  • The determination method for 11 corticosteroids (betamethasone, cortisol. cortisone, cortisone acetate. dexamethasone. cortisol acetate, isoflupredone acetate, methylprednisolone. prednisone, prednisolone, and triamcinolone acetonide) in a herbal medicinal preparation (Sibjeondaibotang) by a gas chromatography-mass spectrometric (GC-MS) method with selected ion monitoring (SIM) mode is described.   • Background: The renal function of individuals is one of the reasons for the variations in therapeutic response to various drugs. Patients with renal impairment are often exposed to drug toxicity, even with drugs that are usually eliminated by hepatic metabolism. Previous study has reported an increased plasma concentration of indoxyl sulfate and decreased plasma concentration of $4{\beta}$-hydroxy (OH)-cholesterol in stable kidney transplant recipients, implicating indoxyl sulfate as a cytochrome P450 (CYP) inhibiting factor. In this study, we aimed to evaluate the impact of renal impairment severity-dependent accumulation of indoxyl sulfate on hepatic CYP3A activity using metabolic markers. Methods: Sixty-six subjects were enrolled in this study; based on estimated glomerular filtration rate (eGFR), they were classified as having mild, moderate, or severe renal impairment. The plasma concentration of indoxyl sulfate was quantified using liquid chromatography-mass spectrometry (LC-MS). Urinary and plasma markers ($6{\beta}$-OH-cortisol/cortisol, $6{\beta}$-OH-cortisone/cortisone, $4{\beta}$-OH-cholesterol) for hepatic CYP3A activity were quantified using gas chromatography-mass spectrometry (GC-MS). The total plasma concentration of cholesterol was measured using the enzymatic colorimetric assay to calculate the $4{\beta}$-OH-cholesterol/cholesterol ratio. The correlation between variables was assessed using Pearson's correlation test. Results: There was a significant negative correlation between MDRD eGFR and indoxyl sulfate levels. The levels of urinary $6{\beta}$-OH-cortisol/cortisol and $6{\beta}$-OH-cortisone/cortisone as well as plasma $4{\beta}$-OH-cholesterol and $4{\beta}$-OH-cholesterol/cholesterol were not correlated with MDRD eGFR and the plasma concentration of indoxyl sulfate. Conclusion: Hepatic CYP3A activity may not be affected by renal impairment-induced accumulation of plasma indoxyl sulfate.

  • 1. In the present experiment, necrotic myocarditis were produced only in suckling mice but also in weanlings or young adult mice by intraperitoneal inoculation with the Tyzzer's organism KK of cat origin, whereas the organism MSK from the mouse was not capable of producing such lesions. 2. Significant difference in pathogenicity for 1-to 6-week-old mice was demonstrable between the two organisms after intraperitoneal inoculation without cortisone.

  • To observe the effect of cortison on the oxytalan fibers of the periodontal membrane, the albino rats of each groups were injected intramuscularly with 2.5mg. of cortison/kg. body weight daily for 3,7,10,14, and 21 days. After 14 days of cortison administration, the distributions of the oxytalan fibers were irrgular. In their path from cementum to alveolar bone, the fibers pass wave-like. Their density was loose and their stain ability was lesser than the normal. Prolonged administration of cortison impaired the development and the normal form of the oxytalan fibers.

  • Non traumatic aseptic necrosis of the head of the femur is well documented with respect to its clinical roentgenographic and pathological aspect, but not with respect to etiology. A number of reports have associated various diseases and conditions with necrosis of the femoral head. Prominent among these are alcoholism, steroid therapy and hemostatic disorder. The increased incidence of avascular necrosis of bone associated with long-term corticosteroid therapy has aroused much interest. The pathogenesis of cortisone-induced avascular necrosis is as yet unproved. But cortisone-induced fatty liver with hyperlipidemia, leading to systemic embolization of fat and avascular necrosis of bone has much experimental support. We present a case of bilateral avascular necrosis of the femur head following several epidural steroid injections in a 30 year old male patient who had low back pain with radiation to both legs for 1 year and who had Cushinoid appearance for several months. One week later, he had a total replacement of both hip operation.

  • This study was investigated that effects of steroid hormones on serum sodium and potassium concentration in parathyroidectomized chickens. The results are as followings; 1) Serum sodium and potassium concentration in SHAM-controls were unchanged. 2) In parathyroidectomized chickens, serum potassium level increased more than that of control and serum sodium level appeared no change in each group. 3) In group of administration of cortisone in parathyroidectomized chickens, serum potassium level increased generally more than that of control and serum sodium level appeared no change in each group. 4) In group of administration of testosterone in parathyroidectomized chickens, serum potassium level increased generally more than that of control and serum sodium level revealed no change in each group.

  • 최근 치수염증 생활치수절단술 치수복담 상아질화각과민증 등에 Cortisone제제가 널리 사용되는 바 저자는 항생제에 Dexamethasone을 함유시켜 실험군으로 삼고, 항생제 단독으로 사용한 것은 대조군으로 삼아, 두 군의 살균효과를 비교하기 위하여 한천배양기상에선 백색 포도상구균으로, 혈액가 천한배양기상에선 α-용혈성 연질상구균과 β-용혈성 연질상구균을 사용하여 균성장억제대를 측정 관찰한바 다음과 같은 결론을 얻었다. 1. Dexamethasone이 함유된 항생제에선 항생제 단독으로 사용한 것보다 균이 더 넓게 성장했다. 2. 항생제의 살균효과는 Dexamethasone에 의해 약간 저하되는 것을 볼수 있었다. 3. 따라서 Dexamethasone과 항생제 제제는 균이 존재시 별 효과가 없다고 사료되는 바이다.

Effects of Foot-Reflexology Massage on Fatigue, Stress and Postpartum Depression in Postpartum Women (발 반사마사지가 산욕기 산모의 피로, 스트레스, 산후 우울에 미치는 효과)

  • Purpose: To identify the effects of foot reflexology massage on fatigue, stress and depression of postpartum women. Methods: A nonequivalent control group pre-post design was used. A total of 70 women in a postpartum care center were recruited and were assigned to the experimental group (35) or control group (35). Foot reflexology massage was provided to the experimental group once a day for three days. Data were collected before and after the intervention program which was carried out from December, 2013 to February, 2014. Data were analyzed using Chi-square test, Fisher's exact test, and t-test. Results: The level of fatigue in the experimental group was significantly lower than the control group (t= - 2.74, p=.008). The level of cortisol in the urine of women in the experimental group was significantly lower than the control group (t= - 2.19, p=.032). The level of depression in the experimental group was significantly lower than the control group (t= - 3.00, p=.004). Conclusion: The results show that the foot reflexology massage is an effective nursing intervention to relieve fatigue, stress, and depression for postpartum women.

  • The activity of taurine transporter is affected by various extracellular stimuli such as ion, hormone and stress. To assess effects of steroid hormones antral cyclosporin A (CsA) on the taurine transporter activity, murine monocytic RAW264.7 cell line was stimulated with dexamethasone (DM), triamcinolone (TA), cortisone (CS), hydrocortisone (HCS), prednisone (PSN), prednisolone (PSL) and methylprednisolone (MPSL) in the presence of 12-0-tetradecanoylphorbol-13-acetate(TPA). Treatment of TPA on the cell line led to significant reduction of taurine transporter activity. However, in case of stimulation of the cells with steroid hormones in the presence of TPA, all of them recovered TPA-induced reduction of the taurine transporter activity. Treatment of the cells with CsA led to significant reduction of the taurine transporter activity. Ionomycin (IM) recovered the reduced taurine transporter activity by CsA, but failed in the presence of EDTA, a calcium chelating agent. These results showed that glucocorticoid hormone recovered TPA-induced reduction of taurine transporter activity and that IM recovered CsA-induced reduction of the transporter activity by increasing intracellular free $Ca^{++}$ concentration.n.

  • The application of steroids has steadily increased thanks to their therapeutic effects. However, alternatives are required due their severe side effects; thus, studies on the activities of steroid derivatives are underway. Sugar derivatives of nandrolone, which is used to treat breast cancer, as well as cortisone and prednisone, which reduce inflammation, pain, and edema, are unknown. We linked O-glucose to nandrolone and testosterone using UDP-glucosyltransferase (UGT-1) and, then, tested their bioactivities in vitro. Analysis by NMR showed that the derivatives were 17β-nandrolone β-ᴅ-glucose and 17β-testosterone β-ᴅ-glucose, respectively. The viability was higher and cytotoxicity was evident in PC12 cells incubated with rotenone and, testosterone derivatives, compared to the controls. SH-SY5Y cells incubated with H2O2 and nandrolone derivatives remained viable and cytotoxicity was attenuated. Both derivatives enhanced neuronal protective effects and increased the amounts of cellular ATP.
