decreased tyrosinase, tyrosinase-related protein-1 (TRP-1) protein expression in the melan-a cells. To identify the signaling pathway of MSE, the ability of MSE to influence extracellular signal-regulated protein kinase (ERK) activation was investigated. MSE induced ERK protein expression in a dose-dependent manner. In addition, MSE presented inhibition of the body pigmentation in vivo zebrafish model. These results suggest that MSE may be an effective anti-melanogenesis agent regulating the expression of ERK protein and melanogenic enzymes.

To develop a new functional agent for cosmetics, we investigated the anti-aging activities in fibroblasts using sweroside isolated from Nymphoides indica. The anti-aging effect of sweroside was validated in CCD-986sk cells. Results showed that sweroside inhibited expressions of UVB-induced ROS and increased pro-collagen. Furthermore, sweroside had inhibited MMP-1 expression. Taken together, the results suggested that sweroside has considerable potential as a new cosmetics ingredient with an anti-aging effect.

${\beta}$-Carotene has been known as an effective quenching agent of singlet oxygen and the carotenoid pigments in general are expected to protect cells against photosensitized oxidations. We are determined the quenching rate constants of several Ketocarotenoids including capsanthin, capsanthin diester, astaxanthin and fucoxanthin, and the relative quenching actiyities against singlet oxygen were compared with those of ${\beta}$-carotene and reported carotenoids. Nevertheless the ketocarotenoids exhibited lower quenching rate constants than ${\beta}$-carotene, they showed more pronounced protective activitives than ${\beta}$-carotene against photohemlysis induced by singlet oxygen. Among the ketocarotenoids investigated, fucoxanthin indicated a significant protective activity for the cell. The results suggested that. 1) 1O2 may be alikely initiator of photohemolysis, but this reaction is followed by slow dark reactions involving secondary reactive species. 2) For protection of RBC against photodynamic action with carotenoids, carotenoids having functional groups such as -C=0 and -OH groups are most efficient. This suggests that partition of carotenoids between the buck and the mombrane and/or their specific binding to membrane proteins are more critical for the photo-protection by carotenoids than is a diffusional quenching of 1O2.
The inner shell of the chestnut (Castanea crenata S. et Z., Fagaceae) has been used as an anti-wrinkle/skin firming agent in East Asia, and preliminary experiments have found that a 70% ethanol extract from this plant material can prevent cell detachment of skin fibroblasts from culture plates. In order to examine the molecular mechanisms underlying this phenomenon, its effects on the expression of adhesion molecules, such as fibronectin and vitronectin, were investigated using the mouse skin fibroblast cell line, NIH/3T3. Using fixed-cell ELISA, Western blotting and immunofluorescence cell staining, it was clearly demonstrated that the chestnut inner shell extract enhanced the expression of the cell-associated fibronectin and vitronectin. Scoparone (6,7-dimethoxycoumarin), isolated from the extract, also possessed similar properties. These findings suggest that the enhanced expression of the adhesion molecules may be one of the molecular mechanisms for how the chestnut inner shell extract preventing cell detachment and may be also responsible for its anti-wrinkle/skin firming effect.
Lysosome organelle extract (LOE) was derived from egg whites. The lysosome is an intracellular organelle that contains several hydrolysis enzymes. Previous studies have reported that LOE performs important functions, such as melanin de-colorization and anti-melanin production in B16F10 melanoma cells. However, its principal molecular and cellular mechanisms have not been elucidated till date. In non-cytotoxic conditions, LOE significantly inhibited α-MSH induced melanin synthesis of murine B16F10 cells. The anti-melanogenic activity of LOE was mediated by suppressing the mRNA expression of tyrosinase enzyme, tyrosinase related protein-1/2 (TRP-1/2), and microphthalmia-associated transcription factor (MITF) genes. By performing western blot analysis, we found that LOE significantly attenuated melanogenesis. In this case, LOE helped in increasing extracellular receptor kinase (ERK) phosphorylation in α-MSH induced B16F10 cells. Furthermore, MITF is found to be a key regulatory transcription factor in melanin synthesis; it was down-regulated by LOE through ERK phosphorylation. In this experiment, PD98059 (MEK inhibitor) was used to check whether LOE directly regulated the activity of ERK. Although LOE exerted inhibitory effect on melanin synthesis, we could not observe this effect in PD98059-treated α-MSH induced B16F10. These results strongly indicate that LOE is related to ERK activation and MITF degradation in anti-skin pigmentation. Hence, LOE should be utilized as a whitening agent of skin in the near future.

The fruit of the Crataegus pinnatifida bunge has been known to have a role as a digestive stimulant, and is used for postpartum abdominal pain and women's menstrual pain. It is used for coronary artery disease, angina pectoris, hypertension and hyperlipidemia. Ascorbic acid, hydroquinone, retinoids, alpha-hydroxy acids, kojic acid, azelaic acid have been used for cosmetic whitening and medical depigmentation. To determine whether Crataegus pinnatifida bunge fruit also has whitening and depigmentation effect, tyrosinase inhibition assay was performed with American Crataegus pinnatifida Bunge ethanol extracts, Korean Crataegus pinnatifida Bunge (Cra) ethanol extracts and Arbutin as a positive control as previously described by Korean FDA guideline. Korean Cra fruit ethanol extracts were 1.87 fold more inhibitory function to tyrosinase activity than American Cra in the experimental condition that inhibitory function to tyrosinase activity of Korean Cra Arbutin is 81.8% when compared to that of the standard control Arbutin as 100%. These results suggest that ethanol extracts of Crataegus pinnatifida bunge have significant whitening effects and may provide the basis for development of cosmetic whitening agent and medical depigmentation applications.
