  • Gentisic acid is a skin-whitening agent which inhibits the tyrosinase activity, an essential enzyme in the process of biological synthesis of melanin. Since melanin is synthesized in melanocytes located between the viable epidermis and dermis layer, drug amount delivered into the epidermis/dermis layer can provide valuable information for the biological effect of skin-whitening agents. The purpose of this study was to prepare the gentisic acid patches with 2% dodecylamine as enhancer, and to observe the in vitro skin permeation and in vivo skin deposition of gentisic acid. Gentisic acid in DuroTak 87-2510 patch formulation permeated across hairless mouse skin at the rate of $40.79\;{\mu}g/cm^2/hr$. In vivo study showed that the gentisic acid amount in both the stratum corneum and the viable epidermis/dermis increased with the increase of application time. The amount of gentisic acid in the stratum corneum was higher than that in the epidermis/dermis layer, and was expected to provide a reservoir effect even after removing the patches. Thus, the patch formulation seems to be useful for the topical delivery of skin-whitening agent into the epidermis/dermis layer, the target site.

Gelatinase, a Possible Etiologic Factor of Photoaging, is Present in Healthy Human Facial Skin and is Inhibited by Turmeric Extract

  • Influence of gelatinase on basement membrane (BM) structure was investigated by using a skin equivalent (SE) model. The results showed that (1) gelatinase produced by cells degraded the BM and (2) the addition of matrix metalloproteinase-specific inhibitor to the SE medium accelerated the formation of BM structure, indicating that gelatinase is involved in BM impairment. The activity of gelatinase was also studied in healthy human facial skin tissues. The result of in situ zymography revealed gelatinase activity around the basal layer of the epidermis, where BM integrity was severely compromised. Therefore, this enzyme was suggested to be associated with BM decomposition in human facial skin. To assess the behavior of gelatinase in stratum corneum (SC) non-invasively, an immunological study was performed. Since positive immunostaining of pro-gelatinase B was observed in SC stripped from sun-exposed skin, whereas no positive staining detected in SC of non-irradiated skin, gelatinase in the epidermis could be non-invasively detected by measuring gelatinase in SC. Gelatinase in SC of healthy female volunteers was monitored using a special film that sensitively and conveniently detects gelatinase. Ninetr percent of SC from facial skin (l00 women, 40's-50's) was gelatinase-positive. On the other hand, SC from non-irradiated skin was negative. These results strongly suggest that (1) gelatinase is constantly produced in the facial epidermis of most middle-aged woman during their daily life, and (2) the enzyme might be involved in the aging-related degeneration of both BM and the matrix fibers of the upper layer of the dermis, acting as a very important aging factor. Strong inhibitory activity against gelatinase was found in turmeric extract and identified curcumin as the major ingredient. Topical application of cream containing turmeric extract significantly decreased the number of gelatinase-positive SC clusters in human facial skins. These results indicated that turmeric is an effective ingredient to prevent skin from photo aging by suppressing chlonically upregulated gelatinase activity by UV and to improve skin condition.

Dermal Stability and In Vitro Skin Permeation of Collagen Pentapeptides (KTTKS and palmitoyl-KTTKS)

  • Collagen pentapeptide (Lys-Thr-Thr-Lys-Ser, KTTKS) and its palmitoylated derivative (pal-KTTKS) have received a great deal of attention as cosmeceutical ingredients for their anti-wrinkle effects. The objective of this study was to evaluate stability and permeability of KTTKS and pal-KTTKS in hairless mouse skin. In this study, a liquid chromatography-tandem mass spectrometric method was developed for the quantification of pal-KTTKS, and used for stability and permeability studies. Stability studies were performed using skin extracts and homogenates. Both KTTKS and pal-KTTKS were rapidly degraded, but pal-KTTKS was more stable than KTTKS. When protease inhibitors were added, the stability of both compounds (KTTKS and pal-KTTKS) improved significantly. In the skin permeation study, neither KTTKS nor pal-KTTKS was detected in the receptor solution, which indicates that neither compound could permeate through the full-thickness hairless mouse skin in the experimental conditions of this study. While KTTKS was not detected in any of the skin layers (the stratum corneum, epidermis, and dermis), pal-KTTKS was observed in all skin layers: $4.2{\pm}0.7{\mu}g/cm^2$ in the stratum corneum, $2.8{\pm}0.5{\mu}g/cm^2$ in the epidermis, and $0.3{\pm}0.1{\mu}g/cm^2$ in the dermis. In conclusion, this study indicated that pal-KTTKS had greater stability and permeability than that of un-modified KTTKS, and may be a useful anti-wrinkle and anti-aging cosmeceutical agent.

Effect of Skin Lipid Barrier Formation on Hataedock Treatment with Douchi

  • Objectives: Hataedock is indigenous treatment of Korean medicine that administers herbal extracts orally to newborn infants for remove the fetal heat. The purpose of this study was to evaluate skin lipid barrier formation effect of Hataedock treatment with Douchi. Methods: We measured the Western blot to observe the expression of protein such as involucrin and loricrin. Moreover, we observed immunohistochemical changes in NC/Nga mice. The 3-week-old NC/Nga mice were divided into 3 groups: the 3-week-old control group (3w-Ctrl), 5-week-old control group (5w-Ctrl), and the Hataedock-treated group (5w-FGT). Only the 5w-FGT group was treated with Douchi at the 3rd week. We identified the changes of the lipid skin barrier and protein differentiation through immunohistochemical changes of involucrin, loricrin, filaggrin and acid sphingomyelinase (ASM) in the stratum corneum. Results: The expression of involucrin and loricrin was increased in the Western blot that was treated with concentration of Douchi extracts. In 5w-FGT group, loricrin-positive reaction was increased by 54.0%, involucrin-positive reaction was increased by 84.0%, filaggrin-positive reaction was increased by 108.0% and ASM-positive reaction was increased by 91.0% in the stratum corneum. Conclusions: These results suggest that Hataedock treatment with Douchi promoted skin lipid barrier formation by promoting differentiation of keratinocytes.

THE SYNTHESIS, PHYSICAL PROPERTY, AND THE BIOLOGICAL ACTIVITY OF NOVEL NEO-CERAMIDES

  • Ceramides are currently emerging as the major skin care ingredients due to !heir barrier properties in the stratum corneum of the human skin. Thus, major cosmetic companies have developed synthetic ceramide analogs for their own use. In this study, several ceramide mimic compounds , new skin barrier lipids, were designed and synthesized, and their physical and biological properties were investigated to evaluate their skin care capability. Several structures were designed from the variation of hydrophobic alkyl chain and hydrophilic moiety by the use of molecular modeling software. The selected targets were synthesized, and their properties and activities were studied as the pure form, in the emulsion, or in the lamellar mixture containing cholesterol and fatty acid. Some compounds, such as 1,3-bis(N-(2-hydroxyethyl)-palmitoylamino)-2-hydroxypropane, enhanced the restoration of skin barrier damaged by SDS(sodium dodecyl sulfate), and by acetone treatment. The rate of restoration was comparable to that of natural ceramides. The synthesized compounds alleviated SDS induced skin irritation and facilitated lamellar phase liquid crystal formation. The treatment of 1,3-Dis(N-(2-hydroxyethyl)-palmitoylam ino)-2-hyd roxypropane on the acetone damaged skin revealed that the compound promoted the recovery of intercellular lipid lamellar structure of stratum corneum layer. The replacement of palmitoyl groups of the compound with shorter alkyl chain gave lower emulsion viscosity and liquid crystal density, suggesting easier formulation and poorer barrier activity. Most of the synthesized compounds were non-irritable in various toxicological tests proving that they can be safely introduced to the skin care formulations.

Deformable Liposomes for Topical Skin Delivery of Arbutin

  • The aim of this study was to investigate the effect of deformable liposomes with sodium cholate on the skin permeation and skin deposition of arbutin, a hydrophilic skin-whitening agent. Various compositions of liposomes were prepared by the extrusion method. Particle size distribution and entrapment efficiency were determined by the laser light scattering and the gel permeation chromatography, respectively. The in vitro rat skin permeation and deposition of arbutin in various skin layers were investigated using the Keshary-Chien diffusion cells at $37^{\circ}C$. The average particle size of the deformable liposomes ranged from 217.4 to 117.4 nm, depending on the composition. The entrapment efficiency was dependent on surfactant concentration and loading dose of arbutin. The permeation rate of 5% arbutin in deformable liposomes was $8.91({\pm}1.33){\mu}g/cm^2/h$, and was not significantly different from 5% arbutin aqueous solution $[9.82({\m}0.86){\mu}g/cm^2/h]$. The deposition of arbutin was $43.34({\pm}12.13)$ and $16.99({\pm}7.83){\mu}g/cm^2$ in stratum corneum layer and epidermis/dermis layer, respectively, after 12 h of permeation study. These results are consistent with several earlier studies for the localization effect of liposomal formulations in stratum corneum, and demonstrated the feasibility of the deformable liposomes as a promising carrier for the skin deposition of hydrophilic skin-whitening compounds.

  • Purpose: The study was done to explore variations in physiologic parameters of the stratum corneum (SC) of the dorsal hand and cord area during early days of life in high-risk newborns. Methods: In this longitudinal study, a total of 77 high-risk newborns were assessed for pH, temperature and hydration of the SC of the dorsal hand and the cord area on the $1^{st}$, $2^{nd}$, $3^{rd}$, $5^{th}$ and $7^{th}$ days of life. Results: Results showed that an acid mantle (AM) was formed in 57.1% for dorsal hand and 35.1% for cord area, implying significant delay regardless of decrease in pH (F=103.60, p<.001), and hydration (F=4.00, p=.003) across days in both areas. Peripheral hypothermia with low hydration level was also observed in both areas. There was a positive relation between hydration and temperature (.14

표피항상성과 건조피부의 관리 (Epidermal Homeostasis and Dry Skin Management)

  • 피부장벽을 포함한 표피층은 인체의 조직 가운데에서도 가장 역동적인 기관이다. 다시 말해서 끊임없이 새로운 표피세포의 형성, 분화 및 탈각과정이 반복되면서 표피항상성(epidermal homeostasis)을 유지한다. 표피항상성은 피부기능 가운데 가장 주요한 기능인 permeability barrier homeostasis의 확립으로 연결된다. Permeability barrier homeostasis는 각질층에서 이루어지며 이를 형성하고 유지하기 위해 매우 정교하게 조절되어야 한다. 표피항상성을 조절하는 핵심 조절인자로서 nuclear hormone receptor(NHR)가 중심에 있음이 최근 다양한 연구를 통해 입증되었다. 이들은 각질세포 특이적인 단백질, 즉, involucrin, loricrin 및 trans-glutaminase 1(TG 1) 등의 발현을 유전자 수준에서 조절할 뿐 아니라 표피 지질성분의 생합성을 증가시키는 등 피부장벽을 구성하는 brick 및 mortar의 생성과 유지에 핵심적 역할을 하는 것으로 알려졌다. NHR 가운데 peroxisome proliferator activator receptor(PPAR)와 liver X receptor(LXR)의 activator/ligands가 리놀레인산 등 지방산, leukotriene, prostanoid 및 oxygenated sterol 등이 지질대사과정에서 형성된 지질 종류인 까닭에 liposensor로도 알려지고 있다. 따라서 liposensor들을 비롯한 PPAR과 LXR activator/ligands들은 피부장벽기능이 저해된 아토피성 피부를 포함하여 건조피부를 관리하는 epidermotherapy의 수단으로서 잠재적 가능성이 있다고 생각된다.

양격산화탕(凉膈散火湯)이 Atopy 피부염(皮膚炎)에 미치는 영향(影響) (Yangkyuksanhwa-tang effected to Atopic Dermatitis)

  • 1. Objectives Yangkyuksanhwa-tang is used mush in pruritus and dermatopathy of Soyangin. It is suggested this prescription is effective on atopy dermatitis. 2. Methods For observation of Yangkyuksanhwa-tang effected to atopic dermatitis, extract of Yangkyuksanhwa-tang has been dispensed to the stratum corneum of epithelium in dermatome of murine after making damage to its defense mechanism against fat and causing atopic dermatitis artificially. After that, the change in outer dermatome and minute mechanism of epidermis, the change of eosinophil, the change in distribution of soybean agglutinin, the change in distribution of fat and ceramide in stratum corneum, the change in inflammation in dermatome, the change of cell accrementition and apoptosis, and the effect on anaphylaxis and Staphylococcus aureus was observed. 3. Results After administration of Yangkyuksanhwa-tang, severe skin damage such as eczema and psoriasis, that was observed in the case of atopy dermatitis, was decreased and the increase of eosinophil in serum was suppressed. Lipid lamella was recovered, so epidermal demage was relieved. The distribution of HSP70 in the outer skin was decreased. Yangkyuksanhwa-tang suppressed activation of $NF-_{\kappa}B$ p50, induced CD11/18b not to be generated, and suppressed inflammatory response of skin. Anaphylaxis and groth of Staphylococcus aureus was suppressed. 4. Conclusions Yangkyuksanhwa-tang decreased skin damage of atopy dermatitis. It has antibiosis about Staphylococcus aureus, it can be medicinal substances on atopy dermatitis. In addition, it is possible that it can be medicinal substances on regional skin allergy.
