Chemical Compositions of Corni Fructus and Separating Properties of Its Flesh by drying (산수유 열매의 화학성분과 건조에 따른 과육분리특성)

  • An attempt was made to investigate the some constituents and separation of flesh by drying of Corni Fructus for utilization as processing foods of Cornus officianalis S. et Z. Moisture and reducing sugar contents of Corni Fructus flesh were 40.4% and 18.6%, respectively. The content of anthocyanin was 183.0 mg/100g(dry base). The free sugars of Corni Fructus were consisted of glucose and fructose and their contents were 7.9% and 9.0%, respectively. The compositions of organic acids were malic and succinic acids which contained 1.9% and 0.5%, respectively. The titrable acidity as malic acid and pH were 3.6% and 3.19. The decrease ratio of weight and the drying temperature as the optimum conditions for the separation of flesh and seed of Corni Fructus were $30{\sim}35%$ and $60^{\circ}C$.

Pharmacological Studies on Ether Fraction of Corni Fructus (산수유 에텔분획물의 약리작용에 관한 연구)

  • The ether fraction obtained from dry fruit of Cornus officinalis was investigated for the anti-inflammatory action, acute toxicity and central nervous system activities. From several pharmacological examinations, it was found that the ether fraction suppressed considerably carrageenin edema at the dose of 600mg/kg p.o. in rats, inhibited the granuloma formation in rats as given 100mg/kg p.o. for 7 days and decreased the swelling of both of complete adjuvant injected and noninjected (contralateral) paws of the rats at the dose of 100mg/kg p.o. given for 14 days. The $LD_{50}$ of the fraction are estimated to be more than 2,000mg/kg p. o. and 642mg/kg i. p. in mice. And the fraction did not show any sedative, stimulative, analgesic and anticonvulsant action but exhibited hypothermic action. These results might be concluded that the ether fraction of Corni fructus showed anti-inflammatory action in both of acute and chronic type models without any considerable central nervous depressant activity and exhibited very weak acute toxicity in mice.

The protective effect of methanol extract of Corni Fructus on brain injury caused by unilateral common carotid artery occlusion in mice (산수유(山茱萸) 메탄올 추출물이 편측 경동맥 폐색으로 유도된 생쥐의 허혈성 뇌손상에 미치는 영향)

  • Objectives : This study was conducted to evaluate the effects of Corni Fructus, the dried fruits of Cornus officinalis Sieb., on unilateral common carotid artery occlusion (UCCAO) in mouse model. Methods : The Corni Fructus used in the experiment was extracted with anhydrous methanol, then filtered and freeze-dried. C57BL/6 mice used in the experiments were conducted left UCCAO surgery to set up UCCAO rodent model for mice. The mice were divided into five groups for evaluate the effect of methanol extract of Corni Fructus (COM) on UCCAO induced ischemic brain injury. The expression levels of nitric oxide in cerebrum and serum, body weight change were measured. To determine the effect of UCCAO and COM administration on brain neurons, morphological changes of the cerebrum through a microscope was conducted. And western blot was performed to confirm the underlying mechanism of neuroprotective effect of COM administration. Results : COM administered UCCAO groups (CO50, CO150, and CO500) had no significant effects on nitric oxide production in ipsilateral hemisphere proteins and sera. The CO500, 500 mg/kg COM administration, attenuated UCCAO-induced p38 inflammatory signaling pathway and inflammatory mediators such as iNOS and COX-2. The CO500 group showed resilient morphological changes of hippocampus neuronal cells about brain damage caused by decreased flow of blood. These group also showed decreased inflammation and cellular stress response in neuronal cells. Conclusions : From these results, COM has a neuroprotective property via moderating inflammatory factors and cellular stress inducing factors in brain cells.

Anti-Glycemic and Anti-Lipid Effect of Ethanol Extract of Supungsunkihwan-gagambang in High Fat Diet-induced Obese Mice (수풍순기환가감방(搜風順氣丸加減方) 투여가 비만형 제2형 당뇨병 동물모델의 당대사 및 지질대사에 미치는 영향)

  • Objectives: This study was undertaken to evaluate anti-glycemic and anti-lipid effects of Supungsunkihwan-gagambang, which is composed of Cornus officinalis, Dioscorea Batatas Decaisne, Auantii fructus and Platicodon grandiflorum. Methods: Diabetes was induced in ICR male mice ($24{\pm}1g$) with Surwit's high fat and high sucrose diet. Mice were divided into 3 groups (n=10) of normal, control and Supungsunkihwan-gagambang. The Supungsunkihwan-gagambang group was given 5% herbal medicine in their diet. The animals were fed on each experimental diet for 8 weeks. Body weight was assessed every week. At the 7th week, the fasting blood sugar (FBS) and oral glucose tolerance test (OGTT) were conducted in all experimental groups. After 8 weeks, fructosamine, lipid profile, epididymal fat weight, liver weight and white adipose tissue (WAT) size were measured. Results: Supungsunkihwan-gagambang significantly reduced FBS, OGTT and fructosamine. It also increased high density lipoprotein (HDL) cholesterol and significantly reduced triglyceride/HDL cholesterol ratio, total cholesterol/HDL cholesterol ratio and WAT size. Conclusions: These results show that Supungsunkihwan-gagambang improves anti-glycemic and anti-lipid effect in high fat diet-induced obese mice. Therefore we suggest that Supungsunkihwan-gagambang could be an effective treatment for patients with type 2 diabetes.

Antioxidant Effects of Corni Fructus in GC-1 Cells (산수유(山茱萸)가 남성 생식세포 GC-1의 항산화에 미치는 영향)

  • The purpose of this study is to examine the antioxidant activity in the germ cells of the extract of Corm fructus. The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, GC-1 cell viability by a modified MTT assay, the effects on $H_2O_2$-induced cytotoxicity by MTT assay and lipid perixidation by malondialdehyde (MDA) formation, respectively. The results showed that the extract scavenged DPPH radical with the IC50 being $200{\mu}g/mL$. The extract at concentrations of $10-500{\mu}g/ml$ showed dose-dependent in growth of GC-1 cell. $H_2O_2$-induced cytotoxicity (63.0%) was blocked by the extract (10, 50, 100, 250 and $500{\mu}g/ml$) concentration-dependently. Furthermore, the extract (50, 100 and $250{\mu}g/ml$) also displayed a dose-dependent reduction of MDA formation on $H_2O_2$-induced lipid peroxidation. In conclusion, the extract of Corm fructus has potent antioxidant activity.

The comparisons of Lycii Radicis Cortex and Corni Fructus water extract effects on streptozotocin-induced diabetes in rats (지골피, 산수유의 streptozotocin 유발 당뇨흰쥐에서의 효과 비교 연구)

  • Objectives : This study aimed to compare the anti-diabetic efficacy of Lycii Radicis Cortex (Lycium chinense Mill.) and Corni Fructus (Cornus officinalis) on streptozotocin (STZ)-induced diabetes in rats. Methods : Male Sprague-Dawley rats were divided into four groups; normal, STZ-control, Lycii Radicis Cortex extract-administrated group (LRC) and Corni Fructus extract-administrated group(CF). Diabetes in rats was induced by intraperitonal injection with streptozotocin (STZ) at doses of, 30 mg/kg (body weight) for 5 days (once per a day). STZ-induced diabetic rats were orally administrated LRC and CF extract daily for 4 weeks at doses of 300 mg/kg. Fasting blood glucose, total cholesterol (TC), triglyceride (TG), blood urea nitrogen (BUN) and creatinine were measured in sera of rats. Histopathological changes of kidney, liver and lung tissues were observed by microscope after H&E staining. Results : There were no differences in body and kidney weights, food intake and water intake in LRC- and CF-administrated groups compared with STZ control group. However, glucose, TC and TG levels in serum were significantly decreased in LRC-administrated groups compared with STZ-control group. In histopathological analysis of kidney, liver and lung, both LRC- and CF-administrated groups showed the inhibition of morphological damage. Conclusions : These results suggest that LRC and CF have a biological action on STZ-induced diabetes in rats via decreasing the serum TG and TG levels and may protect the morphological changes of kidney, liver and lung.

Mixture of Corni Fructus and Schisandrae Fructus improves testosterone-induced benign prostatic hyperplasia through regulating 5α-reductase 2 and androgen receptor

  • BACKGROUND/OBJECTIVES: Benign prostatic hyperplasia (BPH) characterized by an enlarged prostate gland is common in elderly men. Corni Fructus (CF) and Schisandrae Fructus (SF) are known to have various pharmacological effects, including antioxidant and anti-inflammatory activities. In this study, we evaluated the inhibitory efficacy of CF, SF, and their mixture (MIX) on the development of BPH using an in vivo model of testosterone-induced BPH. MATERIALS/METHODS: Six-week-old male Sprague-Dawley rats were randomly divided into seven groups. To induce BPH, testosterone propionate (TP) was injected to rats except for those in the control group. Finasteride, saw palmetto (SP), CF, SF, and MIX were orally administered along with TP injection. At the end of treatment, histological changes in the prostate and the level of various biomarkers related to BPH were evaluated. RESULTS: Our results showed that BPH induced by TP led to prostate weight and histological changes. Treatment with MIX effectively improved TP-induced BPH by reducing prostate index, lumen area, epithelial thickness, and expression of BPH biomarkers such as 5α-reductase type 2, prostate-specific antigen, androgen receptor, and proliferating cell nuclear antigen compared to treatment with CF or SF alone. Moreover, MIX further reduced levels of elevated serum testosterone, dihydrotestosterone, and prostate-specific antigen in BPH compared to the SP, a positive control. BPH was also improved more by MIX than by CF or SF alone. CONCLUSIONS: Based on the results, MIX is a potential natural therapeutic candidate for BPH by regulating 5α-reductase and AR signaling pathway.

Biological Activities of Extracts from Corni fructus, Astragalus membranaceus and Glycyrrhiza uralensis (산수유, 황기, 감초 추출물의 생리활성)

  • Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of the water and ethanol extracts from medicinal herbs. Methods : Three kinds of medicinal herbs(Corni fructus, Astragalus membranaceus and Glycyrrhiza uralensis) were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidative and anticancer activities. Results : The highest polyphenol contents of the water and ethanol extracts from medicinal herbs were 342.14 mg and 435.62 mg per 100 g of Cornus officinalis, respectively. The highest electron donating abilities (EDA) of the water and ethanol extracts from Glycyrrhiza uralensis were 88% and 91% at 1,000 ppm, respectively. The water and ethanol extracts from Astragalus membranaceus had the highest nitrite scavenging abilities (NSA) at 1,000 ppm. The highest anticancer activity of the extracts were from Glycyrrhiza uralensis against both of MDA and A549 cells. Conclusions : These results suggest that the medicinal herbs can be used as natural antioxidant to prevent oxidative damage in normal cells probably because of their antioxidant characteristics.

Protective Effect of Ursolic Acid from Corni fructus on the Hydrogen Peroxide-induced Damage of HEI-OC1 Auditory Cells (산수유로부터 추출한 ursolic acid가 과산화수소로 손상된 HEI-OC1 청각세포보호에 미치는 영향)

  • The fruits of Cornus officinalis have been used in traditional Oriental medicine for treatment of inner ear diseases, such as tinnitus and hearing loss. In the present study, we showed that the ursolic acid obtained from Corni fructus protected HEI-OC1 auditory cells from hydrogen peroxide cytotoxicity in a dose-dependent fashion. In addition, to investigate the protection mechanism of ursolic acid on hydrogen peroxide cytotoxicity toward HEI-OC1, we measured the effects of ursolic acid on lipid peroxidation and activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) in hydrogen peroxide treated cells. Ursolic acid (0.05 - 2 ${\mu}g/ml$) had protective effect against the hydrogen peroxide-induced HEI-OC1 cell damage and reduced lipid peroxidation in a dose-dependent manner. Pre-treatment with ursolic acid significantly attenuated the decrease in activities of CAT and GPX, but SOD activity was not affected by the ursolic acid or hydrogen peroxide. These results indicate that ursolic acid protects hydrogen peroxide-induced HEI-OC1 cell damage through inhibition of lipid peroxidation and induce the antioxidant enzymes CAT and GPX.

Effect of HTE001, an Herbal Formulation, on Electric Stimulation-induced Penile Erection in Rats (전기 자극으로 유발한 음경 발기력 측정 흰쥐 모델에 대한 HTE001의 발기력 상승효과)

  • Objectives : This study was conducted to investigate the effect of HTE001, a multi-herbal mixture consisting of 10 herbs, Cornus Frutus, Schizandrae Fructus, Rubi Fructus, Cnidi Fructus, Acanthopanacis senticosi Radix, Cinnamomi Cortex, Eucommiae Cortex, Allii Bulbus, Rehmanniae Radix and Ginseng Radix, on electrostimulation-induced penile erection in rats. Methods : Intracavernous pressure (ICP) and mean arterial blood pressure (MAP) were simultaneously monitored through electric stimulation of the cavernous nerve after the oral administration of HTE001 (30, 100, 300 mg/kg) in normal rats. Statistical analysis was performed on maximal intracavernous pressure (ICP), maximal intracavernous pressure/mean arterial blood pressure (ICP/MAP) ratio, and the area under the curve (AUC) of ICP/MAP ratio. Results : Oral administration of HTE001 300 mg/kg caused the ICP to increase in a frequency-dependent manner. And HTE001 300 mg/kg treatment group showed the highest value in the ICP/MAP ratio and the AUC value of the ICP/MAP ratio compared to the control group at 2 Hz, 6 Hz and 10 Hz, respectively without an effect on the mean arterial blood pressure under the same stimulation of the cavernous nerve. Conclusions : These results show that HTE001 improve penile erection and prolong the decay period in normal rats without affecting mean arterial blood pressure, and suggest that HTE001 could be a good therapeutic candidate to treat erectile dysfunction.
