Bacillus sp. 의 Cyclodextrin Glucanotransferase 생산 및 이용에 관한 연구 (The Production of Cyclodextrin Glucanotransferase by Bacillus sp. and Its Utilization)

  • 본 연구에서는 CGTase 생산성이 높은 균주인 Bacillus sp.를 자연계에서 분리하였고 이 균주의 특성, 분비효소의 특성, 효의 생산조건 및 효소이용 가능성에 대한 기초연구를 실시하였다. 분리균주는 운동성이 있는 내성포자형성 간균이었고 분비효소의 최적작용조건은 pH 6.0, 45$^{\circ}C$이며 pH 6-10 범위에서 안정하였다. 효소생산조건은 탄소원으로 corn starch 1%, 질소원으로서 corn steep liquor 5%, urea 0.1%, ammonium sulfate 0.25%를 동시에 첨가하는 것이 가장 양호하였으며 본 생산균을 30$\ell$ jar fermentor로 3$0^{\circ}C$, 200rpm, 0.6vvm에서 60시간 정도 배양하였을 때 최대의 효소 생산력을 나타내었다. 또한 본 효소를 이용하여 stevioside를 acceptor로 하고 전분가수분해물을 donor로 하여 당전이반응을 실시한 결과 상당한 전이효과가 나타났으므로 기타 전이제품 및 cyclodextrin 생산에 응용이 가능할 것으로 기대된다.

Effects of Alfalfa and Brown Mid-rib Corn Silage and Level of Forage Neutral Detergent Fiber on Animal Performance of Lactating Cows in Michigan

  • Alfalfa silage and corn silage are the major dairy feeds in most dairy operations in Michigan, USA. In recent years, the need to improve digestible fiber and dry matter intake of forages to meet the nutrient requirements of high yielding dairy cows and the willingness to plant corn specifically for silage has led plant breeders to focus on the brown mid-rib (BMR) trait. The objective of this study was to investigate the effect of different ratio of alfalfa to BMR corn silage and ration level of forage neutral detergent fiber (NDF) on animal performance of lactating cows in the Upper Peninsula of Michigan. This study was conducted at the Upper Peninsula Experiment Station of Michigan State University in Chatham, Michigan, USA. Two different ratios of forage type (high alfalfa silage/low BMR corn silage, AS, and high BMR corn silage/low alfalfa silage, BMRCS) and two different dietary NDF contents (27% NDF, 27 = low forage/high grain feeding, and 33% NDF, 33 = high forage/low grain feeding) were used. The experimental design was a $4{\times}4$ Latin Square with 20 milking cows (12 multiparous and 8 primiparous). This trial had four 21-day periods with 14 d adaptation and 7 d data collection. Milk yield and body condition score (BCS) on the AS-27, BMRCS-27 and BMRCS-33 treatments were significantly (p<0.05) higher than on the AS-33 treatment. Dry matter intake of the AS-27 and BMRCS-27 treatments was significantly (p<0.05) higher than for the AS-33 and BMRCS-33 treatments. Milk urea nitrogen (MUN) on the AS-33 treatment was significantly (p<0.05) higher than on the other diet treatments. A key finding of this study was that the BMRCS-33 (higher amounts of brown mid-rib corn silage than alfalfa silage, high forage and low grain feeding diet at 33% NDF) led to the equal highest milk production whilst having the equal lowest dry matter intake. This study demonstrated that the diet with higher ratio of highly digestible NDF forage such as brown mid-rib corn silage to alfalfa silage could lower grain feeding in the ration.

Evaluation of forage production, feed value, and ensilability of proso millet (Panicum miliaceum L.)

  • Whole-plant corn (Zea may L.) and sorghum-sudangrass hybrid [Sorghum bicolor (L.) Moench] are major summer crops that can be fed as direct-cut or silage. Proso millet is a short-season growing crop with distinct agronomic characteristics that can be productive in marginal lands. However, information is limited about the potential production, feed value, and ensilability of proso millet forage. We evaluated proso millet as a silage crop in comparison with conventional silage crops. Proso millet was sown on June 8 and harvested on September 5 at soft-dough stage. Corn and sorghum-sudangrass hybrid were planted on May 10 and harvested on September 10 at the half milk-line and soft-dough stages, respectively. The fermentation was evaluated at 1, 2, 3, 5, 10, 15, 20, 30, and 45 days after ensiling. Although forage yield of proso millet was lower than corn and sorghum-sudangrass hybrid, its relative feed value was greater than sorghum-sudangrass hybrid. Concentrations of dry matter (DM), crude protein, and water-soluble carbohydrate decreased commonly in the ensiling forage crops. The DM loss was greater in proso millet than those in corn and sorghum-sudangrass hybrid. The in vitro dry matter digestibility declined in the forage crops as fermentation progressed. In the early stages of fermentation, pH dropped rapidly, which was stabilized in the later stages. Compared to corn and sorghum-sudangrass hybrid, the concentration of ammonia-nitrogen was greater in proso millet. The count of lactic acid bacteria reached the maximum level on day 10, with the values of 6.96, 7.77, and 6.95 Log10 CFU/g fresh weight for proso millet, corn, and sorghum-sudangrass hybrid, respectively. As ensiling progressed, the concentrations of lactic acid and acetic acid of the three crops increased and lactic acid proportion became higher in the order of sorghum-sudangrass hybrid, corn, and proso millet. Overall, the shorter, fast-growing proso millet comparing with corn and sorghum-sudangrass hybrid makes this forage crop an alternative option, particularly in areas where agricultural inputs are limited. However, additional research is needed to evaluate the efficacy of viable strategies such as chemical additives or microbial inoculants to minimize ammonia-nitrogen formation and DM loss during ensiling.

옥수수의 생장에 미치는 Azospirillum amagonense Y1의 영향 (Effects of Azospirillum amagonense Y1 on the Growth of Corn)

  • In order to clarify the effect of the associative nitrogen fixing bacteria on the growth of corn(Zea mays L. Suwon19), we inoculated corn seedlings with Azospirillum amazonense Y1, a micro-aerobic and acid-tolerant nitrogen fixer, and analyzed the growth and dry matter production and changes of nitrogen and phosphorus quantity of the plant during the growing period. The inoculation of associative N-fixing bacgteria increased the growth of height, leaf area, dry weight and total nitrogen quantity of the plant by 15.4%, 65.4%, 33.7% and 38.0%, respectively, on the day of 56th after sowing. Especially the inoculation of A. amazonense Y1 showed two to three-fold accumulation of phosphorus in each organ of plant. The associative bacteria accelerated the growth of the under ground parts more than those of the upper ground parts of corn plant, and caused decrease in T/R ratios.

Lovastatin Production in Solid-state Fermention by Aspergillus terreus and Its Application for Animal Feed Additive

  • Solid-state fermentation of lovastatin by Aspergillus terreus was investigated using commercially available 1.2 L polypropylene bottle designed for mushroom cultivation. Moist solid raw materials such as com, rice, and soy bean were tested and com was found to be most suitable for an economical production of lovastatin. 50% or higher water addition prior to the sterilization of com was effective for the maximal lovastatin production. About 0.5% (w/w) lovastatin content in dried cells and corn mass was obtained after 20 days of solid-state fermentation at 30$^{\circ}C$. For safety concerns, aflatoxin Bl and citrinin levels after fermentation were assayed but they were not detected. Lovastatin containing cells and corn residue after fermentation were autoclaved, dried, crushed, and fed to chicken for a period of 3 weeks. Approximately 20% reduction of blood cholesterol level of chicken was observed.

  • In this study, batch production of lactic acid from cheap law material such as barley, wheat, and corn, was tried to lower the total production cost of lactic acid. Although no nutrients were supplemented, lactic acid productivities were achieved up to 0.88 $g/(L{\cdot}hr)$ from barley, wheat and com enzymatic hydrolysate. By adding corn steep liquor to com and wheat hydrolysate media, realtively high lactic acid $productivities(4.14\;g/(L{\cdot}hr))$ were obtained.

인산염이 Fusarium sporotrichioides M-1-1 성장과 T-2 toxin 생성에 미치는 영향 (Effects of Polyphosphates on the Growth and T-2 Toxin Production of Fusarium sporotrichioides M-1-1)

  • The antifungal effects of polyposphates on the growth and T-2 toxin production of Fusarium sporotrichioides M-1-1 were investigated. The growth of the strain was significantly inhibited in the potatoes dextrose agar medium treated with 1.5% polyphosphates or more. When we checked T-2 toxin by the indirect competitive ELISA, the strain produced 11.25 ug/ml and 10.90 ug/ml levels of T-2 toxin rice and corn containing 50% moisture contents, respectively. However, T-2 toxin was little detected in rice medium and corn medium with 1.5% polyphosphates addition for short(14 days) and prolonged incubation time(45 days). We also observed the destruction of cell wall and outflow of cell ingredients with 1% polyphosphates treatment to the strain. Therefore, moisture and polyphosphates greatly effected on the growth and T-2 toxin production of the strain.

  • 1. Amylase producing bacteria have been isolated from natural sources and was identified out. The above strain is identical to Bacillus subtillis in every details of physiological and morphorogical characteristics by this investigations. 2. Sikworm chrysalis nad chlorella extracts are prove to be further effective for the stimulation of amylase production than any other sources. The former silkworm extract is more stronger in its activity. 3. It is found that effective results had been accepted by a less soybean meal addition per that of wheat bran in the mixture ratio of them, also less addition of corn is needed in the mixture ratio of between corn and whear bran. In the mixing ratio of wheat bran per silkworm or chlorella extracts, the addition of wheat bran is necessary for stimulation of amylase production. 4. Fro the stimulation of amylase production, the effective substances from natural sources and its order of activity in cultral media cultral media is proved as follows, silkworms ext.>chlorella ext.>alcohol>skim-milk and peptone>fermented milk>radish leaf ext.

Effects of Fat Sources on Growth Performance, Nutrient Digestibility, Serum Traits and Intestinal Morphology in Weaning Pigs

  • This experiment was conducted to investigate the effects of fat sources on growth performance, nutrient digestibility, serum traits and intestinal morphology in weaning pigs. A total of 128 weaning pigs (Landrace${\times}$Large White${\times}$Duroc, $21{\pm}2$ days of age, $5.82{\pm}0.13kg$ of average initial body weight) were allotted in a randomized complete block (RCB) design with four treatments: 1) corn oil, 2) soybean oil, 3) tallow and 4) fish oil. Each treatment had 8 replicates with 4 pigs per pen. During phase I period (d 0 to 14), pigs fed corn oil or soybean oil diet tended to show higher ADG and FCR than any other treatments although there was no significant difference. During phase II period (d 15 to 28), pigs fed corn oil diet showed better ADG and ADFI than pigs fed soybean oil, tallow or fish oil. For overall period, growth performance of weaning pigs was improved (p<0.05) when pigs were fed soybean oil or corn oil. Apparent digestibility of energy and fat was improved when pigs were fed corn oil diet (p<0.05). Supplementation of corn oil resulted in higher serum triglyceride concentration than the other treatments (p<0.05). However, there was a lower cholesterol concentration when corn oil was provided compared to tallow or fish oil. Pigs fed corn oil tended to have increased villus height compared with soybean oil, tallow or fish oil treatment (p<0.05). This experiment suggested that vegetable oils such as corn oil or soybean oil, were much better fat source for improving growth performance of weaning pigs.

Energy and Standardized Ileal Amino Acid Digestibilities of Chinese Distillers Dried Grains, Produced from Different Regions and Grains Fed to Growing Pigs

  • Two experiments were conducted to determine the digestibility of crude protein (CP), amino acids and energy in three Chinese corn distillers dried grains with solubles (DDGS), one rice DDGS, one American corn DDGS and one American high protein distillers dried grains (HP-DDG). In Exp. 1, the apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of CP and amino acids in the six samples were determined using cannulated barrows (initial BW: $43.3{\pm}1.7$ kg). In Exp. 2, the digestible energy (DE) and metabolizable energy (ME) content of these six samples were determined using crossbred barrows (initial BW: $46.0{\pm}2.5$ kg). The results of the two experiments indicated that Chinese corn DDGS is generally similar to American DDGS in chemical composition, digestibility of amino acids, DE and ME. However, Chinese DDGS had a lower Lys concentration (0.50% vs. 0.74%) and SID Lys (52.3% vs. 57.0%, p<0.01). The DE and ME values in Chinese corn DDGS were 3,427 and 3,306 kcal/kg, respectively. Rice DDGS had a similar DE and ME (3,363 and 3,228 kcal/kg) but higher Lys concentration (0.64% vs. 0.50%) to corn DDGS, while the SID of Lys was quite low (61.8%, p<0.01). HP-DDG had high value of SID of Lys, DE and ME (79.8%, 3,899 and 3,746 kcal/kg). In conclusion, except for a lower Lys concentrations and availability, the chemical composition, digestibility of amino acids, DE and ME values in Chinese corn DDGS are similar to American corn DDGS. Additionally, the rice DDGS had lower Lys content and digestible Lys values than that in corn DDGS. Thirdly, HP-DDG has higher levels of digestible amino acids and energy than DDGS.
