Mortierella alpina를 이용한 아라키돈산의 생산에서 유기질소원의 선정과 배양 조건의 최적화 (Selection of organic Nitrogen Source and Optimization of Culture Conditions for the Production of Arachidonic Acid from Mortierella alpina)

  • 곰팡이인 Mortierella alpina DSA-12를 이용한 arachidonic acid의 생산을 위한 유기질소원의 선정과 배양 조건의 결정에 연구를 수행하였다. Corn steep powder (CSP)를 원료의 가격과 arachidonic acid의 생산을 기준으로 유기질소원으로 선정하였다. 탄소원으로 glucose와 질소원으로 CSP를 사용한 경우에 최적의 C/N ratio는 15-17 범위이다. Arachidonic acid의 생산을 위한 최적의 배양조건은 $25^{\circ}C$에서 500 rpm의 교반과 0.5 vvm의 통기이며, 이 때 50 g/L의 glucose와 18 g/L의 CSP로부터 21.8 g/L의 균사체량에 10.2 g/L의 총 지질을 얻을 수 있었으며, arachidonic acid의 농도는 3.70 g/L이었다. 500 L의 발효조에서 0.5 vvm과 200 rpm의 교반으로 실험을 수행한 결과 19.8 g/L의 균사체량과 9.1 g/L의 총 지질 및 3.67 g/L의 arachidonic acid를 얻었다. 이러한 결과는 bench-scale의 발효조에서도 질소원으로 CSP를 이용하여 arachidonic acid의 생산이 가능함을 보여주었다.

  • 강력(强力)한 inulin 분해력(分解力)을 가진 Streptomyces chibaensis균주(菌株)를 이용하여, 효소생산(酵素生産)에 효과적인 배양조건 및 배지조성을 검토(檢討)하여 다음과 같은 결과를 얻었다. 1. 효소생산배지(酵素生産培地)의 최적 초기 pH는 7.5였으며, 이 조건하에서 $30^{\circ}C$에서 84시간 배양했을 때 가장 높은 효소역가를 나타냈다. 2. Inulase를 다량생산(多量生産)하기 위한 배지의 탄소원(炭素源)으로서는 다른 탄소원(炭素源)에 비하여 inulin이 가장 효과적이었다. 3. 무기질소원(無機質素源)으로는 $(NH_4)_2HPO_4$가 inulase생산(生産)에 가장 효과적(效果的)이었고, 유기질소원(有機窒素源)으로는 corn steep liquor가 효과적(效果的)이었다. 4. 무기염(無機鹽)으로 KCI 0.01%, $MgSO_4$ 0.05%, 그리고 $FeSO_4$ 0.0001% 첨가하였을 때 가장 효과가 컸다. 5. 탄소원(炭素源)(inulin) 1%와 질소원(窒素源)(corn steep liquor) 2%의 비율(比率)로 혼합(混合)하여 만든 배지(培地)에서 가장 효소생산(酵素生産)이 높았다.

Influence of Replacing Corn Grain by Enzose (Corn Dextrose) on Nutrient Utilization, Thyroid Hormones, Plasma Metabolites, and Weight Gain in Growing Lambs

  • The study was conducted to evaluate enzose (corn dextrose), a corn milling byproduct, as substitute for corn grain as energy in growing lambs. Five iso-caloric and iso-nitrogenous diets were formulated. The control diet (E0) had no enzose whereas enzose replaced 20, 40, 60 and 80% corn grain in E20, E40, E60 and E80 diets on the basis of energy supply, respectively. Fifty growing lambs were divided into 5 groups, 10 animals in each, in a randomized complete block design. Nutrients (dry matter, crude protein, neutral detergent fiber and acid detergent fibre) intake and digestibilities increased with gradual replacement of corn grain by enzose. Lambs fed E80 diet also retained higher nitrogen (N) than those fed E0 diet. Plasma glucose, $T_3$ and $T_4$ increased while urea N decreased in lambs receiving higher enzose content. Maximum weight gain was recorded in lambs fed diets containing maximum concentration of E as a replacement for corn grains. A better feed conversion ratio was recorded in lambs fed E80 compared with those fed E0 diet. The study suggests that enzose can be used as an economical feed ingredient to replace corn grain upto 80%, without any adverse effects on growth performance of growing lambs.

Influence of Transgenic Corn on the In vitro Rumen Microbial Fermentation

  • In this study, the comparative effects of transgenic corn (Mon 810 and Event 176) and isogenic corn (DK729) were investigated for their influence on in vitro rumen fermentation. This study consisted of three treatments with 0.25 g rice straw, 0.25 g of corn (Mon810/Event176/DK 729) mixed with 30 ml rumen fluid-basal medium in a serum bottle. They were prepared in oxygen free conditions and incubated at $39^{\circ}C$ in a shaking incubator. The influence of transgenic corn on the number of bacterial population, F. succinogenes (cellulolytic) and S. bovis (amylolytic), was quantified using RT-PCR. Fermentative parameters were measured at 0, 2, 4, 8, 12 and 24 h and substrate digestibility was measured at 12 and 24 h. No significant differences were observed in digestibility of dry matter, NDF, ADF at 12 and 24 h for both transgenic and isogenic form of corns (p>0.05) as well as in fermentative parameters. Fluid pH remained unaffected by hybrid trait and decreased with VFA accumulation as incubation time progressed. No influence of corn trait itself was seen on concentration of total VFA, acetic, propionic, butyric and valeric acids. There were no significant differences (p<0.05) in total gas production, composition of gas (methane and hydrogen) at all times of sampling, as well as in NH3-N production. Bacterial quantification using RT-PCR showed that the population number was not affected by transgenic corn. From this study it is concluded that transgenic corn (Mon810 and Event 176) had no adverse effects on rumen fermentation and digestibility compared to isogenic corn. However, regular monitoring of these transgenic feeds is needed by present day researchers to enable consumers with the option to select their preferred food source for animal or human consumption.

Comparison of in vitro ruminal fermentation incubated with different levels of Korean corn grains with total mixed ration as a basal

  • The present study was conducted to investigate the effect of different levels of Korean corn grain on in vitro ruminal fermentation with total mixed ration (TMR) as a basal feed. Three ruminal cannulated Holstein steers (Body Weight $479{\pm}33.0kg$) were used as rumen fluid donors. Treatments for in vitro fermentation were TMR only (control, 3.0 g), TMR substituted partially with high level (HC, TMR 1.5 and corn 1.5 g), and with low level of Korean corn grain (LC, TMR 2.25 and corn 0.75 g), respectively. To measure in vitro ruminal pH, gas production, ammonia N and volatile fatty acids (VFA), the in vitro fermentation incubation was triplicated at $39^{\circ}C$, 120 rpm for 0, 1, 3, 6, 12, 24 and 48 h, respectively. Mean ruminal pH was significantly lower (p < 0.05) for HC than control. Changes in rumen pH was rather similar between the groups till 6 h after incubation, but the lowest pH for HC (pH 5.10) appeared at 48 h compared with control and LC. Total gas production was tended (p < 0.09) to be higher and ammonia N was significantly lower (p < 0.05) for HC than control and LC. Total VFA was higher (p < 0.05) for HC and LC than control but no differences appeared between HC and LC. Overall, the present data indicate that feeding different levels of Korean domestic corn grain may lead to high and sustainable starch degradation in the rumen.

Optimization of Solid-State Fermentation for Improved Conidia Production of Beauveria bassiana as a Mycoinsecticide

  • The production of conidia of entomopathogenic Beauveria bassiana by solid-state fermentation was studied for the development of a biocontrol agent against aphid Myzus persicae. The optimal conditions for conidia production on polished white rice were 40% moisture content, $25^{\circ}C$ culture temperature, 2-day-old seeding culture grown in 3% corn meal, 2% rice bran, 2% corn steep powder medium, initial conidia concentration of $10^7$ conidia/g in the wet rice, 10% inoculum size, and use of a polyethylene bag as a container. The polyethylene bag containing inoculated rice was hand-shaken every 12 hr during fermentation. Using optimal conditions, the maximum conidia production obtained was 4.05 g conidia/100 g dry rice after 14 days of cultivation, a rate 2.83 times higher than conidia yield of pre-optimization.

Additivity of values for phosphorus digestibility in corn, soybean meal, and canola meal in diets fed to growing pigs

  • Objective: This study was conducted to determine the apparent and standardized total tract digestibility (ATTD and STTD) of phosphorus (P) in corn, soybean meal (SBM), and canola meal (CM), and additivity of values for ATTD and STTD of P in corn, SBM, and CM in diets fed to growing pigs. Methods: Thirty-six growing barrows (initial body weight of $21.6{\pm}1.7kg$) were placed in metabolism crates and allotted to a completely randomized design with 6 diets and 6 pigs per diet. Six diets were formulated using corn, SBM or CM as the sole source of P, or corn and SBM, or corn and CM, or corn, SBM, and CM as the P source in each diet, respectively. Fecal samples were collected for 5 d following a 7 d adaptation period to the diets. Results: Values for ATTD and STTD of P in corn, SBM, and CM in growing pigs were 33.12% and 37.76%, 50.19% and 56.62%, 34.93% and 39.45%, respectively. The ATTD and STTD of P in SBM were greater (p<0.05) than those in corn and CM. However, there were no differences in the ATTD or STTD of P between corn and CM. The determined STTD of P in the mixture of corn and SBM, corn and CM, and corn, SBM, and CM is not different from the calculated STTD values. Conclusion: Values for STTD of P in corn, SBM, and CM are additive in their mixture fed to growing pigs.

가뭄에 따른 논 재배 사일리지용 옥수수의 생육특성, 생산성 및 품질 비교 (Effect of Drought Conditions on Growth, Forage Production and Quality of Silage Corn at Paddy Field)

  • 본 시험은 논에서 가뭄조건에 따른 여름 사료작물인 옥수수의 생육특성과 수량성을 알아보기 위해서 2008년부터 2009년까지 2년간에 걸쳐 전남 영광과 천안의 농가 포장에서 수행하였다. 국내육성 5품종과 도입품종 5품종 등 10품종을 시험한 결과 강다옥 품종이 생육특성과 수량성에서 우수한 결과를 보였고 가뭄에 대해 비교적 수량 감소가 적은 것으로 나타났다. 표준조건 재배에서는 강다옥의 건물수량은 21.8 톤/ha이었고 가뭄조건에서 국산품종인 강다옥의 건물수량이 5,548 kg/ha 으로 공시품종 중에서 최고의 수량을 보였다. 또한 사료가치는 표준조건보다 가뭄조건에서 떨어지는 경향을 보였는데 ADF 함량은 17.8% 증가하였으나 조단백질 함량은 27.4%로 최고의 감소를 보였다. 따라서 본 연구결과를 토대로 볼 때 사일리지용 옥수수를 논에 재배하기 위해서는 가뭄시에 충분히 관수를 하고 가뭄에 강한 품종을 재배하는 것이 중요하다는 사실이다.

Estimating vegetation index for outdoor free-range pig production using YOLO

  • The objective of this study was to quantitatively estimate the level of grazing area damage in outdoor free-range pig production using a Unmanned Aerial Vehicles (UAV) with an RGB image sensor. Ten corn field images were captured by a UAV over approximately two weeks, during which gestating sows were allowed to graze freely on the corn field measuring 100 × 50 m2. The images were corrected to a bird's-eye view, and then divided into 32 segments and sequentially inputted into the YOLOv4 detector to detect the corn images according to their condition. The 43 raw training images selected randomly out of 320 segmented images were flipped to create 86 images, and then these images were further augmented by rotating them in 5-degree increments to create a total of 6,192 images. The increased 6,192 images are further augmented by applying three random color transformations to each image, resulting in 24,768 datasets. The occupancy rate of corn in the field was estimated efficiently using You Only Look Once (YOLO). As of the first day of observation (day 2), it was evident that almost all the corn had disappeared by the ninth day. When grazing 20 sows in a 50 × 100 m2 cornfield (250 m2/sow), it appears that the animals should be rotated to other grazing areas to protect the cover crop after at least five days. In agricultural technology, most of the research using machine and deep learning is related to the detection of fruits and pests, and research on other application fields is needed. In addition, large-scale image data collected by experts in the field are required as training data to apply deep learning. If the data required for deep learning is insufficient, a large number of data augmentation is required.

옥수수 종자의 발육 중 ethylene 발생과 내부형태 변화 (Ethylene Production and Internal Structure of Developing Maize Seeds)

  • 옥수수의 종자발육에 ethylene이 미치는 영향을 알기 위하여 마치종, 단옥수수, 초당옥수의 배와 배유의 발육과 ethylene 발생을 조사하였다. 그리고 초당옥수수에 ethylene 발생과 작용을 조절하는 aminoethoxyvinyl glycine(AVG), silver thiosulfate(STS), ethephon(2-chloroethylphosphonic acid, CEPA)를 출사 후 9 및 21일에 전 식물체와 포엽에 처리하여 ethylene 발생양상과 배와 배유의 발육을 조사하여 채종 시 종자발달을 촉진할 수 있는 방법을 찾고자 하였으며, 그 결과를 요약하면 다음과 같다. 1. 마치종은 배와 배유 모두 출사 후 56일까지 건물중이 증가하였다. 그러나 단옥수수와 초당옥수수의 배는 출사 후 각각 56일 및 49일까지, 배유는 출사 후 35일 및 28일까지 증가하였다. 2. 종자의 ethylene 발생량은 초당옥수수가 단옥수수나 마치종보다 많았다. 3. 배유조직의 변화는 출사 후 21일부터 배의 끝 부분에서 동공이 생기기 시작하여 마치종은 동공이 유지되나 단옥수수는 출수 후 42일에는 동공이 붕괴되었고, 초당옥수수는 배 위쪽 및 옆쪽까지 동공이 생겼다가 수축되었다. 4. 초당옥수수 품종 Xtrasweet 82에서 AVG와 STS는 ethylene 발생량을 감소시키고 100립중을 다소 증가시켰으나, CEPA는 ethylene 발생량을 증가시키고 100립중을 감소 시키는 경향이었다. 5. CEPA 처리는 배유에 동공이 발생하는 시기가 다소 빨랐으나 AVG와 STS 처리는 큰 효과가 없었다.
