The Production of Glucose-1-phosphate from Sucrose by Leuconostoc sp. (Leuconostoc sp.에 의한 Sucrose로부터 Glucoes-1-phosphate의 생산)

  • For the production of glucose-1-phosphate from sucrose, bacteria having sucrose phosphorylase were isolated from Kimchi. Among them, JS-05, newly isolated strain having high activity of sucrose phosphorylase was selected and identified as Leuconostoc sp. The specific activity of sucrose phosphorylase of Leuconostoc sp. JS-05 was the highest when the strain was cultured at 25C for 20 hrs in the medium (pH 7.5) containing 10 g sucrose, 5g corn steep liquor, and 2.5g yeast extract per liter.

Comparative Studies on Streptomycin Producing Strains and Media (스트렙토마이신 생성균주들과 배지들에 대한 비교연구)

  • When various strains of Streptomyces griseus and S. galbus were examined for the ability on the production of streptomycin in tryptic soy(TS) broth, S. griseus ATCC 27001 was found to be the best. S. griseus ATCC 12475 and ATCC 23345 showed also good growth and favorable production of streptomycin. Examination of various complex media reported in fermentation literatures for the industrial production of streptomycin indicated that glucose-soybean media-sodium chloride (GSS) broth and K (Chucken) broth gave higher yields of streptomycin than others studied by us. Examination of the ingredients of media producing streptomycin in high yield indicated that some components in soybean activated the production of streptomycin. Addition of meat extract enhanced the yield of streptomycin but it could be substituted with distillers solubles without much effect on the yield. Addition of corn steep liquor decreased the production of streptomycin.

Production of Erythromycin Using a Carrier-Spported Mycelial Growth in a Fluidized-Bed Bioreactor (균사 증식 담체를 이용한 유동층 생물반응기에서 Erythromycin의 생산)

  • A carriersupported mycelial growth of Sreptomyces erythreus was applied to erythromycin fermentation sistem using celite as a support material. Hyphal growth through the pore matrices of the materials showed anchorages and provided a stable biofilm growth. When the phospate concentration was limited to 0.8g corn steep liquor/L(corresponding to 40mg KH2PO4/L), the specific production rate of erythromycin was increased from 557$\mu$g/g-cell.hr under unlimited condition to 2, 898 $\mu$g/g-cell.hr. A fluidized-bed bioreactor was operated for erythromycin production by a repeated fed-batch mode. The control of free mycelial concentration and the extension of production phase were considered important to maintain the reactor productivity at a desired level. The erythromycin production under phosphate-limited condition could be maintained for at least 600hrs.

  • To overproduce the cyclodextrin glucanotransferase(CGTase) of Bacillus stearothermophilus NO2 in B. subtilis, the pJH-CGTl plasmid (8.14 kb) was constructed, in which the ORF of CGTase gene could be transcribed by strong constitutive promoter, P$\_$JH/. To overproduce CGTase from a recombinant B. subtilis, the effect of media on the cell growth and expression level of CGTase were investigated with various media (LB, 2${\times}$LB, 5% molasses+2% CSL, CS, LBG) in the flask culture. Among them, [5% molasses+2% CSL] medium revealed the maximum expression level of CGTase with 1.8 unit/$m\ell$ at 9 hr culture. In the batch culture on [10% molasses+5% corn steep liquor] medium the expression level of CGTase, the secretion efficiency, and plasmid stability were about 4.2 unit/$m\ell$, 90% and 90%, respectively, at 30 hr culture. The cell growth and expression level in the fermenter culture with the industrial molasses medium were increased by 2-folds over the flask culture.

Production and Characterization of Cholesterol Oxidase from Streptomyces sp. No.4 (방선균으로부터 Cholesterol Oxidase의 생산 및 특성)

  • An actinomycetes strain No.4 which produce the cholesterol oxidase(EC 1.1.3.6), was isolated from soil and identified as Streptomyces sp. based on taxonomic studies. The conditions of cholesterol oxidase production and enzymatic properties were investigated. The optimum composition of medium for production of the enzyme was 1% soluble starch, 2% corn steep liquor, 0.1% $KH_2PO_4$, 0.1% $NaNO_3$ and 0.05% $MgSO_4$ (pH 7.0). The optimum pH and temperature of the cholesterol oxidase were pH 6.0~7.5 and $37^{\circ}C$, respectively. The enzyme was stable in the range of pH 6.0~9.0. The isoelectric point determined by multichambered electrofocusing unit was in the range of pH 6.0~6.5.

  • Approximately 50% of people in the world experience abdominal flatulence after the intake of foods containing galactosides such as lactose or soybean oligosaccharides. The galactoside hydrolyzing enzymes of ${\alpha}$- and ${\beta}$-galactosidases have been shown to reduce the levels of galactosides in both the food matrix and the human gastrointestinal tract. This study aimed to optimize the production of ${\alpha}$- and ${\beta}$-galactosidases of Bifidobacterium longum subsp. longum RD47 with a basal medium containing whey and corn steep liquor. The activities of both enzymes were determined after culturing at $37^{\circ}C$ at pH 6.0 for 30 h. The optimal production of ${\alpha}$- and ${\beta}$-galactosidases was obtained with soybean oligosaccharides as a carbon source and proteose peptone no. 3 as a nitrogen source. The optimum pH for both ${\alpha}$- and ${\beta}$-galactosidases was 6.0. The optimum temperatures were $35^{\circ}C$ for ${\alpha}$-galactosidase and $37^{\circ}C$ for ${\beta}$-galactosidase. They showed temperature stability up to $37^{\circ}C$. At a 1 mM concentration of metal ions, $CuSO_4$ inhibited the activities of ${\alpha}$- and ${\beta}$-galactosidases by 35% and 50%, respectively. On the basis of the results obtained in this study, B. longum RD47 may be used for the production of ${\alpha}$- and ${\beta}$-galactosidases, which may reduce the levels of flatulence factors.

Production of an Antihyperlipemial HMG-CoA Reductase Inhibitor from Bacillus cereus D-3 (Bacillus cereus D-3로부터 항고지혈증 HMG-CoA Reductase 저해제의 생산)

  • For the purpose of production of a novel antihyperlipemial HMG-CoA reductase inhibitor from bacteria, a bacterium which showed the highest HMG-CoA reductase inhibitory activity was isolated from traditional Doenjang. This strain was identified as Bacillus cereus (D-3) based on its microbiological characteristics and 165 rRNA sequence analysis. The maximal HMG-CoA reductase inhibitor production from Bacillus cereus D-3 was obtained by cultivation in a Glucose-CSL broth containing 2% glucose, 0.6% corn steep liquor, $0.04%\;K_{2}HPO_4$ and $0.05%\;KH_{2}PO_4$ at $30^{\circ}C$ for 36 h. The final HMG-CoA reductase inhibitory activity under the above conditions was 39.4%.

Isolation and Characterization of Cyclodextrin Glycosyl Transferase Producing Alkalophilic Bacillus sp. (Cyclodextrin glycosyltransferase를 생산하는 호알칼리성 Bacillus속 미생물)

  • A strain of alkalophilic Bacillus sp. YC-335 has been isolated from soil. The strain was capable of producing large amount of cyclodextrin glycosyl transferase (CGTase) in the culture broth. The preferable medium composition has been determined to be as follows : 1.5% soluble starch, 5% corn steep liquor, 0.1% $K_2$HPO$_4$, 0.02%mgSO$_4$.7$H_2O$, 1% CaCO$_3$and 1% Na$_2$CO$_3$(pH 10.3). The highest enzyme production was observed after 48 hours of cultivation at 31$^{\circ}C$. The optimum pH and temperature for the activity of crude enzyme were 6.0 and 5$0^{\circ}C$, respectively. The enzyme was stable between pH 5 and 9, and upto 5$0^{\circ}C$. The enzyme converted starch into $\alpha$-, $\beta$- and ${\gamma}$-CD in the relative amounts of 1:10:1.5, respectively.
