Effects of opioid and non-opioid antagonists, pH and enzymes on Corchorus olitorius antinociception in mice

  • The present study was carried out to determine the involvement of opioid and non-opioid receptor and the effect of pH and enzymes on the recently reported antinociceptive activity of aqueous extract of Corchorus olitorius (AECO) leaves using the abdominal constriction test. The extract was prepared by soaking the dried powdered leaves of Corchorus (C.) olitorius in distilled water overnight, and the supernatant obtained was considered as a stock solution with 100% concentration/ strength. The extract, administered subcutaneously in the concentrations/ strength of 10, 50 and 100%, was found to show a significant concentration-independent antinociception. The 50% concentration AECO were further used to study on the above mentioned parameters. The extract exhibited: significant (P < 0.05) decreased in activity when pre-treated (s.c.) against 10 mg/kg naloxonazine, bicuculine (10 mg/kg), phenoxybenzamine (10 mg/kg), 10 mg/kg pindolol, and 5 mg/kg mecamylamme, but not 10 mg/kg naltrindole, 10 mg/kg atropine, respectively; significant (P < 0.05) decreased in activity after pre-treatment against 10% a-amylase, but not 1 % protease or 10% lipase and; significant (P < 0.05) decreased in activity after exposure to alkaline condition (pH between 9 and 13) while maintaining the activity at acidic condition, respectively. The C. olitorius leaves antinociception, which involved, at least in part, activation of $\mu-opioid,\;\alpha-and\;\beta-adrenergic$, and nicotinic receptors, was found to decrease under alkaline condition and in the presence of $\alpha-amylase$.

Effects of Corchorus olitorius Powder on the Quality Characteristics of Emulsion-type Sausage (몰로키아 분말 첨가가 유화형 소시지의 품질특성에 미치는 영향)

  • In this study, the effects of Corchorus olitorius powder on the quality characteristics of emulsion type sausages during storage at $4^{\circ}C$ for 28 days was evaluated. Sausages were produced containing 0.5%, 1.0% and 1.5% C. olitorius powder. The pH values of sausage containing C. olitorius powder were higher than the control during 28 days of storage. The L and a values of sausage containing C. olitorius powder significantly decreased with increasing C. olitorius powder content; however, the b values significantly increased. The volatile basic nitrogen (VBN) and nitrite content of sausage containing C. olitorius powder were lower than that of the control. Especially, the VBN and nitrite content of sausages containing 1.5% C. olitorius powder were significantly lower compared to the control. In addition, sausages with $0.5{\sim}1.0%$ C. olitorius powder had higher hardness, cohesiveness, gumminess and brittleness than the control. In regards to the sensory evaluation, sausages containing 0.5% C. olitorius powder had the highest overall acceptability. Therefore, these results suggest that it may be possible to manufacture sausages containing 0.5% C. olitorius powder to help improve the overall quality of sausage.

  • A simple cultivation method using Jew's mallow (Corchorus olitorius L.) was examined to evaluate the biological quality of 72 upland soils collected from greenhouses in Korea. Each soil was mixed with 4-fold volume of sterilized basal soil having no indigenous chemical and physical problems to lessen the chemical and physical variations among original sample soils. This study assumes that the biological characteristics of each mixture 2 weeks after treatment reflect those of individual sample soil prior to treatment. Jew's mallow (Corchorus olitorius L.) was cultivated for 4 weeks in a pot filled with 250g of each experimental soil and the biological quality of each soil was estimated by the aboveground plant growth. The results indicated that microbial floral quality was explicitly deteriorated in 18 soils having EC above or equal to $5dS\;m^{-1}$. However, in soils having EC below $5dS\;m^{-1}$, some showed a sign of deterioration, while others did not. As this simple method is a kind of bioassay, it dose not require the measurements of microbial numbers and activities.

Effects of methanol extracts of Cuscuta reflexa Roxb. stem and Corchorus olitorius Linn. seed on male reproductive system of mice

  • The antifertility activity of methanol extract of Cuscuta reflexa Roxb. stem (MECR) and Corchorus olitorius Linn. seed (MECO) were studied on male Swiss albino mice. The extracts were found to decrease sperm count, percentage of motile sperm and testosterone level in treated mice when compared with vehicle control after 17 days of treatment. The weight of gonads, epididymis were decreased whereas no significant changes of the body weight of mice were observed after methanol extract treatments. The fertility test showed 100% negative result in MECR and MECO treated mice at medium and high dose level of treatment. MECR and MECO in low (25 mg/kg and 15 mg/kg, respectively), medium (50 mg/kg and 20 mg/kg, respectively) and high (75 mg/kg and 25 mg/kg, respectively) dose level caused a simultaneous fall in testicular ${\Delta}5$-$3{\beta}$-hydroxy steroid dehydrogenase and glucose-6-phosphate dehydrogenase activities which are involved in testicular steroidogenesis. Total cholesterol and ascorbic acid content in testis were increased significantly in gonads. The activities of lactate dehydrogenase, malic dehydrogenase and ascorbic acid oxidase were reduced whereas that of carbonic anhydrase was increased significantly in the testis of MECR and MECO treated mice. All these observations indicate that the methanol extract of C. reflexa stem and C. olitorius seed produced antifertility activity in sexually matured male mice, which may be due to inhibition of gonadal steroidogenesis. This activity may be attributed due to the presence of flavonoids and steroids, respectively.

Physicochemical Properties of Mucilage from Domestic Molokhia (Corchorus olitorius) (몰로키아(Corchorus olitorius) 잎에서 추출한 mucilage의 이화학적 특성)

  • Proximate composition, phenolic compounds, and amino acid composition of molokhia (Corchorus olitorius) and molecular weight distribution of its mucilage were determined. Concentrations of dietary fibers and protein were 37.4 and 24.4% of the total dry weight, respectively. Mucilage was obtained by extracting dry molokhia leaves with hot water, followed by the addition of 55% ethanol. Although about 50% of the mucilage was lost during the extration, it was still rich in polyphenol compounds. In mucilage, proteins were composed of glutamic acid, glycine, alanin, and aspartic acid, and molecular weight of most polysaccarides ranged from $40,000{\sim}500,000$ daltons based on gel filtration chromatography.

DNA Fingerprinting of Jute Germplasm by RAPD

  • The genotype characteristic of cultivars was investigated, along with varieties of both of the jute species, Corchorus olitorius and Corchorus capsularis, in the germplasm collection at the Bangladesh Jute Research Institute (BJRI). DNA fingerprinting was generated for 9 different varieties and 12 accessions of jute cultivars by using random amplified polymorphic DNA(RAPD). A total of 29 arbitrary oligonucleotide primers were screened. Seven primers gave polymorphism within the varieties, and 6 primers detected polymorphism within the accessions that were tested. A dendrogram was engendered from these data, and this gave a distinct clustering of the cultivated species of jute. Therefore, we generated RAPD markers, which are species-specific. These primers can distinguish between C. olitorius and C. capsularis. From the dendrogram that we generated between the various members of these two species, we found the existing genetic classification that agrees with our molecular marking data. A different dendrogram showed that jute accessions could be clustered into three groups. These data will be invaluable in the conservation and utilization of the genetic pool in the germplasm collection.

Effect of Drying on the Nutritional and Organoleptic Characteristics of African Leafy Vegetables, Jute Mallow (Corchorus olitorius L.) and Cowpea (Vigna unguiculata)

  • Purpose: The present study investigated the nutritional and organoleptic characteristics of two African leafy vegetables (ALVs)-jute mallow (Corchorus olitorius L) and cowpea (Vigna unguiculata)-at various drying temperatures. Methods: The thin-layer drying of cowpea leaves and jute mallow was studied at various temperatures ($40-100^{\circ}C$) in a convective laboratory dryer, and the nutrient profiles of the dried vegetables were determined. The nutrients considered were vitamins B2 and C, and ${\beta}$-carotene. The level of vitamin C was determined by high-performance liquid chromatography (HPLC), whereas the levels of ${\beta}$-carotene and vitamin B2 were determined by titration. Results: ${\beta}$-carotene was the most stable nutrient, whereas vitamin C was the least stable nutrient in both cowpea leaves and jute mallow. The drying parameters-temperature and time-revealed that temperature had the most profound effect on vegetable nutrient stability. Organoleptic tests were carried out on the fresh and dried vegetable; there were no significant differences in preference between the fresh and dried ALVs (95% confidence interval). Conclusions: The present study revealed that the vegetables can be preserved by drying, and the study could be used as a guide for effective drying of those vegetables.

Cholesterol Improvement Effects of Fermented Defatted Soybean Grits Added to Corchorus olitorius (몰로키아 첨가 탈지대두grit(defatted soybean grit) 발효물의 콜레스테롤 개선 효과)

  • This study was performed to investigate cholesterol improvement effects of adding fermented defatted soybean grit (FD) and FD added to 5 or 10% Corchorus olitorius (FDC). Cholesterol adsorption in the FD and FDC group was more than 70%. Apolipoprotein AI and CIII improved in HepG2 cells, and a greater improvement effect was shown in FDC than that in FD. We also investigated the effect of FDC on body lipid metabolism and a high-fat diet for 4 weeks. Rats were divided into control (Con), high-fat (HF), HF treated with 20% FD (HF-FD), and HF treated with 20% FDC (HF-FDC) groups. Plasma total cholesterol and low density lipoprotein-cholesterol concentrations, hepatic total cholesterol, and triglyceride contents were significantly lower in the HF-FDC group than those in the HF group. Additionally, fecal total cholesterol and triglyceride contents increased in rats treated with FDC. Hepatic 3-hydroxy-3-methylglutaryl-coenzyme A reductase activities were significantly lower in the HF-FDC group than those in the HF group.

  • 본 연구에서는 국내산 몰로키아 잎에서 추출한 mucilage의 이화학적 특성, 동물실험을 통한 콜레스테롤 대사에 미치는 영향가 임상실험에서의 비만 및 체지방대사에 미치는 영향을 검토하였다. 몰로키아 잎은 식이섬유의 함량이 37.4%, 단백질의 함량은 24.4%를 차지하고 있으며, 칼슘의 함량이 2,000mg% 이상으로 대단히 높았다. 몰로키아 앞에서 추출된 mucilage의 경우 단백질의 총당에 대한 비율이 $50^{\circ}C$에서 57.6%를 차지하였으며, 구성다당류의 분자량은 약 500,000 dalton의 크기를 나타내었고 40,000 dalton 이하의 것도 일부 존재하는 것으로 나타났다. 동물실험에서는 혈장의 총 콜레스테롤 함량은 대조구인 셀룰로오스 첨가군과 유의적인 차이가 나타나지 않았지만, 간에서의 지방 농도는 몰로키아 분말과 mucilage 첨가군이 유의적으로 감소되었고 변으로의 콜레스테롤 배설량도 몰로키아 분말 첨가군과 mucilage 첨가군 순으로 높았다. 몰로키아의 섭취가 다이어트와 변비개선에 미치는 효과를 검토한 결과, 체중의 감소는 아주 낮은 유의성을 보였고, 혈액 내 생화학적 수치에서 초기치와 8주 후의 값에는 유의한 차이를 보이지 않았지만, 체지방율의 경우에는 현저히 줄어들었다. 또한 몰로키아의 섭취로 배변이 2배 이상 증가하였으며, 배변시의 통증도 상당히 완화시키는 것으로 나타났다.
