Inhibition of HIV-1 Replication by Extract of Coptis chinensis (황련추출물에 의한 HIV-1 복제 저해)

  • Natural products, total number of 175, were screened to test for their effect on the replication of human immunodeficiency virus type 1 (HIV-1). Five of them, such as Eriobotrya japonica, Eugenia caryphyllata, Cuscuta chinensis, Glycyrrhiza uralensis, and Coptis chinensis were shown to be effective in inhibiting the replication of HIV-1 in tissue culture and their selectivity indexes were 42, 40, 14, 18 and 65, respectively. To further fractionate Coptis chinensis, which is shown to be highest anti-HIV-1 activity, methanol extracts of Coptis chinensis were fractionated into methylene chloride at pH3, pH10 and water residue. The selectivity Indexes of CH$_2$C1$_2$(pH 3), CH$_2$C1$_2$(pH 10) and water residue were 50, 22 and 98 respectively. Our results show that the water residue of Coptis chinensis was the most effective for anti-HIV-1 activity.

Antimicrobial Activity of Coptis chinensis and Sophora flavescens against Streptococcus mutans ATCC 25175 (Streptococcus mutans ATCC 25175에 대한 황련과 고삼의 항균활성)

  • As part of our screening of anticariogenic agents from medicinal plants, the ethanol extracts of 25 herbs widely used in the fork medicine were tested for the antimicrobial activity against a cariogenic bacterium Streptococcus mutans ATCC 25175. The ethanol extracts of Coptis chinensis, Sophora flavescens, Glycyrrhiza uralensis, Mentha arvensis, and Saururus chinensis showed antimicrobial activities against S. mutans. The extracts of Coptis chinensis and Sophora flavescens among these medicinal plants showed significant antomicrobial activity against S. mutans. These results suggested that the extracts of Coptis chinensis and Sophora flavescens could be the potential source of antimicrobial agent against S. mutans ATCC 25175.

Inhibitory Activities of Palmatine from Coptis chinensis Against Helicobactor pylori and Gastric Damage

  • Helicobacter pylori (H. pylori) is the most important factor of gastric disease in clinical practice. Moreover, smoking, stress and a poor diet may be additive factors for gastric damage. With these factors, increasing infection of H. pylori triggers gastritis, gastric ulcers and gastric cancer. To develop a new protective agent, we are concerned with plant-derived extract. The extract of Coptis chinensis (C. chinensis) and its constituents were investigated to assess their protective activities against gastric damage. The C. chinensis extract showed a scavenging effect against 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and superoxide radicals, inhibition of H. pylori colonization and antiulcerogenic activities in rat. In particular, palmatine derived from C. chinensis was found to be the novel protective agent. It is better than the C. chinensis extract, berberine, a well-known constituent of C. chinensis. We suggest that palmatine from the root cortex of C. chinensis may be a good candidate for the development of new pharmaceuticals to prevent gastric disease.

  • $Coptis$ $chinensis$ Franch rhizome is one of the important traditional Korea medicines with anti-inflammatory, anti-bacterial, anti-hypertensive and anti-cancer properties. The methanol extract of rhizome from the $Coptis$ $chinensis$ rhizome was purified by using preparative centrifugal partition chromatography (CPC) and HPLC method. The optimum two-phase CPC solvent system was composed of n-butanol:acetic acid:water at a ratio of 4:1:5. Berberine (16.8 mg) was successfully isolated by CPC and HPLC. The chemical structure of the compound was identified by (1 H)$^1H$, $^{13}C$-NMR and ESI-MS spectral data analysis.

Effect of Reducing Inflammation of Coptis chinensis Extract -Ceramide Complex through ECS Control in Atopic Dermatitis

  • Objectives: The purpose of this study was to confirm effect of reducing inflammation of Coptis chinensis extract-ceramide complex through the endocannabinoid system (ECS) control in atopic dermatitis. Methods: 8-week-old ICR mice were divided into normal group (Ctrl), lipid barrier elimination group (ADE), palmitoylethanolamide treated group after lipid barrier elimination (PEAT), and Coptis chinensis extract-ceramide complex applied group after lipid barrier elimination (CRA). After inducing atopic dermatitis, cannabinoid receptor (CB) 1, CB2, CD68, p-I𝜅B, iNOS, substance P and serotonin were observed to confirm the regulation of the ECS, macrophage activity and mast cell activity. Results: CB1 and CB2 showed higher positive reactions in the CRA than in the ADE and PEAT. CD68, p-I𝜅B and iNOS showed higher positive reaction in the ADE, PEAT and CRA than in the Ctrl, but the increase in the positive reaction was lower in the CEA compared to the ADE and PEAT. Substance P and serotonin showed higher positive reaction in the ADE, PEAT and CRA than in the Ctrl, but the increase in the positive reaction was lower in the CEA compared to the ADE and PEAT. Conclusions: The effects of Coptis chinensis extract -ceramide complex were confirmed on the regulation of the ECS, macrophage activity and mast cell activity.

Dyeing of Pig Skin with Coptis chinensis Franch (황련을 이용한 돈피 염색에 관한 연구)

  • Dyeability and antimicrobial property of dyed pig skin with Coptis chinensis Franch was investigated. Dyestuff was extracted with water and concentrated. Pig skin was dyed and mordanted according to various dyeing temperature, dyeing time, dyestuff concentration, mordanting methods and kinds of mordants. The results of this experiment were as follows: 1. Dyeability of pig skin with Coptis chinensis Franch was best at 200%(owf) dyestuff concentration, 50${^{\circ}C}$ dyeing temperature, and 20min. dyeing time. 2. Pre-mordanting by Cu improved the K/S values of dyed pig skin. 3. Surface color and color difference of dyed pig skin showed various results according to the mordants used : generally their color was yellow, Al post-mordanted pig skin showed the greatest color difference. 4. Color fastness to drycleaning was considerably high in case of staining compared to fading. Fastness to light was improved by Cu-mordanting while other treatments were not effective. 5. Antimicrobial property of samples was not different (99.9%) between untreated and treated.

Coptis chinensis Extract Inhibits the Production of Inflammatory Mediators and Delayed Type Hypersensitivity in Mice

  • Background: Coptis chinensis rhizome has been used as a medicinal herb in traditional Oriental medicine. We investigated the effects of Coptis chinensis extract on inflammatory mediators and delayed type hypersensitivity in mice. Methods: The inhibitory effect of ethanolic extract of Coptis chinensis (CCE) on cell proliferation was evaluated using MTS assay. The lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages and the Con A-activated mouse splenocytes were cultured with various concentrations of CCE. Total nitric oxide (NO) production was determined by Griess reaction. The amounts of secreted prostaglandine E2 ($PGE_2$), interleukin (IL)-2 and IFN-${\gamma}$ were measured by ELISA. To investigate the in vivo anti-inflammatory effect of CCE, oxazolone-induced delayed type hypersensitivity (DTH) model was used. Results: The CCE at $100{\mu}g/ml$ significantly blocked the LPS-induced production of pro-inflammatory mediators (NO and PGE) in RAW264.7 macrophages. Also, it significantly inhibited cell proliferation and cytokine (IL-2 and IFN-${\gamma}$) production in splenocytes. Furthermore, when splenocytes from CCE fed mice (200 mg/kg for 2 weeks) were activated with Con A, cell proliferation and cytokine production were significantly inhibited. In addition, CCE decreased in vivo inflammation in oxazolone-induced DTH model mice. Conclusion: We suggest that Coptis chinensis can be used as an anti-inflammatory drug by exerting an inhibitory effect in inflammatory mediator- and cell-mediated inflammation.

Antioxidant Activity of Water-Extract from Coptis chinensis Franch (황련 열수추출물의 항산화 활성 효과)

  • This study was carried out to investigate the antioxidant activity of the water extract from Coptis chinensis Franch. Coptis chinensis Franch. exhibited antioxidative activity at all concentration tested. The extract was as good as antioxidative action of the synthetic antioxidant, butylated hydroxytoluene and ascorbic acid; Also, the one was superior to that of natural antioxidant, $\alpha$ -tocopherol. With heavy metal ions (Fe3+, Zn2+) present, Coptis chinensis Franch. showed strong antioxidative effect.

Antibacterial Activity of Caesalpinia sappan and Coptis chinensis Extracts against Bacillus cereus and Vibrio parahaemolyticus (소목(蘇木)과 황련(黃連) 추출물(抽出物)의 Bacillus cereus와 Vibrio parahaemolyticus 에 대한 항세균활성(抗細菌活性))

  • Objectives : This experimental study was performed to investigate the antibacterial effect of Caesalpinia sappan and Coptis chinensis extract against B. cereus and V. parahaemolyticus. Methods : Methanol extracts of C. sappan and C. chinensis was tested against B. cereus and V. parahaemolyticus by paper disc method. Results : The growth of B. cereus and V. parahaemolyticus was inhibited by C. sappan and C. chinensis extract among 6 kinds of medicinal plant extracts. The extract of C. sappan and C. chinensis extract inhibited the growth of V. parahaemolyticus and B. cereus, respectively. The growth of B. cereus and V. parahaemolyticus had a tendency to increase depend on the concentration of the extract. EtOEt and EtOAc fractions and EtOEt and BuOH fractions of the C. sappan extract had a high antibacterial activity against B. cereus and V. parahaemolyticus, respectively. And, BuOH and $H_2O$ fractions of the C. chinensis extract showed antibacterial activity against B. cereus highly. Conclusions : C. sappan and C. chinensis extract efficiently inhibited the growth of B. cereus and V. parahaemolyticus.

Antimicrobial Effect of Natural Plant Extracts against Periodontopathic Bacteria (치주염 원인균에 대한 천연 식물 추출물의 항균효과)

  • In this study, we examined the antimicroboal effect against Actinobacillus actinomycetemcomitans and Prevotella intermedia which were the bacteria causing the Periodontopathic by using 34 types of natural plant extracts. Therefore, this study measures growth inhibition activity and Minimum Inhibition Concentration (MIC) of a sample extract with the use of organic solvent extracts in order to analyze the antibacterial effect of natural plant extracts on periodontopathic bacteria. Each of the 34 types of natural plant extracts were extracted by using the ethanol, and subsequently, the size of growth inhibition zone(clear zone, ㎜) of respective extracts were measured through the disk diffusion method. As a result, it was found that the growth inhibitory activity was found for A. actinomycetemcomitans, which is the bacteria causing the Periodontitis, in 13 types of natural plant extracts such as Raphanus sativus, Akebia quinata, Paeonia lactiflora, Belamcanda chinensis, Inula britannics, Houttuynia cordata, Forsythia saxatilis, Gentiana macrophylla, Melia azedarach, Scutellaria baicalensis, Coptis chinensis, Phellodendron amurense, Kalopanax Pictus, etc. In the case of P. intermedia, the growth inhibitory activity was found in 13 types of natural plant extracts such as Raphanus sativus, Angelica acutiloba, Akebia quinata, Belamcanda chinensis, Inula britannics, Houttuynia cordata, Cinnamomum cassia, Aster tataricus, Melia azedarach, Scutellaria baicalensis, Coptis chinensis, Phellodendron amurense, Kalopanax Pictus etc. For A. actinomycetemcomitans, anti-bacterial effect was exhibited in Belamcanda chinensis, Cinnamomum cassia, Kalopanax Pictus, Phellodendron amurense, Coptis chinensis. The Coptis chinensis showed the most excellent growth inhibitory activity in all organic solvent fragment, while P. intermedia showed the growth inhibitory activity in Belamcanda chinensis, Cinnamomum cassia, Meliaazedarach, Phellodendron amurense, and Coptis chinensis.
